Com o objetivo de fornecer dados para o esclarecimento do controvertido problema da natureza de Cytamoeba bacterifera freqüentemente encontrada nos eritrócitos de Leptodactylus ocellatus, realizamos alguns testes citoquímicos. Demonstramos a presença do ácido ribonucleico e polissacarídeos não digeríveis pela ptialina em sua estrutura. Com o método de Feulgen, teste de referência para a caracterização do ácido desoxirribonucleico, obtivemos principlemnte resultados negativos; porém, um parasito com fraca e difusa positividade e algumas reações duvidosas também foram encontrados. Ao emrpegarmos o Verde Metila-Pironina, mesmo após o tratamento pela ribonuclease, e o Azul de Toluidina, também depois da ação desta enzima, não conseguimos confimar a presença de ADN. Como os elementos constituintes de C. bacterifera são minúsculos e, às vezes, não evidenciáveis, é possível que seu teor de ADN, porventura existente, seja muito pequeno e, conseqüentemente, de difícil demonstração por métodos cujos resultados são observados sob microscopia ótica, além de poder ficar facilmente encoberto por outras substâncias. Não estamos propensos a admitir uma provável natureza virótica para Cytamoeba baseados, principalmente, em alguns de seus aspectos estruturais (figs. 8, 15, 17 e 18) e na ausência de alteração no núcleo das células parasitadas. Apesar de não termos comprovado a presença de ADN, achamos possível que C. bacterifera seja um aglomerado intracitoplasmático de organismos modificados, cujas dimensões situam-se nas proximidades do limite de resolução do microscópio ótico, relacionados com as bactérias, assim como são, por exemplo, os Clamídios e as Riquétsias. Observamos o desenvolvimento de Cytamoeba em rã mantida em cativeiro por três meses e semanalmente examinada; constatamos decréscimo paulatino da parasitemia inicial e também que os seus tipos estruturais e medidas não estavam relacionadas com a etapa da infecção. Não conseguimos transmitir, por inoculação intra-peritoneal, Cytamoeba de L. ocellatus para Bufo crucifer.

Specimens of Leptodactylus mystacinus from Brazil were karyotyped with conventional and differential staining. The 2n = 22 karyotype is similar to that found for the majority of the Leptodactylus, the karyotypic conservatism also confirmed by the similarity of the replication banding patterns with those previously described. L. mystacinus has a small amount of C-banded heterochromatin, located mainly at the centromeres, although telomeric or interstitial bands have also been noticed. With DA/CMA(3) some chromosome regions showed slightly bright fluorescence, and with DA/DAPI, no particular AT-rich repetitive region was observed. Silver staining showed an extensive inter- and intraindividual variation in the number and position of Ag-positive regions, in 1p, 4p, 8p, 8q, and 11p. Nevertheless, FISH using rDNA probes confirmed only the signals on the short arms of chromosomes 4 and 8 as true NORs. The remaining silver stained regions are probably due to the heterochromatin with some affinity to the Ag-staining. Phylogenetic analysis based on partial cytochrome b sequence revealed that L. mystacinus forms a basal branch, so that the presence of multiple NORs in pairs 4 and 8 in this species indicates an autapomorphy.

The advertisement calls of Leptodactylus geminus, Leptodactylus gracilis, Leptodactylus plaumanni, and Leptodactylus fuscus are analyzed and compared with published reports. Based on calling data and examination of type material, Leptodactylus geminus is synonymized with L, plaumanni. To differentiate between the sibling species L. gracilis and L. plaumanni, fieldwork is needed, including recording of advertisement calls. Reliable identification of red specimens is not possible based on morphology and coloration alone.

We report herein a case of predation by the ctenid spider Ctenus medius on a leptodactylid frog Leptodactylus marmoratus, observed in a remnant of the Atlantic Forest, municipality of Sao Vicente, São Paulo state, southeast Brazil. This is the first record of predation by C. medius upon L. marmoratus. Nevertheless, due to the high abundance of both groups, we suggest that the interaction between spiders and amphibians could be very common on the floor of the Atlantic Forest.

Capture/recapture studies significantly increase our knowledge of the natural history of anuran amphibians. Many different methods have been employed in these studies, but a number of new techniques still require experimental validation. During two reproductive seasons in a Cerrado remnant in southeastern Brazil, we investigated the movement patterns and habitat use of the pepper frog, Leptodactylus labyrinthicus, using a spool-and-line device. This low-cost device did not appear to interfere with the activities of the frogs and allowed for constant monitoring, showing precise routes of movement and great predictability of relocations. Both sexes were active at night. During the day, males and females made use of retreat sites under vegetation or in burrows constructed by small- and medium-sized mammals. Males and females did not use standardized routes; there were no significant differences between their movements, and movements were not correlated with body size or environmental conditions. Individuals are able to move further than 100 m per day, a characteristic that may enable this species to colonize or recolonize open areas.

Cytogenetic studies were carried out on five species of Leptodactylus, namely L. fuscus, L, notoaktites, L. labyrinthicus, L. ocellatus, and L. podicipinus, after standard staining, Ag-NOR and C-banding as well as BrdU incorporation for three of them. The species had 2n = 22 chromosomes and two basic karyotype patterns. Chromosome 8 was a marker bearing a secondary constriction. In all species, this secondary constriction corresponded to the Ag-NOR site. The species had centromeric C-bands in all chromosomes of the complement, but some interstitial or telomeric bands seemed to differentiate some karyotypes, either at the species or the population level. In L. ocellatus, the C-banding pattern confirmed the occurrence of a heteromorphic pericentric inversion in chromosome 8 in specimens from one of the populations. The BrdU incorporation technique showed no detectable difference in the replication patterns of the major bands in the chromosomes of L. noroaktites, L. labyrinthicus, and L. ocellatus.

Forty-three specimens of Leptodactylus podicipinus (Anura: Leptodactylidae) were collected in the south-eastern Pantanal, municipality of Corumba, State of Mato Grosso do Sul, Brazil in February and July 2007, and examined for endoparasites. Forty (93%) specimens were infected with at least one helminth species. The predominant parasites were nematodes (Aplectana sp., Cosmocerca podicipinus, Oswaldocruzia lopesi, Physalopteroides venancioi, Rhabdias sp.), but the trematode Catadiscus propinquus also showed high prevalence. The trematodes Infidum infidum and Travtrema stenocotyle were also found, but in only one specimen. Adult frogs showed higher parasite diversity than subadults. Leptodactylus podicipinus was preferentially infected by direct life-cycle parasites and was reported as a new host record for seven helminth species.
