CXCR4, a chemokine receptor involved in metastasis and homing of hematopoietic stem cells, signals through two major pathways: Gαi and β-arrestin2. β-arrestin2 terminates G-protein signaling and targets the receptor to endocytosis. This project proposed to study the effect of a previously described set of CXCR4 mutants on both these signaling pathways, as well as their localization. These mutants were assayed by different Bioluminescence Resonance Energy Transfer (BRET) systems. Using these systems, we confirmed that N119S is a constitutively active mutant (CAM), spontaneously activating Gαi. As well, we found that R134A is a constitutively inactive mutant (CIM), devoided of G-protein signaling, but spontaneously recruiting β-arrestin2. In addition, we studied the dependency of β-arrestin2 recruitment on the Gαi activity. By targeting R134A and N119S with pertussis toxin, an inhibitor of the Gαi activation, we showed efficient blocking of the Gαi pathway, while maintaining the constitutive recruitment of β-arrestin2. This demonstrated that for CXCR4, β-arrestin2 recruitment is independent of the Gαi pathway. Finally, two synthetic ligands of CXCR4, AMD3100 and TC14012 were tested for their ability to recruit β-arrestin2. AMD3100 is a clinically approved drug used for stem cell transplantation, with considerable side effects. We found it to be an antagonist on both Gαi and β-arrestin2 recruitment. On the other hand, TC14012 was found to be an inverse agonist on Gαi and an antagonist on β-arrestin2 recruitment. Based on this finding, it would be preferable to use of TC14012 as it will further reduce any basal Gαi activity, without affecting β-arrestin2 recruitment. These results support the development of TC14012 for stem cell mobilization trials.

Purpose: there are many studies reporting the benefits of pulmonary rehabilitation, but few of them exhibit the behavior and activities of these services. This article presents the characteristics of services, parts management and training level of team members, in addition to the variables or instruments used to measure the effectiveness and impact in these programs. Method: it was made a cross sectional convenience sample which included seven pulmonary rehabilitation services in four Colombian cities (Bogotá, Medellín, Manizales and Cali), selected by the coverage, for having at least one year of experience and for being formally established and recognized nationwide. The interdisciplinary team of each service answered a survey that was validated through a pilot test and expert consensus. Participation was voluntary. Results: labor onset pulmonary rehabilitation services correspond to an average of a decade, with COPD and asthma pathologies of attention. The programs are characterized by an outpatient treatment with an average duration of eight to twelve weeks, with a frequency of an hour three times a week. Also, the director of the service is regularly a pulmonologist and the coordinator a physiotherapist (57.14%). The posgradual training of these professionals is notable, and they report to have procedural, administrative and communicative skills, but qualify regular there research skills. The physical and technological resources are well tested. 71.42% have done impact studies, but only 28.57% have been published. All have in common training in upper limbs, lower limbs, respiratory muscles, counseling, functional assessment and quality of life. The effectiveness and impact of programs is measured by the walking test, quality of life questionnaires and activities of daily living.

Este estudio explora el estado actual y el potencial competitivo de las exportaciones agrícolas colombianas a los mercados de Estados Unidos y la Unión Eurpea. Los mercados de estos paises industrializados se constituyen en los más importantes para Colombia, pero al mismo tiempo tienen varias restricciones en el sector agrícola. Este estudio mostrará que la competitividad no es solamente una función de los impedimentos locales o internacionales tales como la pobreza institucional, la baja capacidad tecnológica, la disponibilidad de capital, capital humano y mano de obra, sino también de impedimentos externos como el acceso a los mercados. Por lo tanto, éste es un importante objetivo del presente estudio con el objeto de proyectar posibles escenarios para el comercio colombiano, a fin de investigar la eliminación de estas restricciones. En la parte empírica se enfatizará en los impactos de las restricciones externas en el sentido de un estudio econométrico, sin embargo, la influencia de las restricciones internas será discutida como tal.

A fundamental principle of democracy is citizenship freedom. We suggest that a fair electoral contest is possible if a relationship between free will, electoral preferences and respect to the public institutions (constitutionalism) (section 1) exists. We focus on three illiberal practices that perturb the voter's decision: political clientelism and political markets (sections 2 to 4), media influence (which feeds on the voter's limited rationality and limited information) (section 5), and the suppression of opposition options (section 6). Later (section 8), we provide a brief balance and, additionally, we show how in Colombia the political system has missed opportunities to expand the voters' freedom. Our interpretation of the electoral process in Colombia is an appeal, supported on theoretical arguments and empirical evidence, to doubt about the voters' freedom. Also we make a call for more etudies.

We aim to contribute to the assessment of poverty impacts on the rural sector arising from agricultural policy adjustments in Colombia. For this we use an agriculture specialized static CGE model, jointly (sequentially) with a microsimulation model that allows for effective job relocation. Results indicate that the sectoral impact of the program implemented tends to be small and has considerable variability across crops. They also show that the highest impacts come from the irrigation and land improvements component of the program. Lastly, although it reduces poverty, poverty impacts are small and tend to concentrate in rural households toward the middle of the income distribution ladder.

In this paper we find that the diffusion pattern of mobile telephony in Colombia can be best characterised as following a Logistic curve. Although in recent years the rate of growth of mobile phone subscribers has started to slow down, we find evidence that there is still room for further expansion as the saturation level is expected to be reached in five years time. The estimated saturation level is consistent with some individuals possessing more than one mobile device.
Body mass index as a standard of living measure: a different interpretation for the case of Colombia

We analyze the Body Mass Index (BMI) in a distinct way of its traditional use and it lets us use it as a proxy of standard of living for the case of Colombia. Our approach is focused on studying how far the people are from the normal range and not on the score of each one and this lets us to treat equally extreme cases as severe thinness and obesity. We use a probabilistic model (Ordered Probit) that evaluates the probability of being within the normal range or another level. We found that socioeconomic variables have a significant effect on the dependent variable and that there are no linear effects. Besides, people with difficulties for walking and adults have less probability of having a normal BMI.

In a recent investigation, Landsat TM and ETM+ data were used to simulate different resolutions of remotely-sensed images (from 30 to 1100 m) and to analyze the effect of resolution on a range of landscape metrics associated with spatial patterns of forest fragmentation in Chapare, Bolivia since the mid-1980s. Whereas most metrics were found to be highly dependent on pixel size, several fractal metrics (DLFD, MPFD, and AWMPFD) were apparently independent of image resolution, in contradiction with a sizeable body of literature indicating that fractal dimensions of natural objects depend strongly on image characteristics. The present re-analysis of the Chapare images, using two alternative algorithms routinely used for the evaluation of fractal dimensions, shows that the values of the box-counting and information fractal dimensions are systematically larger, sometimes by as much as 85%, than the "fractal" indices DLFD, MPFD, and AWMFD for the same images. In addition, the geometrical fractal features of the forest and non-forest patches in the Chapare region strongly depend on the resolution of images used in the analysis. The largest dependency on resolution occurs for the box-counting fractal dimension in the case of the non-forest patches in 1993, where the difference between the 30 and I 100 m-resolution images corresponds to 24% of the full theoretical range (1.0 to 2.0) of the mass fractal dimension. The observation that the indices DLFD, MPFD, and AWMPFD, unlike the classical fractal dimensions, appear relatively unaffected by resolution in the case of the Chapare images seems due essentially to the fact that these indices are based on a heuristic, "non-geometric" approach to fractals. Because of their lack of a foundation in fractal geometry, nothing guarantees that these indices will be resolution-independent in general. Within the framework of scattering integral equations in momentum space, we present numerical results of scattering of three identical bosons at low energies in two dimensions for short-range separable potentials. An analysis of the present numerical results reveals the three-particle scattering observables to be independent of potential shape provided the low-energy two-particle binding energy and scattering length are held fixed throughout the investigation. We think that the present conclusion of model independence will be valid for any potential, local or nonlocal, whose range is much smaller than the size of the two-particle bound state.
