The mushroom Agaricus blazei has been extensively investigated because of evidence of its antimutagenic, antitumor, and anticarcinogenic activities. This study investigated the clastogenic and/or anticlastogenic activity of aqueous extract of Agaricus blazei (10% w/v) in drug-metabolizing rat hepatoma tissue cells (HTCs), with continuous treatment and treatment during different phases of the cell cycle. DNA damage was induced utilizing two directacting agents-methyl methane sulfonate and ethyl methane sulfonate-and two indirect-acting agents-2-aminoanthracene and cyclophosphamide. The aqueous extract of A. blazei with either continuous treatment or treatment during different phases of the cell cycle showed clastogenic activity. The results with continuous treatment showed that A. blazei does not protect against DNA damage-inducing agents that are direct acting. Meanwhile, when combined with indirect-acting agents, a protective effect was demonstrated. A protective effect was also found during different phases of the cell cycle when cells were treated with indirect-acting agents. The protective effects against indirect-acting agents (continuous treatment and during the different phases of the cell cycle) suggest that A. blazei may provide some health benefits to the public when used as a functional food.

The effects of crude extracts of the mushroom Agaricus blazei Murrill (Agaricaceae) on both DNA damage and placental form glutathione S-transferase (GST-P)-positive liver foci induced by diethylnitrosamine (DEN) were investigated. Six groups of adult male Wistar rats were used. For two weeks, animals of groups 3 to 6 were treated with three aqueous solutions of A. blazei (mean dry weight of solids being 1.2, 5.6, 11.5 and 11.5 mg/ml, respectively). After this period, groups 2 to 5 were given a single ip injection 200 mg/kg DEN and groups 1 and 6 were treated with 0.9% NaCl. All animals were subjected to 70% partial hepatectomy at week five and sacrificed 4, 24 and 48 h or 8 weeks after DEN or 0.9% NaCl treatments (10th week after the beginning of the experiment). The alkaline comet assay and GST-P-positive liver foci development were used to evaluate the influence of the mushroom extracts on liver cell DNA damage and on the initiation of liver carcinogenesis, respectively. Previous treatment with the highest concentration of A. blazei (11.5 mg/ml) significantly reduced DNA damage, indicating a protective effect against DEN-induced liver cytotoxicity/genotoxicity. However, the same dose of mushroom extract significantly increased the number of GST-P-positive liver foci.

Prophylactic or therapeutic treatments administered with medicinal plants and natural products are used in popular medicine of diverse people since prehistoric times to the present days. Species with medicinal properties are increasingly studied in an attempt to understand their possible effects on organisms and their functioning. This study aimed to analyze the effect of the mushroom Agaricus blazei (aqueous extract) in rabbits subjected to experimental hypercholesterolemia. The animals were divided into two groups (control and treated with the mushroom) whose experimental protocol was divided into three phases: Phase 1, the animals were fed a normal diet to evaluate the physiological level of cholesterol; phase 2, the animals were fed a supplemented diet to induce hypercholesterolemia and in phase 3, the animals of control group continued to take high-cholesterol diet and the animals of treated group high-cholesterol diet including treatment with the mushroom. Weekly, after fasting of 14 hours, blood samples were collected from the animals and its plasma was stored for later measurement of plasmatic cholesterol. In the first phase, the cholesterol level was, on average, 31,30  7,34 mgdL-1. In the second phase, there was a significant increase (p<0,05) in cholesterol level of both groups. During the last phase of the experiment, the mushrooms didn’t cause reduction in plasmatic cholesterol of treated rabbits, however, prevent disease progression, maintaining the cholesterol level established at the beginning of treatment, whereas, in the control group, total serum cholesterol increased significantly at this stage

Agaricus blazei Murrill extracts have previously been shown to have anticarcinogenic and antimutagenic proper-ties. These results suggest that antimutagenic activity, besides the modulation of the immune system, might be involved in the anticarcinogenic action of A. blazei. To investigate the possible antimutagenic effect of A. blazei in vivo, we evaluated its effect on clastogenicity induced by cyclophosphamide (CP) in mice, using the micronucleus test in bone marrow (MNPCE) and in peripheral blood (MNRET). Male Swiss mice were treated with CP (25 or 50 mg/kg i.p.) or with CP plus mushroom solution at three different temperatures: 4, 21, and 60 degreesC. Aqueous solution of a mixture from various lineages of the mushroom inhibited induction of micronuclei by CP in bone marrow and in peripheral blood of mice. In contrast to the mixture of lineages, a single isolated lineage did not lead to a reduction of CP-induced MN frequencies in either bone marrow or blood cells of mice. The results suggest that under certain circumstances these mushrooms exhibit antimutagenic activities that might contribute to an anticarcinogenic effect. (C) 2001 Elsevier B.V. B.V. Agaricus blazei (Ab) has become popularly known for its medicinal properties. Scientifically, it has been tested with regard to its capacity to protect genetic material against damage. We examined different organic extracts (methanolic extract-ME, hexanic extract-HE and n-butanolic extract-BE) and an aqueous extract (AE) of Ab, for their capacity to induce DNA damage as well as for their protective effect. Genetic damage was determined by the chromosomal aberration assay (CA) in CHO-k1 cells for all extracts and the cytokinesis block micronucleus assay (CBMN) in non drug-metabolizing (CHO-k1) and drug-metabolizing (HTC) cell lines for extract BE only. The extracts did not show clastogenicity but showed anticlastogenicity. The greatest percent reduction obtained were with BE (105%) and AE (126%) treatments in CA. BE treatment did not display genotoxicity in CHO-k1, but was genotoxic in HTC. However, BE was shown to be antigenotoxic causing decreased micronucleus frequency in HTC and CHO-k1 cells. These results suggest that all the extracts contained protective substances, but in some cases they could show a genotoxic effect with regard to metabolism. Therefore, these findings warrant caution in the use of this mushroom by the population. (c) 2005 Elsevier Ltd. All rights reserved.

Determinou-se a divergência genética entre cinco isolados do fungo Agaricus blazei pela técnica de RAPD. Dos cinco isolados três não apresentaram qualquer divergência, sendo caracterizados como isolados de uma mesma origem, embora obtidos em locais diferentes do país. Outros dois isolados mostraram-se diferentes do primeiro grupo e divergentes entre si.

Foi avaliado o efeito dos fungos contaminantes Trichoderma sp. e C. olivacearum na produtividade, eficiência biológica e número de cogumelos da produção do A. blazei em composto (mistura de cana-de-açúcar, palha de capim coast-cross, farelo de soja, gesso e calcário calcítico). O delineanamento foi inteiramente casualizado com três tratamentos (Trichoderma sp., C. olivacearum e testemunha) e oito repetições (caixa com 12 kg de composto colonizado com A. blazei). Após a colonização do composto pelo A. blazei, adicionou-se 150g de inóculo à base de triticale de cada um dos fungos contaminantes na superfície do composto seguido da camada de cobertura. O experimento foi conduzido em estufa com cobertura plástica, umidade relativa entre 60-90% e temperatura de 20-34ºC. A produtividade foi determinada pela relação entre a massa fresca de basidiomas e a massa úmida do composto. A eficiência biológica foi determinada pela relação entre a massa fresca de basidiomas e a massa seca do composto ao final do período de colheita. de acordo com os resultados obtidos, os fungos contaminantes C. olivacearum e Trichoderma sp. não afetaram a produtividade, eficiência biológica e número de cogumelos da produção do A. blazei em compostos previamente colonizados.

This work aimed to evaluate the antioxidant activity of Lentinus edodes and Agaricus blazei mushrooms, as well as to measure the content of total phenolic compounds of mushroom extracts and verify the oxidative stability of soybean oil added with mushroom extracts that showed higher antioxidant activity according to the methods of the 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging and the beta-carotene/linoleic acid system. According to the DPPH method, the maximum antioxidant activity for L. edodes and A. blazei methanol extracts was 92.84 and 95.10%, respectively. For the beta-carotene/linoleic acid system, the highest values of antioxidant activity were 93.06% for L. edodes and 78.96% for A. blazei. The content of total phenolic compounds ranged from 7.21 to 128.44 and 26.67 to 134.67 mg gallic acid equivalent/g for L. edodes and A. blazei, respectively. The oxidative stability values provided by the Rancimat method indicated that the two varieties presented similar induction period of 19.85 h.
