This paper reports the results of a survey intended to discover the manner in which Soft Systems Methodology (SSM) is understood and used in Brazil. The focus is upon SSM papers published in national and international journals, and conferences, incorporating a refereeing procedure. To be included in the survey, publications had to meet at least one of the following criteria: (a) authorship is clearly of Brazilian nationality; or, (b) authorship is affiliated to a Brazilian institution; or (c) application is set in Brazil. Similar surveys reporting on the United Kingdom, Australia and, to a lesser extent, Spain have been published previously in the literature. This paper, therefore, contributes to the growing international understanding and usage of SSM. Ultimately the paper serves as an initial map of the developing context in Brazilian SSM theory and practice.

Este trabalho utiliza a metodologia six sigma com o objetivo de aumentar a produtividade da Linha de LCD (Liquid Crystal Display) em uma fábrica do Pólo Industrial de Manaus - PIM e um sistema de inferência fuzzy para mensurar o aumento dessa produtividade, onde foram identificados vários parâmetros baseados na metodologia six sigma. Dentre os quais, conforme grau de relevância dos especialistas deste estudo, pode-se destacar: desperdícios, capacidade produtiva e estudo de tempos. Ressaltando ainda que o sistema proposto seja de grande importância para profissionais e pesquisadores da gestão da produção, os quais desejam resultados que reduzam custos e conseqüentemente aumente os lucros da organização.

The United States National Ice Center (NIC) provides weekly ice analyses of the Arctic and Antarctic using information from ice reconnaissance, ship reports and high-resolution satellite imagery. In cloud-covered areas and regions lacking imagery, the higher-resolution sources are augmented by ice concentrations derived from Defense Meteorological Satellite Program (DMSP) Special Sensor Microwave/Imager (SSMII) passive-microwave imagery. However, the SSMII-derived ice concentrations are limited by low resolution and uncertainties in thin-ice regions. Ongoing research at NIC is attempting to improve the utility of these SSMII products for operational sea-ice analyses. The refinements of operational algorithms may also aid future scientific studies. Here we discuss an evaluation of the standard operational ice-concentration algorithm, Cal/Val, with a possible alternative, a modified NASA Team algorithm. The modified algorithm compares favorably with CallVal and is a substantial improvement over the standard NASA Team algorithm in thin-ice regions that are of particular interest to operational activities.

The presence of the schizont stage of the obligate intracellular parasites Theileria parva or T. annulata in the cytoplasm of an infected leukocyte results in host cell transformation via a mechanism that has not yet been elucidated. Proteins, secreted by the schizont, or expressed on its surface, are of interest as they can interact with host cell molecules that regulate host cell proliferation and/or survival. The major schizont surface protein is the polymorphic immunodominant molecule, PIM, which contains a large glutamine- and proline-rich domain (QP-rd) that protrudes into the host cell cytoplasm. Analyzing QP-rd generated by in vitro transcription/translation, we found that the signal peptide was efficiently cleaved post-translationally upon addition of T cell lysate or canine pancreatic microsomes, whereas signal peptide cleavage of a control protein only occurred cotranslationally and in the presence of microsomal membranes. The QP-rd of PIM migrated anomalously in SDS-PAGE and removal of the 19 amino acids corresponding to the predicted signal peptide caused a decrease in apparent molecular mass of 24kDa. The molecule was analyzed using monoclonal antibodies that recognize a set of previously defined PIM epitopes. Depending on the presence or the absence of the signal peptide, two conformational states could be demonstrated that are differentially recognized, with N-terminal epitopes becoming readily accessible upon signal peptide removal, and C-terminal epitopes becoming masked. Similar observations were made when the QP-rd of PIM was expressed in bacteria. Our observations could also be of relevance to other schizont proteins. A recent analysis of the proteomes of T. parva and T. annulata revealed the presence of a large family of potentially secreted proteins, characterized by the presence of large stretches of amino acids that are also particularly rich in QP-residues.

Proviral integration site for Moloney murine leukemia virus (Pim) kinases are Ser/Thr/Tyr kinases. They modulate B-cell development but become oncoproteins and promote cancer development once overexpressed. Containing three isoforms, Pim-1, -2 and -3 are known to phosphorylate various substrates that regulate transcription, translation, cell cycle, and survival pathways in both hematological and solid tumors. Mantle cell lymphoma (MCL) is an aggressive B-cell lymphoma. Elevated Pim kinase levels are common in MCL, and it negatively correlates with patient outcome. SGI-1776 is a small molecule inhibitor selective for Pim-1/-3. We hypothesize that SGI-1776 treatment in MCL will inhibit Pim kinase function, and inhibition of downstream substrates phosphorylation will disrupt transcriptional, translational, and cell cycle processes while promoting apoptosis. SGI-1776 treatment induced moderate to high levels of apoptosis in four MCL cell lines (JeKo-1, Mino, SP-53 and Granta-519) and peripheral blood mononuclear cells (PBMCs) from MCL patients. Phosphorylation of transcription and translation regulators, c-Myc and 4E-BP1 declined in both model systems. Additionally, levels of short-lived Mcl-1 mRNA and protein also decreased and correlated with decline of global RNA synthesis. Collectively, our investigations highlight Pim kinases as viable drug targets in MCL and emphasize their roles in transcriptional and translational regulation. We further investigated a combination strategy using SGI-1776 with bendamustine, an FDA-approved DNA-damaging alkylating agent for treating non-Hodgkin’s lymphoma. We hypothesized this combination will enhance SGI-1776-induced transcription and translation inhibition, while promoting bendamustine-triggered DNA damage and inducing additive to synergistic cytotoxicity in B-cell lymphoma. Bendamustine alone resulted in moderate levels of apoptosis induction in MCL cell lines (JeKo-1 and Mino), and in MCL and splenic marginal zone lymphoma (a type of B-cell lymphoma) primary cells. An additive effect in cell killing was observed when combined with SGI-1776. Expectedly, SGI-1776 effectively decreased global RNA and protein synthesis levels, while bendamustine significantly inhibited DNA synthesis and generated DNA damage response. In combination, intensified inhibitory effects in DNA, RNA and protein syntheses were observed. Together, these data suggested feasibility of using Pim kinase inhibitor in combination with chemotherapeutic agents such as bendamustine in B-cell lymphoma, and provided foundation of their mechanism of actions in lymphoma cells.

Remote sensing instruments are key players to map land surface temperature (LST) at large temporal and spatial scales. In this paper, we present how we combine passive microwave and thermal infrared data to estimate LST during summer snow-free periods over northern high latitudes. The methodology is based on the SSM/I-SSMIS 37 GHz measurements at both vertical and horizontal polarizations on a 25 km × 25 km grid size. LST is retrieved from brightness temperatures introducing an empirical linear relationship between emissivities at both polarizations as described in Royer and Poirier (2010). This relationship is calibrated at pixel scale, using cloud-free independent LST data from MODIS instruments. The SSM/I-SSMIS and MODIS data are synchronized by fitting a diurnal cycle model built on skin temperature reanalysis provided by the European Centre for Medium-Range Weather Forecasts (ECMWF). The resulting temperature dataset is provided at 25 km scale and at an hourly time step during the ten-year analysis period (2000-2011). This new product was locally evaluated at five experimental sites of the EU-PAGE21 project against air temperature measurements and meteorological model reanalysis, and compared to the MODIS LST product at both local and circumpolar scale. The results giving a mean RMSE of the order of 2.2 K demonstrate the usefulness of the microwave product, which is unaffected by clouds as opposed to thermal infrared products and offers a better resolution compared to model reanalysis.
