This study focused on the development of a sensitive enzymatic biosensor for the determination of pirimicarb pesticide based on the immobilization of laccase on composite carbon paste electrodes. Multi- walled carbon nanotubes(MWCNTs)paste electrode modified by dispersion of laccase(3%,w/w) within the optimum composite matrix(60:40%,w/w,MWCNTs and paraffin binder)showed the best performance, with excellent electron transfer kinetic and catalytic effects related to the redox process of the substrate4- aminophenol. No metal or anti-interference membrane was added. Based on the inhibition of laccase activity, pirimicarb can be determined in the range 9.90 ×10- 7 to 1.15 ×10- 5 molL 1 using 4- aminophenol as substrate at the optimum pH of 5.0, with acceptable repeatability and reproducibility (relative standard deviations lower than 5%).The limit of detection obtained was 1.8 × 10-7 molL 1 (0.04 mgkg 1 on a fresh weight vegetable basis).The high activity and catalytic properties of the laccase- based biosensor are retained during ca. one month. The optimized electroanalytical protocol coupled to the QuEChERS methodology were applied to tomato and lettuce samples spiked at three levels; recoveries ranging from 91.0±0.1% to 101.0 ± 0.3% were attained. No significant effects in the pirimicarb electro- analysis were observed by the presence of pro-vitamin A, vitamins B1 and C,and glucose in the vegetable extracts. The proposed biosensor- based pesticide residue methodology fulfills all requisites to be used in implementation of food safety programs.

Titanate nanotubes (TNT) with different sodium contents have been synthesised using a hydrothermal approach and a swift and highly controllable post-washing processes. The influence of the sodium/proton replacement on the structural and morphological characteristics of the prepared materials was analysed. Different optical behaviour was observed depending on the Na+/H+ samples’ content. A band gap energy of 3.27±0.03 eV was estimated for the material with higher sodium content while a value of 2.81±0.02 eV was inferred for the most protonated material, which therefore exhibits an absorption edge in the near visible region. The point of zero charge of the materials was determined and the influence of the sodium content on the adsorption of both cationic and anionic organic dyes was studied. The photocatalytic performance of the TNT samples was evaluated in the rhodamine 6G degradation process. Best photodegradation results were obtained when using the most protonated material as catalyst, although this material has shown the lowest R6G adsorption capability.

The incorporation of small amount of highly anisotropic nanoparticles into liquid crystalline hydroxypropylcellulose (LC-HPC) matrix improves its response when is exposed to humidity gradients due to an anisotropic increment of order in the structure. Dispersed nanoparticles give rise to faster order/disorder transitions when exposed to moisture as it is qualitatively observed and quantified by stress-time measurements. The presence of carbon nanotubes derives in a improvement of the mechanical properties of LC-HPC thin films.

Using low cost portable devices that enable a single analytical step for screening environmental contaminants is today a demanding issue. This concept is here tried out by recycling screen-printed electrodes that were to be disposed of and by choosing as sensory element a low cost material offering specific response for an environmental contaminant. Microcystins (MCs) were used as target analyte, for being dangerous toxins produced by cyanobacteria released into water bodies. The sensory element was a plastic antibody designed by surface imprinting with carefully selected monomers to ensure a specific response. These were designed on the wall of carbon nanotubes, taking advantage of their exceptional electrical properties. The stereochemical ability of the sensory material to detect MCs was checked by preparing blank materials where the imprinting stage was made without the template molecule. The novel sensory material for MCs was introduced in a polymeric matrix and evaluated against potentiometric measurements. Nernstian response was observed from 7.24 × 10−10 to 1.28 × 10−9 M in buffer solution (10 mM HEPES, 150 mM NaCl, pH 6.6), with average slopes of −62 mVdecade−1 and detection capabilities below 1 nM. The blank materials were unable to provide a linear response against log(concentration), showing only a slight potential change towards more positive potentials with increasing concentrations (while that ofthe plastic antibodies moved to more negative values), with a maximum rate of +33 mVdecade−1. The sensors presented good selectivity towards sulphate, iron and ammonium ions, and also chloroform and tetrachloroethylene (TCE) and fast response (<20 s). This concept was successfully tested on the analysis of spiked environmental water samples. The sensors were further applied onto recycled chips, comprehending one site for the reference electrode and two sites for different selective membranes, in a biparametric approach for “in situ” analysis.

Microcystin-LR (MC-LR) is a dangerous toxin found in environmental waters, quantified by high performance liquid chromatography and/or enzyme-linked immunosorbent assays. Quick, low cost and on-site analysis is thus required to ensure human safety and wide screening programs. This work proposes label-free potentiometric sensors made of solid-contact electrodes coated with a surface imprinted polymer on the surface of Multi-Walled Carbon NanoTubes (CNTs) incorporated in a polyvinyl chloride membrane. The imprinting effect was checked by using non-imprinted materials. The MC-LR sensitive sensors were evaluated, characterized and applied successfully in spiked environmental waters. The presented method offered the advantages of low cost, portability, easy operation and suitability for adaptation to flow methods.

A novel artificial antibody for troponin T (TnT) was synthesized by molecular imprint (MI) on the surface of multiwalled carbon nanotubes (MWCNT). This was done by attaching TnT to the MWCNT surface, and filling the vacant spaces by polymerizing under mild conditions acrylamide (monomer) in N,N′-methylenebisacrylamide (cross-linker) and ammonium persulphate (initiator). After removing the template, the obtained biomaterial was able to rebind TnT and discriminate it among other interfering species. Stereochemical recognition of TnT was confirmed by the non-rebinding ability displayed by non-imprinted (NI) materials, obtained by imprinting without a template. SEM and FTIR analysis confirmed the surface modification of the MWCNT. The ability of this biomaterial to rebind TnT was confirmed by including it as electroactive compound in a PVC/plasticizer mixture coating a wire of silver, gold or titanium. Anionic slopes of 50 mV decade−1 were obtained for the gold wire coated with MI-based membranes dipped in HEPES buffer of pH 7. The limit of detection was 0.16 μg mL−1. Neither the NI-MWCNT nor the MWCNT showed the ability to recognize the template. Good selectivity was observed against creatinine, sucrose, fructose, myoglobin, sodium glutamate, thiamine and urea. The sensor was tested successfully on serum samples. It is expected that this work opens new horizons on the design of new artificial antibodies for complex protein structures.

Concrete is the primary construction material for civil infrastructures and generally consists of cement, coarse aggregates, sand, admixtures and water. Cementitious materials are characterized by quasi-brittle behaviour and susceptible to cracking [1]. The cracking process within concrete begins with isolated nano-cracks, which then conjoin to form micro-cracks and in turn macro-cracks. Formation and growth of cracks lead to loss of mechanical performance with time and also make concrete accessible to water and other degrading agents such as CO2, chlorides, sulfates, etc. leading to strength loss and corrosion of steel rebars. To improve brittleness of concrete, reinforcements such as polymeric as well as glass and carbon fibers have been used and microfibers improved the mechanical properties significantly by delaying (but could not stop) the transformation of micro-cracks into macro forms [2]. This fact encouraged the use of nano-sized fillers in concrete to prevent the growth of nano-cracks transforming in to micro and macro forms. Nanoparticles like SiO2, Fe2O3, and TiO2 led to considerable improvement in mechanical performance and moreover, nano-TiO2 helped to remove organic pollutants from concrete surfaces [3].

La diarrea neonatal representa uno de los problemas sanitarios de mayor relevancia en las primeras semanas de vida del cerdo. Provoca importantes pérdidas económicas por morbilidad y mortalidad. El cultivo de enterocitos primarios representa una herramienta valiosa para el estudio de patologías causadas por agentes infecciosos que afectan la integridad del epitelio intestinal. La producción de anticuerpos extraídos a partir de la yema de huevo de gallinas inmunizadas (IgY), es una tecnología innovadora, que ha mostrado ser protectiva contra diarreas causadas por agentes víricos y bacterianos. La nanotecnología permite mejorar la eficiencia en la administración de distintas drogas. Los nanotubos de carbono han ganado una enorme popularidad por sus propiedades y aplicaciones únicas. La investigación sobre los aspectos toxicológicos de estas nanopartículas es escasa. Una vez dentro de la célula, las nanopartículas pueden inducir estrés oxidativo intracelular por perturbar el equilibrio oxidativo. Las hipótesis de trabajo es: La administración de IgY anti-Escherichia coli a través de nanotubos protegerá in vitro e in vivo a los enterocitos de una infección por E. coli previniendo la diarrea neonatal porcina. Los objetivos del trabajo son: Evaluar la protección por un anticuerpo aviario IgY anti-E. coli aplicado mediante nanotubos de carbono a cultivo de enterocitos porcinos primarios sometidos a una post-infección con E. coli; Analizar los efectos secundarios de los nanotubos con IgY anti-E coli en la citotoxicidad, el balance oxidativo y la apoptosis de los enterocitos porcinos cultivados in vitro y Evaluar la acción terapeútica de la IgY anti-E coli aplicada a porcinos y efectos secundarios de la administración con nanotubos. Se implementará un diseño experimental in vitro con diferentes grupos de cultivos con nanotubos, con IgY anti-E. coli e inespecifica y con exposición a E. coli. Se realizará cultivo de enterocitos porcinos primarios con una técnica de disgregación enzimática con colagenasa según protocolo de Bader et al. (2000). Se evaluará la viabilidad por la prueba de azul tripan. Para la obtención del anticuerpo anti-E. coli aviario se aplicarán un total de 3 dosis de E. coli (109 UFC/ml de adyuvante) a gallinas Legorhn en condiciones fisiológicas. Se recolectarán los huevos diariamente. Se purificará la IgY según método de Polson et al. (1985) utilizando PEG 6000. La concentración de IgY se medirá por ELISA de alta sensibilidad. La IgY será incorporada a nanotubos según protocolo de Acevedo et al. 2006. Para analizar los posibles efectos secundarios de los nanotubos se evaluará: 1. Citotoxicidad por técnica de MTT 2. Estrés oxidativo por técnica de TBARS y 3. Apoptosis por técnica de TUNEL.Además, se implementará un diseño experimental in vivo para probar la acción terapeútica de este nutraceútico aplicados a lechones destetados y los efectos secundarios de la administración con nanotubos. Se realizará un cultivo de enterocitos de lechones que previamente fueron tratados con la IgY anti-E. coli administrada mediante nanotubos y efectuarán las técnicas descriptas anteriormente. Los resultados esperados son: Elaboración de un Ac aviario IgY anti-E. coli para prevenir infección de enterocitos, Profundización en el conocimiento acerca de los efectos citotóxicos de los nanotubos de carbono multilamelares, Generación de tratamiento alternativo para enfermedades entéricas porcinas.

The enhanced flow in carbon nanotubes is explained using a mathematical model that includes a depletion layer with reduced viscosity near the wall. In the limit of large tubes the model predicts no noticeable enhancement. For smaller tubes the model predicts enhancement that increases as the radius decreases. An analogy between the reduced viscosity and slip-length models shows that the term slip-length is misleading and that on surfaces which are smooth at the nanoscale it may be thought of as a length-scale associated with the size of the depletion region and viscosity ratio. The model therefore provides a physical interpretation of the classical Navier slip condition and explains why `slip-lengths' may be greater than the tube radius.

BACKGROUND: carbon nanotubes (CNT) can have adverse effects on health. Therefore, minimizing the risk associated with CNT exposure is of crucial importance. The aim of this work was to evaluate if coating multi-walled CNT (MWCNT) with polymers could modify their toxicity, thus representing a useful strategy to decrease adverse health effects of CNT. We used industrially-produced MWCNT uncoated (NT1) or coated (50/50 wt%) with acid-based (NT2) or polystyrene-based (NT3) polymer, and exposed murine macrophages (RAW 264.7 cell line) or Balb/c mice by intratracheal administration. Biological experiments were performed both in vitro and in vivo, examining time- and dose-dependent effects of CNT, in terms of cytotoxicity, expression of genes and proteins related to oxidative stress, inflammation and tissue remodeling, cell and lung tissue morphology (optical and transmission electron microscopy), and bronchoalveolar lavage fluid content analysis.RESULTS: extensive physico-chemical characterization of MWCNT was performed, and showed, although similar dimensions for the 3 MWCNT, a much smaller specific surface area for NT2 and NT3 as compared to NT1 (54.1, 34 and 227.54 m(2)/g respectively), along with different surface characteristics. MWCNT-induced cytotoxicity, oxidative stress, and inflammation were increased by acid-based and decreased by polystyrene-based polymer coating both in vitro in murine macrophages and in vivo in lung of mice monitored for 6 months.CONCLUSIONS: these results demonstrate that coating CNT with polymers, without affecting their intrinsic structure, may constitute a useful strategy for decreasing CNT toxicity, and may hold promise for improving occupational safety and that of general the user.

We studied the feasibility of using halloysite clay nanotubes (HNTs) and carboxyl-functionalised multi-walled carbon nanotubes (COOH-MWCNTs) as antigen carriers to improve immune responses against a recombinant LipL32 protein (rLipL32). Immunisation using the HNTs or COOH-MWCNTs significantly increased the rLipL32-specific IgG antibody titres (p < 0.05) of Golden Syrian hamsters. None of the vaccines tested conferred protection against a challenge using a virulent Leptospira interrogans strain. These results demonstrated that nanotubes can be used as antigen carriers for delivery in hosts and the induction of a humoral immune response against purified leptospiral antigens used in subunit vaccine preparations.

The aim of this study was to evaluate adverse effects of multiwalled carbon nanotubes (MWCNT), produced for industrial purposes, on the human epithelial cell line A549. MWCNT were dispersed in dipalmitoyl lecithin (DPL), a component of pulmonary surfactant, and the effects of dispersion in DPL were compared to those in two other media: ethanol (EtOH) and phosphate-buffered saline (PBS). Effects of MWCNT were also compared to those of two asbestos fibers (chrysotile and crocidolite) and carbon black (CB) nanoparticles, not only in A549 cells but also in mesothelial cells (MeT5A human cell line), used as an asbestos-sensitive cell type. MWCNT formed agglomerates on top of both cell lines (surface area 15-35 μm2) that were significantly larger and more numerous in PBS than in EtOH and DPL. Whatever the dispersion media, incubation with 100 μg/ml MWCNT induced a similar decrease in metabolic activity without changing cell membrane permeability or apoptosis. Neither MWCNT cellular internalization nor oxidative stress was observed. In contrast, asbestos fibers penetrated into the cells, decreased metabolic activity but not cell membrane permeability, and increased apoptosis, without decreasing cell number. CB was internalized without any adverse effects. In conclusion, this study demonstrates that MWCNT produced for industrial purposes exert adverse effects without being internalized by human epithelial and mesothelial pulmonary cell lines. The aim of this study was to determine whether multiwalled carbon nanotubes (MWNCT) are taken up by and are toxic to human intestinal enterocytes using the Caco-2 cell model. Caco-2 cells were exposed to 50 ?g/ml MWCNT (oxidized or pristine) for 24 h, and experiments were repeated in the presence of 2.5 mg/L natural organic matter. Cells displayed many of the properties that characterize enterocytes, such as apical microvilli, basolateral basement membrane, and glycogen. The cell monolayers also displayed tight junctions and electrical resistance. Exposure to pristine and oxidized MWCNT, with or without natural organic matter, did not markedly affect viability, which was assessed by measuring activity of released lactate dehydrogenase (LDH) and staining with propidium iodide. Ultrastructural analysis revealed some damage to microvilli colocalized with the MWCNT; however, neither type of MWCNT was taken up by Caco-2 cells. In contrast, pristine and oxidized MWCNT were taken up by the macrophage RAW 264.7 line. Our study suggests that intestinal enterocytes cells do not take up MWCNT. [Authors]
