Os cogumelos do gênero Pleurotus são cultivados em diversos substratos lignocelulósicos, dada a atividade decompositora desses organismos proveniente de seu metabolismo enzimático. O presente estudo teve como objetivo analisar a composição mineral de Pleurotus ostreatus e dos substratos de cultivo preparados à base de resíduos madeireiros e agroindustriais da região amazônica. Foram analisados macro (P, K, Ca e Mg) e micronutrientes (Fe, Zn, Cu, Mn e Na) dos cogumelos e dos substratos. Os substratos foram formulados a partir da serragem de Simarouba amara Aubl. (marupá), Ochroma piramidale Cav. ex. Lam. (pau de balsa) e de bagaços de Bactris gasipaes Kunth (pupunheira) e de Saccharum officinarum (cana-de-açúcar). As amostras foram solubilizadas mediante digestão ácida (nítrico-peridrol). Os elementos Ca, Mg, Fe, Cu, Zn e Mn foram determinados por espectrometria de absorção atômica; o Na e K, por emissão atômica e o P, por colorimetria. A composição mineral do cogumelo variou com o substrato de cultivo. Os diferentes substratos possibilitaram a produção de um cogumelo rico em K, P, Mg e Fe, essenciais à nutrição e à saúde humana. O potássio foi o mineral de maior teor no cogumelo em todos os substratos testados (36,83-42,18 g.kg-1), seguido de fósforo (6,95-10,60 g.kg-1) e do magnésio (1,57-2,50 g.kg-1).

The objective of the present work was to evaluate the nutritional composition of mushrooms produced in alternative substrates in agricultural and agro-industrial residues from the Amazon. C, N, pH, moisture, soluble solids, protein, lipids, total fibers, ashes, carbohydrates and energy were determined. Substrates were formulated from Simarouba amara Aubl. and Ochroma piramidale Cav. ex. Lam. Sawdust and from Bactris gasipaes Kunth and Saccharum officinarum stipe. Results showed that the nutritional composition of P. ostreatus varied according to the cultivation substrate and that it can be considered important food due to its nutritional characteristics such as: high protein content; metabolizable carbohydrates and fiber; and low lipids and calories contents.

Les systèmes cholinergique et dopaminergique jouent un rôle prépondérant dans les fonctions cognitives. Ce rôle est exercé principalement grâce à leur action modulatrice de l’activité des neurones pyramidaux du cortex préfrontal. L’interaction pharmacologique entre ces systèmes est bien documentée mais les études de leurs interactions neuroanatomiques sont rares, étant donné qu’ils sont impliqués dans une transmission diffuse plutôt que synaptique. Ce travail de thèse visait à développer une expertise pour analyser ce type de transmission diffuse en microscopie confocale. Nous avons étudié les relations de microproximité entre ces différents systèmes dans le cortex préfrontal médian (mPFC) de rats et souris. En particulier, la densité des varicosités axonales en passant a été quantifiée dans les segments des fibres cholinergiques et dopaminergiques à une distance mutuelle de moins de 3 µm ou à moins de 3 µm des somas de cellules pyramidales. Cette microproximité était considérée comme une zone d’interaction probable entre les éléments neuronaux. La quantification était effectuée après triple-marquage par immunofluorescence et acquisition des images de 1 µm par microscopie confocale. Afin d’étudier la plasticité de ces relations de microproximité, cette analyse a été effectuée dans des conditions témoins, après une activation du mPFC et dans un modèle de schizophrénie par déplétion des neurones cholinergiques du noyau accumbens. Les résultats démontrent que 1. Les fibres cholinergiques interagissent avec des fibres dopaminergiques et ce sur les mêmes neurones pyramidaux de la couche V du mPFC. Ce résultat suggère différents apports des systèmes cholinergique et dopaminergique dans l’intégration effectuée par une même cellule pyramidale. 2. La densité des varicosités en passant cholinergiques et dopaminergiques sur des segments de fibre en microproximité réciproque est plus élevée comparé aux segments plus distants les uns des autres. Ce résultat suggère un enrichissement du nombre de varicosités axonales dans les zones d’interaction. 3. La densité des varicosités en passant sur des segments de fibre cholinergique en microproximité de cellules pyramidales, immunoúactives pour c-Fos après une stimulation visuelle et une stimulation électrique des noyaux cholinergiques projetant au mPFC est plus élevée que la densité des varicosités de segments en microproximité de cellules pyramidales non-activées. Ce résultat suggère un enrichissement des varicosités axonales dépendant de l’activité neuronale locale au niveau de la zone d'interaction avec d'autres éléments neuronaux. 4. La densité des varicosités en passant des fibres dopaminergiques a été significativement diminuée dans le mPFC de rats ayant subi une déplétion cholinergique dans le noyau accumbens, comparée aux témoins. Ces résultats supportent des interrelations entre la plasticité structurelle des varicosités dopaminergiques et le fonctionnement cortical. L’ensemble des donneès démontre une plasticité de la densité locale des varicosités axonales en fonction de l’activité neuronale locale. Cet enrichissement activité-dépendant contribue vraisemblablement au maintien d’une interaction neurochimique entre deux éléments neuronaux.

The aim of this study was to identify molecular pathways involved in audiogenic seizures in the epilepsy-prone Wistar Audiogenic Rat (WAR). For this, we used a suppression-subtractive hybridization (SSH) library from the hippocampus of WARs coupled to microarray comparative gene expression analysis, followed by Northern blot validation of individual genes. We discovered that the levels of the non-protein coding (npc) RNA BC1 were significantly reduced in the hippocampus of WARs submitted to repeated audiogenic seizures (audiogenic kindling) when compared to Wistar resistant rats and to both naive WARs and Wistars. By quantitative in situ hybridization, we verified lower levels of BC1 RNA in the GD-hilus and significant signal ratio reduction in the stratum radiatum and stratum pyramidale of hippocampal CA3 subfield of audiogenic kindled animals. Functional results recently obtained in a BC1-/- mouse model and our current data are supportive of a potential disruption in signaling pathways, upstream of BC1, associated with the seizure susceptibility of WARs. O presente estudo teve como objetivo avaliar a composição nutricional dos cogumelos produzidos em substratos alternativos à base de resíduos agrícolas e agroindustriais da Amazônia. Determinou-se C, N, pH, umidade, sólidos solúveis, proteína, lipídios, fibra total, cinzas, carboidratos e energia. Os substratos foram formulados a partir de serragem de Simarouba amara Aubl. (marupá), Ochroma piramidale Cav. ex. Lam. (pau de balsa) e do estipe de Bactris gasipaes Kunth (pupunheira) e de Saccharum officinarum (cana-de-açúcar). Os resultados demonstraram que: a composição nutricional do P. ostreatus variou com o substrato de cultivo e; O P. ostreatus pode ser considerado um importante alimento devido suas características nutricionais: altos teores de proteínas, carboidratos metabolizáveis e fibras; baixos teores de lipídios e de calorias.

We studied the effects of a wasp toxin beta-pompilidotoxin (beta-PMTX) on rat hippocampal CA1 interneurons by the current-clamp technique. The firing patterns of pyramidal neurons and pyramidale interneurons were not affected by beta-PMTX, but in oriens and radiatum interneurons, beta-PMTX converted the action potentials to prolonged depolarizing potentials by slowing the inactivation of Na+ channels. In lacunosum moleculare interneurons, beta-PMTX induced initial bursting spikes followed by block of succeeding spikes. Comparison of beta-PMTX with a sea anemone toxin, ATX 11, revealed that ATX 11 altered the firing properties of pyramidal neurons and pyramidale interneurons that were unchanged by beta-PMTX. Our results suggest that beta-PMTX modulates Na+ currents in CAl interneurons differently in various CAl neurons and the toxin is useful to classify Na+ channel subtypes. (C) 2002 Elsevier B.V. In a culture of a Pleurotus ostreatus (oyster mushroom) strain, macro and micronutrients of the raw material and the initial and spent substrates were evaluated. Substrates were formulated with sawdust from Simarouba amara Aubl. and Ochroma piramidale Cav. ex. Lam., crushed Bactris gasipaes Kunth and crushed Saccharum officinarum (sugar cane). Samples were solubilized by means of acid digestion (nitric-peridrol). Ca, Mg, Fe, Cu, Zn and Mn were determined by atomic absorption spectrophotometry, Na and K by atomic emission, and P by colorimetry. The mineral composition of the fruiting body varied with the substrates, which made possible the production of a fruiting body rich in K, P Mg and Fe. Potassium was the mineral with the highest content in the fruiting body in all substrates tested (36.83-42.18g.kg(-1)). There was an increase of protein and mineral content in the spent substrate in relation to the initial one.

The aim of the present study was to carry out the physical, chemical and nutritional analyses of the alternative substrates formulated from wood and agroindustrial residues of the Amazon for Pleurotus ostreatus (Jacq.:Fr.) Kummer cultivation. The determination of C, N, pH, humidity, soluble solids, protein, fat, total fiber ash, carbohydrates (total and available) and energy were carried out. The substrates were formulated from sawdust of Simarouba amara Aubl. (marupa), Ochroma piramidale Cav. ex. Lam. (pau de balsa) and from the stein of Bactris gasipaes Kunth (pupunheira palm tree), and from Saccharum officinarum L. (sugar cane bagasse). The results demonstrated that the nutritional composition of the substrate is variable and the improvement of the quality of the spent substrate (energy and protein increment) promoted by the metabolism of the fungus during the cultivation, contributed for a more nutritive substrate than the initial one, which could be used as a compost for Agaricus sp. cultivation, as organic fertilizer and for bioremediation for contaminated soils.

Para estudar a resposta imune inata produzida especificamente no interior do SNC em desenvolvimento, evitando a influencia do sistema imune, empregamos modelo de infeccao viral induzida pela inoculacao intracerebral do virus da dengue em camundongos neonatos. Oito camundongos lactentes de dois dias de idade da espécie Mus musculus e variedade suica albina foram inoculados por via intracerebral com homogenado cerebral infectado com a especie Flavivirus (DENV3 genotipo III). Outro conjunto de animais foi utilizado como controle (nao infectado) e inoculado com igual volume de homogenado cerebral nao infectante e mantidos nas mesmas condicoes dos infectados. Decorridos 7 dias apos a infeccao os camundongos doentes foram sacrificados e tiveram seus cerebros processados para imunomarcacao de astrocitos e microglias. Quantificou-se a resposta imune glial no stratum lacunosum molecular (Lac Mol), radiatum (Rad) e pyramidale (Pir) de CA1-2 do hipocampo e na camada molecular do giro denteado (GDMol) usando o fracionador optico para estimar o numero de microglias e astrocitos em animais infectados e controles. Intensa astrocitose reativa e intensa ativacao microglial foram encontradas em animais neonatos com sinais clinicos de meningoencefalite. Entretanto, embora tenham sido maiores as estimativas do numero de microglias ativadas nos infectados (Inf) do que nos animais controles (Cont) nas camadas GDMol (Inf: 738,95 } 3,07; Cont: 232,73 } 70,38; p = 0,0035), Rad (Inf: 392,49 } 44,13; Cont: 62,76 } 15,86; p = 0,0004), em relacao ao numero total de microglias (ativadas ou nao) apenas o stratum radiatum mostrou diferença significante (Inf: 6.187,49 } 291,62; Cont: 4.011,89 } 509,73; p = 0,01). Por outro lado apenas a camada molecular do giro denteado mostrou diferenca no numero de astrocitos (Inf: 8.720,17 } 903,11; Cont: 13.023,13 } 1.192,14; p = 0,02). Tomados em conjunto os resultados sugerem que a resposta imune inata do camundongo neonato a encefalite induzida pelo virus da dengue (sorotipo 3, genotipo III) esta associada a um maior aumento do numero de microglias do que de astrocitos reativos e essa mudanca e dependente da camada e da regiao investigada. As implicacoes fisiopatologicas desses achados permanecem por ser investigadas.

Avaliou-se a composição centesimal e análise físico-química do Lentinus strigosus, um cogumelo comestível de ocorrência na Amazônia brasileira, produzidos em substratos alternativos à base de resíduos madeireiros e agroindustriais. Com este objetivo, determinou-se C, N, pH, sólidos solúveis, atividade de água, proteína, lipídios, fibra total, cinzas, carboidratos e energia. Os substratos foram formulados a partir de serragem de Simarouba amara Aubl. (marupá), Ochroma piramidale Cav. ex. Lam. (pau-de-balsa) e Anacardium giganteum (cajuí); e do estipe de Bactris gasipaes Kunth (pupunheira) e de Saccharum officinarum (cana-de-açúcar). Os resultados demonstraram que: a composição nutricional do L. strigosus variou com o substrato de cultivo; os valores de proteína encontrados nos cogumelos cultivados nos diferentes substratos (18 - 21,5%) variaram de acordo com o substrato, sendo considerados elevados; os sólidos solúveis presentes nos cogumelos podem ter relação com vitaminas hidrossolúveis do complexo B; o L. strigosus pode ser considerado um importante alimento devido suas características nutricionais: alto teor de proteína, carboidratos metabolizáveis e fibras; baixos teores de lipídios e de calorias.

Organylhalogenide RX reagieren formal gemäß einer oxidativen 1,1-Addition mit Dihypersilylplumbylen) PbHyp2 zu Dihypersilyl-halogenorganylplumbanen PbHyp2RX. Diese Umsetzung gelingt mit nahezu allen untersuchten Organylresten, lediglich beispielsweise der Mesitylrest erweist sich als zu sperrig (R = Me, Et, nPr, iPr, tBu, Hx, cHx, Ad, Ph, C6F5, oTo, mTo, pTo, Naph, Anthr).rnrnFür einige Halogenorganyle wird eine analoge Addition auch an das zinnhomologe Dihypersilylstannylen beschrieben.rnrnDie untersuchten Addukte sind thermisch und gegenüber UV-Strahlung, Sauerstoff und Wasser deutlich weniger empfindlich als vergleichbare andere blei- und zinnorganische Verbindungen.rnrnUmfangreiche NMR-Datensätze beschreiben eine markante Hoch-feldverschiebung der Hypersilylprotonen beim Übergang von PbHyp2 hin zu PbHyp2RX, die für Arylreste stärker ausfällt als für Alkylreste.rnrnEin Großteil der Addukte wurde einkristallin erhalten und wird anhand der röntgendiffraktometrisch ermittelten Strukturparameter detailliert charakterisiert. rnrnEs finden sich gegenüber dem idealen Tetraderwinkel auffällig stark aufgeweitete Si Pb Si-Winkel von 130-143°, während die anderen Winkel mehrheitlich unter dem theoretischen Idealwert bleiben (Si Pb X: 94 103°; Si Pb C: 98-112°; C Pb X: 95-103°). Insbesondere größere, weichere Reste und planare Arylplumbane rücken näher an das Halogen heran.rnDie insgesamt recht hohen Blei-Halogen-Abstände nehmen von den Chloriden hin zu den Iodiden zu. Die Iodoplumbane zeigen dabei generell die kleinsten Pb Si und die größten Pb C Abstände. Alkylplumbane weisen längere Pb C Bindungen auf als ansonsten vergleichbare Arylplumbane.rnrnViele der gefundenen Molekülstrukturen zeigen Anzeichen hoher sterischer Spannung in Form von Substituentenverzerrungen. rnrnDie Bildungsgeschwindigkeit der Addukte ist auch bei tiefen Temperaturen hoch. Sie nimmt von X = Cl über Br hin zu I zu und ist für Alkylreste höher als für Aryle. Die Zerfallsgeschwindigkeiten verhalten sich genau entgegengesetzt. Bei den Thermolysen wird regelmäßig Hypersilylhalogenid eliminiert. Dabei entstehen in Abwesenheit koordinierender Solventien Dihypersilyl-diorganylplumbane PbHyp2R2.rnAndere, unbekannte Zerfallskanäle führen zu unerwarteten Produkten, wie dem Iodonium-verbrückten, cyclischen Tetraplumbetan Pb4I(C6F5)Hyp3.rnrnIn Anwesenheit von Lewis-Basen hingegen können sich hetero-leptische Plumbylene bilden, wie am Beispiel eines Bitolyldiylbisplumbylens gezeigt wird. Dieses zeigt auch, dass prinzipiell eine zweifache Addition von Dihalogenorganylen an zwei Äqui-valente Dihypersilylplumbylen möglich ist. Entsprechende Untersuchungen beschäftigen sich ausführlich mit dafür geeigneten und ungeeigneten Organdiylresten.rnrnUnter günstigen reduktiven Bedingungen lassen sich mittels Metal-lierungsreagenz aus den Dihypersilylhalogenorganylplumbanen unter Halogenidentzug Plumbanide erhalten, die in Form getrennter Ionenpaare isolierbar sind. Diese lassen sich in Ana-logie zu den zuvor beschriebenen Plumbanen ebenfalls als Addukt aus Dihypersilylplumbylen und Lithiumorganylen darstellen.rnrnUnter geeigneten speziellen Bedingungen sind neben metallierten auch formal hydridierte Halogenplumbanide zugänglich.rnrnEntsprechend einer geringen Hybridisierung am zentralen Blei-atom, also eines hohen p-AO-Charakters der bindenden Molekülorbitale und s-AO-Charakters des LEP-Orbitals ergeben sich keine trigonal-planaren, sondern Strukturen mit Substituenten-winkeln sogar nahe bei 90°. Die Bindungslängen zum Blei sind deutlich größere als bei den entsprechenden Halogenplumbanen.rnrnEin besonderes Augenmerk der Arbeit liegt auf der Betrachtung von langlebigen und persistenten heteroleptischen Plumbyl-Radikalen, die durch milde Oxidation mittels PbNsi23) aus den Dihypersilylorganylplumbaniden erhalten werden. Während bislang nur homoleptische Vertreter bekannt waren, die aufgrund des sterischen Anspruchs der Substituenten und aufgrund von Hyperkonjugation von axialsymmetrischer nahezu planarer Geometrie sind, findet sich für die heteroleptischen Plumbyle dieser Arbeit eine stärker pyramidale Geometrie. Ausführlich diskutierte EPR-Experimente liefern Spektren, die gut mit Simulationen für die entsprechenden Radikale übereinstimmen.rnrnDie im Zentrum der Betrachtungen dieser Arbeit stehenden Dihypersilylhalogenorganylplumbane stellen somit einen aussichtsreichen und darüber hinaus persistenten und gut zu handhabenden Ausgangspunkt bei der Darstellung neuartiger und interessanter Spezies dar.

We showed that when CA3 pyramidal neurons in the caudal 80% of the dorsal hippocampus had almost disappeared completely, the efferent pathway of CA3 was rarely detectable. We used the mouse pilocarpine model of temporal lobe epilepsy (TLE), and injected iontophoretically the anterograde tracer phaseolus vulgaris leucoagglutinin (PHA-L) into gliotic CA3, medial septum and the nucleus of diagonal band of Broca, median raphe, and lateral supramammillary nuclei, or the retrograde tracer cholera toxin B subunit (CTB) into gliotic CA3 area of hippocampus. In the afferent pathway, the number of neurons projecting to CA3 from medial septum and the nucleus of diagonal band of Broca, median raphe, and lateral supramammillary nuclei increased significantly. In the hippocampus, where CA3 pyramidal neurons were partially lost, calbindin, calretinin, parvalbumin immunopositive back-projection neurons from CA1-CA3 area were observed. Sprouting of Schaffer collaterals with increased number of large boutons in both sides of CA1 area, particularly in the stratum pyramidale, was found. When CA3 pyramidal neurons in caudal 80% of the dorsal hippocampus have almost disappeared completely, surviving CA3 neurons in the rostral 20% of the dorsal hippocampus may play an important role in transmitting hyperactivity of granule cells to surviving CA1 neurons or to dorsal part of the lateral septum. We concluded that reorganization of CA3 area with its downstream or upstream nuclei may be involved in the occurrence of epilepsy.

The purpose of this study was to determine the effects of benzodiazepine in area CA1 of the hippocampus, and to explore possible mechanisms of action for these agents in this brain area. Two distinctly different benzodiazepine-induced changes in hippocampal physiology have been identified. First, benzodiazepine depresses the population spike recorded in stratum pyramidale, indicating a decrease in action potential generation. Second, benzodiazepine decreases the magnitude of post-tetanic potentiation of the population EPSP recorded in stratum radiatum, and shortens the duration. The effect of benzodiazepine on pyramidal cell excitation was reversed by the GABA antagonis bicuculline, and mimicked by GABA itself. Thus the available evidence is consistent with the hypothesis that benzodiazepine acts by enhancing the effect of GABA in this area. In stratum radiatum, on the other hand, the effect of benzodiazepine on post tetanic potentiation of the population EPSP was not altered by bicuculline although bicuculline did antagonize GABA in this area. In addition, application of GABA, while it caused profound changes in the population EPSP,p, did not cause the same changes that were induced by benzodiazepine. Thus the evidence does not support the hypothesis that benzodiazepine is acting in stratum radiatum by enhancing the effects of GABA. ^
