Fiber membranes prepared from jute fragments can be valuable, low cost, and renewable. They have broad application prospects in packing bags, geotextiles, filters, and composite reinforcements. Traditionally, chemical adhesives have been used to improve the properties of jute fiber membranes. A series of new laccase, laccase/mediator systems, and multi-enzyme synergisms were attempted. After the laccase treatment of jute fragments, the mechanical properties and surface hydrophobicity of the produced fiber membranes increased because of the cross-coupling of lignins with ether bonds mediated by laccase. The optimum conditions were a buffer pH of 4.5 and an incubation temperature of 60 °C with 0.92 U/mL laccase for 3 h. Laccase/guaiacol and laccase/alkali lignin treatments resulted in remarkable increases in the mechanical properties; in contrast, the laccase/2,2-azino-bis-(3-ethylthiazoline-6-sulfonate) (ABTS) and laccase/2,6-dimethoxyphenol treatments led to a decrease. The laccase/ guaiacol system was favorable to the surface hydrophobicity of jute fiber membranes. However, the laccase/alkali lignin system had the opposite effect. Xylanase/laccase and cellulase/laccase combined treatments were able to enhance both the mechanical properties and the surface hydrophobicity of jute fiber membranes. Among these, cellulase/laccase treatment performed better; compared to mechanical properties, the surface hydrophobicity of the jute fiber membranes showed only a slight increase after the enzymatic multi-step processes.

Microtubule-associated protein 2 (MAP2), a protein linked to the neuronal cytoskeleton in the mature central nervous system (CNS), has recently been identified in glial precursors indicating a potential role during glial development. In the present study, we systematically analyzed the expression of MAP2 in a series of 237 human neuroepithelial tumors including paraffin-embedded specimens and tumor tissue microarrays from oligodendrogliomas, mixed gliomas, astrocytomas, glioblastomas, ependymomas, as well as dysembryoplastic neuroepithelial tumors (DNT), and central neurocytomas. In addition, MAP2-immunoreactive precursor cells were studied in the developing human brain. Three monoclonal antibodies generated against MAP2A-B or MAP2A-D isoforms were used. Variable immunoreactivity for MAP2 could be observed in all gliomas with the exception of ependymomas. Oligodendrogliomas exhibited a consistently strong and distinct pattern of expression characterized by perinuclear cytoplasmic staining without significant process labeling. Tumor cells with immunoreactive bi- or multi-polar processes were mostly encountered in astroglial neoplasms, whereas the small cell component in neurocytomas and DNT was not labeled. These features render MAP2 immunoreactivity a helpful diagnostic tool for the distinction of oligodendrogliomas and other neuroepithelial neoplasms. RT-PCR, Western blot analysis, and in situ hybridization confirmed the expression of MAP2A-C (including the novel MAP2+ 13 transcript) in both oligodendrogliomas and astrocytomas. Double fluorescent laser scanning microscopy showed that GFAP and MAP2 labeled different tumor cell populations. In embryonic human brains, MAP2-immunoreactive glial precursor cells were identified within the subventricular or intermediate zones. These precursors exhibit morphology closely resembling the immunolabeled neoplastic cells observed in glial tumors. Our findings demonstrate MAP2 expression in astrocytic and oligodendroglial neoplasms. The distinct pattern of immunoreactivity in oligodendrogliomas may be useful as a diagnostic tool. Since MAP2 expression occurs transiently in migrating immature glial cells, our findings are in line with an assumed origin of diffuse gliomas from glial precursors.

State-of-the-art production technologies for conjugate vaccines are complex, multi-step processes. An alternative approach to produce glycoconjugates is based on the bacterial N-linked protein glycosylation system first described in Campylobacter jejuni. The C. jejuni N-glycosylation system has been successfully transferred into Escherichia coli, enabling in vivo production of customized recombinant glycoproteins. However, some antigenic bacterial cell surface polysaccharides, like the Vi antigen of Salmonella enterica serovar Typhi, have not been reported to be accessible to the bacterial oligosaccharyltransferase PglB, hence hamper development of novel conjugate vaccines against typhoid fever. In this report, Vi-like polysaccharide structures that can be transferred by PglB were evaluated as typhoid vaccine components. A polysaccharide fulfilling these requirements was found in Escherichia coli serovar O121. Inactivation of the E. coli O121 O antigen cluster encoded gene wbqG resulted in expression of O polysaccharides reactive with antibodies raised against the Vi antigen. The structure of the recombinantly expressed mutant O polysaccharide was elucidated using a novel HPLC and mass spectrometry based method for purified undecaprenyl pyrophosphate (Und-PP) linked glycans, and the presence of epitopes also found in the Vi antigen was confirmed. The mutant O antigen structure was transferred to acceptor proteins using the bacterial N-glycosylation system, and immunogenicity of the resulting conjugates was evaluated in mice. The conjugate-induced antibodies reacted in an enzyme-linked immunosorbent assay with E. coli O121 LPS. One animal developed a significant rise in serum immunoglobulin anti-Vi titer upon immunization.

Au cours des dernières années, la mondialisation des marchés a radicalement modifié les activités économiques des entreprises et le tissu productif à travers la planète. Les restructurations deviennent dès lors des options avantageuses pour les entreprises afin de demeurer concurrentielles. Alors qu’auparavant les restructurations consistaient essentiellement en une réduction d’effectifs dans un site considéré peu rentable, elles sont aujourd’hui polymorphes: fermeture de site, délocalisations, sous-traitance, ouverture de nouvelles unités au pays et hors pays, relocalisation et fusions-acquisitions (Rouleau, 2000) et (AgirE, 2008). Or, les restructurations posent un problème pour l’action syndicale qui doit composer avec ces processus complexes en raison de leur caractère multidimensionnel, multi niveaux et multi acteurs. Les réponses syndicales aux restructurations s’élaborent toujours dans un contexte d’asymétrie de pouvoirs dans la mesure où l’employeur est maître des dimensions spatiales et temporelles des restructurations. L'anticipation syndicale des restructurations apparaît être une stratégie innovante qui permet aux syndicats de réduire ou mieux, de prévenir les conséquences négatives qui découlent des restructurations. Cette recherche a pour objectif d’examiner les facteurs influençant l’anticipation des restructurations en vue d’en dégager les conditions se rattachant à un tel exercice pour les syndicats. À cette fin, deux modèles d'analyse ont été mobilisés. En premier lieu, les ressources de pouvoir syndicales de Lévesque et Murray (2003) permettent d'expliquer l'anticipation syndicale des restructurations dans la mesure où la capacité d'action du syndicat local constitue un facteur qui ne dépend que du syndicat lui-même. Puis, le modèle d'analyse d'AgirE (2008) permet d'expliquer l'espace et le temps d'action alloués au syndicat par l'employeur pour anticiper les restructurations. Les variables indépendantes provenant de ce modèle d'analyse sont les suivantes: les caractéristiques de l'entreprise, les caractéristiques du territoire et la stratégie patronale de restructuration. Cette présente recherche a été réalisée sous la forme d'une étude de cas. Il s'agit ici de l'étude d'un cas critique d'anticipation syndicale d'une restructuration ayant eu cours en 2004-2005 dans une usine du secteur manufacturier québécois. Des entrevues ont été effectuées auprès de représentants syndicaux de l'usine, d'un ex-cadre de l'usine et de deux intervenants importants du milieu communautaire. Les résultats de notre recherche montrent sans équivoque que la capacité d'action du syndicat local est un facteur-clé qui a permis d'expliquer l'anticipation stratégique et opérationnelle de la restructuration interne ayant eu lieu dans l'usine de fabrication et d'emballage de verre. En dépit des caractéristiques de l'entreprise et de la stratégie patronale de restructuration défavorable à l'implication syndicale, le syndicat local a su lui-même se créer un espace d'action lui ayant permis de sauver l'établissement d'une possible fermeture à moyen-long terme. Les caractéristiques territoriales favorables à une action concertée entre les différents acteurs du territoire où est située l'usine se sont également avérées une condition importante pour l'anticipation syndicale de la restructuration. The use of light microscopy and DMACA staining strongly suggested that plant and animal cell nuclei act as sinks for flavanols [1, 2]. Detailed uv-vis spectroscopic titration experiments indicated that histone proteins are the likely binding sites in the nucleus [2]. Here we report the development of a multi-photon excitation microscopy technique combined with fluorescent lifetime measurements of flavanols. Using this technique, (+) catechin, (-) epicatechin and (-) epigallocatechin gallate (EGCG) showed strikingly different excited state lifetimes in solution. Interaction of histone proteins with flavanols was indicated by the appearance of a significant τ2-component of 1.7 to 4.0ns. Tryptophan interference could be circumvented in the in vivo fluorescence lifetime imaging microscopy (FLIM) experiments with 2-photon excitation at 630nm. This enabled visualisation and semi-quantitative measurements that demonstrated unequivocally the absorption of (+)catechin, (-)epicatechin and EGCG by nuclei of onion cells. 3D FLIM revealed for the first time that externally added EGCG penetrated the whole nucleus in onion cells. The relative proportions of EGCG in cytoplasm: nucleus: nucleoli were ca. 1:10:100. FLIM experiments may therefore facilitate probing the health effects of EGCG, which is the major constituent of green tea.

Distillers’ Dried Grains with Solubles (DDGS) is the major by-product of bioethanol and distillery plants. Due to its high content of proteins, water-soluble vitamins and minerals, DDGS has been long marketed as animal feed for livestock. EU legislation on liquid biofuels could raise the demand on bioethanol production in Europe, with a resulting increase in DDGS availability. DDGS contains a spectrum of complex organic macromolecules, particularly polysaccharides, in addition to proteins and vitamins, and its use as a starting raw material within a biomass-based biorefining strategy could lead to the development of multi-stream processes for the production of commodities, platform molecules or speciality chemicals, with concomitant economic benefits and waste reduction for bioethanol plants. The present review aims to outline the compositional characteristics of DDGS and evaluate its potential utilisation as a starting material for the production of added-value products. Parameters of influence on the chemical and physical characteristics of DDGS are discussed. Moreover, various pre-treatment strategies are outlined in terms of efficient DDGS fractionation into several added value streams. Additional processing steps for the production of medium and high added value compounds from DDGS are evaluated and their potential applications in the food and chemical industry sector are identified.

Background: Current classifications of Mental Disorders are centered on Westernized concepts and constructs. Cross-cultural sensitivity emphasizes culturally-appropriate translations of symptoms and questions, assuming that concepts and constructs are applicable.Methods: Groups and individual psychiatrists from various cultures from Asia, Latin America, North Africa and Eastern Europe prepared descriptions of main symptoms and complaints of treatment-seeking women in their cultures, which are interpreted by clinicians as a manifestation of a clinically-relevant dysphoric disorder. They also transliterated the expressions of DSM IV criteria of main dysphoric disorders in their cultures.Results: In many non-western cultures the symptoms and constructs that are interpreted and treated as dysphoric disorders are mostly somatic and are different from the Western-centered DSM or ICD systems. In many cases the DSM and ICD criteria of depression and anxieties are not even acknowledged by patients.Limitations: the descriptive approach reported here is a preliminary step which involved local but Westernized clinicians-investigators following a biomedical thinking. It should be followed by a more systematic-comprehensive surveys in each culture.Conclusions: Westernized concepts and constructs of mental order and disorders are not necessarily universally applicable. Culturallysensitive phenomena, treatments and treatment responses may be diversified. Attempts at their cross-cultural harmonization should take into consideration complex interactional multi-dimensional processes. (c) 2006 Elsevier B.V. All rights reserved.

The nonlinear (NL) optical properties of glassy xBi2O 3-(1-x) GeO2 with x = 0.72 and 0.82 were investigated. The experiments were performed with lasers at 800 nm (pulses of 150 fs) and 532 nm (pulses of 80 ps and 250 ns). Using the Kerr gate technique, we observed that the NL response of the samples at 800 nm is faster than 150 fs. NL refraction indices, | n 2 | ≈ 5 × 10-16 cm2/W, and two-photon absorption coefficients, α 2, smaller than 0.03 cm/GW, were measured at 800 nm. At 532 nm, we measured the NL transmittance of the samples. From the results obtained, we determined α 2 ≈1 cm/GW and excited-state absorption cross-sections of ≈10-22 cm2 due to free-carriers. © 2013 AIP Publishing LLC.

Ba1-xCaxTiO3, Ba1-xSrxTiO3 and Sr1-xCaxTiO3 (x = 0, 0.25, 0.50, 0.75 and 1) nanoparticles were synthesized using the microwave-assisted hydrothermal method. Samples were prepared for 40 minutes at 140°C under a pressure of 3 MPa using an adapted domestic microwave oven. The samples were characterized by X-Ray diffraction (XRD), scanning electron microscopy (FE-SEM), and Raman, photoluminescence (PL) and ultraviolet-visible (UV-Vis) spectroscopies. XRD data show that ceramic powders have crystalline phases associated with a short-range structural disorder. This structural disorder is confirmed by Raman spectral bands indicating multi-phonon processes and the presence of defects or impurities. Such defects account for a broad band in the photoluminescence spectrum in the green light (460 nm) region for all samples. Gap energy variation, obtained from UV-Vis spectra, suggest a non-uniform band structure of these titanates in accordance with the PL results. The morphology of each sample is changed with doping and varies from a spherical to cubic appearance for energy minimization.

This paper reports the synthesis of Au nanoparticles by 30-fs pulses irradiation of a sample containing HAuCl4 and chitosan, a biopolymer used as reducing agent and stabilizer. We observed that it is a multi-photon induced process, with a threshold irradiance of 3.8 x 10(11) W/cm(2) at 790 nm. By transmission electron microscopy we observed nanoparticles from 8 to 50 nm with distinct shapes. Infrared spectroscopy indicated that the reduction of gold and consequent production of nanoparticles is related to the fs-pulse induced oxidation of hydroxyl to carbonyl groups in chitosan. In dieser Arbeit wurden Untersuchungen mit verschiedenenMethoden derMikrospektroskopie unter Verwendung einesPhotoemissions-Elektronenmikroskops(PEEM) durchgeführt. Es wurde ein Energiefilterfür den Einsatzmit dem PEEM aufgebaut und getestet. Dieses Instrument wurdezusammen miteinem Multilayer-Monochromator am Synchrotron eingesetzt.Der Energiefilterwurde unter anderem für Photoelektronenspektroskopie anAuxCs1-xund Pb/Si-Schichten, sowie zur Charakterisierung desMonochromators eingesetzt.
Die Transmissionsfunktion eines abbildendenGegenfeldanalysators fürdas PEEM wurde berechnet und mit Messungen verglichen. InÜbereinstimmungmit der Berechnung zeigt sich ein charakteristischesasymmetrisches Profilder mit dem Analysator gemessenen Photoelektronenpeaks. Eswurde eine Verbesserungder Ortsauflösung durch energieselektive Abbildungnachgewiesen.
Mit dem PEEM wurde erstmals Mehr-Photonen Photoemissionzur Abbildungausgenutzt. Es wurde ein Verfahren gezeigt, mit dem sich dieOrdnung dernichtlinearen Photoemission aus zwei mit unterschiedlicherLaserleistungaufgenommenen Bildern lateral abbilden läßt. DiePolarisationsabhängigkeitder Photoemission von glatten Probenstellen läßtsich mit einercos4-Funktion anpassen, wie es fürZwei-Photonen Photoemissionzu erwarten ist. Alle Proben wiesen Zentren erhöhterPhotoemissionsintensitätauf, welche durch Anregung lokalisierter Plasmonen auf derOberflächean kleinen Partikeln entstehen. An diesen erstmalig sichtbargemachten'Hot Spots' zeigt sich eine charakteristischePolarisationsabhängigkeitder Photoemission mit bevorzugter Anregung vonPhotoelektronen bei s-polarisiertemLicht.

In this work, metal nanoparticles produced by nanosphere lithography were studied in terms of their optical properties (in connection to their plasmon resonances), their potential application in sensing platforms - for thin layer sensing and bio-recognition events -, and for a particular case (the nanocrescents), for enhanced spectroscopy studies. The general preparation procedures introduced early in 2005 by Shumaker-Parry et al. to produce metallic nanocrescents were extended to give rise to more complex (isolated) structures, and also, by combining colloidal monolayer fabrication and plasma etching techniques, to arrays of them. The fabrication methods presented in this work were extended not only to new shapes or arrangements of particles, but included also a targeted surface tailoring of the substrates and the structures, using different thiol and silane compounds as linkers for further attachment of, i.e. polyelectrolyte layers, which allow for a controlled tailoring of their nanoenvironment. The optical properties of the nanocrescents were studied with conventional transmission spectroscopy; a simple multipole model was adapted to explain their behaviour qualitatively. In terms of applications, the results on thin film sensing using these particles show that the crescents present an interesting mode-dependent sensitivity and spatial extension. Parallel to this, the penetrations depths were modeled with two simplified schemes, obtaining good agreement with theory. The multiple modes of the particles with their characteristic decay lengths and sensitivities represent a major improvement for particle-sensing platforms compared to previous single resonance systems. The nanocrescents were also used to alter the emission properties of fluorophores placed close to them. In this work, green emitting dyes were placed at controlled distances from the structures and excited using a pulsed laser emitting in the near infrared. The fluorescence signal obtained in this manner should be connected to a two-photon processes triggered by these structures; obtaining first insight into plasmon-mediated enhancement phenomena. An even simpler and faster approach to produce plasmonic structures than that for the crescents was tested. Metallic nanodiscs and nanoellipses were produced by means of nanosphere lithography, extending a procedure reported in the literature to new shapes and optical properties. The optical properties of these particles were characterized by extinction spectroscopy and compared to results from the literature. Their major advantage is that they present a polarization-dependent response, like the nanocrescents, but are much simpler to fabricate, and the resonances can be tailored in the visible with relative ease. The sensing capabilities of the metallic nanodiscs were explored in the same manner as for the nanocrescents, meaning their response to thin layers and to bio-recognition events on their surface. The sensitivity of these nanostructures to thin films proved to be lower than that of the crescents, though in the same order of magnitude. Experimental information about the near field extension for the Au nanodiscs of different sizes was also extracted from these measurements. Further resonance-tailoring approaches based on electrochemical deposition of metals on the nanodiscs were explored, as a means of modifying plasmon resonances by changing surface properties of the nanoparticles. First results on these experiments would indicate that the deposition of Ag on Au on a submonolayer coverage level can lead to important blue-shifts in the resonances, which would open a simple way to tailor resonances by changing material properties in a local manner.

In dieser Arbeit wird die Entwicklung und experimentelle Umsetzung einer kontinuierlichen, kohärenten Lichtquelle im vakuumultravioletten Wellenlängenbereich um 122 nm präsentiert. Diese basiert auf der nichtlinearen optischen Summenfrequenzmischung dreier Fundamentallaserstrahlen in einer mit Quecksilberdampf gefüllten Hohlfaser. Die Wellenlängen der fundamentalen Laser sind dabei an der Niveaustruktur des Quecksilbers orientiert, um eine mehrfach resonante Überhöhung der nichtlinearen Suszeptibilität zu erreichen. Der transversale Einschluss der Lichtfelder in der Faser verlängert die Wechselwirkungszone mit dem nichtlinearen Medium um mehrere Größenordnungen gegenüber dem Regime fokussierter Strahlen und erlaubt so signifikante Steigerungen der Mischeffizienz.rnrnIm Zuge dieser Arbeit wurde neben einer umfassenden mathematischen Analyse des nichtlinearen Mischprozesses unter Einfluss der Fasercharakteristika eine Apparatur zur Erzeugung und Detektion vakuumultravioletter Strahlung entwickelt. Die Generierung ausreichend hoher Dampfdichten innerhalb des 50 µm durchmessenden Faserkerns konnten spektroskopisch nachgewiesen werden.rnrnDas erste erfolgreiche Summenfrequenzmischen zu 121,26 nm in der Faser wurde demonstriert. Die erzielten Mischeffizienzen sind bereits mit denen vergleichbar, welche unter Verwendung fokussierter Strahlen erreicht werden, obwohl eine Phasenanpassung in der Faser bisher nicht möglich war. Die Ergebnisse dieser Arbeit markieren damit einen wichtigen Schritt hin zu Leistungssteigerungen kohärenter, kontinuierlicher vakuumultravioletter Lichtquellen.rnrnEine solche Quelle wird für zukünftige Laserkühlung von magnetisch gefangenem Antiwasserstoff auf dem Lyman-Alpha Übergang, sowie die Rydberganregung von Calciumionen in einer Paulfalle zur Implementierung quantenlogischer Operationen benötigt.rnrnFerner hat eine Untersuchung der, für eine effiziente Konversion essentiellen, 6^1S_0 - 7^1S_0 Zwei-Photonen Resonanz in Quecksilber Hinweise auf eine bis dato experimentell nicht beobachtete, auf einer Mehr-Photonen Anregung beruhende Licht-induzierte Drift ergeben.

A study of hadron production by photons opens unique ways to address a number of fundamental problems in strong interaction physics as well as fundamental questions in Quantum Field Theory. In particular, an understanding of two-photon processes is of crucial importance for constraining the hadronic uncertainties in precision measurements and in searches for new physics. The process of
