The Zhikong Scallop, Chlamys farreri, is one of the most Important bivalve mollusks cultured in northern China However, mass mortality of the cultured C farreri has posed a serious threat to the maricultural Industry in recent years. Acute Viral Necrobiotic Virus (AVNV) is believed as an important etiological agent causing the scallop mass mortalities To understand the mechanism behind the AVNV associated scallop disease and mortality, we assessed the physiological and immune responses of C farreri to the virus infection using oxygen consumption rate, ammonium-nitrogen excretion rate, hemocyte copper, zinc superoxide dismutase gene expression, and plasma superoxide dismutase activity and alkaline phosphatase activity as indicators Scallops challenged by AVNV at 25 C developed typical disease signs 2 days after virus injection Before the disease manifested, scallop oxygen consumption and NH4+-N excretion rates rose and then fell back. Real-time PCR revealed that the hemocyte cytosol Cu, Zn SOD gene expression was upregulated followed by recovery The plasma SOD activity, however, augmented consistently following virus injection Moreover, plasma AKP activity first lowered and then elevated gradually to the highest level at 24 h post virus injection Scallops challenged by AVNV at 17 degrees C neither developed notable disease nor showed obvious responses that could be associated with the virus infection. Seasonal cycles in the rates of oxygen consumption, feeding, absorption efficiency and ammonia-nitrogen excretion in two populations of Mytilus edulis were measured in the field under ambient conditions and related to body size, the gametogenic cycle, the concentration of suspended particulate matter in the water and temperature. Relationships between the various physiological variables are also considered and protein and energy budgets estimated. Both the “scope for growth” and the “relative maintenance cost” were seasonally variable, demonstrating a minimum capacity for growth in the winter and a maximum capacity in the summer. In one population subjected to abnormally high temperatures in the winter the scope for growth was negative for four or five months between January and May. These population differences are discussed and the potential for using physiological integrations in intra-specific comparisons of fitness is identified.

The aim of this study was to determine whether breath 13CO2 measurements could be used to assess the compliance to a diet containing carbohydrates naturally enriched in 13C. The study was divided into two periods: Period 1 (baseline of 4 days) with low 13C/12C ratio carbohydrates. Period 2 (5 days) isocaloric diet with a high 13C/12C ratio (corn, cane sugar, pineapple, millet) carbohydrates. Measurements were made of respiratory gas exchange by indirect calorimetry, urinary nitrogen excretion and breath 13CO2 every morning in post-absorptive conditions, both in resting state and during a 45-min low intensity exercise (walking on a treadmill). The subjects were 10 healthy lean women (BMI 20.4 +/- 1.7 kg/m2, % body fat 24.4 +/- 1.3%), the 13C enrichment of oxidized carbohydrate and breath 13CO2 were compared to the enrichment of exogenous dietary carbohydrates. At rest the enrichment of oxidized carbohydrate increased significantly after one day of 13C carbohydrate enriched diet and reached a steady value (103 +/- 16%) similar to the enrichment of exogenous carbohydrates. During exercise, the 13C enrichment of oxidized carbohydrate remained significantly lower (68 +/- 17%) than that of dietary carbohydrates. The compliance to a diet with a high content of carbohydrates naturally enriched in 13C may be assessed from the measurement of breath 13CO2 enrichment combined with respiratory gas exchange in resting, postabsorptive conditions.

Success achieved in seed production and culture of prawns induced researchers to explore the feasibiltiy of culture of other important crustaceans like lobsters and crabs. The CMFRI initiated a project to study the prospects of spiny lobster’ culture and I was associated with this project from the beginning. Compared to shrimp farming, culture of spiny lobsters is an entirely different proposition due to inherent problems in producing their seeds. The emphasis on .obster culture, therefore, shifted to rearing of juvenile lobsters, which form a major portion in commercial lobster landings in India. Commendable success was achieved in enhancing growth rate of four important speices of spiny lobsters by bilateral eyestalk ablation. But due to strong reservations expressed against the "blinding" of lobsters by people from all walks of life, this technique could not be tested commercially for its economic viability. As in prawns, energetics of reproduction and egg development have not yet been documented in spiny lobsters and so these aspects were included in this study. Though some limited informations on food conversion efficiency are availble in spiny lobster, a detailed study on the effect of size and sexual maturity on food conversion was wanting and this also was included in this investigation. The most dominant spiny lobster in south east and south west coast of India, Panulirus homarus (Linnaeus) was selected for this study. Uptake, accumulation and utilization of minerals and trace elements are important events during embryogenesis of aquatic invertebrates. The scope of this study was widened to include these aspects also in order to understand how specific requirements of minerals and trace elements are met in the developing eggs of these two species of crustaceans.

An isotope dilution model for partitioning phenylalanine uptake by the liver of the lactating dairy cow was constructed and solved in the steady state. If assumptions are made, model solution permits calculation of the rate of phenylalanine uptake from portal vein and hepatic arterial blood supply, phenylalanine release into the hepatic vein, phenylalanine oxidation and synthesis, and degradation of hepatic constitutive and export proteins. The model requires the measurement of plasma fow rate through the liver in combination with phenylalanine concentrations and plateau isotopic enrichments in arterial, portal and hepatic plasma during a constant infusion of [1-13C]phenylalanine tracer. The model can be applied to other amino acids with similar metabolic fates and will provide a means for assessing the impact of hepatic metabolism on amino acid availability to peripheral tissues. This is of particular importance for the dairy cow when considering the requirements for milk protein synthesis and the negative environmental impact of excessive nitrogen excretion.

Marine cartilaginous fish retain a high concentration of urea to maintain the plasma slightly hyperosmotic to the surrounding seawater. In adult fish, urea is produced by hepatic and extrahepatic ornithine urea cycles (OUCs). However, little is known about the urea retention mechanism in developing cartilaginous fish embryos. In order to address the question as to the mechanism of urea-based osmoregulation in developing embryos, the present study examined the gene expression profiles of OUC enzymes in oviparous holocephalan elephant fish (Callorhinchus milii) embryos. We found that the yolk sac membrane (YSM) makes an important contribution to the ureosmotic strategy of the early embryonic period. The expression of OUC enzyme genes was detectable in the embryonic body from at least stage 28, and increased markedly during development to hatching, which is most probably due to growth of the liver. During the early developmental period, however, the expression of OUC enzyme genes was not prominent in the embryonic body. Meanwhile, we found that the mRNA expression of OUC enzymes was detected in the extra-embryonic YSM; the mRNA expression of cmcpsIII in the YSM was much higher than that in the embryonic body during stages 28-31. Significant levels of enzyme activity and the existence of mitochondrial-type cmgs1 transcripts in the YSM supported the mRNA findings. We also found that the cmcpsIII transcript is localized in the vascularized inner layer of the YSM. Taken together, our findings demonstrate for the first time that the YSM is involved in urea-based osmoregulation during the early to mid phase of development in oviparous cartilaginous fish.

Fish oil omega-3 fatty acids exert antiinflammatory effects on patients with ulcerative colitis. However, a comparative study in patients with mild to moderate ulcerative colitis receiving only sulfasalazine or omega-3 fatty acids has not been performed. We sought to detect changes in the inflammatory disease activity with the use of either fish oil omega-3 fatty acids or sulfasalazine in patients with ulcerative colitis. Ten patients (five male, five female; mean age = 48 +/- 12 y) with mild to moderate active ulcerative colitis were investigated in a randomized cross-over design. They received either sulfasalazine (2 g/d) or omega-3 fatty acids (5.4 g/d) for 2 mo. Disease activity was assessed by clinical and laboratory indicators, sigmoidoscopy, histology, and whole-body protein turnover (with N-15-glycine). Treatment with w-3 fatty acids resulted in greater disease activity as detected by a significant increase in platelet count, erythrocyte sedimentation rate, C-reactive protein, and total fecal nitrogen excretion. No major changes in protein synthesis and breakdown were observed during either treatment. In conclusion, treatment with sulfasalazine is superior to treatment with omega-3 fatty acids in patients with mild to moderate active ulcerative colitis. Avaliou-se o desempenho de tilápias-do-nilo (Oreochromis niloticus) criadas em tanques-rede e alimentadas com dietas contendo 27,0 (controle); 25,2; 24,3 e 22,7% de proteína digestível. Aminoácidos cristalinos (L-lisina, DL-metionina e L-treonina) foram adicionados à dieta considerando o conceito de proteína ideal e simulando o perfil de aminoácidos da dieta controle. Os peixes (34,63±19 g) foram alimentados manualmente com dietas isoenergéticas (3.075 kcal de energia digestível/kg de dieta) até saciedade aparente, três vezes ao dia, durante 91 dias. Utilizou-se um delineamento inteiramente casualizado, com quatro tratamentos, três repetições e 25 peixes/unidade experimental. Não foram observados efeitos dos níveis de proteína digestível sobre o ganho de peso, a conversão alimentar, a taxa de eficiência protéica, o peso da carcaça eviscerada, o rendimento de carcaça, o peso e o rendimento de filé, a sobrevivência e o hematócrito. Houve efeito quadrático dos níveis de proteína digestível sobre o consumo; o maior valor foi estimado para a dieta contendo 24,41% de proteína digestível e excreção de nitrogênio, na qual o melhor resultado estimado foi obtido com peixes que receberam a dieta contendo 24,92% de PD. Com a redução nos níveis de proteína digestível, observou-se aumento linear na retenção de nitrogênio. É possível reduzir o nível de proteína digestível, de 27 (29,1% de PB) para 24,3% (26,6% de PB), em dietas para tilápias-do-nilo criadas em tanques-rede. Essa redução deve ser feita por meio da suplementação de aminoácidos (com base no conceito de proteína ideal), considerando o desempenho e o custo da dieta/kg ganho em filé.

O experimento foi conduzido com o objetivo de maximizar a produção e a qualidade dos ovos e minimizar a excreção de nitrogênio nas excretas de poedeiras no final do primeiro ciclo de produção, por meio do fornecimento de aporte adequado de proteína bruta (PB) e aminoácidos sulfurados totais (AAST) na dieta. Foram utilizadas 432 poedeiras Isa Brown, com 52 semanas de idade, distribuídas em delineamento inteiramente casualizado, em esquema fatorial 3 x 3 (PB e AAST) e nove tratamentos (14 e 0,57; 14 e 0,64; 14 e 0,71; 15,5 e 0,57; 15,5 e 0,64; 15,5 e 0,71; 17 e 0,57; 17 e 0,64; 17 e 0,71 % de PB e AAST, respectivamente), com seis repetições de oito aves cada. A duração do experimento foi de 140 dias. Foram avaliadas as características de desempenho, qualidade dos ovos e excreção de nitrogênio nas excretas. A única característica de desempenho influenciada pelos tratamentos foi o peso dos ovos, que apresentou os maiores valores para as combinações de 15,5 e 0,71; 17 e 0,71; 15,5 e 0,64; 14 e 0,71 e 17 e 0,64% de PB e AAST, respectivamente. Não foram observadas diferenças significativas para consumo de ração, porcentagem de postura e de ovos quebrados, massa de ovos, conversão alimentar por dúzia e por massa de ovos e mortalidade. Para os parâmetros de qualidade dos ovos, foram observadas diferenças significativas apenas para as porcentagens de gema e de albúmem. A excreção de nitrogênio foi maior nas aves alimentadas com as rações contendo 17% de PB. Pode-se sugerir que a ração contendo 14% de PB e 0,57% de AAST pode ser utilizada, sem prejuízos no desempenho e na qualidade dos ovos, e ainda contribui para a redução da excreção de nitrogênio no ambiente e do custo da ração.

The objective of this study was to evaluate the effect of corn texture and the particle size on broiler performance, carcass yield, nutrient digestibility, and digestive organ morphometrics. In Experiment I, 720 male Cobb chicks were distributed in a completely randomized experimental design with a 2 x 3 factorial arrangement, consisting two corn textures (dented and hard) and three corn particle sizes, was applied, with four replicates of 30 birds each. Corn particle size was classified according to geometric mean diameter (GMD) as fine - 0.46 mm; medium - 0.73 mm, and coarse - 0.87 mm. In Experiment II, 120 broiler chicks were used to evaluate corn digestibility during the periods of 16 to 22 days and 35 to 41 days of age, using the method of total excreta collection. In Experiment I, corn particle size influenced body weight, average weight gain, feed intake and feed conversion ratio of 21-day-old birds. Corn texture and particle size did not affect the performance of 42-day-old broilers or carcass traits. In Experiment II, there was no influence of corn texture and particle size on digestive organ weights. Dented corn increased nitrogen excretion in the first trial, and hard corn improved dry matter digestibility in the second metabolic trial. Corn with fine particle size promotes better performance of broilers at 21 days of age. Hard corn results in higher dry matter digestibility and lower nitrogen excretion, and consequently higher production factor in 42-day-old broilers.

Com o objetivo de verificar alterarações na composição corporal, diferentes níveis protéicos (12 e 18% PB) foram utilizados na dieta de leitoas de reposição, dos 100 aos 221 dias de idade. Durante esse período foram avaliados o peso corporal, espessura de toucinho (ET), manifestação de estro, características do trato reprodutivo e o balanço nitrogenado. Para a determinação do balanço nitrogenado, as leitoas foram submetidas a dois ensaios de digestibilidade (aos 136 e aos 201 dias de idade). em ambos os ensaios, as leitoas alimentadas com 18% PB apresentaram maior ingestão, retenção e excreção de nitrogênio e maiores níveis de uréia plasmática e urinária (P<0,001). A eficiência da utilização de nitrogênio não foi influenciada pelo nível protéico da dieta (P>0,05). Houve redução de 48% na excreção de nitrogênio com a dieta menos protéica. Leitoas alimentadas com 12% PB apresentaram maior ganho de ET (7,4 mm) em comparação às alimentadas com 18% PB (4,5 mm), dos 100 aos 207 dias de idade (P<0,02). O peso corporal, peso do útero, comprimento dos cornos uterinos e número de corpos lúteos não foram afetados pelo nível de proteína na dieta. A taxa de detecção de estro, até 25 dias após a exposição das leitoas ao cachaço, foi semelhante para as fêmeas recebendo 12% (71%) e 18% PB (75%). Os níveis de proteína da dieta afetaram a composição corporal das fêmeas sem que o peso final, o desencadeamento da puberdade e características do trato reprodutivo fossem afetados, indicando que dietas de baixa proteína podem ser utilizadas como estratégia para aumentar a espessura de toucinho de leitoas de reposição.

Objetivou-se avaliar o efeito do fornecimento, no período das águas, de suplementos formulados com diferentes fontes de proteína sobre os parâmetros nutricionais de bovinos de corte em recria. Foram utilizados cinco novilhos mestiços Holandês × Zebu com peso vivo médio inicial de 300 kg, fistulados no esôfago e no rúmen, distribuídos em cinco piquetes de Brachiaria decumbens de 0,3 ha, em delineamento quadrado latino incompleto (5 × 5), com quatro períodos e cinco tratamentos, em quatro períodos experimentais de 14 dias. Como tratamentos, avaliaram-se suplementos à base de farelo de soja (FS), farelo de algodão (FA, 38% PB), farelo de glúten de milho (FGM, 60% PB) e farelo de trigo + uréia (FTU) e um tratamento testemunha, constituído apenas de mistura mineral (MM). A quantidade diária de suplemento fornecida foi fixada para fornecer aproximadamente 180 g de PB/dia. As fontes protéicas afetaram apenas o consumo de carboidratos não-fibrosos (CNF) e o de PB, que foi maior quando fornecido o suplemento à base de farelo de algodão e menor quando fornecida a mistura mineral. Não houve efeito das fontes protéicas sobre as digestibilidades total e parcial dos nutrientes. O pH e os níveis de nitrogênio amoniacal do líquido ruminal (N-NH3) não foram influenciados pelas fontes protéicas avaliadas, mas todos os valores mantiveram-se nos limites favoráveis à digestão da forragem. As fontes de proteína não afetaram a eficiência microbiana, em média 9,96 g PBmic/100g NDT, nem as concentrações de nitrogênio uréico no plasma (NUP), média de 12,78 mg/dL, e a excreção de nitrogênio na urina (NUr), média de 63,14 g/dia.

After up to 21 days without food, adult male quails (Coturnix coturnix japonica) lost about 45% of the initial body weight (100-150 g). As in naturally fast-adapted and larger birds, three phases were identified during prolonged fasting in quails. Phase I lasted 2-3 days and was characterized by a rapid decrease in the rate of body weight loss and high fat mobilization. Phase II was longer and characterized by a slow and steady decline in the rates of body weight loss and of nitrogen excretion. The third (critical) period was marked by an abrupt increase in the rates of body weight loss and of nitrogen excretion. Despite their small size, the duration of phase II in quails was relatively long, a clear advantage for the study of the relationships between the several metabolic events that occur during this crucial adaptative period. Also, the beginning of phase III could be precisely determined. Changes in blood glucose, plasma FFA and triacylglycerols levels, as well as in liver and carcass lipid content were similar to those found in other species of birds. Therefore, quails seem to be a suitable model to investigate the biochemical mechanisms involved in the metabolic adjustments to prolonged food deprivation in non fasting-adapted birds. © 1995.

Heat stress causes significant economic losses on broilers production due to poorer performance and carcass quality. Considering that protein has the highest heat increment among nutrients, it has been suggested that protein levels should be reduced in diets for heat-exposed broilers. Nevertheless, there are no conclusive results on the benefits of such practice, and further studies should be performed to elucidate some reported discrepancies. Thus, a trial was carried out to evaluate the effects of dietary protein levels (17, 20 and 23%) and environmental temperature (22 and 32°C) on the performance, nutrients digestibility, and energy and protein metabolism of broiler chickens from 21 to 42 days of age. Nutrients digestibility was determined by total excreta collection, and energy and protein metabolism was evaluated by comparative slaughter method. It was concluded that (1) heat exposure impairs broilers performance and increases nitrogen excretion, but do not change nutrients digestibility; (2) high-protein diets are technically feasible and promotes lower heat production for broilers reared under thermoneutral or hot environments, however, high-protein diets increases nitrogen excretion. © Asian Network for Scientific Information, 2007.
