Myeloid cell leukemia-1 (Mcl-1) ist ein anti-apoptotisches Mitglied der Bcl-2-Proteinfamilie. Als solches ist es in der Lage, die mitochondriale Aktivierung während der Apoptose zu hemmen. Dadurch schützt es Zellen bei zellulärem Stress (wie z.B. Differenzierung, Proliferation oder Virusinfektion) vor Apoptoseinduktion. Aufgrund dieser Eigenschaft ist es unabkömmlich während der Embryogenese und in verschiedenen hämatopoetischen Zellpopulationen. Des Weiteren ist Mcl-1 als Protoonkogen in verschiedenen humanen Tumorentitäten verstärkt exprimiert und kann so zu einer verminderten Apoptosesensitivität von Tumorzellen beitragen. Auch primäre humane Hepatozyten können nach Mcl-1-Induktion durch Wachstumsfaktorbehandlung gegenüber CD95-vermittelter Apoptose geschützt werden. Daher sollte untersucht werden, welche Bedeutung Mcl-1 im hepatozellulären Karzinom (HCC) und in der gesunden Leber einnimmt. Hierzu wurde zunächst humanes HCC-Gewebe hinsichtlich der Expression von Mcl-1 untersucht. Es konnte gezeigt werden, dass Mcl-1 sowohl auf mRNA- als auch auf Protein-Ebene in HCC-Gewebe verstärkt exprimiert ist im Vergleich zu benachbartem Normalgewebe. Auch in verschiedenen HCC-Zelllinien konnte eine starke Mcl-1-Expression nachgewiesen werden. Diese war vor allem über den PI3K/Akt-Signalweg reguliert. Eine Hemmung dieses Signalwegs führte zu einer Reduktion der Mcl-1-Expression und so zu einer Sensitivierung der Zellen gegenüber verschiedenen Chemotherapeutika und zielgerichteten Therapien. Des Weiteren wurde die Mcl-1-Expression spezifisch durch RNA-Interferenz gehemmt. Auch hier konnte gezeigt werden, dass Zellen mit unterdrückter Mcl-1-Expression deutlich sensitiver gegenüber verschiedenen Apoptose-induzierenden Substanzen reagierten. Eine kombinierte Hemmung der Mcl-1-Expression und der PI3-Kinase führte schließlich zu einer nochmals verstärkten Sensitivierung. Im Gegensatz dazu führte eine Überexpression von Mcl-1 zu einer Hemmung der Apoptoseinduktion. Im zweiten Teil der Arbeit wurde eine Mauslinie etabliert, welche spezifisch in Hepatozyten kein Mcl-1 exprimiert, um so die Bedeutung von Mcl-1 für die Leber in vivo zu untersuchen. Es zeigte sich, dass Mcl-1flox/flox-AlbCre-Mäuse bereits im Alter von acht Wochen eine verminderte Lebergröße aufweisen. Dies wurde verursacht durch spontane Apoptoseinduktion in den Mcl-1 negativen Hepatozyten. Hierdurch kam es zu einer Leberschädigung, ersichtlich durch erhöhte Transaminasenwerte, erhöhte Caspase-3-Aktivierung, und Schädigung der Gewebsstruktur. Zudem war als kompensatorischer Effekt die Zellproliferation erhöht, ohne dass sich jedoch das Lebergewicht an das von Kontrolltieren anglich. Interessanterweise kam es in Mcl-1flox/flox-AlbCre-Mäusen als Folge der chronischen Leberschädigung zur Entwicklung einer Leberfibrose, ersichtlich durch eine verstärkte Collageneinlagerung. Weiterhin reagierten Mcl-1flox/flox-AlbCre-Mäuse wesentlich empfindlicher gegenüber Todesrezeptor-vermittelter Apoptose. Diese Daten zeigen zum einen, dass Mcl-1 zur Apoptoseresistenz von HCC-Zellen beitragen kann. Zielgerichtete Therapien, welche die Expression von Mcl-1 hemmen, könnten folglich für die Therapie des HCCs von Interesse sein. Des Weiteren konnte in dieser Arbeit zum ersten Mal gezeigt werden, dass Mcl-1 ein zentraler anti-apoptotischer Faktor für Hepatozyten in vivo ist.

Scutellaria baicalensis (SB) and SB-derived polyphenols possess anti-proliferative activities in several cancers, including pancreatic cancer (PaCa). However, the precise molecular mechanisms have not been fully defined. SB extract and SB-derived polyphenols (wogonin, baicalin, and baicalein) were used to determine their anti-proliferative mechanisms. Baicalein significantly inhibited the proliferation of PaCa cell lines in a dose-dependent manner, whereas wogonin and baicalin exhibited a much less robust effect. Treatment with baicalein induced apoptosis with release of cytochrome c from mitochondria, and activation of caspase-3 and -7 and PARP. The general caspase inhibitor zVAD-fmk reversed baicalein-induced apoptosis, indicating a caspase-dependent mechanism. Baicalein decreased expression of Mcl-1, an anti-apoptotic member of the Bcl-2 protein family, presumably through a transcriptional mechanism. Genetic knockdown of Mcl-1 resulted in marked induction of apoptosis. The effect of baicalein on apoptosis was significantly attenuated by Mcl-1 over-expression, suggesting a critical role of Mcl-1 in this process. Our results provide evidence that baicalein induces apoptosis in pancreatic cancer cells through down-regulation of the anti-apoptotic Mcl-1 protein.

Lerneinheiten müssen stark variierenden Anforderungen gerecht werden. Neben unterschiedlichen Lerntypen spielen vor allem auch die Umfeldbedingungen eine wesentliche Rolle, in denen Lernprozesse stattfinden. Faktoren wie z. B. die Tagesform führen letztlich dazu, dass nicht einmal für eine einzelne Person konstante Lernpräferenzen herrschen. Mit diesem Beitrag wird vorgeschlagen, zur Lösung des Problems einer Mehrkanalstrategie zu folgen. Allerdings sind spezifische Eigenschaften von Learning-Content-Systemen (LCS) notwendig, um ein sog. Multi-Channel-Learning (MCL) zu ermöglichen. Diese Eigenschaften werden im Beitrag anhand von Informationsmodellen beschrieben werden. Sie sollen als Referenzmodell dienen, das sowohl bei der Entwicklung als auch bei der Auswahl und Anpassung von LCS hilfreich sein kann. Das Referenzmodell wird deduktiv abgeleitet und anhand praktischer Anwendungen geprüft. Vorgestellt werden sowohl Anwendungs- als auch Organisationssysteme, die nach dem Modell realisiert worden sind. Auf dieser Grundlage kann schließlich eine Nutzenabschätzung des Modells für das Multi-Channel-Learnings vorgenommen werden.

Incubating rat aortic smooth muscle cells with either platelet-derived growth factor BB (PDGF) or insulin-like growth factor I (IGF-I) increased the phosphorylation of PHAS-I, an inhibitor of the mRNA cap binding protein, eukaryotic initiation factor (eIF) 4E. Phosphorylation of PHAS-I promoted dissociation of the PHAS-I-eIF-4E complex, an effect that could partly explain the stimulation of protein synthesis by the two growth factors. Increasing cAMP with forskolin decreased PHAS-I phosphorylation and markedly increased the amount of eIF-4E bound to PHAS-I, effects consistent with an action of cAMP to inhibit protein synthesis. Both PDGF and IGF-I activated p70S6K, but only PDGF increased mitogen-activated protein kinase activity. Forskolin decreased by 50% the effect of PDGF on increasing p70S6K, and forskolin abolished the effect of IGF-I on the kinase. The effects of PDGF and IGF-I on increasing PHAS-I phosphorylation, on dissociating the PHAS-I-eIF-4E complex, and on increasing p70S6K were abolished by rapamycin. The results indicate that IGF-I and PDGF increase PHAS-I phosphorylation in smooth muscle cells by the same rapamycin-sensitive pathway that leads to activation of p70S6K.

This study proposes to investigate quercetin antitumor efficacy in vitro and in vivo, using the P39 cell line as a model. The experimental design comprised leukemic cells or xenografts of P39 cells, treated in vitro or in vivo, respectively, with quercetin; apoptosis, cell-cycle and autophagy activation were then evaluated. Quercetin caused pronounced apoptosis in P39 leukemia cells, followed by Bcl-2, Bcl-xL, Mcl-1 downregulation, Bax upregulation, and mitochondrial translocation, triggering cytochrome c release and caspases activation. Quercetin also induced the expression of FasL protein. Furthermore, our results demonstrated an antioxidant activity of quercetin. Quercetin treatment resulted in an increased cell arrest in G1 phase of the cell cycle, with pronounced decrease in CDK2, CDK6, cyclin D, cyclin E, and cyclin A proteins, decreased Rb phosphorylation and increased p21 and p27 expression. Quercetin induced autophagosome formation in the P39 cell line. Autophagy inhibition induced by quercetin with chloroquine triggered apoptosis but did not alter quercetin modulation in the G1 phase. P39 cell treatment with a combination of quercetin and selective inhibitors of ERK1/2 and/or JNK (PD184352 or SP600125, respectively), significantly decreased cells in G1 phase, this treatment, however, did not change the apoptotic cell number. Furthermore, in vivo administration of quercetin significantly reduced tumor volume in P39 xenografts and confirmed in vitro results regarding apoptosis, autophagy, and cell-cycle arrest. The antitumor activity of quercetin both in vitro and in vivo revealed in this study, point to quercetin as an attractive antitumor agent for hematologic malignancies.

The characteristics of municipal solid waste (MSW) play a key role in many aspects of waste disposal facilities and landfills. Because most of a landfill is made up of MSW, the overall stability of the landfill slopes are governed by the strength parameters and physical properties of the MSW. These parameters are also important in interactions involving the waste body and the landfill structures: cover liner, leachate and gas collection systems. On the other hand, the composition of the waste, which affects the geotechnical behavior of the MSW, is dependent on a variety of factors such as climate, disposal technology, the culture and habits of the local community. It is therefore essential that the design and stability evaluations of landfills in each region be performed based on the local conditions and the geotechnical characteristic of the MSW. The Bandeirantes Landfill, BL, in Sao Paulo and the Metropolitan Center Landfill, MCL, in Salvador, are among the biggest landfills in Brazil. These two disposal facilities have been used for the development of research involving waste mechanics in recent years. Considerable work has been made in the laboratory and in the field to evaluate parameters such as water and organic contents, composition, permeability, and shear strength. This paper shows and analyzes the results of tests performed on these two landfills. The authors believe that these results could be a good reference for certain aspects and geotechnical properties of MSW materials in countries with similar conditions. (C) 2010 Elsevier Ltd. The aim of this study was to determine the apoptotic pathways and mechanisms involved in electronegative LDL [LDL(-)]-induced apoptosis in RAW 264.7 macrophages and the role of Nrf2 in this process. Incubation of RAW 264 7 macrophages with LDL(-) for 24 11 resulted in dose-dependent cell death. Activated caspases were shown to be involved in the apoptosis induced by LDL(-): incubation with the broad caspase inhibitor z-VAD prevented apoptosis in LDL(-)-treated cells CD95 (Fas), CD95 ligand (FasL). CD36 and the tumor necrosis factor (TNF) ligand Tnfsf10 were overexpressed in LDL(-)-treated cells However, Bax, Bcl-2 and Mcl-1 protein levels remained unchanged after LDL(-) treatment. LDL(-) promoted hyperpolarization of the mitochondrial membrane, elevated reactive oxygen species (ROS) production and translocation of Nrf2 to the nucleus, a process absent in cells treated with native LDL Elicited peritoneal macrophages from Nrf2-deficient mice exhibited an elevated apoptotic response after challenge with LDL(-), together with an increase in the production of ROS in the absence of alterations in CD36 expression These results provide evidence that CD36 expression induced by LDL(-) is Nrf2-dependent. Also, it was demonstrated that Nrf2 acts as a compensatory mechanism of LDL(-)-induced apoptosis in macrophages. (C) 2009 Elsevier B V. All rights reserved

Two in sacco experiments were conducted to evaluate the impact on the nutritive value of rhodes grass hay (Chloris gayana cv. Callide) of treatment with alkalis or oxidants. In Experiment 1, three alkalis (Ca(OH)(2), NaOH, CaO) and two oxidants (NaOCl and H2O2) were applied at levels of 0, 20, 40, 60 or 80 g/kg of dry matter (DM). NaOH, Ca(OH)(2) and CaO had negative linear effects (P < 0.05) on the neutral detergent fibre (NDF) content and positive linear effects (P < 0.05) on the 48 h in sacco disappearances of DM, organic matter (OM), NDF and acid detergent fibre (ADF). NaOCl reduced (P < 0.05) NDF content but had no effect (P > 0.05) on the in sacco disappearances. H2O2 had no effect (P > 0.05) on the composition or digestibility of rhodes grass hay. In Experiment 2, effects of urea (0, 20, 40, 60 and 80 g urea/kg DM) and water (250, 500 and 750 g/kg DM) treatment of rhodes grass hay were examined in a 5 x 3 factorial experiment. Significant interactions between water and urea (P < 0.05) occurred for concentrations of crude protein (CP) and NDF, and 48 h in sacco disappearances of DM, OM (OMD) and NDE The combinations of water (g/kg DM) and urea (g/kg DM) that resulted in the highest concentrations of CP (281 g/kg DM) and OMD (747 g/kg DM) were 250 + 80 and 500 + 80, respectively. NaOH, Ca(OH)(2), CaO and urea significantly alter the NDF content and digestibility of rhodes grass hay, and urea also increases its CP content. Overall, NaOH was the most efficacious, followed by Ca(OH)(2), CaO, urea, NaOCl and H2O2. Crown Copyright (C) 2002 Published by Elsevier Science B.V. All rights reserved.

Mantle cell lymphoma (MCL) commonly involves extranodal sites, usually as a manifestation of disseminated disease. In rare cases, MCLs may arise as a primary tumor in the skin. Blastoid mantle cell lymphoma (BV-MCL) is a rare variant and has a more aggressive clinical course. The phenotype of BV-MCL is characterized as CD20(+), CD5(+), cyclin D1(+), CD23(-), and CD10(-). Interphase fluorescence in situ hybridization shows a characteristic t(11; 14) fusion pattern. We report a case of a BV-MCL arising in skin as primary cutaneous MCL with the characteristic immunophenotype and translocation.
