Germination of pelleted seeds of different lettuce cultivars was evaluated. The experimental design used was completely randomized, with four replications, in a factorial outline 5 x 4. Seeds of 'Elisa', 'Veronica AF 259', 'Vera' and 'Tainá' were tested. The sodium chloride (NaCl) concentrations used were 0; 50 Mm(2,9222 g/L); 100 Mm (5,8443 g/L); 150 Mm (8,7665 g/L); and 200 Mm (11,6886 g/L). Germination percentage, germination rate and number of normal seedling were evaluated. The concentration of 200 Mm of NaCl resulted in the lowest number of germinated plants. 'Veronica AF 259' germinated in the shortest time using a concentration of 150 Mn of NaCl.

The objective of this study was to evaluate the behavior of morphological Eucalyptus platyphylla when subjected to five levels of soil salinity. The research project was conducted in a greenhouse at the experimental area of the Department of Agricultural Engineering of the FCA / UNESP, Botucatu-SP, utilizing plastic pots filled with soil containing NaCl concentrations, in sufficient quantities to raise the level of electrical conductivity. The experiment was conducted in randomized blocks in factorial scheme 5 x 4, 5 electrical conductivities (1.41, 2.50, 4.50, 6.45 and 8.33 dS m-1) and four repetitions. The results showed that soil salinity did not significantly affect the following parameters: height, green mass and dry matter of shoot, and root and leaf area of the plants.

The influence of ethanol, sulfuric acid and chloride on the corrosion resistance of 316L stainless steel was investigated by means of polarization curves and electrochemical impedance spectroscopy measurements. Over the studied range, the steel corrosion potential was independent of H2SO 4 and NaCl concentrations in aqueous solution. On the other hand, in solution containing 65 wt.% ethanol and 35 wt.% water, the corrosion potentials were higher than those obtained in aqueous solution. Besides, the steel corrosion potential was affected by the addition of H2SO4 and NaCl in solution. In solutions with and without ethanol, plus 0.35 wt.% NaCl, the presence of 1 wt.% H2SO4 inhibited the appearance of pitting corrosion. © 2013 Sociedade Brasileira de Química.

The lipase produced by a newly isolate Sporidiobolus pararoseus strain has potential catalysis ability for esterification reactions. In order to improve its synthetic activity, this work aimed at optimizing 'synthetic lipase' production by submerged fermentation of a conventional media based on peptone, yeast extract, NaCl and olive oil using experimental design technique. According to the results obtained in the first experimental design (2(4-1)), yeast extract and NaCl concentrations were tested to further optimization by response surface methodology. The maximum 'synthetic lipase' activity obtained was 26.9 U/mL in the optimized media (5.0, 6.8, 7.0 and 1.0% (wt/v) of peptone, yeast extract, NaCl and olive oil, respectively), representing a 6.36-fold increase compared to the initial medium. The time course of 'synthetic lipase' production in the optimized condition was evaluated in terms of synthetic activity, protease activity, biomass and total carbon and the maximum synthetic activity was observed during the stationary phase of growth.

Methyl chloride transferase, a novel enzyme found in several fungi, marine algae, and halophytic plants, is a biological catalyst responsible for the production of atmospheric methyl chloride. A previous paper reports the purification of this methylase from Batis maritima and the isolation of a cDNA clone of the gene for this enzyme. In this paper, we describe the isolation of a genomic clone of the methylase gene and the expression of recombinant methyl chloride transferase in Escherichia coli and compare the kinetic behavior of the wild-type and recombinant enzyme. The recombinant enzyme is active and promotes the production of methyl chloride by E. coli under in vivo conditions. The kinetic data indicate that the recombinant and wild-type enzymes have similar halide (Cl−, Br−, and I−)-binding capacities. Both the recombinant and wild-type enzymes were found to function well in high NaCl concentrations. This high salt tolerance resembles the activity of halobacterial enzymes rather than halophytic plant enzymes. These findings support the hypothesis that this enzyme functions in the control and regulation of the internal concentration of chloride ions in halophytic plant cells.

As águas residuárias provenientes da indústria do charque são conhecidas por apresentarem elevado teor de cloreto de sódio, aliado a grandes concentrações de matéria orgânica proveniente do sangue liberado ao longo do processo industrial. Esse tipo de água residuária apresenta potencial para degradação biológica, contudo, o cloreto de sódio, em concentração elevada, pode inibir a atividade dos microrganismos e, em alguns casos, levar sistemas biológicos à falência. No presente trabalho, foi avaliada a viabilidade de degradação anaeróbia de efluente sintético de Charqueada contendo elevado teor de cloreto de sódio, em reator anaeróbio tipo UASB (Upflow Anaerobic Sludge Blanket), em escala de laboratório. Foram utilizados 4 reatores, alimentados com água residuária sintética com características similares à água residuária de Charqueada. O reator 1 foi utilizado como controle, o reator 2 recebeu NaCl e os demais (3 e 4) foram operados na presença de NaCl acrescidos de: betaína e potássio com cálcio, respectivamente. Os compostos citados são conhecidos como antagonizantes, por possuirem capacidade de minimizar o efeito inibitório do sódio sobre o processo de digestão anaeróbia. Os reatores foram inoculados com lodo de reator UASB e submetidos à concentração de 5000 mg/L de matéria orgânica, como DQO. A carga orgânica aplicada foi de 5 Kg/m3.d e os reatores não suportaram tal carga. Reiniciou-se a operação com aumento progressivo da DQO de 500 a 2000 mg/L resultando em cargas orgânicas de 0,5 a 2,0 Kg/m3.d, respectivamente. Após estabilização dos reatores, iniciou-se a fase de introdução de cloreto de sódio (1.500 a 13.500 mg/L) e antagonizantes com aumento progressivo a cada fase. Na presença ou ausência de antagonizantes, os reatores 2, 3 e 4 não tiveram o desempenho alterado até a concentração de NaCl de 6000 mg/L. Na presença de 9000 mg/L de NaCl, a betaína se mostrou pouco efetiva como soluto compatível no reator 3 e os antagonizantes do reator 4, potássio e cálcio, apresentaram efeitos estimulatórios. As morfologias encontradas ao longo do experimento foram cocos, víbrios, bacilos, sarcinas, além de morfologias semelhantes a Methanosarcina sp. e Methanosaeta sp. O aumento da concentração de cloreto de sódio provocou a redição da população de Arqueas.

用动态光散射法研究了不同浓度NaCl对液—液扩散法生长溶菌酶晶体的影响,并测量了晶体生长前后体系的Zeta电势.结果表明,NaCl浓度较高时,在溶菌酶溶液—凝胶界面处会发生液液分层现象,溶液中一直存在较大的聚集体,生长出的晶体质量较差.而在合适的NaCl浓度下,随着溶液Zeta电势降低,溶液中溶菌酶的大的聚集体发生解聚集,生长出的晶体质量较高.

采用不同浓度NaCl溶液(100 mmol/L、200 mmol/L)胁迫处理甜高粱幼苗,测定了叶片中叶绿素含量、脯氨酸含量及三种保护酶活性等生理指标。结果表明:100 mmol/L和200 mmol/L处理的甜高粱幼苗质膜相对透性、脯氨酸和丙二醛含量升高;可溶性蛋白和叶绿素含量降低;保护酶系统中叶片超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性均升高,且在100 mmol/L浓度处理下达到最大,而过氧化物酶(POD)活性呈先升高再降低趋势。NaCl胁迫对甜高粱幼苗的一些生理特性产生一定影响。

One-year-old carob (Ceratonia siliqua L.) rootstock was grown in fertilised substrate to evaluate the effects of NaCl salinity stress. The experiment consisted of seven treatments with different concentrations of NaCl in the irrigation water: 0 (control), 15, 30, 40, 80, 120 and 240 (mmol L(-1)), equivalent to electrical conductivities of 0.0, 1.5, 2.9, 3.9, 7.5, 10.9 and 20.6 dS m(-1), respectively. Several growth parameters were measured throughout the experimental period. At the end of the experiment, pH, extractable P and K, and the electrical conductivity of the substrate were assessed in each salinity level. On the same date, the mineral composition of the leaves was compared. The carob rootstock tolerated 13.4 dS m(-1) for a period of 30 days but after 60 days the limit of tolerance was only 6.8 dS m(-1). Salt tolerance indexes were 12.8 and 4.5 for 30 and 60 days, respectively. This tolerance to salinity resulted from the ability to function with concentrations of Cl(-) and Na(+) in leaves up to 24.0 and 8.5 g kg(-1), respectively. Biomass allocation to shoots and roots was similar in all treatments, but after 40 days the number of leaves was reduced, particularly at the larger concentrations (120 and 240 mmol NaCl L(-1)). Leaves of plants irrigated with 240 mmol NaCl L(-1) became chlorotic after 30 days exposure. However, concentrations of N, P. Mg and Zn in leaves were not affected significantly (P > 0.05) by salinity. Apparently, K(+) and Ca(2+) were the key nutrients affected in the response of carob rootstocks to salinity. Plants grown with 80 and 120 mmol L(-1) of NaCl contained the greatest K. concentration. Na(+)/K(+) increased with salinity, due to an elevated Na(+) content but K(+) uptake was also enhanced, which alleviated some Na. stress. Ca(2+) concentration in leaves was not reduced under salinity. Salinization of irrigation water and subsequent impacts on agricultural soils are now common problems in the Mediterranean region. In the present report and for the first time in the international literature, the impact of the addition of NaCl upon growth and lipid production on the oleaginous yeast Rhodosporidium toruloides was studied. Moreover, equally for first time, lipid production by R. toruloides was performed under non-aseptic conditions. Therefore, the potentiality of R. toruloides DSM 4444 to produce lipid in media containing several initial concentrations of NaCl with glucose employed as carbon source was studied. Preliminary batch-flask trials with increasing amounts of NaCl revealed the tolerance of the strain against NaCl content up to 6.0% (w/v). However, 4.0% (w/v) of NaCl stimulated lipid accumulation for this strain, by enhancing lipid production up to 71.3% (w/w) per dry cell weight. The same amount of NaCl was employed in pasteurized batch-flask cultures in order to investigate the role of the salt as bacterial inhibiting agent. The combination of NaCl and high glucose concentrations was found to satisfactorily suppress bacterial contamination of R. toruloides cultures under these conditions. Batch-bioreactor trials of the yeast in the same media with high glucose content (up to 150 g/L) resulted in satisfactory substrate assimilation, with almost linear kinetic profile for lipid production, regardless of the initial glucose concentration imposed. Finally, fed-batch bioreactor cultures led to the production of 37.2 g/L of biomass, accompanied by 64.5% (w/w) of lipid yield. Lipid yield per unit of glucose consumed received the very satisfactory value of 0.21 g/g, a value amongst the highest ones in the literature. The yeast lipid produced contained mainly oleic acid and to lesser extent palmitic and stearic acids, thus constituting a perfect starting material for “second generation” biodiesel

One of the major current challenges for oilwell companies is the extraction of oil from evaporitic zones, also known as pre-salt basins. Deep reservoirs are found under thick salt layers formed from the evaporation of sea water. Salt layers seal the flow of oil from underneath rock formations, which store hydrocarbons and increase the probability of success in oil and gas exploration. Oilwells are cemented using Portland-based slurries to promote mechanical stability and zonal isolation. For pre-salt oilwells, NaCl must be added to saturate the cement slurries, however, the presence of salt in the composition of slurries affects their overall behavior. Therefore, the objective of the present study was to evaluate the effect of the addition of 5 to 25% NaCl on selected properties of Portland-based slurries. A series of tests were carried out to assess the rheological behavior, thickening time, free water and ultrassonic compressive strength. In addition, the slurries were also characterized by thermal analysis, X ray diffraction and scanning electron microscopy. The results showed that the addition of NaCl affected the thickening time of the slurries. NaCl contents up to 10% shortened the thickening time of the slurries. On the other hand, concentrations in excess of 20% not only extended the thickening time, but also reduced the strength of hardened slurries. The addition of NaCl resulted in the formation of a different crystalline phase called Friedel´s salt, where free chlorine is bonded to tricalcium aluminate

One of the major challenges faced nowadays by oil companies is the exploration of pre-salt basins. Thick salt layers were formed in remote ages as a consequence of the evaporation of sea water containing high concentrations of NaCl and KCl. Deep reservoirs can be found below salt formations that prevent the outflow of oil, thus improving the success in oil prospection. The slurries used in the cement operations of salt layers must be adequate to the properties of those specific formations. At the same time, their resulting properties are highly affected by the contamination of salt in the fresh state. It is t herefore important to address the effects of the presence of salt in the cement slurries in order to assure that the well sheath is able to fulfill its main role to provide zonal isolation and mechanical stability. In this scenario, the objective of the present thesis work was to evaluate the effect of the presence of NaCl and KCl premixed with cement and 40% silica flour on the behavior of cement slurries. Their effect in the presence of CO2 was also investigated. The rheological behavior of slurries containing NaCl and KCl was evaluated along with their mechanical strength. Thermal and microstructural tests were also carried out. The results revealed that the presence of NaCl and KCl affected the pozzolanic activity of silica flour, reducing the strength of the hardened slurries containing salt. Friedel´s salt was formed as a result of the bonding between free Cl- and tricalcium aluminate. The presence of CO2 also contributed to the degradation of the slurries as a result of a process of carbonation/bicarbonataion

Objetivou-se nesse estudo avaliar o papel atenuador da espermidina exógena sobre a germinação, vigor de sementes e crescimento de plântulas de cultivares das forrageiras alfafa, guandu e labe-labe submetidas ao estresse salino. A semeadura foi realizada em caixas de plástico tipo gerbox forradas com papel de filtro umedecidos com soluções salinas nas concentrações de 0, 20, 40, 60, 80 e 120 mM de NaCl contendo 0 ou 0,5 mM de espermidina. O delineamento experimental foi inteiramente casualizado em esquema fatorial 3 x 6 x 2 (forrageiras x salinidade x espermidina) com cinco repetições de 25 sementes. As avaliações da germinação foram realizadas no quarto e décimo dias, juntamente com o índice de velocidade de germinação (IVG), avaliado até o nono dia após a semeadura. Determinou-se a concentração salina que reduz em 50% a germinação e a massa seca da parte aérea e raiz no décimo dia após a semeadura. A aplicação de espermidina exógena proporcionou maior acúmulo de massa seca das raízes de labe-labe cv. Rongai, além de ter possibilitado a germinação de 50% das sementes de guandu cv. Caqui até a concentração salina de 72,09 mM de NaCl, assim como maior IVG nas sementes das leguminosas alfafa cv. Crioula e labe-labe cv. Rongai.
