Degradation of a synthetic tanning agent CNSF (a condensation product of 2-naphthatenesulfonic acid (2-NSA) and formaldehyde) by four activated sludges, two previously characterised bacterial strains, Arthrobacter sp. 2AC and Comamonas sp. 4BC, and the fungus Cunninghamella polymorpha, was studied in batch culture at 25 degrees C by determining the changes in the concentrations of CNSF and its component monomers and oligomers (n2-n11). The loss of individual oligomers was correlated with the length of the NSA-CH2 chain. Approximately 25% of the total CNSF was degraded (i.e. mineralised) by the microbes contained in the four activated sludges and by the two bacterial isolates but with different lag phases and at different overall rates. The decline in CNSF concentration was due almost entirely to the biodegradation of the monomers (34.3% of CNSF) and, in particular, 2-NSA (27% of CNSF). There was no change in the n2-n 11 components. The growth of C. polymorpha, on the other hand, arose from extracellular depolymerisation of CNSF oligomers and the biodegradation of the lower molecular mass products. Between 38% and 42% of total CNSF was degraded by C. polymorpha at 25 degrees C. The order of oligomer degradation was inversely related to degree of polymerisation. Eighty percent and 90% of the n4 and n5 and 100% oligomers n6-n11 were degraded after 120 h. At a higher temperature (37 degrees C) oligomers n4-n11 were degraded completely after 120 h. A combination of biodegradation (75%) and sorption to fungal biomass (25%) accounted for the measured loss of all oligomers from the solution phase. The CNSF degradation rates and the volume of fungal biomass produced (and therefore the extent of biosorption) were dependent on the presence of a second carbon source (both optimum at glucose 5 g/l). This is the first report that identifies and distinguishes between depolymerisation, sorption and biodegradation processes in the removal of CNSF and its component oligomers. The use of combinations of the depolymerising fungus C. polymorpha, and the monomer-degrading bacteria, Arthrobacter sp. 2AC and Comamonas sp. 4BC, have potential for wastewater treatment.

Fifteen years ago subterranean clover (Trifolium subterraneum) and annual medics (Medicago spp.) dominated annual pasture legume sowings in southern Australia, while limited pasture legume options existed for cropping areas of subtropical Australia. Since then a number of sustainability and economic challenges to existing farming systems have emerged, exposing shortcomings in these species and the lack of legume biodiversity. Public breeding institutions have responded to these challenges by developing 58 new annual and short-lived perennial pasture legumes with adaptation to both existing and new farming systems. This has involved commercialisation of new species and overcoming deficiencies in traditional species. Traits incorporated in legumes of Mediterranean Basin origin for the Mediterranean, temperate and southern subtropical climates of Australia include deeper root systems, protection from false breaks (germination-inducing rainfall events followed by death from drought), a range of hardseed levels, acid-soil tolerant root nodule symbioses, tolerance to pests and diseases and provision of lower cost seed through ease of seed harvesting and processing. Ten new species, French serradella (Ornithopus sativus), biserrula (Biserrula pelecinus), sulla (Hedysarum coronarium), gland (Trifolium glanduliferum), arrowleaf (Trifolium vesiculosum), eastern star (Trifolium dasyurum) and crimson (Trifolium incarnatum) clovers and sphere (Medicago sphaerocarpos), button (Medicago orbicularis) and hybrid disc (Medicago tornata x Medicago littoralis) medics have been commercialised. Improved cultivars have also been developed of subterranean (T. subterraneum), balansa (Trifolium michelianum), rose (Trifolium hirtum), Persian (Trifolium resupinatum) and purple (Trifolium purpureum) clovers, burr (Medicago polymorpha), strand (M. littoralis), snail (Medicago scutellata) and barrel (Medicago truncatula) medics and yellow serradella (Ornithopus compressus). New tropical legumes for pasture phases in subtropical cropping areas include butterfly pea (Clitoria ternatea), burgundy bean (Macroptilium bracteatum) and perennial lablab (Lablab purpureus). Other species and cultivars of Mediterranean species are likely to be released soon. The contributions of genetic resources, rhizobiology, pasture ecology and agronomy, plant pathology, entomology, plant chemistry and animal science have been paramount to this success. A farmer survey in Western Australia has shown widespread adoption of the new pasture legumes, while adoption of new tropical legumes has also been high in cropping areas of the subtropics. This trend is likely to increase due to the increasing cost of inorganic nitrogen, the need to combat herbicide-resistant crop weeds and improved livestock prices. Mixtures of these legumes allows for more robust pastures buffered against variable seasons, soils, pests, diseases and management decisions. This paper discusses development of the new pasture legumes, their potential use and deficiencies in the current suite. 脂肪酸是生物体内普遍存在、具重要生理功能的物质,亦是重要的化工原料。研究脂肪酸生物合成及其调控,既是揭示生命活动基本规律的需要,又具巨大的经济价值。多形汉逊氏酵母(Hansenula polymorpha)是一种甲基嗜热酵母,能合成多聚不饱和脂肪酸,是研究脂肪酸生物合成的理想材料之一。为阐明多形汉逊氏酵母细胞中脂肪酸生物合成途径、关键步骤、调节机理,并利用此系统生产有用脂肪酸,我们开展了不饱和脂肪酸生物合成关键酶基因--△9-脂肪酸去饱和酶基因研究。 以P. angusta IFO 1475的P-OLE1基因为探针,Southern杂交分析,发现在亲缘关系很近的不同种类的甲基嗜热酵母如H. pofymorpha、Pichia angusta、P. pastoris、P. methanolica和Candida boMinii中Δ9-脂肪酸去饱和酶基因的结构多形性。 构建了H. polymorpha CBS 1976染色体Δ9-脂肪酸去饱和酶基因座位的限制性酶切图谱,进而分离了3.4 kb BamHI-XhoI基因片段并进行全序列分析,结果表明这个片段含1个与已克隆的酵母Δ59-脂肪酸去饱和酶基因高度同源的、由1353 bp组成的ORF。推导的H-OLE1多肽具脂肪酸去饱和酶的一些基本特征,如含2个结构域:1个位于N一端、含3个保守的组氨酸簇、具催化功能,另1个位于C-端、参与脱饱和反应中电子传递、类似细胞色素b5。将这个序列申报DDBJ,获得Accession number为:AB024576,推导的蛋白的氨基酸序列的Accession number为:BAA75902。 为验证H-OLE1基因的功能,建立了多形汉逊氏酵母DNA电穿孔实验系统,进行了遗传互补测验。发现完整的H-OLE1基因可互补缺乏Δ59-脂肪酸去饱和酶活性的多形汉逊氏酵母营养缺陷型fadl突变体,却不能互补相应的酿酒酵母olel突变体,而由酿酒酵母GAP表达框架和H-OLE1 ORF组成的嵌合基因可互补上述olel突变体。说明H-OLE1基因编码Δ9-脂肪酸去饱和酶,多形汉逊氏酵母的Δ9-脂肪酸去饱和酶和酿酒酵母的脂肪酸脱饱和系统相亲和,而H-OLE1基因的启动子在异源细胞中没有活性。 为研究H-OLE1基因的转录及其调节规律,通过一系列实验,首次找到了可在研究多形汉逊氏酵母基因表达时用作内标的GAP基因。Northern杂交发现,H-OLE1基因在细胞中以较低水平表达,产生1.5 kb的转录子;基因表达略受不饱和脂肪酸的抑制;在多形汉逊氏酵母HOLE1基因的转录调节中,Choi等在酿酒酵母OLE1基因中发现的脂肪酸调节元件FAR可能不是关键的。 利用基因敲除技术,通过转化H-OLE1∷S-LEU2线性DNA到多形汉逊氏酵母二倍体细胞(fadl/FADl)中,首次构建了多形汉逊氏酵母H-OLE1基因的破坏株。遗传学和分子生物学研究表明,破坏株细胞中线性DNA定位串联多拷贝整合到染色体中并置换了fadl突变部位。利用气相色谱分析了ΔH-OLE1破坏株、fadl-2突变株、野生型菌株及含H-OLE1基因转化子的细胞总脂肪酸,发现多形汉逊氏酵母细胞中除18:0→18:1(Δ9)→18:2(Δ9,12)→18:3(Δ9,12,15)这个脂肪酸去饱和主路外,还可能存在其它几个脱饱和反应与延长反应,如16:1(Δ9)→16:2(Δ9,12)→18:2(Δ11,14);16:1(Δ9)→18:1(Δ11)→18:2(Δ11,15)等。 近年维管组织分化研究进展迅速,取得大量可喜结果,也存在许多不足,如细胞分化调节机理,特别是激素诱导的分子机理研究比较薄弱。为建立研究维管组织分化的理想系统,研究嫁接体发育的激素调节机制,在Parkinson和Yeoman发明的离体茎段嫁接系统的基础上,研究了激素对嫁接体发育特别是维管组织分化的影响。 采用不同的嫁接方法,用试管苗对黄瓜离体茎段自体嫁接、亲和性的黄瓜/黑籽南瓜与不亲和性的黄瓜/绿豆离体茎段嫁接组合进行研究,建立了嫁接过程简单、污染率低的试管苗离体茎段嫁接系统。利用往培养基中添加或不加植物激素研究嫁接体发育,发现通过改变培养基中的植物激素,可使亲和的嫁接体难以形成贯通砧木和接穗的维管束桥,也可诱导非亲和性的嫁接体产生维管束桥。初步研究证明利用植物激素可以克服嫁接不亲和性,这一结果是嫁接基础理论研究的一个重要进展,对揭示嫁接亲和性机制具重要意义。由于黄瓜绿豆嫁接组合中,砧木绿豆是可以固氮的豆科植物,研究结果具有潜在的应用前景。 详细地研究了外源IAA和玉米素(ZT)对黄瓜自体嫁接系统中维管束桥形成时间和数目特别是贯通砧木和接穗的管状分子数的影响。当砧木和接穗培养基中都没有添加植物激素时,嫁接接合部难以产生维管束桥,也难以产生贯通的管状分子。当培养基中添加植物激素时,维管束桥数和贯通的管状分子数随激素浓度和种类的不同而不同。本实验的最佳激素条件是:在接穗培养基中加IAA 1.0 mg/L和ZT 0.25 mg/L,在砧木培养基中加ZT 0.25 mg/L。研究表明在试管苗离体茎段自体嫁接系统中,外源激素是嫁接成功的必要条件。试管苗离体茎段嫁接系统是一个理想的研究植物维管组织分化的新系统。 通过对嫁接体发育期接合部及嫁接体各部分IAA、玉米素及玉米素核苷(Z+ZR)的ELISA分析,发现嫁接接合部维管束的再生受IAA和Z+ZR含量的共同调节;连接接穗和砧木维管束桥的分化比维管束的网联要求更高的IAA水平及LAA(Z+ZR)比率。 上述结果为利用嫁接系统研究维管组织分化机理奠定了基础,使进一步研究嫁接体发育的激素调节机理成为可能。

This project was done during a one-year period (2006-2007) with the aim of assessing and evaluating the susceptible and vulnerable habitat of Tajan River estuarine region as well as identifying its ecological features. This region consists of Tajan estuarine region as one of the sub-basins of the Caspian Sea basin which covers a surface of 2km2. In this assessment, 6 riverine estuarine and marine stations were chosen in which non-biotic parameters such as temperature, salinity, dissolved oxygen, pH and nutrients, and biotic parameters such as variation, density, plankton, primary production by chlorophyll-a. Benthos variation density, silt and the organic materials of the sediments were sampled and measured monthly. The amount of chlorophyll-a concentration and primary production showed a lot of seasonal changes at these stations which ranged from0.3 to 96 mg/m3. The results from the primary productions indicated that the eastern station of the estuary had high concentrations of chlorophyll-a during all seasons (96mg/m3). The most important and dominant planktonic groups in this region included Bacillariophyta from plankton and copepoda from zooplankton. The most important Benthos communities consisted of Driessena polymorpha.Cerastoderma lamarki in estuarine region,Chironomus plumosus in riverine region and Hypaniola sp. In marine region. Assessing the annual variation in these three riverine, estuarine and marine regions, phytoplankton with 3.1, Zooplankton with 2.7 and Benthos with 1.9 Showed the most density in the estuarine region. Assessing the annual density, phytoplanktonic (6118967 no . in m3) and zooplanktonic (7272 no . in m3) communities showed the most density in the marine region. Assessing the statistical tests showed that the estuarine and riverine regions had a significant difference in planktonic density (p<0.005) compared with the marine region. Moreover, The zeoplanktonic density in the marine region had a significant difference (p<0.005) with estuarine and riverine regions. Tooki test and one-way variance Analysis showed that in assessing the planktonic groups (p<0.005) and Benthos (p<0.005), there was a significant difference in variation index between river with estuary, and estuary with the sea. The amount of the total annual live biomass of the Benthos resource in Tajan river estuarine region was estimated 757.66 g/m2.

During January 2003 the bathymetric distribution of the cephalopod fauna of the South Georgia and Shag Rocks slope (100-900 m) was investigated using a commercial bottom trawl. Forty-four trawl stations caught 193 cephalopod specimens including six species of octopod and seven of squid. The benthic octopods Pareledone turqueti and Adelieledone polymorpha were abundant in shallow water at South Georgia, being replaced by Thaumeledone gunteri in greater depths. However, neither A. polymorpha nor T gunteri were caught on the adjacent Shag Rocks area. Two specimens of the deep-sea genus Graneledone were caught on the South Georgia slope. The most abundant squid species caught were Moroteuthis knipovitchi, Psychroteuthis glacialis and Slosarczykovia circumantarctica, which are primarily pelagic and may have been taken when their vertical migrations impinged on the slope.

Feeding in Dikerogammarus villosus (Sowinsky, 1894) males was observed in the field and recorded on video in the laboratory. The following feeding modes were recognized: detritus feeding, grazing, particle feeding, coprophagy, predation on benthic and free swimming invertebrates, predation on fish eggs and larvae, as well as feeding on byssus threads of the zebra mussel, Dreissena polymorpha (Pallas, 1771). The feeding methods are described and illustrated with screenshots of video recordings. The very flexible feeding modes of D. villosus, which make diet switches possible, form a trait that must be an important factor in the invasion success of this Ponto-Caspian gammaridean species, and may thus explain for a great deal its high ecosystem impact.

Cannibalism and intraguild predation (IGP) are common amongst freshwater amphipod crustacean aswsemblages, particularly between individuals of different body size, with IGP of smaller by larger species. The decline of Gammarus tigrinus Populations in mainland Europe has been accompanied by the arrival of the Ponto-Caspian invader Dikerogammarus villosus and previous studies have implicated IGP of G. tigrinus by the larger D. villosus as the principal driving force in this replacement. We examined how factors such as microhabitat and body size may mediate both cannibalism within G. tigrinus populations and IGP by D. villosus and thus contribute to field patterns of coexistence and exclusion. A field Survey of an invaded Dutch fake indicated that G. tigrinus and D. villosus differed in distribution. with D. villosus being the numerically dominant amphipod (80-96 %) on the rocky boulder Substrate of the shoreline and G. tigrinus being the dominant amphipod (100 %) in the crushed shell/sand matrix immediately adjacent to this. Laboratory microcosm experiments indicated that G. tigrinus cannibalism, particularly of smaller by larger size classes, may be common. In addition, although D. villosus predation of all G. tigrinus size classes was extreme, the smallest size classes Suffered the highest predation. Indeed, when exposed to D. villosus, predation of larger G. tigrinus was lowest when smaller G. tigrinus were also present. Increasing microhabitat complexity from a simple bare substrate littered with Dreissena polymorpha zebra mussels to a Crushed shell/sand matrix significantly reduced both cannibalism and IGP. Our Study emphasizes the need to consider both life history stages and habitat template, when considering the impacts of biotic interactions and it also emphasizes that complex, interacting factors may be mediating the range expansion of D. villosus.

Invasive alien aquatic species, including marine and freshwater macroinvertebrates, have become increasingly important in terms of both environmental and socio-economic impacts. In order to assess their environmental and economic costs, we applied the Generic Impact Scoring System (GISS) and performed a comparison with other taxa of invaders in Europe. Impacts were scored into six environmental and six socio-economic categories, with each category containing five impact levels. Among 49 aquatic macroinvertebrates, the most impacting species were the Chinese mitten crab, Eriocheir sinensis (Milne-Edwards, 1853) and the zebra mussel, Dreissena polymorpha (Pallas, 1771). The highest impacts found per GISS impact category were, separately; on ecosystems, through predation, as competitors, and on animal production. Eleven species have an impact score > 10 (high impact) and seven reach impact level 5 in at least one impact category (EU blacklist candidates), the maximum score that can be given is 60 impact points. Comparisons were drawn between aquatic macroinvertebrates and vertebrate invaders such as fish, mammals and birds, as well as terrestrial arthropods, revealing invasive freshwater macroinvertebrates to be voracious predators of native prey and damaging to native ecosystems compared with other taxa. GISS can be used to compare these taxa and will aid policy making and targeting of invasive species for management by relevant agencies, or to assist in producing species blacklist candidates.

The Common Tern (Sterna hirundo) is a ground nesting colonial seabird. Terns rely primarily on small prey fishes which they obtain through plunge diving for their survival as well as the survival of their offspring during the breeding season. The zebra mussel (Dreissena polymorpha) is a small bivalve mollusk that invaded North American waters in the late 1980's. Through its suspension feeding, the zebra mussel has the ability to alter the entire aquatic ecosystem, ultimately leading to a reduction in pelagic organisms including small prey fish. The objective of the study was to determine what (if any) indirect effects the invasion of the zebra mussel has had on fish prey captured by terns. The study took place in two separate two-year periods, 1990-91 and 1995-96 on a concrete breakwall off the north shore of Lake Erie near Port Colborne, Ontario. Daily nest checks revealed clutch initiation dates, egg-laying chronology, hatching success and morphological egg characteristics (length and breadth). Behavioural observations included time each sex spent in attendance with its brood, the frequency of feeding chicks and the prey species composition and size fed to chicks as well as to females (courtship feeding). Egg sizes did not differ between study periods, nor did feeding rates to chicks, suggesting that food was not a limiting resource. Terns spent less time with their broods (more time foraging) in the 1995-96 period. However, they also had significantly larger broods and fledged more offspring. The time of each individual foraging trip decreased, suggesting that fish were easier to obtain in 1995 and 1996. Lastly, kleptoparasitism rates decreased, suggesting that the costs of foraging (time, energy) actually decreased as fewer birds adopted this strategy to compensate for what I assumed to be a lack of available food (fish). The only significant difference between the periods of 1990, 1991 and 1995, 1996 was a change in diet. Terns delivered significantly fewer rainbow smelt and more emerald shiner in 1995 and 1996. However, the average size of fish delivered did not change. Thus, there was little impact on prey captured by Common Terns in Lake Erie since the invasion of the zebra mussel.

Foi realizado um estudo da dinâmica de assembléias de Drosofilídeos em oito amostras insulares e continentais de Santa Catarina através de dados coletados em várias visitas ao longo de dois anos. Dentre os resultados obtidos está a estimativa do grau de diversidade destas assembléias. Nossas coletas mostraram que a predição de qual espécie será dominante, num determinado período amostrado, é razoavelmente possível. A análise dos índices de diversidade nos indica que o Morro da Lagoa é o ponto de menor diversidade específica, seguido de Ratones Grande. Contudo, os dois pontos têm um grande número de espécies diferentes, sendo a sua diversidade baixa em função da alta dominância do subgrupo willistoni neles encontrada. Analisando o componente S (número de espécies) da diversidade, nas ilhas pontos de coleta, percebe-se que a ilha maior (Ilha de Santa Catarina) tem realmente um maior número de espécies coletadas – 46 no ponto A além de 10 espécies diferentes coletadas no ponto D (56 espécies no total) – do que as ilhas menores (42, 44, 40 e 50), o que corrobora a teoria da biogeografia de ilhas. No continente, a curva espécie/área se comportou da mesma forma que nas ilhas se considerarmos a diversidade como um todo. Realmente estes pontos foram uns dos que apresentaram maior diversidade, principalmente o ponto F, com um H’ de 2,22, que se manteve com Mata Atlântica Primária até o final do período de coletas, sendo portanto o ponto mais preservado de todos utilizados e, teoricamente, o que apresentava maior diversidade de nichos ecológicos para serem ocupados Outros Drosofilídeos como Zaprionus, Zygotricha, Gitona, Cladochaeta, Diathoneura, Micodrosophila, Leucophenga e Amiota foram coletados. Embora nosso interesse preliminar fosse apenas o gênero Drosophila, a inclusão destes outros gêneros em nosso estudo visou uma maior compreensão das possíveis associações que podem ocorrer entre eles e espécies de Drosophila. Os dois primeiros gêneros foram mais freqüentes nas nossas coletas. Zaprionus indianus foi considerada uma espécie invasora, pois surgiu com freqüências baixíssimas que aumentaram gradualmente nas coletas subsequentes, superando em freqüência as espécies nativas. Isto confirma o caráter generalista e polifágico deste tipo de espécie. Neste trabalho, é relatado o primeiro registro do Gênero Zaprionus (Diptera, Drosophilidae) para o Estado de Santa Catarina, na região litorânea central que inclui as Ilhas de Santa Catarina, Arvoredo, Ratones Grande, Ratones Pequeno e Campeche. Drosophila roerhae, D. unipunctata, D. schineri, D. bifilum, D. fuscolineata, D. meridionalis, D. neosaltans, D. bocainoides e D. platitarsus foram pela primeira vez registradas para a região Sul do Brasil, aumentando, portanto o limite meridional de suas distribuições. Como um ponto de partida para estudar o polimorfismo para inversões cromossômicas em D. neocardini, foi construído um fotomapa de referência dos cromossomos politênicos de glândulas salivares de larvas de terceiro estágio. Pelo menos 258 indivíduos (aproximadamente três núcleos por glândula) de sete diferentes localidades (Sertão do Peri, Ilha do Arvoredo, Serra do Tabuleiro, Ilha de Ratones Grande, Ilha de Ratones Pequeno, Morro da Lagoa da Conceição e Ilha do Campeche, todos no Estado de Santa Catarina) foram analisados e fotomicrografias foram obtidas, até se chegar a um consenso sobre a identidade dos elementos cromossômicos. Uma nova inversão no braço cromossômico IIIL foi registrada e denominada de IIILA. A variabilidade cromossômica encontrada nas espécies de Drosophila do grupo cardini em todas as localidades também foi pesquisada, e foi comparada visando contribuir para uma melhor compreensão da evolução destas comunidades. Analisando o polimorfismo cromossômico de D. polymorpha encontramos nove inversões diferentes pela primeira vez descritas. Uma das inversões novas foi encontrada no cromossomo X, duas outras foram encontradas para o braço IIL; quatro foram catalogadas para o braço cromossômico IIIR e duas inversões novas foram achadas no braço cromossômico IIIL. Com relação ao polimorfismo, em D. neocardini foi encontrada apenas uma nova inversão no braço IIIL e para D. cardinoides uma nova inversão no braço IIIL. O estudo discute as implicações ecológicas e evolutivas deste tipo de polimorfismo, para um maior entendimento da evolução deste grupo de espécies.

Apesar de assembléias de drosofilídeos já terem sido estudadas em diversos ambientes, os manguezais ainda surgem como um ambiente inexplorado. Este trabalho teve como objetivo caracterizar as assembléias de drosofilídeos encontradas nos manguezais da ilha de Santa Catarina, sul do Brasil. Para isso, foram realizadas 28 coletas nos três principais manguezais da ilha – Itacorubi (13 coletas), Tavares (8 coletas) e Ratones (7 coletas) - entre o período de julho de 2002 e julho de 2005. Um total de 82.942 espécimes foi analisado, distribuídos em 69 espécies de seis gêneros. Foi encontrada uma grande dominância de Drosophila simulans Sturtevant, seguida por D. malerkotliana Parshad & Paika, Zaprionus indianus Gupta, D. mediostriata Duda, D. willistoni Sturtevant, D. paulistorum Dobzhansky & Pavan, D. repleta Wollaston, D. polymorpha Dobzhansky & Pavan e D. mercatorum Patterson & Wheeler. As demais espécies não atingiram 1% de abundância relativa. Não foram encontradas diferenças importantes entre os locais, mas as diferenças sazonais foram relevantes. A dinâmica populacional de várias espécies pareceu estar relacionada, experimentando picos de abundância no outono, embora haja algumas exceções importantes O número de indivíduos e a riqueza de espécies observada também foram mais elevados nesta estação, mas a eqüitabilidade e a riqueza de espécies estimada por rarefação se mostraram mais elevadas no inverno. No entanto, algumas importantes variações temporais pareceram não ser relacionadas a fatores que operam sazonalmente. A composição das assembléias pareceu sofrer uma pequena modificação quando são comparadas as amostras de verão e outono com as de inverno e primavera. As assembléias de outono apresentaram uma estrutura típica, caracterizada pela abundância aumentada de D. malerkotliana, enquanto as demais estações se assemelharam mais aos períodos adjacentes do que aos mesmos períodos de anos diferentes.

O gênero Gochnatia é comumente encontrado em diferentes fitofisionomias do Cerrado do Estado de São Paulo, crescendo desde ambientes mais abertos até áreas florestais mais fechadas. Aqui foram comparadas a anatomia foliar e alguns parâmetros ecofisiológicos de duas espécies do gênero Gochnatia, uma arbustiva (Gochnatia barrosii Cabrera) e a outra arbórea (Gochnatia polymorpha (Less.) Cabrera), ambas ocorrendo em área de cerradão na Estação Ecológica de Assis, SP. Encontraram-se diferenças estruturais qualitativas entre as espécies, com G. barrosii apresentando folhas anfiestomáticas, com epiderme unisseriada e G. polymorpha apresentando folhas hipoestomáticas, com epiderme múltipla ou hipoderme, na face adaxial. Além disso, as folhas de G. barrosii apresentaram menores valores para a espessura dos tecidos (com exceção da epiderme na face abaxial) e da folha em relação a G. polymorpha. Foram observadas diferenças na assimilação de CO2 tanto em base de área quanto de massa seca foliar, além de diferenças na área foliar específica, sendo esta maior em G. barrosii. Apesar das folhas de G. barrosii possuírem estrutura bem menos escleromorfa do que as folhas de G. polymorpha, não foram encontradas diferenças na eficiência do uso de água. Os resultados sugerem que espécies de formas distintas de crescimento de um mesmo gênero possuem características foliares diferenciadas para lidar com as variações ambientais a que são submetidas.

The flotation capacity was determined for cells of yeasts strains belonging to the genera Hansenula, Candida and Saccharomyces. A heterogeneous group of yeasts, comprising strains from the three genera, was identified as showing high flotation capacities (degrees of flotation above 50%), which were practically not affected by variations in medium pH in both the synthetic medium and 2% molasses. Thus, the flotation capacity of the cells in this yeast group seemed strongly dependent on the liquid phase properties and/or growth medium composition, more than on the simple variation in pH of the cell suspensions. A second group of strains, belonging to the Saccharomyces genus, including also brewing yeast strains, was identified as having lower flotation capacities (degrees of flotation below 50% at pH 1.5), which showed no alterations or variations significantly affected by the medium pH. Foam volumes obtained with Saccharomyces strains were greater in synthetic media than in molasses owing to the higher air flow rates required for flotation in molasses. The flotation efficiency decreased in molasses in all cases as well as the foam volume, except in the case of Hansenula cells, which showed an increased foam volume. This was probably due to variations in product excretion by the different yeasts and/or differences in cell wall composition.

Flotation is a process of cell separation based on the affinity of cells to air bubbles. In the present work, flotability and hydrophobicity were determined using cells from different yeasts (Hansenulla polymorpha, Saccharomyces cerevisiae, Candida albicans), which were propagated in different media and at different temperatures. Alterations to the supernatant of the cells were also carried out before the flotation assays. The results described here indicate that supernatants of the yeast cells can play a more important role on flotation than cell-wall hydrophobicity. For example, wall-hydrophobicity of strain FLT-01 of S. cerevisiae was high but flotation did not occur when their washed cells were resuspended in water. Additions of neopeptone to cultures of S. cerevisiae and H. polymorpha repressed flotation and increased the volume of foam. An additional task of the present work was to show that the relationship between cell-wall hydrophobicity and flotation performance was dependent on the method used for the measurement of hydrophobicity. Based on the assay procedure, two types of hydrophobicity were distinguished: (a) the apparent hydrophobicity for cells suspended in the medium and expressed by the degree of cell affinity to the organic solvent in the two-phase system supernatant/hexane; (b) the standard hydrophobicity, which was determined for cells suspended in a standard solution (acetate buffer, in the present work) within the acetate buffer/hexane system. Flotation of cells of S. cerevisiae and C albicans were best related to the degree of apparent hydrophobicity (varying with the supernatant composition at the cell/medium interface) rather than to the degree of standard hydrophobicity (varying with the alterations in the wall components, since the liquid phase was constant in the assay). However, depending on the yeast unpredictable results can be obtained. For example, cells of H. polymorpha exhibited good flotation associated to a high degree of standard hydrophobicity while having a lower degree of apparent hydrophobicity. Concerning growth temperature, flotation of cells of C albicans was strongly repressed when the temperature was raised from 30 to 38 degreesC while a similar effect was not observed in cultures of S. cerevisiae and H. polymorpha. It is difficult to understand and predict flotation of yeast cells but simple modifications made to the supernatant of cultures can activate or repress flotation. (C) 2003 Elsevier B.V. B.V.
