Wnt1 signaling has been implicated as one factor involved in neural crest-derived melanocyte (NC-M) development. Mice deficient for both Wnt1 and Wnt3a have a marked deficiency in trunk neural crest derivatives including NC-Ms. We have used cell lineage-directed gene targeting of Wnt signaling genes to examine the effects of Wnt signaling in mouse neural crest development. Gene expression was directed to cell lineages by infection with subgroup A avian leukosis virus vectors in lines of transgenic mice that express the retrovirus receptor tv-a. Transgenic mice with tva in either nestin-expressing neural precursor cells (line Ntva) or dopachrome tautomerase (DCT)-expressing melanoblasts (line DCTtva) were analyzed. We overstimulated Wnt signaling in two ways: directed gene transfer of Wnt1 to Ntva+ cells and transfer of β-catenin to DCTtva+ NC-M precursor cells. In both methods, NC-M expansion and differentiation were effected. Significant increases were observed in the number of NC-Ms [melanin+ and tyrosinase-related protein 1 (TYRP1)+ cells], the differentiation of melanin− TYRP1+ cells to melanin+ TYRP1+ NC-Ms, and the intensity of pigmentation per NC-M. These data are consistent with Wnt1 signaling being involved in both expansion and differentiation of migrating NC-Ms in the developing mouse embryo. The use of lineage-directed gene targeting will allow the dissection of signaling molecules involved in NC development and is adaptable to other mammalian developmental systems.

Identification of individual major genes affecting quantitative traits in livestock species has been limited to date. By using a candidate gene approach and a divergent breed cross involving the Chinese Meishan pig, we have shown that a specific allele of the estrogen receptor (ER) locus is associated with increased litter size. Female pigs from synthetic lines with a 50% Meishan background that were homozygous for this beneficial allele produced 2.3 more pigs in first parities and 1.5 more pigs averaged over all parities than females from the same synthetic lines and homozygous for the undesirable allele. This beneficial ER allele was also found in pigs with Large White breed ancestory. Analysis of females with Large White breed background showed an advantage for females homozygous for the beneficial allele as compared to females homozygous for the other allele of more than 1 total pig born. Analyses of growth performance test records detected no significant unfavorable associations of the beneficial allele with growth and developmental traits. Mapping of the ER gene demonstrated that the closest known genes or markers were 3 centimorgans from ER. To our knowledge, one of these, superoxide dismutase gene (SOD2), was mapped for the first time in the pig. Analysis of ER and these linked markers indicated that ER is the best predictor of litter size differences. Introgression of the beneficial allele into commercial pig breeding lines, in which the allele was not present, and marker-assisted selection for the beneficial allele in lines with Meishan and Large White background have begun.

Wolbachia bacteria are common intracellular symbionts of arthropods and have been extensively studied in Drosophila. Most research focuses on two Old Word hosts, Drosophila melanogaster and Drosophila simulans, and does not take into account that some of the Wolbachia associations in these species may have evolved only after their fast global expansion and after the exposure to Wolbachia of previously isolated habitats. Here we looked at Wolbachia of Neotropical Drosophila species. Seventy-one lines of 16 Neotropical Drosophild species sampled in different regions and at different time points were analyzed. Wolbachia is absent in lines of Drosophild willistoni collected before the 1970s, but more recent samples are infected with a strain designated wWiL Wolbachia is absent in all other species of the willistoni group. Polymorphic wWil-related strains were detected in some saltans group species, with D. septentriosaltans being coinfected with at least four variants. Based on wsp and ftsZ sequence data, wWil of D. willistoni is identical to wAu, a strain isolated from D. simulans, but can be discriminated when using a polymorphic minisatellite marker. In contrast to wAu, which infects both germ line and somatic tissues of D. simulans, wWil is found exclusively in the primordial germ line cells of D. willistoni embryos. We report on a pool of closely related Wolbachia strains in Neotropical Drosophila species as a potential source for the wAu strain in D. simulans. Possible evolutionary scenarios reconstructing the infection history of wAu-like Wolbachia in Neotropical Drosophild species and the Old World species D. simulans are discussed.

In recent years, haying has extended to Iberian Mediterranean dry grasslands potentially impacting on grassland bird ecology. We evaluated the impact of haying on a grassland bird community of South Portugal. Our main goals were: (1) to investigate the exposure of different species to haying, (2) to investigate potential removal of nests and dead birds from hayed fields by haying machinery using the ratio (REC) between the expected number of records and the number of records collected and (3) to link clutch destruction and bird mortality with haying management practices. Hayed fields were surveyed for signs of breeding and birds censused prior to mowing. Linear models were computed, linking the REC with haying machinery and sward properties. GLMs and model averaging were used to obtain models linking clutch destruction, bird mortality and haying management variables. Only 4 % of records evidenced successful nesting attempts (N = 177). REC evaluation suggested high nest or dead bird removal by the machinery, particularly in fields with lower vegetation biomass prior to cutting. Sickle bar mowers and one-rotor rotary rakes returned higher REC but lower probability of found nests removed from the original nesting sites comparatively to discs mowers and wheel rakes. Higher probabilities of mortality events were found in fields mown earlier (but not in all years). On the other hand, lower mortality was found in fields raked with two-rotor rotary rakes. Delayed haying, silage production in temporary crops and the use haying machinery enabling simultaneously mowing and gathering hay in lines are discussed as management alternatives.

Maize streak virus (MSV; Genus Mastrevirus, Family Geminiviridae) occurs throughout Africa, where it causes what is probably the most serious viral crop disease on the continent. It is obligately transmitted by as many as six leafhopper species in the Genus Cicadulina, but mainly by C. mbila Naudé and C. storeyi. In addition to maize, it can infect over 80 other species in the Family Poaceae. Whereas 11 strains of MSV are currently known, only the MSV-A strain is known to cause economically significant streak disease in maize. Severe maize streak disease (MSD) manifests as pronounced, continuous parallel chlorotic streaks on leaves, with severe stunting of the affected plant and, usuallly, a failure to produce complete cobs or seed. Natural resistance to MSV in maize, and/or maize infections caused by non-maize-adapted MSV strains, can result in narrow, interrupted streaks and no obvious yield losses. MSV epidemiology is primarily governed by environmental influences on its vector species, resulting in erratic epidemics every 3-10 years. Even in epidemic years, disease incidences can vary from a few infected plants per field, with little associated yield loss, to 100% infection rates and complete yield loss. Taxonomy: The only virus species known to cause MSD is MSV, the type member of the Genus Mastrevirus in the Family Geminiviridae. In addition to the MSV-A strain, which causes the most severe form of streak disease in maize, 10 other MSV strains (MSV-B to MSV-K) are known to infect barley, wheat, oats, rye, sugarcane, millet and many wild, mostly annual, grass species. Seven other mastrevirus species, many with host and geographical ranges partially overlapping those of MSV, appear to infect primarily perennial grasses. Physical properties: MSV and all related grass mastreviruses have single-component, circular, single-stranded DNA genomes of approximately 2700 bases, encapsidated in 22 × 38-nm geminate particles comprising two incomplete T = 1 icosahedra, with 22 pentameric capsomers composed of a single 32-kDa capsid protein. Particles are generally stable in buffers of pH 4-8. Disease symptoms: In infected maize plants, streak disease initially manifests as minute, pale, circular spots on the lowest exposed portion of the youngest leaves. The only leaves that develop symptoms are those formed after infection, with older leaves remaining healthy. As the disease progresses, newer leaves emerge containing streaks up to several millimetres in length along the leaf veins, with primary veins being less affected than secondary or tertiary veins. The streaks are often fused laterally, appearing as narrow, broken, chlorotic stripes, which may extend over the entire length of severely affected leaves. Lesion colour generally varies from white to yellow, with some virus strains causing red pigmentation on maize leaves and abnormal shoot and flower bunching in grasses. Reduced photosynthesis and increased respiration usually lead to a reduction in leaf length and plant height; thus, maize plants infected at an early stage become severely stunted, producing undersized, misshapen cobs or giving no yield at all. Yield loss in susceptible maize is directly related to the time of infection: Infected seedlings produce no yield or are killed, whereas plants infected at later times are proportionately less affected. Disease control: Disease avoidance can be practised by only planting maize during the early season when viral inoculum loads are lowest. Leafhopper vectors can also be controlled with insecticides such as carbofuran. However, the development and use of streak-resistant cultivars is probably the most effective and economically viable means of preventing streak epidemics. Naturally occurring tolerance to MSV (meaning that, although plants become systemically infected, they do not suffer serious yield losses) has been found, which has primarily been attributed to a single gene, msv-1. However, other MSV resistance genes also exist and improved resistance has been achieved by concentrating these within individual maiz genotypes. Whereas true MSV immunity (meaning that plants cannot be symptomatically infected by the virus) has been achieved in lines that include multiple small-effect resistance genes together with msv-1, it has proven difficult to transfer this immunity into commercial maize genotypes. An alternative resistance strategy using genetic engineering is currently being investigated in South Africa. Redesenham-se artes de dizer em um papel xerocado, artes que se traduzem em traços, rabiscos, versos, gritos, modos de fazer. Fanzine é assim, mais do que uma revista de papel sulfite, um espaço recortado, colado para produção de saberes ou, se preferir, uma revista independente na tela do computador. Este trabalho, portanto, propõe um mergulho entre a linha do underground e da pedagogia de criação de fanzines em sala de aula. Compreender os usos e as artes de dizer significa entender historicamente o que as publicações não-oficiais representam para a contemporaneidade. Desta forma, a lógica dos fanzines é bem mais do que trafegar em um circuito alternativo, define-se por si só como uma forma independente de fazer uso de uma expressão. Assim, o fazer fanzine como filofanzine de vida é um exemplo de que esta forma de expressão ultrapassa as paredes institucionais.

The influence of GaAS(1 0 0)2 degrees substrate misorientation on the formation and optical properties of InAs quantum dots (QDs) has been studied in compare with dots on exact GaAs(1 0 0) substrates. It is shown that, while QDs on exact substrates have only one dominant size, dots on misoriented substrates are formed in lines with a clear bimodal size distribution. Room temperature photoluminescence measurements show that QDs on misoriented substrates have narrower FWHM, longer emission wavelength and much larger PL intensity relative to those of dots on exact substrates. However, our rapid thermal annealing (RTA) experiments indicate that annealing shows a stronger effect on dots with misoriented substrates by greatly accelerating the degradation of material quality. A novel line-order of InAs quantum dots (QDs) along the [1, 1, 0] direction on GaAs substrate has been prepared by self-organized growth. After 2.5 monolayer InAs deposition, QDs in the first layer of multi-layer samples started to gather in a line. Owing to the action of strong stress between layers, almost all the dots of the fourth layer gathered in lines. The dots lining up tightly are actually one-dimensional superlattice of QDs, of which the density of electronic states is different from that of isolated QDs or quantum wires. The photoluminescence spectra of our multi-layer QD sample exhibited a feature of very broad band so that it is suitable for the active medium of super luminescent diode. The reason of dots lining up is attributed to the hill-and-valley structure of the buffer, anisotropy and different diffusion rates in the different directions on the buffer and strong stress between QD layers. The growth of ordered self-assembled nanoislands on stepped substrates is studied systematically by kinetic Monte Carlo simulations. As the terrace width is small, the formation of nanoislands is confined in the steps and nanoislands ordered in lines or nanowires can be obtained. The Schwoebel barrier at the step edges has a great influence on the evolution of both the size and space distributions of the islands. When the terrace width is relatively large, self-ordering of nanoislands in the center regions of the terraces happens. An unexpected trend of the nanoisland self-ordering is found as the deposition thickness is larger than 0.2 ML, which can be related to the attractive migrations between nearby islands.

We consider the problem of train planning or scheduling for large, busy, complex train stations, which are common in Europe and elsewhere, though not in North America. We develop the constraints and objectives for this problem, but these are too computationally complex to solve by standard combinatorial search or integer programming methods. Also, the problem is somewhat political in nature, that is, it does not have a clear objective function because it involves multiple train operators with conflicting interests. We therefore develop scheduling heuristics analogous to those successfully adopted by train planners using ''manual'' methods. We tested the model and algorithms by applying to a typical large station that exhibits most of the complexities found in practice. The results compare well with those found by traditional methods, and take account of cost and preference trade-offs not handled by those methods. With successive refinements, the algorithm eventually took only a few seconds to run, the time depending on the version of the algorithm and the scheduling problem. The scheduling models and algorithms developed and tested here can be used on their own, or as key components for a more general system for train scheduling for a rail line or network.Train scheduling for a busy station includes ensuring that there are no conflicts between several hundred trains per day going in and out of the station on intersecting paths from multiple in-lines and out-lines to multiple platforms, while ensuring that each train is allowed at least its minimum required headways, dwell time, turnaround time and trip time. This has to be done while minimizing (costs of) deviations from desired times, platforms or lines, allowing for conflicts due to through-platforms, dead-end platforms, multiple sub-platforms, and possible constraints due to infrastructure, safety or business policy.

Spectroscopic measurements of NOAA AR 10871, obtained with the Extreme Ultraviolet Normal Incidence Spectrograph (EUNIS) sounding rocket instrument on 2006 April 12, reveal velocity oscillations in the He II 303.8 angstrom emission line formed at T approximate to 5; 10(4) K. The oscillations appear to arise in a bright active region loop arcade about 25 '' wide which crosses the EUNIS slit. The period of these transition region oscillations is 26 +/- 4 s, coupled with a velocity amplitude of +/- 10 km s(-1), detected over four complete cycles. Similar oscillations are observed in lines formed at temperatures up to T approximate to 4; 10(5) K, but we find no evidence for the coupling of these velocity oscillations with corresponding phenomena in the corona. We interpret the detected oscillations as originating from an almost purely adiabatic plasma, and infer that they are generated by the resonant transmission of MHD waves through the lower active region atmospheres. Through the use of seismological techniques, we establish that the observed velocity oscillations display wave properties most characteristic of fast body global sausage modes.

Hoje em dia, e com os avanços tecnológicos a surgirem de forma constante, existem novas áreas que têm de ser consideradas com um foco importante por parte de todas as organizações, sendo uma delas a robótica industrial. Motivada em aumentar o seu output, as condições de trabalho para os seus colaboradores, bem como todas as condições de organização da logística interna, a Grohe Portugal, mais especificamente o departamento da montagem, achou relevante fazer um estudo de métodos e tempos, calculando os ganhos potenciais de aumento de output com introdução de robótica nas linhas dos cartuchos. Os objetivos principais seriam então, fazer uma restruturação de todo o layout dessas linhas, tendo como foco automatizar alguma(s) operação(ões), conseguindo assim uma melhoria significativa do output dessas linhas com o menor payback possível. Posto isto, esta dissertação pretende apresentar o trabalho desenvolvido junto da Grohe Portugal, que teve como objetivo fazer os estudos e a automatização de linhas de montagem de cartuchos, bem como melhorar algumas linhas de montagem tendo em conta fatores ergonómicos. Relativamente à automatização da linha dos cartuchos, foi importante estudar todos os seguintes aspetos: utilização de unidades robóticas; ergonomia; ganhos de produtividade; automatizar ou semi-automatizar operações; simplificar processos de montagem; simplificar setups; solicitar orçamentos; elaborar caderno de encargos. Para a realização deste projeto, o trabalho desenvolvido foi decomposto em várias etapas, entre as quais se destacam: análise e estudo dos métodos e sequências de montagem; levantamento de todos os componentes e operações de montagem até à obtenção do cartucho final; estudo de tempos de todas essas operações de montagem; caraterização de um novo layout para as linhas com a introdução de unidades robóticas, tanto quanto possível, mais adequadas; elaboração de caderno de encargos para ser enviado a empresas, para estas poderem apresentar uma orçamentação, bem como indicar as unidades robóticas mais adequadas para as tarefas pretendidas; automatização da linha dos cartuchos. Relativamente ao projeto de novas linhas de montagem tendo em conta fatores ergonómicos, de forma a melhorar os sistemas de abastecimento e as condições de trabalho por parte dos operadores, foram postos em prática diferentes passos, nomeadamente: identificação de todos os processos de montagem realizados pelos operadores na linha a melhorar; estudo e definição da disposição dos componentes na nova linha, bem como a sua forma de abastecimento; projeto da nova linha de montagem em 3D com recurso ao software SolidWorks; realização prática da linha, acompanhando e ajudando a equipa da ferramentaria. O balanço final do trabalho foi bastante positivo, tanto na automatização das linhas de montagem dos cartuchos, em que todo o seu estudo foi alcançado com sucesso, como no melhoramento ergonómico das linhas de montagem, tendo-se alcançado melhorias em alguns índices de qualidade, tempos de abastecimento, organização das linhas, e nas condições de iluminação, resultando essas melhorias numa avaliação positiva por parte dos colaboradores que nelas trabalham todos os dias.

La presente investigación tiene como objetivo analizar en qué medida la estrategia de liderazgo regional de la India ha sido impulsada a través de los programas y proyectos de cooperación sur-sur ofrecidos por este país en el periodo de 2003-2012. De igual forma se pretende indagar sobre el papel histórico que ha jugado la India en el establecimiento y posterior evolución de esta nueva forma de cooperación que ha sido vista por la mayoría de los académicos como un complemento de la cooperación norte-sur, y que responde a una serie de intereses particulares. En este caso en particular, la cooperación sur-sur ha sido utilizada por India como una herramienta para incrementar sus capacidades de poder que le permitan consolidarse como líder regional.

A toxoplasmose congénita é uma doença infecciosa, causada pelo parasita Toxoplasma gondii e, adquirida por transmissão materno-fetal, a qual pode acarretar sequelas neurológicas e oculares muito graves, no recém-nascido. O presente estudo incide sobre as linhas de prevenção da doença, em Portugal. A base da prevenção define-se como primária, através da determinação do estatuto imunológico da mulher, do aconselhamento e adopção de medidas higiénico-dietéticas das mulheres seronegativas, de forma a evitar a infecção materna. A vigilância serológica, na detecção de uma possível infecção materna, e a instituição da terapêutica de profilaxia, constituem a prevenção secundária, de modo a evitar a infecção fetal. A prevenção terciária recai, sobre o estabelecimento de um novo esquema terapêutico, dotado de alguma teratogenicidade, com o intuito de minimizar as sequelas da infecção. Em Portugal, existem muitas mulheres seronegativas, mal informadas acerca da doença, e que não tomam medidas preventivas correctas, para evitar a infecção. Esta problemática é decrescente, de norte para sul do país. A prevenção da doença pode ser bem-sucedida, através da implementação de directrizes específicas, dirigidas aos diferentes grupos de risco e da orientação correcta, pelos profissionais de saúde. A realização de estudos, em várias áreas de intervenção da doença, optimiza a sua prevenção e a sua relação de custo-benefício.

O trabalho teve como objetivo avaliar diferentes épocas de poda, correspondentes aos meses de julho, agosto, setembro e outubro dos anos de 2004/05 e 2005/06, com e sem o uso de irrigação, em figueira 'Roxo de Valinhos', no município de Botucatu-SP. Para atingir tal objetivo, adotou-se o delineamento experimental em parcelas subdivididas, com 5 repetições em blocos, onde as parcelas corresponderam aos tratamentos com e sem irrigação, e as subparcelas foram constituídas pelas podas realizadas nos quatro meses do ano. Foram avaliadas características de produção e alguns atributos de qualidade, que serviram para indicar a necessidade do uso de irrigação. Os resultados obtidos permitiram concluir que, na avaliação do desdobramento da interação entre épocas de poda e irrigação complementar, o mês de agosto, com o uso de irrigação complementar, foi o mais favorável para a realização da poda da figueira, proporcionando as maiores produções médias verificadas no ensaio, tanto no ciclo agrícola de 2004/05 (3.513,8 g planta-1), quanto em 2005/06 (4.110,7 g planta-1). em condições não-irrigadas, não houve diferença estatística entre os meses de julho, agosto e setembro de 2004/05 e entre julho e agosto de 2005/06.
