The interference of microwave-induced resistance oscillations and magneto-intersubband oscillations in double quantum wells exposed to a continuous microwave irradiation is under study. By comparing experimental and theoretical magnetoresistance traces at different temperatures, we confirm that the inelastic mechanism of photoresistance explains our observations up to T similar or equal to 4 K. For higher temperatures, our results suggest a deviation of the inelastic scattering time tau(in) from the predicted T(-2) dependence. (C) 2009 Elsevier B.V. All rights reserved.

Resistance to melarsoprol and pentamidine was induced in bloodstream-form Trypanosoma brucei rhodesiense STIB 900 in vitro, and drug sensitivity was determined for melarsoprol, pentamidine and furamidine. The resistant populations were also inoculated into immunosuppressed mice to verify infectivity and to monitor whether rodent passage selects for clones with altered drug sensitivity. After proliferation in the mouse, trypanosomes were isolated and their IC(50) values to the three drugs were determined. To assess the stability of drug-induced resistance, drug pressure was ceased for 2 months and the drug sensitivity was determined again. Resistance was stable, with a few exceptions that are discussed. Drug IC(50)s indicated cross-resistance among all drugs, but to varying extents: resistance of the melarsoprol-selected and pentamidine-selected trypanosomes to pentamidine was the same, but the pentamidine-selected trypanosome population showed lower resistance to melarsoprol than the melarsoprol-selected trypanosomes. Interestingly, both resistant populations revealed the same intermediate cross-resistance to furamidine. Resistant trypanosome populations were characterised by molecular means, referring to the status of the TbAT1 gene. The melarsoprol-selected population apparently had lost TbAT1, whereas in the pentamidine-selected trypanosome population it was still present.

Roots respond dynamically to belowground herbivore attack. Yet, little is known about the mechanisms and ecological consequences of these responses. Do roots behave the same way as leaves, or do the paradigms derived from aboveground research need to be rewritten? This is the central question that we tackle in this article. To this end, we review the current literature on induced root defenses and present a number of experiments on the interaction between the root herbivore Diabrotica virgifera and its natural host, maize. Currently, the literature provides no clear evidence that plants can recognize root herbivores specifically. In maize, mild mechanical damage is sufficient to trigger a root volatile response comparable to D. virgifera induction. Interestingly, the jasmonate (JA) burst, a highly conserved signaling event following leaf attack, is consistently attenuated in the roots across plant species, from wild tobacco to Arabidopsis. In accordance, we found only a weak JA response in D. virgifera attacked maize roots. Despite this reduction in JA-signaling, roots of many plants start producing a distinct suite of secondary metabolites upon attack and reconfigure their primary metabolism. We, therefore, postulate the existence of additional, unknown signals that govern induced root responses in the absence of a jasmonate burst. Surprisingly, despite the high phenotypic plasticity of plant roots, evidence for herbivore-induced resistance below ground is virtually absent from the literature. We propose that other defensive mechanisms, including resource reallocation and compensatory growth, may be more important to improve plant immunity below ground.

Aphids are important herbivores of both wild and cultivated plants. Plants rely on unique mechanisms of recognition, signalling and defence to cope with the specialized mode of phloem feeding by aphids. Aspects of the molecular mechanisms underlying aphid-plant interactions are beginning to be understood. Recent advances include the identification of aphid salivary proteins involved in host plant manipulation, and plant receptors involved in aphid recognition. However, a complete picture of aphid-plant interactions requires consideration of the ecological outcome of these mechanisms in nature, and the evolutionary processes that shaped them. Here we identify general patterns of resistance, with a special focus on recognition, phytohormonal signalling, secondary metabolites and induction of plant resistance. We discuss how host specialization can enable aphids to co-opt both the phytohormonal responses and defensive compounds of plants for their own benefit at a local scale. In response, systemically induced resistance in plants is common and often involves targeted responses to specific aphid species or even genotypes. As co-evolutionary adaptation between plants and aphids is ongoing, the stealthy nature of aphid feeding makes both the mechanisms and outcomes of these interactions highly distinct from those of other herbivore-plant interactions. © 2016 Macmillan Publishers Limited.

Cells of the monocyte/macrophage lineage play a central role in both innate and acquired immunity of the host. However, the acquisition of functional competence and the ability to respond to a variety of activating or modulating signals require maturation and differentiation of circulating monocytes and entail alterations in both biochemical and phenotypic profiles of the cells. The process of activation also confers survival signals essential for the functional integrity of monocytes enabling the cells to remain viable in microenvironments of immune or inflammatory lesions that are rich in cytotoxic inflammatory mediators and reactive free-radical species. However, the molecular mechanisms of activation-induced survival signals in monocytes remain obscure. To define the mechanistic basis of activation-induced resistance to apoptosis in human monocytes at the molecular level, we evaluated the modulation of expression profiles of genes associated with the cellular apoptotic pathways upon activation and demonstrate the following: (i) activation results in selective resistance to apoptosis particularly to that induced by signaling via death receptors and DNA damage; (ii) concurrent with activation, the most apical protease in the death receptor pathway, caspase-8/FLICE is rapidly down-regulated at the mRNA level representing a novel regulatory mechanism; and (iii) activation of monocytes also leads to dramatic induction of the Bfl-1 gene, an anti apoptotic member of the Bcl-2 family. Our findings thus provide a potential mechanistic basis for the activation-induced resistance to apoptosis in human monocytes.

Based on the observation that removal of tumors from metastatic organs reversed their chemoresistance, we hypothesized that chemoresistance is induced by extracellular factors in tumor-bearing organs. By comparing chemosensitivity and proteins in different tumors (primary vs. metastases) and different culture systems (tumor fragment histocultures vs. monolayer cultures derived from the same tumor), we found elevated levels of acidic (aFGF) and basic (bFGF) fibroblast growth factors in the conditioned medium (CM) of solid and metastatic tumors. These CM induced broad spectrum resistance to drugs with diverse structures and action mechanisms (paclitaxel, doxorubicin, 5-fluorouracil). Inhibition of bFGF by mAb and its removal by immunoprecipitation resulted in complete reversal of the CM-induced chemoresistance, whereas inhibition/removal of aFGF resulted in partial reversal. Using CM that had been depleted of aFGF and/or bFGF and subsequently reconstituted with respective human recombinant proteins, we found that bFGF but not aFGF induced chemoresistance whereas aFGF amplified the bFGF effect. aFGF and bFGF fully accounted for the CM effect, indicating these proteins as the underlying mechanism of the chemoresistance. The FGF-induced resistance was not due to reduced intracellular drug accumulation or altered cell proliferation. We further showed that an inhibitor of aFGF/bFGF (suramin) enhanced the in vitro and in vivo activity of chemotherapy, resulting in shrinkage and eradication of well established human lung metastases in mice without enhancing toxicity. These results indicate elevated levels of extracellular aFGF/bFGF as an epigenetic mechanism by which cancer cells elude cytotoxic insult by chemotherapy, and provide a basis for designing new treatment strategies.

Natural killer (NK) cells are inhibited from killing cellular targets by major histocompatibility complex (MHC) class I molecules. In the mouse, this can be mediated by the Ly-49A NK cell receptor that specifically binds the H-2Dd MHC class I molecule, then inhibits NK cell activity. Previous experiments have indicated that Ly-49A recognizes the alpha 1/alpha 2 domains of MHC class I and that no specific MHC-bound peptide appeared to be involved. We demonstrate here that alanine-substituted peptides, having only the minimal anchor motifs, stabilized H-2Dd expression and provided resistance to H-2Dd-transfected, transporter associated with processing (TAP)-deficient cells from lysis by Ly-49A+ NK cells. Peptide-induced resistance was blocked only by an mAb that binds a conformational determinant on H-2Dd. Moreover, stabilization of "empty" H-2Dd heavy chains by exogenous beta 2-microglobulin did not confer resistance. In contrast to data for MHC class I-restricted T cells that are specific for peptides displayed MHC molecules, these data indicate that NK cells are specific for a peptide-induced conformational determinant, independent of specific peptide. This fundamental distinction between NK cells and T cells further implies that NK cells are sensitive only to global changes in MHC class I conformation or expression, rather than to specific pathogen-encoded peptides. This is consistent with the "missing self" hypothesis, which postulates that NK cells survey tissues for normal expression of MHC class I.

The necrotrophic fungal pathogen Fusarium pseudograminearum (F. pseudograminearum) causes crown rot disease (CR) in wheat. This host-pathogen interaction has not been studied previously at the molecular level. In this study. using real-time quantitative PCR, the expression of 26 selected wheat genes was examined 1, 2 and 4 days after inoculation of wheat seedlings of the CR susceptible cultivar Kennedy and the partially field-resistant cultivar Sunco. Reproducible induction of eight defence genes consisting of PR1.1, PR2 (beta,1-3 glucanase), PR3 (chitinase), PR4 (wheativin), PR5 (thaumatin-like protein). TaPERO (peroxidase), PR10 and TaGLP2a (germin-like) was observed. These genes were induced in both cultivars, however. some genes were induced more rapidly in Sunco than in Kennedy. MJ treatment also induced the above pathogen responsive defence genes in both cultivars while benzo(1,2,3)thiadiazole-7-carbothionic acid S-methyl ester (BTH) treatment weakly induced them in Kennedy only. Similarly. treatment with MJ before inoculation significantly delayed the development of necrotic symptoms for 2 weeks in both wheat cultivars, while BTH pre-treatments delayed symptom development in Kennedy only. The chemically induced protection, therefore, correlated with induction of the F. pseudograminearum-responsive genes. These results support the emerging role of jasmonate signalling in defence against necrotrophic fungal pathogens in monocots and future manipulation of this pathway may improve CR resistance in wheat. Several Eph receptor tyrosine kinases (RTKs) are commonly over-expressed in epithelial and mesenchymal cancers and are recognized as promising therapeutic targets. Although normal interaction between Eph receptors and their ephrin ligands stimulates kinase activity and is generally tumor suppressive, significant Eph over-expression allows activation of ligand- and/or kinase-independent signaling pathways that promote oncogenesis. Single-agent kinase inhibitors are widely used to target RTK-driven tumors but acquired and de novo resistance to such agents is a major limitation to effective clinical use. Accumulating evidence suggests that Ephs can be inhibited by “leaky” or low-specificity kinase inhibitors targeted at other RTKs. Such off-target effects may therefore inadvertently promote ligand- and/or kinase-independent oncogenic Eph signaling, thereby providing a new mechanism by which resistance to the RTK inhibitors can emerge. We propose that combining specific, non-leaky kinase inhibitors with tumor-suppressive stimulators of Eph signaling may provide more effective treatment options for overcoming treatment-induced resistance and clinical failure.

The goal of this thesis was to examine the ecophysiological responses of Scots pine (Pinus sylvestris L.), with an emphasis on the oxidative enzyme peroxidase and plant phenolics to environmental stresses like elevated levels of nickel (Ni) and copper (Cu), and herbivory. The effects of Ni and Cu were studied in a gradient survey at a sulphur dioxide contaminated site in the Kola Peninsula, and with experiments in which seedlings were exposed to Ni mist or to Ni and Cu amended into the soil. In addition, experimental Ni exposure was combined with disturbance of the natural lichen cover of the forest ground layer. Pine sawfly attack was simulated in the early season defoliation experiment, in which mature Scots pine were defoliated (100 %) during two successive years in a dry, nutrient-poor Scots pine stand. In addition, the effect of previous defoliation on the growth of sawfly (Diprion pini L.) larvae was studied. Apoplastic peroxidase activity was elevated in the needles of pine in a Ni- , Cu- and SO2- polluted environment, which indicated an increased oxidative stress. Increased foliar peroxidase activity due to Ni contamination was shown in the experiment, in which Ni was added as mist. No such response was found in peroxidase acitivity of the roots exposed to elevated Ni and/or Cu in the soil. Elevated Ni in the soil increased the concentration of foliar condensed tannins, which are able to bind heavy metals in the cells. Addition of low levels of Ni in the soil appeared to benefit pine seedlings, which was seen as promoted shoot growth and better condition of the roots. Wet Ni deposition of 2000 mg m-2 reduced growth and survival of pine seedlings, whereas deposition levels 200 mg m-2 or 20 mg m-2 caused no effects in a 2-y lasting experiment. The lichen mat on the forest floor did not act as an effective buffer against the adverse impacts of heavy metals on pine seedlings. However, some evidence was found indicating that soil microbes profited from the lichen mat. Artificial defoliation increased peroxidase activity in the Scots pine needles. In addition, defoliation decreased nitrogen, diamine putrescine and glucose concentrations in the needles and increased the concentrations of several phenolic compounds, starch and sucrose. Previous artificial defoliation led to poor growth of sawfly larvae reared on the pines, suggesting delayed induced resistance in Scots pine. However, there was no consistent relationship between inducibility (proportional increase in a compound following defoliation) and adverse effects on the growth of pine sawfly larvae. The observed inducible responses in needle phenolics due to previous defoliation thus appear to represent non-specific responses against sawflies.

真菌病害是造成采后新鲜水果损失的一个主要原因。生物拮抗菌能有效地防治果实采后腐烂,降低杀菌剂的用量,从而增加了食品安全性和降低了潜在的环境危害。然而,与化学杀菌剂相比,单独使用生物拮抗菌对果实采后病害的控制效果有时不如化学杀菌剂明显。因此,为了提高拮抗菌的生防效力,有效控制果实的采后病害,本文主要研究了拮抗菌与化学物质使用的防病机理,并从冬枣果实中克隆β-1,3-葡聚糖酶基因并对其特性进行了初步分析。研究结果表明: 1. 酵母菌Cryptococcus laurentii和枯草芽孢杆菌Bacillus subtilis能够有效的防治冬枣果实采后青霉病和黑霉病的发生,而且C. laurentii对病害的防治效果比B. subtilis好。拮抗菌的抑病效果与使用浓度成正比。在接种C. laurentii的伤口上再接种病原菌可以显著刺激酵母菌的生长。然而,在接种B. subtilis的伤口上接种病原菌则不增加拮抗细菌的群体数量。 2. 不同酵母拮抗菌对四种杀菌剂(Deccozil,Sportak,Iprodine和Stroby)的敏感程度不同。其中,R. glutinis对Deccozil,Iprodione和Stroby最敏感。将低剂量的杀菌剂与酵母菌配合能显著增强酵母菌对采后病菌的抑制作用。C. laurentii与100 µl/L的Stroby配合能完全抑制青霉和黑霉病菌的孢子萌发。2%(w/v)的碳酸氢钠(SBC)与C. laurentii或T. pullulans配合使用显著抑制采后病菌(Penicillium expansum或Alternaria alternata)的孢子萌发和芽管伸长。SBC显著增强拮抗菌对梨果实采后青霉病和黑霉病的防治能力。C. laurentii对采后病害的防治效果好于T. pullulans的防治效果。 3. C. laurentii和B. subtilis对冬枣果实抗病性的诱导与接种距离和接种时间密切相关。距接种拮抗菌近的部位,抗性诱导就越强。酵母菌诱导果实的这种抗病性与诱导果实几丁质酶,β-1,3-葡聚糖酶, PAL,POD和PPO活性有关。 4. 采前喷施2 mM的水杨酸(SA)和0.2 mM的茉莉酸甲酯(MeJA)显著降低甜樱桃果实采后褐腐病的病斑直径, 并能诱导甜樱桃果实β-1,3-葡聚糖酶, PAL, POD和PPO活性以及乙烯含量的增加。采前处理对果实抗病性的诱导效果要好于采后处理。采前和采后SA或MeJA处理,贮藏于25C的甜樱桃果实β-1,3-葡聚糖酶和PAL活性显著高于贮藏于0C的甜樱桃果实的酶活性。2 mM的SA显著抑制了Monilinia fructicola的孢子萌发和菌丝扩展;而0.2 mM的MeJA则对M. fructicola几乎没有抑制作用。在贮藏早期,MeJA对果实β-1,3-葡聚糖酶和PAL活性的诱导作用要强于SA的诱导作用。 5. 1 × 108CFU/ml的C. laurentii,以及5 × 107CFU/ml的C. laurentii与0.2 mM的MeJA 配合使用均可诱导桃果实的抗性,并显著降低果实青霉病和褐腐病的病斑直径。0.2 mM的MeJA能促进C. laurentii生长,抑制P. expansum的菌丝扩展, 但对M. fructicola基本没有抑制作用。在25和0C,MeJA和C. laurentii单独或配合使用都诱导了桃果实几丁质酶,β-1,3-葡聚糖酶,PAL和POD活性的升高。这些抗病相关酶活性的升高可能与病斑扩展的程度是直接相关的。 6. 通过设计简并引物,采用降落PCR,扩增出β-1,3-葡聚糖酶基因的同源片段,分别克隆到两个彼此间同源性很低的β-1,3-葡聚糖酶的cDNA全长(Glu-1和Glu-2)。RT-PCR结果表明,Glu-1基因的表达受酵母拮抗菌C. laurentii处理所诱导,这一结果与酵母拮抗菌诱导果实β-1,3-葡聚糖酶活性的增加相呼应;而Glu-2基因的表达则不受C. laurentii处理所诱导。

用生物和非生物因子来进行采后病害的防治,是一个非常有效的方法。诱导抗性作为控制果蔬采后病害的生物技术,已成为该领域的一个研究热点。然而诱导抗性的机制非常复杂,涉及到寄主、病原菌、激发子之间的相互作用关系。本研究主要利用酵母拮抗菌Pichia membranefaciens和SA处理果实,观察其抗性诱导表达和对采后青霉病菌(Penicillium expansum)的抑制作用,并从蛋白质组学水平上对诱导抗性的机理进行了分析。研究结果表明: 1、酵母拮抗菌P. membranefaciens (5 × 107 cells·ml-1)和SA(0.5 mM)处理采后甜樱桃果实,能够明显地降低病害的发病率和病斑直径。酵母菌和SA处理影响到了果实抗氧化酶的活性,同时还改变了POD同工酶谱和甜樱桃果实的总蛋白含量,并诱导了新的蛋白质条带产生。用光学显微镜和扫描电子显微镜技术观察发现,在in vitro条件下P. membranefaciens能够紧密地结合与病原菌的菌丝,而在in vivo条件下这种结合较为松散。 2、借鉴其它模式植物的方法,我们建立了一整套适用于多汁类植物材料的蛋白质组学研究方法。对于芒果,桃,甜樱桃、苹果以及冬枣等果实,都取得了重复性非常好的2-D图谱。我们应用该技术进一步研究了P. membranefaciens (1 × 108 cells·ml-1)以及SA (0.5 mM)处理对桃果实蛋白质组的诱导影响。结果显示,两种激发子处理都能够诱导桃果实产生抗性,从而减轻青霉病引起的腐烂。在诱导处理1 d以后,酵母拮抗菌和SA分别诱导22和16个蛋白的差异表达。质谱鉴定的蛋白属于6大类:代谢,防御反应,转录,能量途径以及细胞结构。有6个蛋白受到两种激发子的共同调控。其中,4种蛋白(包括glutathione peroxidase, polyphenol oxidase precursor, catalase和methionine sulfoxide reductase) 属于抗氧化蛋白,涉及到活性氧代谢。另2个蛋白(Major allergen Pru av 1和peroxidase)是病程相关蛋白,直接参与植物的防御反应。同时一些磷酸化酶和转录因子也受到两种激发子的调节从而参与果实的抗病反应。酶学测定和Northern杂交的结果表明,拮抗菌与SA处理均能影响过氧化氢酶活性及其基因的表达。 3、采前用较高浓度SA (2 mM) 短时间(10s)处理不同成熟期的甜樱桃果实,能够明显降低果实青霉病的病斑直径,并能减轻较低成熟度果实的发病率。在没有接菌的情况下,SA诱导了33个差异表达的蛋白,其中用质谱鉴定出了26个。而在接种病原菌的情况下,SA诱导了19个差异表达的蛋白,并鉴定出了其中的12个。这些蛋白分别涉及到代谢、防御反应、转录、能量途径、信号转导等过程。在没有接种病原菌的情况下,SA处理诱导了Putative DnaJ heat shock protein, PR1-like protein, Peroxidase, Major allergen Pru av 1 (Pru a 1)和Catalase等与抗病有关的蛋白。而在接种病原菌的情况下,诱导了PR1-like protein, Peroxidase和Catalase蛋白的差异表达。通过酶活性测定以及对细胞学定位的研究,我们发现在没有接种病原菌的情况下,POD的活性受到SA的诱导。但是在接种病原菌以后,诱导效果不明显。

果实为开花植物所特有的发育器官,在种子的成熟和传播过程中发挥着重要作用。同时,肉质果实中含有丰富的营养物质,包括纤维素、维生素、抗氧化剂等,成为人们饮食的重要组成部分。由于果实的成熟衰老和抗病性直接影响果品的质量和市场价值,因此,研究果实成熟衰老和抗病性的调控机制具有重要的理论意义和应用前景。本文主要利用蛋白质组学的方法,探讨外源化学物质抑制果实成熟衰老和诱导抗病性的调控机制。 1. 硅对果实的抗病性诱导:用硅酸钠(1%)处理采后的甜樱桃果实,再接种褐腐病原菌(Molinilia fracticola),置于20C下,观测贮藏期间果实的发病率,并分析硅处理后诱导的主要蛋白质及调控机制。研究结果表明:硅酸钠处理可显著抑制贮藏期间褐腐病的发生,其抑病机理与硅诱导PR-蛋白的表达,提高果实的抗氧化水平,减轻由病原菌侵染造成的氧化胁迫相关。同时,硅处理还能保护细胞骨架结构,有利于增强果实对病原菌入侵的抵抗力。 2. 水杨酸对果实的抗病性诱导:用水杨酸(SA,2mM)在果园处理三种成熟度的甜樱桃果实,然后接种青霉病原菌(Penicillium expansum)观察其发病情况,并取样分析参与抗病性应答的主要蛋白质及调控机制。试验结果表明:SA处理能显著降低青霉病的发病率和抑制病斑扩展,而且SA对低成熟度甜樱桃果实的抗性诱导效果更好。在八成熟的果实中,有5个热激蛋白和4个脱氢酶蛋白被SA诱导,这些蛋白参与了糖酵解和三羧酸循环。抗氧化蛋白和PR蛋白主要参与较低成熟度果实的抗性应答,而热激蛋白和脱氢酶在较高成熟度果实的抗性应答中更明显,SA诱导的抗性与代谢途径相关。 3. 草酸对果实的抗性诱导:用5mM的草酸处理冬枣果实后,接种青霉菌(P. expansum),观察果实发病情况,测定果实相关的生理指标,分析参与果实抗性应答的主要蛋白质及调控机制。结果表明:草酸能明显延缓冬枣果实的衰老,提高果实对青霉菌的抗性。草酸处理能抑制果实乙烯的释放量和呼吸强度,延缓叶绿素的降解,减少乙醇积累。利用蛋白质组学的研究方法证实了在25个参与了草酸处理应答的蛋白中,胱硫醚-β-合酶结构域包含蛋白(CBB domain-containing protein)和3个与光合作用相关蛋白[二磷酸核酮糖羧化酶/加氧酶(Ribulose bisphosphate carboxylase/oxygenase activase, chloroplast precursor),二磷酸核酮糖羧化酶/加氧酶大亚基结合蛋白(RuBisCO large subunit-binding protein subunit beta, chloroplast precursor),植物光系统Ⅱ放氧复合蛋白2(PSII oxygen-evolving complex protein 2)]的表达量上调,乙醇脱氢酶的表达量出现下调。草酸处理还提高了与乙烯合成前体相关蛋白的表达,抑制了ACC合成酶的活性。草酸提高果实抗病的机制与延缓果实成熟衰老和保持果实抗性有关。 4. 果实衰老的调控机制:采用高氧(100%)和低氧(2-3%)处理苹果果实,观察果实衰老的进程,并基于蛋白质组学的研究方法,探讨苹果果实衰老与线粒体蛋白质组的关系。结果表明,在苹果衰老过程中有22个蛋白的表达量发生变化,这些蛋白主要参与了三羧酸循环,电子传递,碳代谢和胁迫应答。高氧处理能诱导氧化胁迫,加速了果实的衰老。质谱鉴定结果证明:在高氧胁迫下,超氧化物歧化酶(manganese superoxide dismutase,MnSOD)和线粒体外膜通道蛋白(porin) 的表达量降低,MnSOD的活性受到抑制,由此提高了线粒体中超氧阴离子的含量,增加了蛋白质的氧化损伤。 此外,高氧处理改变了porin的功能,导致了线粒体膜的透势发生变化,从而引起外膜损伤。由此阐明了活性氧在果实的成熟衰老调控中的重要作用。

在果实采后贮藏过程中,病原真菌的侵染会引起果实腐烂,造成巨大的经济损失。利用生物和非生物因子诱导果实抗病性,已经成为采后病害防治领域的一个研究热点。本文主要利用RT-PCR和RACE技术克隆果实抗病相关基因,通过分子杂交和蛋白羰基化免疫检测技术,研究了外源SA和酵母拮抗菌诱导果实抗病性机理,结果表明: 1. 通过优化RNA提取方法,能从含有多糖的冬枣、葡萄、甜樱桃、桃、番茄等果实中提取到质量较好的RNA,用于RT-PCR和Northern杂交。 2. 采用RT-PCR和RACE方法,从甜樱桃果实克隆了两个抗氧化相关基因CAT2(Genbank:EF165590)和GPX(Genbank:EF165591)和两个PR基因GLU-1(Genbank:EF177487)和GLU-3(Genbank:EF177488)。其中CAT2全长cDNA序列为1479 bp,编码492个氨基酸;GPX全长cDNA序列为513 bp,编码170个氨基酸;GLU-1全长cDNA序列为1050 bp,编码349个氨基酸;GLU-3部分cDNA序列为454 bp,编码141个氨基酸。 3. 酵母拮抗菌Pichia membranaefaciens处理不同成熟度的甜樱桃果实,能显著降低果实贮藏期间青霉病(Penicillium expansum)的发生,并且对低成熟度果实的病害防治效果更为明显。酵母拮抗菌的抑病机理与减轻了甜樱桃果实蛋白羰基化程度,诱导了果实抗氧化酶基因(CAT和GPX)和PR基因(GLU-1)的表达和提高了抗氧化酶(CAT和GPX)和β-1,3-葡聚糖酶的活性有关。 4. 四种酵母拮抗菌P. membranaefaciens, Cryptococcus laurentii, Candida guilliermondii和Rhodotorula glutinis处理桃果实,可显著降低贮藏期间的褐腐病(Monilinia fructicola)。这是由于酵母拮抗菌能抑制病原菌侵染造成的氧化胁迫和蛋白羰基化。此外,酵母拮抗菌处理还能显著诱导CAT、POD、几丁质酶、β-1,3-葡聚糖酶活性及相应基因的表达。 5. 水杨酸(SA,2 mM)处理采后不同成熟度的甜樱桃果实,能显著降低青霉病的危害。其抑病机理与SA处理能减轻P. expansum侵染引起的果实蛋白羰基化程度,显著提高CAT、GPX和β-1,3-葡聚糖酶基因的表达和相关的酶的活性有关。而2 mM的SA处理对P. expansum的生长没有直接抑制作用。 6. 水杨酸(SA,2 mM)与P. membranaefaciens(1×108 CFU/ml)配合处理能显著降低低温贮藏期间桃果实的褐腐病,并能提高几丁质酶、β-1,3-葡聚糖酶和POD的活性和相关基因的表达。另外,2 mM的SA对拮抗菌P. membranaefaciens的生长没有影响,但能够抑制病原菌M. fructicola的孢子萌发和菌丝扩展。
