The genetic relationships of colony members in the ant Myrmica tahoensis were determined on the basis of highly polymorphic microsatellite DNA loci. These analyses show that colonies fall into one of two classes. In roughly half of the sampled colonies, workers and female offspring appear to be full sisters. The remaining colonies contain offspring produced by two or more queens. Colonies that produce female sexuals are always composed of highly related females, while colonies that produce males often show low levels of nestmate relatedness. These results support theoretical predictions that workers should skew sex allocation in response to relatedness asymmetries found within colonies. The existence of a relatedness threshold below which female sexuals are not produced suggests a possible mechanism for worker perception of relatedness. Two results indicate that workers use genetic cues, not queen number, in making sex-allocation decisions. (i) The number of queens in a colony was not significantly correlated with either the level of relatedness asymmetry or the sex ratio. (ii) Sex-ratio shifts consistent with a genetically based mechanism of relatedness assessment were seen in an experiment involving transfers of larvae among unrelated nests. Thus workers appear to make sex-allocation decisions on the basis of larval cues and appear to be able to adjust sex ratios long after egg laying.

Forest trees, like oaks, rely on high levels of genetic variation to adapt to varying environmental conditions. Thus, genetic variation and its distribution are important for the long-term survival and adaptability of oak populations. Climate change is projected to lead to increased drought and fire events as well as a northward migration of tree species, including oaks. Additionally, decline in oak regeneration has become increasingly concerning since it may lead to decreased gene flow and increased inbreeding levels. This will in turn lead to lowered levels of genetic diversity, negatively affecting the growth and survival of populations. At the same time, populations at the species’ distribution edge, like those in this study, could possess important stores of genetic diversity and adaptive potential, while also being vulnerable to climatic or anthropogenic changes. A survey of the level and distribution of genetic variation and identification of potentially adaptive genes is needed since adaptive genetic variation is essential for their long-term survival. Oaks possess a remarkable characteristic in that they maintain their species identity and specific environmental adaptations despite their propensity to hybridize. Thus, in the face of interspecific gene flow, some areas of the genome remain differentiated due to selection. This characteristic allows the study of local environmental adaptation through genetic variation analyses. Furthermore, using genic markers with known putative functions makes it possible to link those differentiated markers to potential adaptive traits (e.g., flowering time, drought stress tolerance). Demographic processes like gene flow and genetic drift also play an important role in how genes (including adaptive genes) are maintained or spread. These processes are influenced by disturbances, both natural and anthropogenic. An examination of how genetic variation is geographically distributed can display how these genetic processes and geographical disturbances influence genetic variation patterns. For example, the spatial clustering of closely related trees could promote inbreeding with associated negative effects (inbreeding depression), if gene flow is limited. In turn this can have negative consequences for a species’ ability to adapt to changing environmental conditions. In contrast, interspecific hybridization may also allow the transfer of genes between species that increase their adaptive potential in a changing environment. I have studied the ecologically divergent, interfertile red oaks, Quercus rubra and Q. ellipsoidalis, to identify genes with potential roles in adaptation to abiotic stress through traits such as drought tolerance and flowering time, and to assess the level and distribution of genetic variation. I found evidence for moderate gene flow between the two species and low interspecific genetic differences at most genetic markers (Lind and Gailing 2013). However, the screening of genic markers with potential roles in phenology and drought tolerance led to the identification of a CONSTANS-like (COL) gene, a candidate gene for flowering time and growth. This marker, located in the coding region of the gene, was highly differentiated between the two species in multiple geographical areas, despite interspecific gene flow, and may play a role in reproductive isolation and adaptive divergence between the two species (Lind-Riehl et al. 2014). Since climate change could result in a northward migration of trees species like oaks, this gene could be important in maintaining species identity despite increased contact zones between species (e.g., increased gene flow). Finally I examined differences in spatial genetic structure (SGS) and genetic variation between species and populations subjected to different management strategies and natural disturbances. Diverse management activities combined with various natural disturbances as well as species specific life history traits influenced SGS patterns and inbreeding levels (Lind-Riehl and Gailing submitted).

The emergence of less common fungal pathogens has been increasingly reported in the last decade. We describe 25 cases of Rhodotorula spp. isolated from blood cultures at a large Brazilian tertiary teaching hospital from 1996-2004. We also investigated the in vitro activity of four antifungal drugs, using a standardized method. The median age of patients was 43 years. The majority of patients (88%) had a central venous catheter (CVC) and 10 (40%) were recipients of a bone marrow transplant. The episode was classified as a bloodstream infection (BSI) in 80% of the patients. Amphotericin B deoxycholate was the most common antifungal used and CVC was removed in 89.5% of the patients. Death occurred in four patients (17.4%), all classified as BSI. All strains were identified as R. mucilaginosa by conventional methods. Misidentification of the species was observed in 20% and 5% of the strains with the Vitek Yeast Biochemical Card and API 20C AUX systems, respectively. Amphotericin B demonstrated good in vitro activity (MIC(50/90), 0.5 mu g/ml) and the MICs for fluconazole were high for all strains (MIC(50/90), 64 mu g/ml).

A otospongiose é uma osteodistrofia do osso temporal caracterizada pela reabsorção e neoformação óssea desordenada. Clinicamente, caracteriza-se por disacusia condutiva, neurossensorial, e/ou mista, progressiva e zumbidos. O início dos sintomas ocorre entre 30 e 40 anos de idade sendo rara sua manifestação na infância. Descrevemos o caso de um paciente de 11 anos de idade, com quadro de hipoacusia unilateral progressiva há 5 anos. O exame otorrinolaringológico revelou mancha rubra de Schwartze em orelha esquerda. A audiometria, imitanciometria e a tomografia computadorizada demonstraram características sugestivas de otospongiose. Realizamos uma revisão dos aspectos clínicos, diagnósticos e da conduta terapêutica da otospongiose na infância.

Objetivou-se isolar e identificar a microbiota fúngica em ambientes considerados assépticos, através de exposições com meios de cultivo adequados, em três épocas distintas do ano, antes e imediatamente após as manobras técnicas realizadas em três áreas de trabalho: ambiente aberto, ambiente fechado sem filtração de ar e ambiente fechado com filtração de ar, utilizadas em produção de imunobiológicos. Os meios ágar-Sabouraud e ágar-soja, enriquecidos com 0,2% de extrato de levedura e sem cloranfenicol, foram estudados quanto à sua eficácia no isolamento de bolores e leveduras, considerando-se o número de colônias desenvolvidas e a freqüência dos diversos fungos isolados. Isolaram-se 67 espécimens, sendo 64 fungos filamentosos (bolores) e três leveduras. Dos bolores, 54 pertenciam a 22 gêneros da divisão Deuteromycota, famílias Moniliaceae e Dematiaceae, cinco amostras filamentosas foram incluídas na ordem Agonomycetales (Mycelia Sterilia), e uma amostra foi classificada na divisão Deuteromycota, ordem Sphaeropsidales, classe Coelomycetes. Da divisão Zygomycota, ordem Mucorales, família Mucoraceae, um único mucoráceo foi identificado até gênero. As três leveduras pertenciam também à divisão Deuteromycota (Fungi Imperfecti), família Cryptococcaceae, e foram identificadas como sendo duas Rhodotorula rubra e uma Torulopsis candida. Comprovou-se que o número de colônias isoladas aumentou após a realização das monobras técnicas e que a filtração de ar através de filtros tipo HEPA, reduzindo o número de colônias isoladas nos ambientes fechados, aumenta a segurança do trabalho; comumente é recomendada para áreas de atividade técnica cujos resultados satisfatórios estão diretamente relacionados com uma baixa incidência de contaminantes.

In this paper we report a eight-year prospective study designed to further characterize incidence, epidemiology, specific syndromes, treatment and prognosis associated with fungal infections in neutropenic patients. During the study period 30 fungal infections were diagnosed in 30 patients among 313 episodes of fever and neutropenia (10%). There were 15 cases of candidiasis, 5 pulmonary aspergillosis, 3 sinusitis by Aspergillus fumigatus, 5 infections by Fusarium sp., one infection by Trichosporon sp., and one infection due to Rhodotorula rubra. Blood cultures were positive in 18 cases (60%). The predisposing factors for fungal infection in multivariate analysis were the presence of central venous catheter (p<0.001), longer duration of profound (<100/mm³) neutropenia (p<0.001), the use of corticosteroids (p<0.001), gram-positive bacteremia (p=0.002) and younger age (p=0.03). In multivariate analysis only recovery of the neutropenia (p<0.001) was associated with good prognosis whereas the diagnosis of infection by Fusarium sp. (p=0.006) was strongly associated with a poor outcome. The death rate was 43%. There was no statistically significant difference in the death rate between patients who did receive (52%) or did not receive (50%) antifungal treatment. Identifying patients at risk, specific syndromes and prognostic factors may help to reduce the high mortality associated with disseminated fungal infections in neutropenic patients.

Foi amostrada fítossociologicamente 1 ha. de floresta tropical densa de terra-firme, em um trecho da Estrada da Várzea, que liga Manaus aos municípios de Silves c Itapiranga, no estado do Amazonas. O trabalho teve por objetivo analisar a composição florística e os parâmetros fitossociológicos, necessários para a determinação do índice de valor de importância ecológica das espécies (IVI), bem como a estrutura da vegetação. A distribuição da amostra foi efetuada mediante a análise da imagem de satélite. A amostragem foi na forma de transecto de 10 x 1 .000m dividida em 20 subparcelas de 10 x 50m, totalizando 10.000rn2. Foram abordados todos os indivíduos arbóreos, palmeiras, cipós e ervas terrestres com CAP ≥ 30cm, dos quais obteve-se amostras botânicas para posterior identificação, a qual obedeceu ao sistema de classificação de Cronquist. Como resultado foram registrados 527 indivíduos, que estão distribuídos em 47 famílias, 118 gêneros e 196 espécies. As famílias que apresentaram maior diversidade foram Lecythidaceae (20), Lauraceae (19), Sapotaceae (17), Chrysobalanaceae, Burscraceae (12) c Annonaceae (9) representando 47% da diversidade familiar, demonstrando que a diversidade local está concentrada em poucas famílias. As espécies com maior valor de importância (IVI) foram Goupia glabra Aubl. (9,34), Ocotea rubra(Meiss.) Allen (8,71), influenciados pela exuberância dos seus diâmetros, representados na dominância relativa. Contudo, quando da comparação de densidade local com as obtidas em outros trabalhos, sob os mesmos critérios amostrais, concluiu-se que o local é menos abundante quanto ao número de indivíduos por hectare. Todavia, a diversidade por família c espécies não difere dos resultados alcançados em outras áreas da Amazônia, para florestas de terra-firme.

O presente trabalho avaliou o desempenho de seis cultivares de alface em ambiente protegido. O delineamento experimental foi o de blocos ao acaso, com seis tratamentos e quatro repetições. As cultivares de alface avaliadas foram: Hortência, Rubra, Mônica SF1, Regina de Verão, Verônica e Grandes Lagos 659. A temperatura do ar dentro do ambiente protegido oscilou entre 24,4 ºC e 31,5oC durante o cultivo. Rubra e Grandes Lagos 659 apresentaram os melhores desempenhos, com rendimentos médios superiores a 28.000 kg.ha-1 e massa fresca por planta de 176,7 g e 184,4 g, respectivamente.

Este trabalho teve como objetivo agrupar espécies com características de teor de umidade e densidade básica da madeira semelhantes para compor cargas mistas de espécies para secagem em câmaras industriais de secagem da madeira. Foram amostradas nove espécies em floresta de terra firme na Amazônia Central com três repetições por espécie. Em todas as espécies os teores de umidade não diferiram entre base e topo do tronco, o que confere estabilidade da madeira durante o processo de secagem. Os valores de densidade básica variaram de 0,561 a 0,904 g cm-3. A partir do presente estudo foi possível separar as espécies em três grupos com base na densidade básica e teor de umidade, o primeiro com as espécies Minquartia guianensis, Lecythis poiteaui, Mezilaurus itauba, Manilkara huberi e Brosimum rubescens que são consideradas madeiras pesadas (densidade variando de 0,835 a 0,904 g cm-3) , de secagem mais lenta ou difícil. O segundo grupo contem as espécies Clarisia racemosa e Ocotea rubra (densidade média de 0,665 e 0,720 g cm-3, respectivamente) e o terceiro grupo com as espécies Parkia paraensis e Brasimum parinarioides que apresentam densidade média e secagem rápida (densidade de 0,561 e 0,588 g cm-3, respectivamente).

The author has studied the domatia appearing in the Rubiaceae family by examining 278 species distributed among 95 genera; and she has verified that 51 species belonging to 29 genera have domatia fitting following types according to the Chevalier's classification: in the "touffe de poils", "em pertuis" and " enpochette". Fourtheen species showed domatia that has chamber and outlet orifice. The others 29 species present domatia either as aglomerates-hair, clusters-hair or scattered hairs and variations of this types; eight species present domatia "em pochette". On Paurichiantha rubra (Benth.) Brem., Rondelettia purdiei Hook f., and Randia cladantha K. Schum the domatia also appear in the axils nervure of several orders; and also in Psychotria racemosa Aubl., they are located in the axil of the angle toward the leaf base. The author observed for the first time two types of domatia in the same leaf on Psychotria fortuita Standi, and on type of domatia, with hairs, that is formed by a fold on the blade on Chomelia tenuiflora Benth.

Simple and colonial ascidians were collected at different depths at Arraial do Cabo, Rio de Janeiro, between 2000 and 2003. The collection here presented includes representatives of the families Clavelinidae (Clavelina oblonga), Polycitoridae (Cystodytes dellechiajei), Polyclinidae (Polyclinum constellatum and Polyclinum molle sp. nov.), Holozoidae (Distaplia bermudensis), Ascidiidae (Ascidia sydneiensis and Phallusia nigra), Styelidae (Botrylloides giganteum, Botrylloides nigrum, Symplegma brakenhielmi, Symplegma rubra, Polyandrocarpa anguinea, Eusynstyela floridana, Eusynstyela tincta and Styela plicata), Pyuridae (Herdmania pallida and Microcosmus exasperatus). Didemnids were also collected in the area but were not considered here. Of the 17 species found, one (Polyclinum molle sp. nov.) is a new species in the family Polyclinidae and the others are all species with tropical distribution.

The genus Pycnothele Chamberlin, 1917 is revised. The Brazilian species Pycnothele perdita Chamberlin, 1917, P. singularis (Mello-Leitão, 1934) and P. auronitens (Keyserling, 1891) are redescribed, diagnosed and illustrated. The females of P. auronitens and P. singularis are described for the first time and P. auripila (Mello-Leitão, 1946) from Uruguay, currently a junior synonym of P. auronitens, is revalidated. Pycnothele piracicabensis (Piza, 1938) is transferred to Rachias Simon, 1892, with which it shares the morphology of the sexual structures, restoring the original combination. Five new species are proposed for Brazil: Pycnothele rubra sp. nov., P. jatai sp. nov. and P. araraquara sp. nov., from state of São Paulo, P. arapongas sp. nov., from state of Paraná, and P. gauderio sp. nov., from state of Rio Grande do Sul. After this revision the genus includes ten Neotropical species. Maps with distribution of all species is presented for the first time and the occurrence of species of the genus is enhanced for the Brazilian state of Paraná.

One of the main opportunistic fungal infections amongst immunocompromised individuals is oral candidosis, which has been found in up to 90% of human immunodeficiency virus (HIV)-infected patients. This study employed yeasts isolated from the saliva and oral cavities of 114 HIV-infected patients living in Campinas, São Paulo. Of the isolates, 57.8% were identified as Candida albicans and 42.1% as non-C. albicans. The latter isolates were subsequently identified as C. krusei (7.5%), C. lusitaniae (5.2%), C. tropicalis (4.6%), C. parapsilosis (4.6%), C. glabrata (2.8%), C. kefyr (1.7%), C. guilliermondii (1.7%), C. intermedia (1.1%), C. norvegensis (0.5%), and Rhodotorula rubra (1.7%). Susceptibility of the isolates to amphotericin B, fluconazole, miconazole, and itraconazole was also determined by a microdilution method adopted by the National Committee for Clinical Laboratory Standards. The isolates demonstrated various susceptibilities to the antifungal agents. In particular 29 C. albicans and 13 non-C. albicans isolates showed low susceptibility to FLCZ (> 64 µg/ml). This study revealed huge diversity of Candida species, in particular the increasing emergence of non-C. albicans associated with the oral flora of HIV-infected patients.

It has been shown previously that the laticifer fluid of Calotropis procera (Ait.) R.Br. is highly toxic to the egg hatching and larval development of Aedes aegypti L. In the present study, the larvicidal potential of other laticifer fluids obtained from Cryptostegia grandiflora R.Br., Plumeria rubra L. and Euphorbia tirucalli L. was evaluated. We attempted to correlate larvicidal activity with the presence of endogenous proteolytic activity in the protein fraction of the fluids. After collection, the fluids were processed by centrifugation and dialysis to obtain the soluble laticifer protein (LP) fractions and eliminate water insoluble and low molecular mass molecules. LP did not visibly affect egg hatching at the doses assayed. LP from Cr. grandiflora exhibited the highest larval toxicity, while P. rubra was almost inactive. E. tirucalli was slightly active, but its activity could not be correlated to proteins since no protein was detected in the fluid. The larvicidal effects of LP from C. procera and Cr. grandiflora showed a significant relationship with the proteolytic activity of cysteine proteinases, which are present in both materials. A purified cysteine proteinase (papain) from the latex of Carica papaya (obtained from Sigma) was similarly effective, whereas trypsin and chymotrypsin (both serine proteinases) were ineffective. The results provide evidence for the involvement of cysteine proteinase activity in the larvicidal action of some laticifer fluids. C. procera is an invasive species found in areas infested with Ae. aegypti and thus could prove useful for combating mosquito proliferation. This is the first report to present evidence for the use of proteolytic enzymes as chemical agents to destroy Ae. aegypti larvae.
