We present high resolution profiles for the methane concentration and the carbon isotope composition of methane from surface sediments and from the sediment-water transition in the Black Sea. At shallow water sites methane migrates from the sediment into the water column, and the magnitude of this upward migrating flux depends on the depth of the sulfate-methane transition (SMT) in the sediment. The isotope data reveal that the sediments at shallow water sites are a source for methane depleted in 13C relative to the isotope composition of methane in the water column. At deep water sites the methane concentration first decreases with depth in the sediment to reach lowest values at the Unit I to Unit II transition. Below this transition the concentration increases again. Numerical modeling of methane concentration and isotope data shows that high methane oxidation rates occur in the surface sediment layer, indicating that the removal of methane in the surface sediments is not related to the anaerobic oxidation of methane coupled to sulfate reduction that occurs a few meters deep in the sediment, at the SMT. Instead, near-surface methane consumption in the euxinic Black Sea sediments appears to be related to lithological stratification. Furthermore, a map of the diffusive methane fluxes in the Black Sea surface sediments indicates that approximately half of the Black Sea seafloor acts as a sink for methane and thus limits the flux of methane to the atmosphere.

The giant pockmark REGAB (West African margin, 3160 m water depth) is an active methane-emitting cold seep ecosystem, where the energy derived from microbially mediated oxidation of methane supports high biomass and diversity of chemosynthetic communities. Bare sediments interspersed with heterogeneous chemosynthetic assemblages of mytilid mussels, vesicomyid clams and siboglinid tubeworms form a complex seep ecosystem. To better understand if benthic bacterial communities reflect the patchy distribution of chemosynthetic fauna, all major chemosynthetic habitats at REGAB were investigated using an interdisciplinary approach combining porewater geochemistry, in situ quantification of fluxes and consumption of methane, as well bacterial community fingerprinting. This study revealed that sediments populated by different fauna assemblages show distinct biogeochemical activities and are associated with distinct sediment bacterial communities. The methane consumption and methane effluxes ranged over one to two orders of magnitude across habitats, and reached highest values at the mussel habitat, which hosted a different bacterial community compared to the other habitats. Clam assemblages had a profound impact on the sediment geochemistry, but less so on the bacterial community structure. Moreover, all clam assemblages at REGAB were restricted to sediments characterized by complete methane consumption in the seafloor, and intermediate biogeochemical activity. Overall, variations in the sediment geochemistry were reflected in the distribution of both fauna and microbial communities; and were mostly determined by methane flux.

Geological, mineralogical and microbiological aspects of the methane cycle in water and sediments of different areas in the oceans are under consideration in the monograph. Original and published estimations of formation- and oxidation rates of methane with use of radioisotope and isotopic methods are given. The role of aerobic and anaerobic microbial oxidation of methane in production of organic matter and in formation of authigenic carbonates is considered. Particular attention is paid to processes of methane transformation in areas of its intensive input to the water column from deep-sea hydrothermal sources, mud volcanoes, and cold methane seeps.

Sites 1033 and 1034 of ODP Leg 169S in Saanich Inlet have an unusual diagenetic system, that has the appearance of being depth reversed, i.e. a bacterial methane accumulation zone underlain by a sulphate reduction zone. During the late Pleistocene grey, undifferentiated, glacio-marine clays were deposited with low Corg contents (<0.4 wt.%), and interstitial fluids replete in SO4 (ca. 27 mM), devoid of CH4 and low in nutrients. This indicates oxic conditions are present, reflecting the open exchange of waters with Haro Strait during the Pleistocene before the Saanich Peninsula emerged. In the earliest Holocene (ca. 11,000 years BP) the inlet was formed, severely restricting water circulation, and leading to the presence of anoxic bottom waters. The sediments are laminated and show a dramatic rise to high Corg, Norg and Stot contents (up to 2.5, 0.4, 1.4 wt.%, respectively) over a period of ca. 1000 years. The nutrient concentrations are especially high (TA, NH4, PO4 up to 115 meq/l, 20 mM and 400 µM, respectively), SO4 is exhausted and CH4 is prolific. Stable carbon isotope ratio measurements of CH4 and co-existing CO2 indicate that methanogenesis is via carbonate reduction (delta13C-CH4 ca. -60 to - 70 per mil, delta13C-CO2 ca. +10 per mil). At the sulphate-methane interfaces, both at the near-surface and at 50 mbsf (Site 1033) and 80 mbsf (Site 1034) methane consumption by sulphate reducing bacteria is intensive.

High acoustic seafloor-backscatter signals characterize hundreds of patches of methane-derived authigenic carbonates and chemosynthetic communities associated with hydrocarbon seepage on the Nile Deep Sea Fan (NDSF) in the Eastern Mediterranean Sea. During a high-resolution ship-based multibeam survey covering a ~ 225 km**2 large seafloor area in the Central Province of the NDSF we identified 163 high-backscatter patches at water depths between 1500 and 1800 m, and investigated the source, composition, turnover, flux and fate of emitted hydrocarbons. Systematic Parasound single beam echosounder surveys of the water column showed hydroacoustic anomalies (flares), indicative of gas bubble streams, above 8% of the high-backscatter patches. In echosounder records flares disappeared in the water column close to the upper limit of the gas hydrate stability zone located at about 1350 m water depth due to decomposition of gas hydrate skins and subsequent gas dissolution. Visual inspection of three high-backscatter patches demonstrated that sediment cementation has led to the formation of continuous flat pavements of authigenic carbonates typically 100 to 300 m in diameter. Volume estimates, considering results from high-resolution autonomous underwater vehicle (AUV)-based multibeam mapping, were used to calculate the amount of carbonate-bound carbon stored in these slabs. Additionally, the flux of methane bubbles emitted at one high-backscatter patch was estimated (0.23 to 2.3 × 10**6 mol a**-1) by combined AUV flare mapping with visual observations by remotely operated vehicle (ROV). Another high-backscatter patch characterized by single carbonate pieces, which were widely distributed and interspaced with sediments inhabited by thiotrophic, chemosynthetic organisms, was investigated using in situ measurements with a benthic chamber and ex situ sediment core incubation and allowed for estimates of the methane consumption (0.1 to 1 × 10**6 mol a**-1) and dissolved methane flux (2 to 48 × 10**6 mol a**-1). Our comparison of dissolved and gaseous methane fluxes as well as methane-derived carbonate reservoirs demonstrates the need for quantitative assessment of these different methane escape routes and their interaction with the geo-, bio-, and hydrosphere at cold seeps.

实验研究了添加剂对甲烷气体水合物形成过程的影响。发现微量的表面活性剂降低了甲烷气体水合物在静止反应器中形成的诱导时间,并使水合物快速形成和生长,提高了水合物形成过程中的填充密度。阴离子表面活性剂(十二烷基硫酸钠)对水合物生长的促进作用比非离子表面活性剂(烷基多糖苷)强。液态烃环戊烷降低了水合物形成的诱导时间,但环戊烷不能提高水合物的填充密度。


The effect of additives on methane gas hydrate formation was tested. The induction time of methane hydrate formation was reduced, gas hydrate could grow rapidly, and the methane consumption was improved during hydrate formation in a quiescent cell with micella surfactants. The effect of an anionic surfactant ( sodium dodecyl sulfate) on gas hydrate formation is more pronounced compared to a nonionic surfactant (dodecyl polysaccharide glycoside). Cyclopentane reduced the induction time of hydrate formation, but could not improve the methane consumption during gas hydrate formation in a quiescent cell.

La fonte et l’effondrement du pergélisol riche en glace dans la région subarctique du Québec ont donné lieu à la formation de petits lacs (mares de thermokarst) qui émettent des gaz à effet de serre dans l’atmosphère tels que du dioxyde de carbone et du méthane. Pourtant, la composition de la communauté microbienne qui est à la base des processus biogéochimiques dans les mares de fonte a été très peu étudiée, particulièrement en ce qui concerne la diversité et l’activité des micro-organismes impliqués dans le cycle du méthane. L’objectif de cette thèse est donc d’étudier la diversité phylogénétique et fonctionnelle des micro-organismes dans les mares de fonte subarctiques en lien avec les caractéristiques de l’environnement et les émissions de méthane. Pour ce faire, une dizaine de mares ont été échantillonnées dans quatre vallées situées à travers un gradient de fonte du pergélisol, et disposant de différentes propriétés physico-chimiques. Selon les vallées, les mares peuvent être issues de la fonte de palses (buttes de tourbe, à dominance organique) ou de lithalses (buttes de sol à dominance minérale) ce qui influence la nature du carbone organique disponible pour la reminéralisation microbienne. Durant l’été, les mares étaient fortement stratifiées; il y avait un fort gradient physico-chimique au sein de la colonne d’eau, avec une couche d’eau supérieure oxique et une couche d’eau profonde pauvre en oxygène ou anoxique. Pour identifier les facteurs qui influencent les communautés microbiennes, des techniques de séquençage à haut débit ont été utilisées ciblant les transcrits des gènes de l’ARNr 16S et des gènes impliqués dans le cycle du méthane : mcrA pour la méthanogenèse et pmoA pour la méthanotrophie. Pour évaluer l’activité des micro-organismes, la concentration des transcrits des gènes fonctionnels a aussi été mesurée avec des PCR quantitatives (qPCR). Les résultats montrent une forte dominance de micro-organismes impliqués dans le cycle du méthane, c’est-à-dire des archées méthanogènes et des bactéries méthanotrophes. L’analyse du gène pmoA indique que les bactéries méthanotrophes n’étaient pas seulement actives à la surface, mais aussi dans le fond de la mare où les concentrations en oxygène étaient minimales; ce qui est inattendu compte tenu de leur besoin en oxygène pour consommer le méthane. En général, la composition des communautés microbiennes était principalement influencée par l’origine de la mare (palse ou lithalse), et moins par le gradient de dégradation du pergélisol. Des variables environnementales clefs comme le pH, le phosphore et le carbone organique dissous, contribuent à la distinction des communautés microbiennes entre les mares issues de palses ou de lithalses. Avec l’intensification des effets du réchauffement climatique, ces communautés microbiennes vont faire face à des changements de conditions qui risquent de modifier leur composition taxonomique, et leurs réponses aux changements seront probablement différentes selon le type de mares. De plus, dans le futur les conditions d’oxygénation au sein des mares seront soumises à des modifications majeures associées avec un changement dans la durée des périodes de fonte de glace et de stratification. Ce type de changement aura un impact sur l’équilibre entre la méthanogenèse et la méthanotrophie, et affectera ainsi les taux d’émissions de méthane. Cependant, les résultats obtenus dans cette thèse indiquent que les archées méthanogènes et les bactéries méthanotrophes peuvent développer des stratégies pour survivre et rester actives au-delà des limites de leurs conditions d’oxygène habituelles.
