Ce mémoire présente une poursuite de l’étude vers la synthèse de l’hodgsonox, un sesquiterpénoïde naturel possédant des propriétés insecticides contre la mouche verte d’Australie, Lucilia cuprina. L’hodgsonox comporte six centres stéréogènes et trois cycles : un époxyde fusionné à un cycle à cinq chaînons et une fonction éther cyclique à six chaînons doublement allylique. La stratégie de synthèse de l’hodgsonox proposée comporte dix-neuf étapes linéaires. Elle s’appuie sur les travaux préliminaires de Lise Bréthous, étudiante au doctorat, de Nicolas Lévaray, étudiant à la maîtrise, ainsi que du Dr. Ying Dong Lu et de la Dr. Sonia Diab, qui ont tous travaillé précédemment dans le groupe de la Pr. Lebel. La première étape de cette synthèse consiste en une hydrogénation cinétique dynamique de Noyori permettant d’obtenir un seul diastéréoisomère à partir de l’α-acétylbutyrolactone. Une séquence de six étapes linéaires supplémentaires, comprenant l’ouverture de la lactone ainsi qu’une métathèse d’oléfine, permet d’obtenir le cycle à cinq chaînons avec un rendement global de 37%. L’unité isopropyle est par la suite installée par une addition conjuguée pour former un éther d’énol silylé, qui est directement oxydé en la cétone correspondante avec l’acétate de palladium(II). Une réaction d’hydrosilylation subséquente permet d’obtenir la stéréochimie syn attendue de l’unité isopropyle. Par la suite, la carbonylation d’un intermédiaire triflate permet d’obtenir le squelette de base pour la formation de l’éther cyclique. Enfin, le cycle à six chaînons est formé par insertion O−H intramoléculaire d’un diazo avec un rendement global de 2% sur 17 étapes. Les travaux spécifiques de l’auteure comprennent l’évaluation de conditions catalytiques pour l’oxydation de Saegusa de l’éther d’énol silylé. Les trois dernières étapes ont également été explorées par l’auteure. Il s’agit de l’époxydation de la double liaison endocyclique, de l’insertion dans un lien O−H catalysée par un dimère de rhodium, et de la méthylénation. Enfin, l’exploration d’une voie alternative a été entamée. Cette nouvelle voie consiste à former l’éther cyclique par une substitution nucléophile sur un époxyde. La double liaison exo-cyclique serait installée par une simple réaction de déshydratation.

Many insects feed on blood or tissue from mammalian hosts. One potential strategy for the control of these insects is to vaccinate the host with antigens derived from the insect. The larvae of the fly Lucilia cuprina feed on ovine tissue and tissue fluids causing a cutaneous myiasis associated with considerable host morbidity and mortality. A candidate vaccine antigen, peritrophin 95, was purified from the peritrophic membrane, which lines the gut of these larvae. Serum from sheep vaccinated with peritrophin 95 inhibited growth of first-instar L. cuprina larvae that fed on this serum. Growth inhibition was probably caused by antibody-mediated blockage of the normally semipermeable peritrophic membrane and the subsequent development of an impervious layer of undefined composition on the gut lumen side of the peritrophic membrane that restricted access of nutrients to the larvae. The amino acid sequence of peritrophin 95 was determined by cloning the DNA complementary to its mRNA. The deduced amino acid sequence codes for a secreted protein containing a distinct Cys-rich domain of 317 amino acids followed by a mucin-like domain of 139 amino acids. The Cys-rich domain may be involved in binding chitin. This report describes a novel immunological strategy for the potential control of L. cuprina larvae that may have general application to the control of other insect pests.

Resistance to organophosphorus (OP) insecticides is associated with decreased carboxylesterase activity in several insect species. It has been proposed that the resistance may be the result of a mutation in a carboxylesterase that simultaneously reduces its carboxylesterase activity and confers an OP hydrolase activity (the “mutant ali-esterase hypothesis”). In the sheep blowfly, Lucilia cuprina, the association is due to a change in a specific esterase isozyme, E3, which, in resistant flies, has a null phenotype on gels stained using standard carboxylesterase substrates. Here we show that an OP-resistant allele of the gene that encodes E3 differs at five amino acid replacement sites from a previously described OP-susceptible allele. Knowledge of the structure of a related enzyme (acetylcholinesterase) suggests that one of these substitutions (Gly137 → Asp) lies within the active site of the enzyme. The occurrence of this substitution is completely correlated with resistance across 15 isogenic strains. In vitro expression of two natural and two synthetic chimeric alleles shows that the Asp137 substitution alone is responsible for both the loss of E3’s carboxylesterase activity and the acquisition of a novel OP hydrolase activity. Modeling of Asp137 in the homologous position in acetylcholinesterase suggests that Asp137 may act as a base to orientate a water molecule in the appropriate position for hydrolysis of the phosphorylated enzyme intermediate.

The purpose of this work was to determine the diversity and population fluctuations of calliphorid flies in the Biological Reserve of Tinguá (ReBio-Tinguá), Nova Iguaçu, state of Rio de Janeiro, Brazil and to correlate their occurrence with the environmental variables of temperature, rainfall and relative air humidity. Specimens of Diptera were collected monthly between June 2002 and January 2005 using four traps placed at four points along a trail and exposed for 48 hours. The traps were baited with sardines and the trapped insects were stored in 70% alcohol. It was collected 8,528 calliphorids, thirteen species were identified among the blowflies including Laneela nigripes Guimarães 1977, Chrysomya megacephala (Fabricius, 1794), C. albiceps (Wiedemann, 1819), C. putoria (Wiedemann, 1830), Chloroprocta idioidea (Robineau-Devoidy, 1830), Cochliomyia macellaria (Fabricius, 1775), Hemilucilia semidiaphana (Rondani, 1850), H. segmentaria (Fabricius, 1805), Lucilia eximia (Wiedemann,1819), L. cuprina (Wiedemann, 1830), Paralucilia pseudolyrcea (Mello, 1969), Mesembrinella sp. and Eumesembrinella pauciseta (Aldrich, 1922). No significant correlation was found between the abundance of blowflies and the temperature and relative air humidity. Only C. megacephala and C. albiceps showed a positive and significant correlation with rainfall. An analysis of grouping by month (UPGMA) revealed no seasonal difference in the composition of the community, indicating that the community of calliphorid flies is probably more influenced by the ecological niches occupied by each species than by the seasons of the year.

Os dípteros califorídeos são os primeiros indivíduos a compor a sucessão faunística de cadáveres em decomposição, auxiliando assim na estimativa do intervalo pós-morte. Estudos de dispersão pós-alimentar de suas larvas possuem relevância para investigações médico-criminais. Diante disto, uma arena circular, simulando o ambiente natural, foi montada em laboratório a fim de verificar-se a dispersão larval radial pós-alimentar de Lucilia sericata (Meigen, 1826), tentando assim estabelecer relações entre as variáveis peso, distância e profundidade de enterramento na arena. Os resultados demonstraram que 45% das pupas foram recuperadas a uma profundidade compreendida entre 6 cm e 8 cm e a uma distância entre 33 cm e 45 cm do centro da arena. Não houve diferenças significativas quanto à propagação e profundidade de enterramento de machos, fêmeas e indivíduos inviáveis. Com relação ao peso, verificou-se que a média das fêmeas (x = 32,35 mg) foi superior a dos machos (x = 30,28 mg). A análise de correlação e de regressão entre peso e distância percorrida e entre peso e profundidade foram positivas, ou seja, pupas oriundas de larvas mais pesadas propagaram e se aprofundaram mais. O experimento permitiu ainda concluir que uma arena circular possibilita o deslocamento das larvas em todas as direções.

Entre Maio e Junho de 2008 foi observado a sobrevivência das pupas de Lucilia eximia (Diptera, Calliphoridae) após submersão em laboratório. Para este experimento foram utilizadas 480 pupas de mesma idade, sendo estas divididas em oito grupos: o controle não foi submerso e os demais grupos foram submersos de um a sete dias. O aumento do período de submersão diminui a sobrevivência, com um dia de submersão a sobrevivência é de 80%, com dois dias 40%, é de 30% a partir do terceiro dia, no quarto dia cai para 23,34% e no quinto dia fica em 10%. Após este período a mortalidade sobe para 100%. Este padrão pode ser explicado pela curva "U-shaped" que ocorre no consumo de O2 durante o período pupal, onde o consumo é maior no início e no final do período pupal. O tempo de submersão também afeta o tempo de desenvolvimento, aumentando o período pupal. Estes dados têm o potencial para serem utilizados em investigações envolvendo Entomologia Forense, para a estimativa do tempo de submersão de um cadáver.

Under natural environmental conditions, blowflies utilize discrete and ephemeral feeding resources such as decaying carcasses. Competition for food on such feeding substrates is usually very severe, and only the individuals that are capable of attaining the critical larval weight for pupation will be able to survive. This critical weight is hitherto unknown for several blowfly species; therefore, the current work is aimed at obtaining such a critical value for four blowfly species of the genera Chrysomya and Lucilia, deploying two types of feeding substrate, namely, artificial diet and macerated bovine meat. On the whole, the critical weights ranged from 30 to 35 mg. The lowest larval weight which permitted pupation was 30.0 mg for Chrysomya megacephala reared on macerated bovine meat. This species was also the best adapted to pupation at low larval weights in relation to the maximum larval weight for males. Regarding the pupation of females, the best-adapted individual was a C. albiceps specimen exhibiting a critical weight that was equal to 39.20 % of the maximum value obtained. Concerning all the species and diet types, the female individuals exhibited the lowest critical weights that produced viable pupae, probably representing an evolutionary strategy that favoured the survival of females, responsible for the egg formation, contributing to the establishment of future generations. Regarding the loss (in percentage) of adult biomass in relation to the third instar larvae, the females of C. megacephala lost less weight than males in both feeding substrates. On the other hand, such a loss of weight occurred in males of C. albiceps and L. cuprina.

The aim of this study was to evaluate the reliability of insect larvae as samples for toxicological investigations. For this purpose, larvae of Lucilia sericata were reared on samples of minced pig liver treated with different concentrations of codeine: therapeutic, toxic, and potentially lethal doses. Codeine was detected in all tested larvae, confirming the reliability of these specimens for qualitative toxicology analysis. Furthermore, concentrations measured in larvae were correlated with levels in liver tissue. These observations bring new elements regarding the potential use of opiates concentrations in larvae for estimation of drug levels in human tissues. Morphine and norcodeine, two codeine metabolites, have been also detected at different concentrations depending on the concentration of codeine in pig liver and depending on the substance itself. The effects of codeine on the development of L. sericata were also investigated. Results showed that a 29-h interval bias on the evaluation of the larval stage duration calculated from the larvae weight has to be considered if codeine was present in the larvae substrate. Similarly, a 21-h interval bias on the total duration of development, from egg to imago, has to be considered if codeine was present in the larvae substrate.

Introducción: Lucilia sericata es una especie de importancia médica y forense, utilizada en terapialarval para curar heridas crónicas y en estudios médico-legales empleada en la estimacióndel intervalo post mórtem y el traslado de cadáveres. No existen registros de las característicascitogenéticas de esta mosca en el neotrópico. El objetivo principal de este trabajo fue identificarlas características morfométricas cromosómicas y las estructuras primarias del cariotipo, a partirde especímenes de L. sericata de la cepa Bogotá, Colombia. Materiales y métodos: Se tomaronhuevos embrionados, que fueron previamente esterilizados en su superficie, se maceraron y luegofueron sembrados en el medio de cultivo L-15, suplementado con 20% de sfb, e incubados auna temperatura de 28 ºC, sin atmosfera de C02. La preparación de los cromosomas se obtuvo demonocapas celulares semiconfluentes, empleando diversas soluciones: antimitótica (Colchicina),hipotónica (KCl 0,075 M) y fijadora (Carnoy: metanol y ácido acético; 3:1). Se llevó a cabo la técnicade bandeo C para la identificación de regiones cromosómicas de heterocromatina constitutiva.Resultados: Se obtuvieron parámetros morfométricos de cada par cromosómico. El número diploidedel cariotipo obtenido de los cultivos celulares fue 2n = 12; éstos se clasificaron morfológicamente,de acuerdo con patrones previamente establecidos, así: los pares I, II, IV y V fueronmetacéntricos, y el par III fue submetacéntrico. A su vez, el par sexual fue heteromórfico, siendoel cromosoma X metacéntrico y el cromosoma Y submetacéntrico. El bandeo C fue positivo paratodos los pares cromosómicos. Conclusiones: Se establecieron las características citogenéticas deL. sericata, cepa Bogotá, Colombia, relacionadas con número, forma, tamaño, posición del centrómeroy regiones heterocromáticas de los cromosomas.

The main purpose of this study was to obtain primary cell cultures derived from Lucilia sericata (Diptera: Calliphoridae). Necrophagous this fly is used for determination of post-mortem interval and larval therapy. Since explants embryonated eggs were performed in various culture media (Grace Schneider, MM/VP12, DMEM, Grace/L-15 and L-15), supplemented with 20% fetal serum. Sterilization of the biological material was carried out by immersing it in formaldehyde and sodium hypochlorite solutions. The cell growth was initiated in the L-15, MM/VP12, and Schneider Grace/L-15 in an average time of 10 days after completion of planting by the proliferation of groups of colonies scattered on the surface of the boxes crops and also from the endings of larval fragments. The evolution of cell growth to the formation of monolayer semi-confluent was relatively fast, reaching at 3 weeks post-explant. Cellular morphology in cultured cells was heterogeneous, especially epithelioid forms, similar to nerve, giant and irregular. Comparison of the growth characteristics of these cell cultures with those obtained from other species of flies was more favorable in the evolution of those obtained from L. sericata, on the grounds that the cells are better adapted to the physical-chemical conditions of several culture media. This is the first report of a cell culture-fly family Calliphoridae.
