An extracellular glucoamylase produced by Paecilomyces variotii was purified using DEAE-cellulose ion exchange chromatography and Sephadex G-100 gel filtration. The purified protein migrated as a single band in 7% PAGE and 8% SDS-PAGE. The estimated molecular mass was 86.5 kDa (SDS-PAGE). Optima of temperature and pH were 55 degrees C and 5.0, respectively. In the absence of substrate the purified glucoamylase was stable for 1 h at 50 and 55 degrees C, with a t(50) of 45 min at 60 degrees C. The substrate contributed to protect the enzyme against thermal denaturation. The enzyme was mainly activated by manganese metal ions. The glucoamylase produced by P. variotii preferentially hydrolyzed amylopectin, glycogen and starch, and to a lesser extent malto-oligossacarides and amylose. Sucrose, p-nitrophenyl alpha-D-maltoside, methyl-alpha-D-glucopyranoside, pullulan, alpha- and beta-cyclodextrin, and trehalose were not hydrolyzed. After 24 h, the products of starch hydrolysis, analyzed by thin layer chromatography, showed only glucose. The circular dichroism spectrum showed a protein rich in alpha-helix. The sequence of amino acids of the purified enzyme VVTDSFR appears similar to glucoamylases purified from Talaromyces emersonii and with the precursor of the glucoamylase from Aspergillus oryzae. These results suggested the character of the enzyme studied as a glucoamylase (1,4-alpha-D-glucan glucohydrolase).

An alpha-amylase produced by Paecilomyces variotii was purified by DEAE-cellulose ion exchange chromatography, followed by Sephadex G-100 gel filtration and electroelution. The alpha-amylase showed a molecular mass of 75 kDa (SDS-PAGE) and pl value of 4.5. Temperature and pH optima were 60 degrees C and 4.0, respectively. The enzyme was stable for 1 h at 55 degrees C, showing a t(50) of 53 min at 60 degrees C. Starch protected the enzyme against thermal inactivation. The a-amylase was more stable in alkaline pH. It was activated mainly by calcium and cobalt, and it presented as a glycoprotein with 23% carbohydrate content. The enzyme preferentially hydrolyzed starch and, to a lower extent, amylose and amylopectin. The K(m) of alpha-amylase on Reagen (R) and Sigma (R) starches were 4.3 and 6.2 mg/mL, respectively. The products of starch hydrolysis analyzed by TLC were oligosaccharides such as maltose and maltotriose. The partial amino acid sequence of the enzyme presented similarity to alpha-amylases from Bacillus sp. An extracellular polygalacturonase (PG) produced from Paecilomyces variotii was purified to homogeneity through two chromatography steps using DEAE-Fractogel and Sephadex G-100. The molecular weight of P. variotii PG was 77,300 Da by gel filtration and SDS-PAGE. PG had isoelectric point of 4.37 and optimum pH 4.0. PG was very stable from pH 3.0 to 6.0. The extent of hydrolysis of different pectins by the purified enzyme was decreased with an increase in the degree of esterification. PG had no activity toward non-pectic polysaccharides. The apparent K (m) and V (max) values for hydrolyzing sodium polypectate were 1.84 mg/mL and 432 A mu mol/min/mg, respectively. PG was found to have temperature optimum at 65 A degrees C and was totally stable at 45 A degrees C for 90 min. Half-life at 55 A degrees C was 50.6 min. Almost all the examined metal cations showed partial inhibitory effects under enzymatic activity, except for Na(+1), K(+1), and Co(+2) (1 mM) and Cu(+2) (1 and 10 mM).

Com o objetivo de demonstrar a eficácia do fungo Paecilomyces lilacinus sobre ovos de Taenia saginata em condições laboratoriais, foi montado ensaio em placas de Petri com agar - água 2%. Houve atividade ovicida (p<0,05) em relação ao grupo controle no décimo dia de interação e colonização interna dos ovos de 25,5%.

Twenty four strains of the entomopathogenic fungi (Hyphomycetes) Beauveria bassiana, Metarrhizium anisopliae, Nomuraea rileyi, Paecilomyces farinosus, P. fumosoroseus and P. lilacinus, maintained in the culture collection of Embrapa-Centro Nacional de Pesquisa de Recursos Genéticos e Biotecnologia (Cenargen) and preserved by lyophilization and in liquid nitrogen, had their conidial viability assessed. Germination rates of 16- to 84-month-old cultures stored in liquid nitrogen decreased, on average, less than 13.3%. For 29- to 49-month-old cultures preserved by lyophilization, the viability loss ranged, on average, from 28.6 to 94.5%. The results demonstrated the efficiency of the tested methods, especially liquid nitrogen, in preserving the viability of entomopathogenic fungi.

The effectiveness of a formulated product containing spores of the naturally occurring fungus Paecilomyces lilacinus, strain 251, was evaluated against root-knot nematodes in pot and greenhouse experiments. Decrease of second-stage juveniles hatching from eggs was recorded by using the bio-nematicide at a dose of 4 kg ha(-1), while a further decrease was recorded by doubling the dose. However, the mortality rate decreased by increasing the inoculum level. Application of P. lilacinus and Bacillus firmus, singly or together in pot experiments, provided effective control of second-stage juveniles, eggs or egg masses of root-knot nematodes. In a greenhouse experiment, the bio-nematicide was evaluated for its potential to control root-knot nematodes either as a stand-alone method or in combination with soil solarization. Soil was solarized for 15 d and the bio-nematicide was applied just after the removal of the plastic sheet. Soil solarization for 15 d either alone or combined with the use of P. lilacinus did not provide satisfactory control of root-knot nematodes. The use of oxamyl, which was applied 2 weeks before and during transplanting, gave results similar to the commercial product containing P. lilacinus but superior to soil solarization. A rare monophialidic fungus, Taifanglania hechuanensis gen. & sp. nov., was isolated from soil oil the banks of Jialin River, Hechuan, Chongqing City during a survey of soil-borne filamentous fungi from different phytogeographical areas in China. It is described and illustrated in this paper. A further eight monophialidic species of Paecilomyces are transferred to the genus. Diagnosis features of the new genus are white, grey, straw yellow or brown to black colonies on Czapek agar. Conidiophores are always absent or simple. Phialides are solitary, consisting of a cylindrical or ellipsoidal swollen basal portion, tapering into a thin neck, directly arising on vegetative hyphae or prophialides, sometimes consisting of a whorl of 2 to 3 phialides oil simple conidiophores. Conidia arc one-celled, hyaline, smooth-walled, subglobose, ellipsoidal or fusiform, having or no the connective between conidia and being thermotolerant. The new species is characterized by pale yellow to grey-yellow colonies, solitary phialides with ail ellipsoidal or fusiform basal portion that arise directly from the vegetative hyphae, big conidia (3.1-)3.9-8.7 x ( 1.7-)2.1-4.7(-5.1) mu m with the connective, and thermotolerant growth. A molecular study based oil the nucleotidic sequences of the SSU rDNA and ITS regions support the status of T. hechuanensis as a new species and Taifanglania as a new genus.

The lace bug, Leptopharsa heveae is an insect that causes serious damage on rubber trees. In laboratory condition, the strains of Verticillium lecanii (ARSEF 6430, 6431 e 6432) and Aphanocladium album (ARSEF 6433) were tested on third-and fifth-instar nymphs and adults of L. heveae to evaluate their virulence using 2.4 x 10(5) and 2.4 x 10(7) conidia/mL. The bioassays were carried out using Petri dishes whose inner bottoms were covered with damp filter papers. Each Petri dish contained five insects and one rubber tree leaflet. The plates were covered with PVC film to provide high relative humidity, maintained at 26 +/- 0.5 degrees C and a photophase of 14 hours. The Probit analysis was calculated from mortality date of insects killed by fungi. In the highest concentrations, ARSEF 6430 was more virulent for third instar nymphs, and the LT50 was 1.9 days. For the fifth instar, the strains ARSEF 6430, 6433 and 6432 showed similar virulence with LT50 of 2.6, 2.6 and 3.2 days, respectively. For adults, ARSEF 6431 was the most virulent strain with the LT50 recorded at 2.0 days. The smallest concentration did not always cause more than 50% mortality.

This study reports on the influence of heat and hydrogen peroxide combination on the inactivation kinetics of two heat resistant molds: Neosartorya fischeri and Paecilomyces variotii. Spores of different ages (1 and 4 months) of these molds were prepared and D-values (the time required at certain temperature/hydrogen peroxide combination to inactivate 90% of the mold ascospores) were determined using thermal death tubes. D-values found for P. variotii ranged from 1.2 to 25.1 s after exposure to different combinations of heat (40 or 60 degrees C) and hydrogen peroxide (35 or 40% w/w) while for N. fischeri they varied from 2.7 to 14.3 s after exposure to the same hydrogen peroxide concentrations and higher temperatures (60 or 70 degrees C). The influence of temperature and hydrogen peroxide concentration on the d-values varied with the genus of mold and their ages. A synergistic effect of heat and hydrogen peroxide in reducing D-values of Paecilomyces variotti and N. fischeri has been observed. In addition to strict control of temperature, time and hydrogen concentration, hygienic storage and handling of laminated paperboard material must be considered to reduce the probability of package's contamination. All these measures together will ensure package's sterility that is imperative for the effectiveness of aseptic processing and consequently to ensure the microbiological stability of processed foods during shelf-life. [ES]El estudio químico de un organismo, y la elucidación de las estructuras moleculares de sus componentes, puede dar lugar a descubrimientos que aconsejen una posterior producción industrial del mismo. Con este propósito, se seleccionaron tres microorganismos para ser investigadas químicamente, siendo estos: un hongo zoospórico, Schizochytrium aggregatum, y dos hifomicetos, Paecilomyces variotii y Penicillium roqueforti, estos últimos fueron aislados del medio marino del litoral canario. Los resultados demuestran que los tres hongos estudiados no son, en nuestras condiciones de cultivo, productores de micotoxinas; al contrario, producen importantes componentes nutricionales, lo cual supone que se pueden utilizar directamente para la nutrición animal en acuicultura. Por otra parte, también se pueden usar para producir industrialmente productos de alto valor añadido como oleína, peróxido de ergosterol, (9,11)-dehidroperóxido del ergosterol, p-hidroxi-benzaldehido, D- (-)-manitol y 2-deoxi-2-fosfamino-?-D-glucopyranosa. Esta última es nueva en la bibliografía
