On July 15, 2014 the European Parliament confirmed the new European Commission President. An absolute majority was needed for this purpose, and the 422 votes “For” cleared the 376-vote threshold in the legislative body of 751 members. A Grand Coalition has been formed among the three largest political parties: the European People’s Party (EPP), the Progressive Alliances of Socialists Democrats (S&D), and the Alliance of Liberals and Democrats for Europe (ALDE). Considering policy decisions going forward, the European Union (EU) faces the pressing question: Will there be more, less, or similar power from the EU? There are a greater number voices from across the political spectrum contributing to the democratic plurality. European leaders may regain trust by acknowledging that future governance will not be “business as usual” as the reform agenda gets underway. 2014 has been an exciting and important year in European politics. “This time is different” was the motto for the European Parliament’s election campaign. This essay analyzes recent EU political trends with the new Commission leadership and the Parliamentary elections results. The Parliamentary elections, held in late May, and the new European Commission, planned to be in place in the autumn, influence the leadership direction of the 28-member bloc. Additionally, this year on July 1 Croatia celebrated the first anniversary of joining the EU in 2013. Leading the way for candidate countries, Croatia embraces the democratic politics and capitalist market economics embodied by the EU. The greater number of seats held by newer political parties in the European Parliament demonstrates increasing plurality in the EU democracy. The Parliamentary elections have taken place every 5 years since 1979. In this eighth legislative session, the EPP and the S&D remain the largest parties represented, with 221 and 191 seats respectively. As the EU has evolved, a greater number of voices influence politics. The ongoing point of contention on a host of policies is national sovereignty in relation to pooled sovereignty in the EU. The European Parliament is important for democracy in EU governance since it is the direct link from the national citizens to their elected leaders at the supranational level. The representatives of the European Commission are appointed by the national governments of Member States, and their heads of government are the representatives to the European Council. These three political institutions – the European Parliament, the European Commission, and the European Council – together with other important institutions, including the European Court of Justice Luxembourg, form the EU. The new European Commission President is Jean-Claude Juncker, former Prime Minister and Minister of Finance of Luxembourg (1995-2013). After being nominated by the European Council on June 27, his candidacy was voted on by the European Parliament on July 15, according to the guidelines of the Lisbon Treaty. The leadership for the President of the European Commission has been an important issue, considering Britain’s deliberations on whether or not to stay in the EU in the face of a future national referendum. Voting on June 27, among the European Council on the nomination of Commission President-Designate Juncker, was 26 in favor and 2 opposed. Only Viktor Orbán, the prime minister of Hungary, joined David Cameron, the prime minister of the United Kingdom (UK), with a negative vote (Spiegel and Parker 2014). The UK had not been supportive, being concerned that Juncker embraces the policies of a federalist, prioritizing an ever-closer union above the interests of individual Member States. Historically, since joining the predecessor institution of the European Economic Community in 1973, the UK has had a relatively independent attitude about participation in the EU.

On June 15, 2014, Jean-Claude Juncker, the lead candidate of the European People’s Party, was elected President of the European Commission, with the support of the Alliance of Liberals and Democrats for Europe, and some of the European Socialists and Greens. Amid unprecedented Euroscepticism, the media and many pundits predicted a record-low voter turnout and record-high results for Europhobic parties. The aforementioned parties then decided that the political outcome of these 2014 European elections would also be unprecedented. For the first time in EU history, the European political parties agreed to nominate candidates to chair the institution, which they justified by putting forward Article 17 of the Lisbon Treaty. The European Parliament has often characteristically used political discourse - the logos, to influence the EU’s institutional framework, even though it entails grappling with Member States. It took the form of reports and resolutions, like the official use of the phrase “European Parliament” in 1962, direct universal suffrage elections in 1975 and a European Union in 1984. Nominating contenders to chair the European Commission is no exception. It requires a specific political discourse whose origins can be traced back to the early years of the European Parliament, when it was still the “Common Assembly”. This political discourse is one of the elements thanks to which the European Parliament acquired visibility and new prerogatives, in pursuit of its legitimacy. However, the executive branch in all member states is not intent on yielding such prerogatives to the European Parliament. As a matter of fact, the European Parliament has often ended up strengthening the heads of state and governments, since MEPs are forced to resort to self-discipline. The symbolic significance of its logos and, consequently, its own politicisation as a source of legitimacy, is thus undermined. For instance, in 2014, Jean-Claude Juncker’s election actually strengthened German Chancellor Angela Merkel. First she questioned the fact that the candidate whose party holds the parliamentary majority after the election should be appointed President of the Commission. Then she seemed strongly intent on democratising the Union, when she confronted David Cameron, who openly opposed Juncker, believed to be too federalist and old-fashioned a candidate. By doing so, she eventually reduced the symbolic dimension of the European Parliament’s initiative, and Juncker’s election. She also unquestionably embodied EU leadership. This paper aims at analysing Juncker’s election to the Presidency of the European Commission, as well as other questions it raises. In the first part, I lay out some thoughts about the sociohistorical context of voting in European elections in order to make the readers understand why the European Parliament should be bolder. Secondly, I try to explain how the European Parliament has used the logos as a weapon to grapple with member states for more power, as was the case during the 2014 European elections. Last but not least, I seek to show how Angela Merkel got hold of that weapon and took advantage of it, thus proving that despite MEPs’ best efforts, Juncker’s task will be all the more complicated as he was not the consensual candidate of all the governments.

There is evidence across several species for genetic control of phenotypic variation of complex traits1, 2, 3, 4, such that the variance among phenotypes is genotype dependent. Understanding genetic control of variability is important in evolutionary biology, agricultural selection programmes and human medicine, yet for complex traits, no individual genetic variants associated with variance, as opposed to the mean, have been identified. Here we perform a meta-analysis of genome-wide association studies of phenotypic variation using ~170,000 samples on height and body mass index (BMI) in human populations. We report evidence that the single nucleotide polymorphism (SNP) rs7202116 at the FTO gene locus, which is known to be associated with obesity (as measured by mean BMI for each rs7202116 genotype)5, 6, 7, is also associated with phenotypic variability. We show that the results are not due to scale effects or other artefacts, and find no other experiment-wise significant evidence for effects on variability, either at loci other than FTO for BMI or at any locus for height. The difference in variance for BMI among individuals with opposite homozygous genotypes at the FTO locus is approximately 7%, corresponding to a difference of ~0.5 kilograms in the standard deviation of weight. Our results indicate that genetic variants can be discovered that are associated with variability, and that between-person variability in obesity can partly be explained by the genotype at the FTO locus. The results are consistent with reported FTO by environment interactions for BMI8, possibly mediated by DNA methylation9, 10. Our BMI results for other SNPs and our height results for all SNPs suggest that most genetic variants, including those that influence mean height or mean BMI, are not associated with phenotypic variance, or that their effects on variability are too small to detect even with samples sizes greater than 100,000.

Ten different tRNAGly1 genes from the silk worm, Bombyx mori, have been cloned and characterized. These genes were transcribed in vitro in homologous nuclear extracts from the posterior silk gland (PSG) or nuclear extracts derived from the middle silk gland or ovarian tissues. Although the transcription levels were much higher in the PSG nuclear extracts, the transcriptional efficiency of the individual genes followed a similar pattern in all the extracts. Based on the levels of in vitro transcription, the ten tRNAGly1 genes could be divided into three groups, viz., those which were transcribed at very high levels (e.g., clone pR8), high to medium levels (e.g., pBmil, pBmpl, pBmhl, pBmtl) and low to barely detectable levels (e.g., pBmsl, pBmjl and pBmkl). The coding sequences of all these tRNA genes being identical, the differential transcription suggested that the flanking sequences modulate their transcriptional efficiency. The presence of positive and negative regulatory elements in the 5' flanking regions of these genes was confirmed by transcription competition experiments. A positive element was present in the immediate upstream A + T-rich sequences in all the genes, but no consensus sequences correlating to the transcriptional status could be generated. The presence of negative elements on the other hand was indicated only in some of the genes and therefore may have a role in the differential transcription of these tRNAGly genes in vivo.

Multilayers of poly(diallyldimethylammonium chloride) (PDDA) and citrate capped Au nanoparticles (AuNPs) anchored on sodium 3-mercapto-1-propanesulfonate modified gold electrode by electrostatic layer-by-layer assembly (LbL) technique are shown to be an excellent architecture for the direct electrochemical oxidation of As(III) species. The growth of successive layers in the proposed LbL architecture is followed by atomic force microscopy, UV-vis spectroscopy, quartz crystal microbalance with energy dissipation, and electrochemistry. The first bilayer is found to show rather different physico-chemical characteristics as compared to the subsequent bilayers, and this is attributed to the difference in the adsorption environments. The analytical utility of the architecture with five bilayers is exploited for arsenic sensing via the direct electrocatalytic oxidation of As(III), and the detection limit is found to be well below the WHO guidelines of 10 ppb. When the non-redox active PDDA is replaced by the redoxactive Os(2,2'-bipyridine)(2)Cl-poly(4-vinylpyridine) polyelectrolyte (PVPOs) in the LbL assembly, the performance is found to be inferior, demonstrating that the redox activity of the polyelectrolyte is futile as far as the direct electro-oxidation of As(III) is concerned. The molecular mechanism of antimony-resistant Leishmania donovani ((SbLD)-L-R)-driven up-regulation of IL-10 and multidrug-resistant protein 1 (MDR1) in infected macrophages (M phi s) has been investigated. This study showed that both promastigote and amastigote forms of (SbLD)-L-R, but not the antimony-sensitive form of LD, express a unique glycan with N-acetylgalactosamine as a terminal sugar. Removal of it either by enzyme treatment or by knocking down the relevant enzyme, galactosyltransferase in (SbLD)-L-R (KD (SbLD)-L-R), compromises the ability to induce the above effects. Infection of M phi s with KD (SbLD)-L-R enhanced the sensitivity toward antimonials compared with infection with (SbLD)-L-R, and infection of BALB/c mice with KD (SbLD)-L-R caused significantly less organ parasite burden compared with infection induced by (SbLD)-L-R. The innate immune receptor, Toll-like receptor 2/6 heterodimer, is exploited by (SbLD)-L-R to activate ERK and nuclear translocation of NF-kappa B involving p50/c-Rel leading to IL-10 induction, whereas MDR1 up-regulation is mediated by PI3K/Akt and the JNK pathway. Interestingly both recombinant IL-10 and (SbLD)-L-R up-regulate MDR1 in M. with different time kinetics, where phosphorylation of PI3K was noted at 12 h and 48 h, respectively, but M phi s derived from IL-10(-/-) mice are unable to show MDR1 up-regulation on infection with (SbLD)-L-R. Thus, it is very likely that an IL-10 surge is a prerequisite for MDR1 up-regulation. The transcription factor important for IL-10-driven MDR1 up-regulation is c-Fos/c-Jun and not NF-kappa B, as evident from studies with pharmacological inhibitors and promoter mapping with deletion constructs.

The high species richness of tropical forests has long been recognized, yet there remains substantial uncertainty regarding the actual number of tropical tree species. Using a pantropical tree inventory database from closed canopy forests, consisting of 657,630 trees belonging to 11,371 species, we use a fitted value of Fisher's alpha and an approximate pantropical stem total to estimate the minimum number of tropical forest tree species to fall between similar to 40,000 and similar to 53,000, i.e., at the high end of previous estimates. Contrary to common assumption, the Indo-Pacific region was found to be as species-rich as the Neotropics, with both regions having a minimum of similar to 19,000-25,000 tree species. Continental Africa is relatively depauperate with a minimum of similar to 4,500-6,000 tree species. Very few species are shared among the African, American, and the Indo-Pacific regions. We provide a methodological framework for estimating species richness in trees that may help refine species richness estimates of tree-dependent taxa.

A presente dissertação tem como objetivo refletir sobre a imagem do artista na obra de Silviano Santiago, mais precisamente sobre a figura do narrador-escritor da novela Uma história de família (1992), em abordagem comparativa com outros artistas, cuja produção e aparecimento na mídia problematizaram as primeiras décadas do HIV/AIDS, tais como Cazuza e o artista visual Leonilson, que posavam midiaticamente como perigosos. Pretende-se apontar, na obra de Silviano Santiago, uma performance do artista (soropositivamente) perigoso, na qual a relação entre enfermidade, dor, prazer e labor literário é verificável. Outros artistas são estudados, tais como o escritor Jean-Claude Bernardet, a artista visual Teresa Margolles e o performer Ron Athey, assim como os já citados Cazuza e Leonilson. Partindo da tese de Doutorado e da dissertação de Mestrado de Marcelo Secron Bessa, este trabalho procura avançar com algumas questões que, para tanto, recorrem ao conceito de performatividade, em Judith Butler, à noção do corpo como um Outro radical, de Henri-Pierre Jeudy, e à análise da relação entre AIDS e a discursividade empreendida por Susan Sontag. Os textos críticos de Silviano Santiago também fundamentam o presente estudo, mais precisamente na análise daqueles da relação entre dor e prazer na criação literária e da noção de uma homossexualidade astuciosa em resposta às discursividades heteronormativas

Esta dissertação tem como objetivo analisar a trajetória escolar de jovens de origem popular e oriundos de um bairro de periferia da cidade do Rio de Janeiro, que ingressaram em cursos de pós-graduação stricto sensu de importantes universidades públicas. Tais trajetórias serão aqui analisadas a partir das narrativas dos próprios atores sociais em questão, buscando evidenciar as marcas de sua socialização e formação escolar em um contexto social que, do ponto de vista de algumas análises no campo da sociologia da educação, não favoreceriam a aquisição do capital cultural e social necessários ao ingresso na carreira acadêmica. Desta perspectiva, além de não serem muito comuns nos meios populares, essas trajetórias também não seriam reconhecidas pelo grupo de origem que, na maioria das vezes, identifica a formação escolar como porta de entrada no mundo do trabalho, tendo no curso superior o ponto máximo de uma formação escolar bem sucedida. Busca-se aqui, a partir da análise do conjunto dessas trajetórias, apreender os elementos e experiências sociais que possibilitaram esse prolongamento na formação escolar, e, sobretudo, o impacto e o significado da inserção na carreira acadêmica, tanto do ponto de vista da mobilidade social quanto dos conflitos decorrentes dessa experiência subjetiva, quer em relação às expectativas familiares ou do grupo social de origem. O trabalho de campo de caráter etnográfico, baseado na observação participante e realização de entrevistas aprofundadas constituíram as ferramentas metodológicas básicas a partir das quais essa pesquisa foi desenvolvida. Com relação à discussão teórica aqui proposta tomou-se como referência os trabalhos de Pierre Bourdieu, Jean-Claude Passeron, Bernard Lahire, Jailson de Souza e Silva e Maria da Graça Jacintho Setton, entre outros

Dynamin-Related Protein 1 (Drp1), a large GTPase of the dynamin superfamily, is required for mitochondrial fission in healthy and apoptotic cells. Drp1 activation is a complex process that involves translocation from the cytosol to the mitochondrial outer membrane (MOM) and assembly into rings/spirals at the MOM, leading to membrane constriction/division. Similar to dynamins, Drp1 contains GTPase (G), bundle signaling element (BSE) and stalk domains. However, instead of the lipid-interacting Pleckstrin Homology (PH) domain present in the dynamins, Drp1 contains the so-called B insert or variable domain that has been suggested to play an important role in Drp1 regulation. Different proteins have been implicated in Drp1 recruitment to the MOM, although how MOM-localized Drp1 acquires its fully functional status remains poorly understood. We found that Drp1 can interact with pure lipid bilayers enriched in the mitochondrion-specific phospholipid cardiolipin (CL). Building on our previous study, we now explore the specificity and functional consequences of this interaction. We show that a four lysine module located within the B insert of Drp1 interacts preferentially with CL over other anionic lipids. This interaction dramatically enhances Drp1 oligomerization and assembly-stimulated GTP hydrolysis. Our results add significantly to a growing body of evidence indicating that CL is an important regulator of many essential mitochondrial functions.

A presente pesquisa tem como objetivo geral analisar as representações sociais da hierarquia construídas pelos militares da FAB, considerando suas segmentações em termos dos círculos de oficiais e graduados, dos respectivos processos de formação, dos sexos de seus componentes e dos estágios, se formados ou em formação, em que se encontram. A fundamentação teórica discorre sobre a hierarquia, princípio geral das instituições militares, previsto por lei e regulador do comportamento militar, conforme prevê o Estatuto dos Militares (BRASIL, 1980); a respeito do poder, na perspectiva de Max Weber (1999) que trata a obediência, a partir da autoridade de uma pessoa sobre outras, e de acordo com Amitai Etzioni (1974), que entende o poder como a capacidade que uma pessoa tem de induzir outra a seguir suas orientações. Apresenta ainda a Teoria das Representações Sociais, proposta por Serge Moscovici (1961), para quem as representações sociais servem de instrumento de leitura da realidade, e a partir da abordagem estrutural, proposta por Jean-Claude Abric (1976), segundo a qual a representação social é composta pelo sistema central, constituído pelo núcleo central, e pelo sistema periférico, formado por elementos periféricos. A amostra foi composta por 600 militares, oriundos das escolas de formação de oficiais e de graduados, do âmbito do Comando da Aeronáutica (COMAER): Academia da Força Aérea (AFA), cuja finalidade é a formação dos Oficiais da Ativa dos quadros de Aviação, de Intendência e de Infantaria, a Escola de Especialistas da Aeronáutica (EEAR), responsável pela formação e pelo aperfeiçoamento dos graduados e o Centro de Instrução Especializada da Aeronáutica (CIEAR), responsável pela capacitação de oficiais e graduados em áreas afins à sua especialização. A coleta de dados foi realizada por meio da técnica de evocação livre ao termo indutor hierarquia, da aplicação do questionário de caracterização e do questionário geral, composto por questões abertas e fechadas. As evocações foram processadas pelo software EVOC, desenvolvido por Pierre Vergès (2005), as questões fechadas receberam o processamento com o auxílio do software SPSS e as questões abertas sofreram uma categorização para o refinamento das análises. Os resultados encontrados nas comparações do conteúdo temático e da estrutura das representações sociais entre os diferentes segmentos permitem afirmar que os temas respeito e disciplina são os prováveis elementos centrais da hierarquia para o conjunto dos 600 militares pesquisados e caracterizam a base comum e consensual compartilhada na caserna. A hierarquia, norteada por um conjunto de regras que regula e assegura a disciplina e o respeito, direciona o comportamento dos militares, durante a formação e no decorrer da carreira. Da análise, conclui-se que os diferentes segmentos de militares da FAB possuem a mesma representação social de hierarquia.

Commercial longline fishing data were analyzed and experiments were conducted with gear equipped with hook timers and timedepth recorders in the Réunion Island fishery (21°5ʹS lat., 53°28ʹE long.) to elucidate direct and indirect effects of the lunar cycle and other operational factors that affect catch rates, catch composition, fish behavior, capture time, and fish survival. Logbook data from 1998 through 2000, comprising 2009 sets, indicated that swordfish (Xiphias gladius) catch-per unit of effort (CPUE) increased during the first and last quarter of the lunar phase, whereas albacore (Thunnus alalunga) CPUE was highest during the full moon. Swordfish were caught rapidly after the longline was set and, like bigeye tuna (Thunnus obesus), they were caught during days characterized by a weak lunar illumination—mainly during low tide. We found a significant but very low influence of chemical lightsticks on CPUE and catch composition. At the time the longline was retrieved, six of the 11 species in the study had >40% survival. Hook timers indicated that only 8.4% of the swordfish were alive after 8 hours of capture, and two shark species (blue shark [Prionace glauca] and oceanic whitetip shark [Carcharhinus longimanus]) showed a greater resilience to capture: 29.3% and 23.5% were alive after 8 hours, respectively. Our results have implications for current fishing practices and we comment on the possibilities of modifying fishing strategies in order to reduce operational costs, bycatch, loss of target fish at sea, and detrimental impacts on the environment.

Esta tese teve por objetivo compreender a relação da criança com seu próprio aniversário, considerando o aniversário como ―experiência‖ e comemoração. Em um primeiro momento, aborda o tema do aniversário ao longo da história, como este se apresenta em diferentes campos da cultura, analisando de que forma elementos como convite, bolo, tema e brindes tornam-se constituintes dessa efeméride. Tal análise adota como referência os estudos de Régine Sirota, Jean-Claude Schmitt, Christian Heslon, Françoise Lebrun e Alain Montandon. Em um segundo momento, transpondo a perspectiva da ―alteridade‖ tal como proposta por Mikhail Bakhtin e Marília Amorim para o campo da pesquisa com crianças, apresenta sentidos compartilhados com crianças sobre a experiência de seu aniversário. O trabalho de campo adotou como principais estratégias metodológicas entrevistas individuais e em grupo com crianças de idade entre 4 e 10 anos, e observação de diferentes festas de aniversário, cujos relatos são trazidos para o texto como ―imagens dialéticas‖, seguindo a orientação de Walter Benjamin. O aniversário, compreendido na perspectiva da criança primeiro como ―festa‖ e depois como ―condição para crescer‖, é analisado neste trabalho com base nas vozes das crianças, sua relação com o tempo e com o tema do aniversário. É trazido como contínuo de mudanças, manifestado pelo pertencimento a determinada idade cronológica que toma como referencia parâmetros de comportamento socialmente estabelecidos. A interlocução com as crianças e o estudo teórico mais aprofundado apontaram para a complexidade da temática, que se apresenta socialmente situada entre a noção de espetáculo, fenômeno abordado por Guy Debord, e a ideia de simplicidade, isto é, os pequenos gestos que desenham silenciosamente a experiência e buscam formas sutis para comunicá-la, como postula Benjamin. Assentado na premissa de que ―menos pode ser mais‖, este estudo abre mão de estabelecer uma fácil e determinista relação de oposição entre espetáculo e simplicidade. Em vez disso, aceitando o desafio de Walter Benjamin e Win Wenders, aposta na riqueza das contradições que se colocam entre estes como um desvio metodológico que convide a uma reflexão crítica, aguçando o olhar para diferentes comemorações de aniversários e buscando nelas estilhaços de histórias.

A dissertação se propõe a investigar as diferentes formas de estruturas narrativas para o desenvolvimento em um roteiro cinematográfico, adaptado de uma obra literária. Para este trabalho, vamos usar como corpus o romance Ensaio sobre a cegueira de José Saramago que foi adaptado pelo roteirista Don Mckellar, originando o filme dirigido por Fernando Meirelles, em 2008. A finalidade deste recorte é mostrar o processo de construção narrativa no audiovisual, tendo como base o livro. Nessa investigação, o suporte para análise do audiovisual será realizado com conceituados especialistas em roteiros cinematográfico como Linda Hutcheon, Robert Mckee, Jean-Claude Carriére, Flávio de Campos e David Howard. As possibilidades da criação do roteiro seguirão o mote em quatro divisões: narrativa, estrutura, ensaio sobre imagens e da palavra para imagem, procurando responder como a linguagem intervém no sentido da palavra para imagem e como ocorre o conceito de autoria nessa vertente da adaptação. A trama de Ensaio sobre a cegueira, traz elementos clássicos da estrutura dramática e amplia sua dimensão em relação ao conteúdo da estória na roteirização de Don Mckellar. Com esta pesquisa, esperamos contribuir para um melhor entendimento na construção e organização de um roteiro no processo de adaptação em si
