There is substantial international variation in human papillomavirus (HPV) prevalence; this study details the first report from Northern Ireland and additionally provides a systematic review and meta-analysis pooling the prevalence of high-risk (HR-HPV) subtypes among women with normal cytology in the UK and Ireland. Between February and December 2009, routine liquid based cytology (LBC) samples were collected for HPV detection (Roche Cobas® 4800 [PCR]) among unselected women attending for cervical cytology testing. Four electronic databases, including MEDLINE, were then searched from their inception till April 2011. A random effects meta-analysis was used to calculate a pooled HR-HPV prevalence and associated 95% confidence intervals (CI). 5,712 women, mean age 39 years (±SD 11.9 years; range 20-64 years), were included in the analysis, of which 5,068 (88.7%), 417 (7.3%) and 72 (1.3%) had normal, low, and high-grade cytological findings, respectively. Crude HR-HPV prevalence was 13.2% (95% CI, 12.7-13.7) among women with normal cytology and increased with cytological grade. In meta-analysis the pooled HR-HPV prevalence among those with normal cytology was 0.12 (95% CIs, 0.10-0.14; 21 studies) with the highest prevalence in younger women. HPV 16 and HPV 18 specific estimates were 0.03 (95% CI, 0.02-0.05) and 0.01 (95% CI, 0.01-0.02), respectively. The findings of this Northern Ireland study and meta-analysis verify the prevalent nature of HPV infection among younger women. Reporting of the type-specific prevalence of HPV infection is relevant for evaluating the impact of future HPV immunization initiatives, particularly against HR-HPV types other than HPV 16 and 18. J. El cáncer de cuello uterino y su mortalidad en Colombia ha permanecido constantes pese a los esfuerzos institucionales, distritales y nacionales que han buscado fortalecer los programas de prevención; sin embargo las estrategias actuales hacen énfasis en la toma de citología y no en la solicitud oportuna del resultado y el tratamiento de la usuaria en caso de anormalidad. METODOLOGIA: Se realizó un estudio en dos fases, un análisis descriptivo se analizaron 12875 y una segunda que involucro 257 pacientes en un análisis de casos y controles de una muestra aleatoria. RESULTADOS: Se utiliza para análisis bivariado la prueba de Chi cuadrado y regresión logística que muestran diferencias significativas en los siguientes variables: la explicación sobre la importancia del examen (p= 0.0060), importancia de la solicitud de resultado (p= 0.003), explicación sobre cuando reclamarlo (p=0.030), distancia entre residencia y centro de salud (p=0.065) DISCUSIÓN: En nuestro estudio se identificó que los factores como el acceso de la paciente al centro de salud, el tiempo del cual dispone para solicitar el resultado, si el dinero con el que cuenta la paciente para desplazarse al hospital, si es la primera vez que se realiza la citología, si le explicaron la importancia de reclamar el resultado y en que lapso de tiempo debía hacerlo, la información que tiene acerca de la importancia de realizarse la citología contribuyen en el hecho de que una paciente de esta población de un hospital público reclame o no su resultado de su citología

Hormone therapy is an important tool in the treatment of breast cancer and tamoxifen represents one of the most important drugs used in this type of treatment. Recently other drugs based on the inhibition of aromatase had been developed, this enzyme is responsible for the synthesis of estrogenic esteroids from the androgenic ones. The objective of this study would be the development of a quantitative cytological model of murine estral analysis that allowed the characterization of different hormone drugs effect over vaginal epithelium. The technique of monochromatic staining with Evans blue (C.I. 23860) showed to be efficient in the qualitative and quantitative classification of the cycle. It had been observed differences in the cytological standard of animals submitted to the studied drugs; tamoxifen presented a widening of phases of lesser maturation (diestrais), while anastrozole and exemestane increased the duration of the phases of larger maturation (estrais). The data were analysed through a cubical non linear regression (spline) which allowed a better characterization of the drugs, suggesting a proper cytological profile to the antagonism of the estrogen receptor (tamoxifen), aromatase competition (anastrozole) and inhibition of the enzyme (exemestane)

The cancer of the uterine cervix remains a serious public health problem in developing countries with low social-economic conditions. Papanicolaou smear has been used to screen women for cervical cancer since 1940, and it is widely recognized as an effective method for preventing cervical cancer. In Brazil, conventional Papanicolaou is the most common method used in diagnosis cytopathological routine, involving the morphological cell analysis by microscopic examination of cervical samples smeared on glass slides. On the other hand, the liquid cytology is an advantageous method whose specimen collected is put into a fluid alcohol based to guarantee the potential use of residual cellular material in cytobrush. Moreover, this method permits additional diagnostic studies, preserving the morphological cell features as the conventional one does. The aim of the present study was report the historical evolution of different cervical cytologic tests, emphasizing the advantages and disadvantages between conventional Papanicolaou method and new technologies in liquid based cytology.

O HPV (Papilomavírus humano) foi apontado pela OMS (Organização Mundial de Saúde - WHO) como principal fator de risco para o desenvolvimento do câncer de colo uterino, tornando-se assim um importante e gravíssimo problema de saúde pública, especialmente nos países subdesenvolvidos ou em desenvolvimento. A precocidade das atividades sexuais, múltiplos parceiros e sexo casual, o tabagismo, a imunossupressão (por exemplo, na população de pacientes aidéticos), gravidez, doenças sexualmente transmissíveis prévias como herpes e clamídia, além do não cumprimento das medidas já adotadas como prevenção de Doenças Sexualmente Transmissíveis (DST), como por exemplo, o simples uso de preservativos, está reconhecidamente associado à incidência da infecção por HPV. Essa pesquisa teve como objetivo avaliar o desempenho diagnóstico das metodologias de citologia convencional (Exame de Papanicolau) em relação à citologia em base líquida, além de determinar a prevalência dos genótipos 16 e 18 do HPV em mulheres sem efeito citopático compatível com HPV e relacionar a presença de quadros inflamatórios, associados ou não ao HPV, com dados epidemiológicos como idade, escolaridade, condição sociocultural de mulheres provenientes do município de Barcarena – Pará – Brasil. Para tanto, participaram deste estudo, voluntariamente, 50 mulheres atendidas na Unidade de Saúde de Barcarena – Pará, através de campanha para coleta de Exame de Papanicolau como método de prevenção de câncer do colo do útero. Estas mulheres receberam informações referentes a todos os procedimentos realizados pelo corpo de saúde deste estudo e aos resultados desta pesquisa e somente após as voluntárias terem assinado o Termo de Consentimento Livre e Esclarecido, as mesmas foram incluídas para as coletas de amostras. As análises e os resultados dos testes de citologia de base líquida e convencional foram realizados segundo a Classificação de Bethesda e revisados cegamente por dois citopatologistas. Para a análise estatístca foi utilizado o teste exato de Fisher e o "Screening Test" visando determinar a especificidade/ sensibilidade dos métodos, considerando significativo o valor de p ≤ 0,05. Como resultado, observamos que o uso da citologia de base líquida tem demonstrado uma série de vantagens em relação à citologia convencional. No diagnóstico molecular (PCR) foram observadas ocorrências de HPV dos tipos 16 e 18 em 10% das mulheres atendidas. Dentre os casos que apresentaram PCR positivo para os tipos 16 ou 16/18 a maioria das mulheres tinham 27,4 anos de idade em média; com maior escolaridade; que exercem atividades domésticas e rurais; e com ocorrências de co-infecção por agentes infecciosos causadores de outras doenças sexualmente transmissíveis. Os resultados obtidos neste estudo reforçam a importância da manutenção de campanhas gratuitas de prevenção do câncer de colo do útero como uma medida preventiva no combate desta doença, principalmente no Estado do Pará onde, provavelmente, o perfil epidemiológico da doença está associado às grandes distâncias que as mulheres de comunidades ribeirinhas têm que percorrer para realizar este exame de forma gratuita; ao tipo de atividade econômica da região; ao preconceito local ainda existente com o exame; e ao grau de dificuldade de implementação de ações efetivas de retorno das pacientes às consultas médicas após a obtenção do resultado do exame e mesmo o encaminhamento para diagnóstico molecular dos casos positivos para lesões do tipo ASC-H e NIC I, II e III.

We sought to evaluate the performance of diagnostic tools to establish an affordable setting for early detection of cervical cancer in developing countries. We compared the performance of different screening tests and their feasibility in a cohort of over 12,000 women: conventional Pap smear, liquid-based cytology, visual inspection with acetic acid (VIA), visual inspection with Iodine solution (VILI), cervicography, screening colposcopy, and high-risk human papillomavirus (HPV) testing (HR-HPV) collected by physician and by self-sampling. HR-HPV assay collected by the physician has the highest sensitivity (80 %), but high unnecessary referrals to colposcopy (15.1 %). HR-HPV test in self-sampling had a markedly lower (57.1 %) sensitivity. VIA, VILI, and cervicography had a poor sensitivity (47.4, 55, and 28.6 %, respectively). Colposcopy presented with sensitivity of 100 % in detecting CIN2+, but the lowest specificity (66.9 %). Co-testing with VIA and VILI Pap test increased the sensitivity of stand-alone Pap test from 71.6 to 87.1 % and 71.6 to 95 %, respectively, but with high number of unnecessary colposcopies. Co-testing with HR-HPV importantly increased the sensitivity of Pap test (to 86 %), but with high number of unnecessary colposcopies (17.5 %). Molecular tests adjunct to Pap test seems a realistic option to improve the detection of high-grade lesions in population-based screening programs.

BACKGROUND Cytology is an excellent method with which to diagnose preinvasive lesions of the uterine cervix, but it suffers from limited specificity for clinically significant lesions. Supplementary methods might predict the natural course of the detected lesions. The objective of the current study was to test whether a multicolor fluorescence in situ hybridization (FISH) assay might help to stratify abnormal results of Papanicolaou tests. METHODS A total of 219 liquid-based cytology specimens of low-grade squamous intraepithelial lesions (LSIL), 49 atypical squamous cells of undetermined significance (ASCUS) specimens, 52 high-grade squamous intraepithelial lesion (HSIL) specimens, and 50 normal samples were assessed by FISH with probes for the human papillomavirus (HPV), MYC, and telomerase RNA component (TERC). Subtyping of HPV by polymerase chain reaction (PCR) was performed in a subset of cases (n=206). RESULTS There was a significant correlation found between HPV detection by FISH and PCR (P<.0001). In patients with LSILs, the presence of HPV detected by FISH was significantly associated with disease progression (P<.0001). An increased MYC and/or TERC gene copy number (>2 signals in>10% of cells) prevailed in 43% of ASCUS specimens and was more frequent in HSIL (85%) than in LSIL (33%) (HSIL vs LSIL: P<.0001). Increased TERC gene copy number was significantly correlated with progression of LSIL (P<.01; odds ratio, 7.44; area under the receiver operating characteristic curve, 0.73; positive predictive value, 0.30; negative predictive value, 0.94) CONCLUSIONS: The detection of HPV by FISH analysis is feasible in liquid-based cytology and is significantly correlated with HPV analysis by PCR. The analysis of TERC gene copy number may be useful for risk stratification in patients with LSIL.

O linfoma canino é uma das neoplasias mais prevalentes em cães, diagnosticado frequentemente através de exame citológico de aspirados. A imunofenotipagem para diferenciação entre linfomas de células B e T é importante para definição prognóstica e tratamento, entretanto, dificilmente é realizada em materiais provenientes de aspirados citológicos. A citologia em meio líquido (CML) é uma metodologia automatizada de produção de esfregaços citológicos, que permite a conservação de todo o material da aspiração, e utilização do material residual para, por exemplo, produção de emblocado celular (EC) e imunocitoquímica. Este trabalho teve como objetivo aplicar a CML e EC com imunocitoquímica em amostras de linfonodos caninos suspeitos para linfoma. Além disso, pretendeu caracterizar morfologicamente as células linfoides neste tipo de preparado, comparando ao esfregaço convencional. Para isto, foram selecionados 54 cães com linfadenomegalia, 10 deles sorologicamente positivos para leishmaniose canina. Comparou-se a CML com a citologia convencional, quanto às características técnicas e testou-se dois métodos diferentes de EC: Bouin e agarose com formalina. Aplicou-se painel imunocitoquímico de anticorpos anti-CD3, anti-CD79a, anti-Pax-5 e anti-Ki67, para imunofenotipagem e determinação da proliferação celular. Adicionalmente, realizaram-se análises estatísticas para avaliação do desempenho e concordância interobservador comparada entre citologia convencional, CML e o uso conjunto de CML e imunocitoquímica do emblocado celular. Como resultado, as células linfoides apresentaram-se preservadas, com melhor definição de cromatina e nucléolos, porém com tamanho reduzido e pior definição citoplasmática, na CML. O EC de Bouin revelou grupos densos e bem definidos, que permitiram a aplicação da imunocitoquímica. Por outro lado, os emblocados de agarose, apresentaram-se frouxos, com células marcadamente dispersas, revelando-se inadequados para preparados de células linfoides. O anticorpo anti-Pax-5 mostrou-se mais adequado do que o anti-CD79a para marcação de células B em emblocados, por produzir menos fundo e pela marcação nuclear ser mais distinguível do que a citoplasmática. A concordância interobservadores da CML foi moderada (k= 0,434), enquanto a da citologia convencional foi boa (k=0,762). Ambas as técnicas exibiram a mesma acurácia e, com aplicação da CML, houve redução do percentual de amostras insatisfatórias. A CML em conjunto com EC e imunocitoquímica apresentou maior sensibilidade (75%), especificidade (99%) e acurácia (89,47%). Como conclusão, a CML em conjunto com EC e imunocitoquímica pode ser aplicada para diagnóstico de linfoma canino, com maior sensibilidade, especificidade e acurácia

The nucleophilic displacement reaction of n-bromooctane and potassium iodide in ionic liquid based on cyclic guanidinium cation(2) was investigated. The kinetic reasult shows that the rate of the reaction is enhanced in ionic liquid (2). The same reaction in [bmim][PF6](1)(where bmim = 1-butyl-3-methylimidazolium) was also studied. It was found that as a reaction medium ionic liquid (2) is better than (1) for nucelophilic displacement reactions.

A thermodynamic analysis of the experimental conditions of the Beckmann rearrangement reaction of oximes into amides has been undertaken to examine whether the reaction is under thermodynamic or kinetic control. To answer this question, the thermodynamic properties of the typical Beckmann rearrangement reactions in the ideal gaseous state-cyclohexanone oxime to caprolactam and 2-butanone oxime to N-methylpropanarnide-were studied by using the quantum mechanical method. Gibbs energy and equilibrium constants of the Beckmann rearrangement have been assessed in the gaseous and the liquid phases. Results of the thermodynamic analysis have shown that Beckmann rearrange ments are kinetically controlled. Thus, a search for possible active ionic liquid based catalysts for the mild reaction conditions has been performed.

Densities and viscosities of the ionic liquid 1-butyl-3-methylimidazolium octylsulfate, [C4C1Im][C8SO4] were measured as a function of temperature between 313 K and 395 K. Solubilities of hydrogen and carbon dioxide were determined, between 283 K and 343 K, and at pressures close to atmospheric in [C4C1Im][C 8SO4] and in another ionic liquid based on the alkylsulfate anion-1-ethyl-3-methylimidazolium ethylsulfate, [C 2C1Im][C2SO4]. Density and viscosity were measured using a vibrating tube densimeter from Anton Paar and a rheometer from Rheometrics Scientific with accuracies of 10-3 g cm -3 and 1%, respectively. Solubilities were obtained using an isochoric saturation technique and, from the variation of solubility with temperature, the partial molar thermodynamic functions of solvation, such as the standard Gibbs energy, the enthalpy, and the entropy, are calculated. The precision of the experimental data, considered as the average absolute deviation of the Henry's law constants from appropriate smoothing equations, is better than ±1%. © The Royal Society of Chemistry.

Highly stable and crystalline V(2)O(5) nanoparticles with an average diameter of 15 nm have been easily prepared by thermal treatment of a bariandite-like vanadium oxide, V(10)O(24)center dot 9H(2)O. Their characterization was carried out by powder X-ray diffractometry (XRD). Fourier transform infrared (FT-IR) and Raman spectroscopies, and transmission electron microscopy (TEM). The fibrous and nanostructured film obtained by electrophoretic deposition of the V(2)O(5) nanoparticles showed good electroactivity when submitted to cyclic voltammetry in an ionic liquid-based electrolyte. The use of this film for the preparation of a nanostructured electrode led to an improvement of about 50% in discharge capacity values when compared with similar electrodes obtained by casting of a V(2)O(5) xerogel. This paper is concerned with the possibility of a direct second-order transition out of a collinear Neel phase to a paramagnetic spin liquid in two-dimensional quantum antiferromagnets. Contrary to conventional wisdom, we show that such second-order quantum transitions can potentially occur to certain spin liquid states popular in theories of the cuprates. We provide a theory of this transition and study its universal properties in an epsilon expansion. The existence of such a transition has a number of interesting implications for spin-liquid-based approaches to the underdoped cuprates. In particular it considerably clarifies existing ideas for incorporating antiferromagnetic long range order into such a spin-liquid-based approach.

The underwater casting of relatively thin lifts of concrete in water requires the proportioning of highly flowable concrete that can resist water dilution and segregation and spread readily into place. An investigation was carried out to determine the effects of antiwashout admixture concentration, water-cementitious materials ratio, and binder composition on the washout resistance of highly flowable concrete. Two main types of antiwashout admixtures were used: 1) a powdered welan gum at concentrations of 0.07 and 0.15% (by mass of binder); and 2) a liquid-based cellulosic admixture with dosages up to 1.65 L/100 kg of binder. The water-cementitious materials ratios were set at 0.41 and 0.47, corresponding to high-quality underwater concrete. Four binder compositions were used: a standard Canadian Type 10 cement, the same cement with 10% silica fume replacement, the cement with 50% granulated blast-furnace slag replacement, and a ternary cement containing 6% silica fume and 20% Class F fly ash. The concentrations of anti-washout admixture have direct impact on washout resistance. For a given washout loss, greater slump flow consistency can be achieved with the increases in anti-washout admixture concentration and decreases in water-binder ratio. The washout mass loss can be reduced, for a given consistency
