Angiomatoid fibrous histiocytoma (AFH) is a rare soft tissue neoplasm of intermediate biologic potential and uncertain differentiation, most often arising in the extremities of children and young adults. Although it has characteristic histologic features of a lymphoid cuff surrounding nodules of ovoid cells with blood-filled cystic cavities, diagnosis is often difficult due to its morphologic heterogeneity and lack of specific immunoprofile. Angiomatoid fibrous histiocytoma is associated with recurrent chromosomal translocations, leading to characteristic EWSR1-CREB1, EWSR1-ATF1, and, rarely, FUS-ATF1 gene fusions; fluorescence in situ hybridization (FISH), detecting EWSR1 or FUS rearrangements, and reverse transcription-polymerase chain reaction (RT-PCR) for EWSR1-CREB1 and EWSR1-ATF1 fusion transcripts have become routine ancillary tools. We present a large comparative series of FISH and RT-PCR for AFH. Seventeen neoplasms (from 16 patients) histologically diagnosed as AFH were assessed for EWSR1 rearrangements or EWSR1-CREB1 and EWSR1-ATF1 fusion transcripts. All 17 were positive for either FISH or RT-PCR or both. Of 16, 14 (87.5%) had detectable EWSR1-CREB1 or EWSR1-ATF1 fusion transcripts by RT-PCR, whereas 13 (76.5%) of 17 had positive EWSR1 rearrangement with FISH. All 13 of 13 non-AFH control neoplasms failed to show EWSR1-CREB1 or EWSR1-ATF1 fusion transcripts, whereas EWSR1 rearrangement was present in 2 of these 13 cases (which were histopathologically myoepithelial neoplasms). This study shows that EWSR1-CREB1 or EWSR1-ATF1 fusions predominate in AFH (supporting previous reports that FUS rearrangement is rare in AFH) and that RT-PCR has a comparable detection rate to FISH for AFH. Importantly, cases of AFH can be missed if RT-PCR is not performed in conjunction with FISH, and RT-PCR has the added advantage of specificity, which is crucial, as EWSR1 rearrangements are present in a variety of neoplasms in the histologic differential diagnosis of AFH, that differ in behavior and treatment.

We have used interphase fluorescence in situ hybridization (IFISH) to detect trisomy 8, trisomy 9 and 20q deletion in circulating granulocytes from patients with polycythaemia vera (PV). Out of 64 PV patients, 15 (23%) exhibited an abnormality. Two patients had trisomy 9, three had trisomy 8 and 10 patients had hemizygous deletion of D20S108 (a locus in the 20q common deleted region). Aberrant nuclei ranged from 10% to 80% in these 15 cases. There was no correlation between the presence of a marker and sex, age, interval between presentation and IFISH analysis, neutrophil or platelet count or therapy. Conventional marrow cytogenetic karyotype results were available in 23 cases and there was concurrence between these and blood IFISH in 16 cases (13 normal and three with 20q/D20S108 deletion by both methods). Three patients with D20S108 deletion by IFISH were normal by previous marrow cytogenetic testing and four cases with 20q deletion by previous marrow cytogenetics had normal blood granulocytes according to IFISH. Thus, we confirm that trisomies 8 and 9 and deletion of 20q are diagnostically useful markers of PV. IFISH analysis of blood granulocytes is a practical method for detecting these markers, but as an adjunct to, not as a substitute for, conventional marrow cytogenetics.

Determination of HER2 protein expression by immunohistochemistry (IHC) and genomic status by fluorescent in situ hybridisation (FISH) are important in identifying a subset of high HER2-expressing gastric cancers that might respond to trastuzumab. Although FISH is considered the standard for determination of HER2 genomic status, brightfield ISH is being increasingly recognised as a viable alternative. Also, the impact of HER2 protein expression/genomic heterogeneity on the accuracy of HER2 testing has not been well studied in the context of gastric biopsy samples.

Nucleolus organizer regions (NORs) were analysed in two related and geographically close populations of Eigenmannia sp.1 (Pisces, Gymnotoidei, Sternopygidae) using silver staining and fluorescence in situ hybridization (FISH). The two populations differed in their AS-NOR phenotypes, displaying fixed differences in the NOR-bearing chromosome pairs. FISH with rDNA probes showed that these differences were due to the location of rDNA cistrons. This finding, showing fixed NOR differences between two populations belonging to the same species in a connected river system, is highly significant in terms of evolutionary change, possibly indicating an initial step of genetic differentiation. This result also has important implications from the cytosystematic point of view, as NORs usually have a very constant karyotypic location in fish species and have been used as species-specific chromosome markers.

Members of the highly diverse bacterial phylum Verrucomicrobia are globally distributed in various terrestrial and aquatic habitats. They are key players in soils, but little is known about their role in aquatic systems. Thus, we applied newly designed 16S rRNA-targeted probe set for the identification of Verrucomicrobia and of clades within this phylum to a study concerning the seasonal abundance of Verrucomicrobia in waters of the humic lake Große Fuchskuhle (Germany) by catalyzed reporter deposition fluorescence in situ hybridization. The Lake Große Fuchskuhle is located in the large Mecklenburg-Brandenburg lake district near Berlin (53°10'N, 13°02'E). The lake was artificially divided into four basins (northwest, northeast, southwest, and southeast). We chose the two most contrasting basins, the acidotrophic humic southwestern (SW) basin with a high influx of allochthonous dissolved organic carbon (DOC) and the more mesotrophic northeastern (NE) basin, to study abundance and seasonality of Verrucomicrobia. Lake water was collected from depths of 0.5 m (oxic) and 4.5 m (seasonally anoxic) approximately trimonthly in 2000 (March, June, September and December). The lake hosted diverse Verrucomicrobia clades in all seasons. Either Spartobacteria (up to 19%) or Opitutus spp. (up to 7%) dominated the communities, whereas Prosthecobacter spp. were omnipresent in low numbers (<1%). Verrucomicrobial abundance and community composition varied between the seasons, and between more and less humic basins, but were rather stable in oxic and seasonally anoxic waters.

Deletion of the TP53 gene on chromosome 17p13.1 is the prognostic factor associated with the shortest survival in CLL. We used array-based comparative genomic hybridisation (arrayCGH) to identify additional DNA copy number changes in peripheral blood samples from 74 LRF CLL4 trial patients, 37 with >or=5% and 37 without TP53-deleted cells. ArrayCGH reliably detected deletions on 17p, including the TP53 locus, in cases with >or=50%TP53-deleted cells detected by fluorescence in situ hybridisation, plus seven additional cases with deleted regions on 17p excluding TP53. Losses on chromosomal regions 18p and/or 20p were found exclusively in cases with >or=5%TP53-deleted cells (por=5%TP53-deleted cases (p=0.02). In particular, amplification of 2p and deletion of 6q were both more frequent. Cases with >20%TP53-deleted cells had the worst prognosis in the LRF CLL4 trial.

El presente trabajo se realizó para determinar variación fenotípica en un muestreo in situ en pataste o chayote [Sechium edule (Jacq.) Sw.] existente en la comunidad El Castillito, municipio de Las Sabanas, departamento de Madriz. Se muestrearon 35 accesiones de chayote, y se midieron 34 variables cuantitativas y cualitativas, en las que se pueden mencionar el tamaño de fruto y semilla, forma y color del fruto, presencia de espinas y cantidad de fibra, entre otras. Se utilizó un análisis descript ivo, análisis de correlación, análisis de componentes principales y análisis de conglomerados. Se muestrearon frutos de coloración crema hasta verde oscuro, frutos de forma redonda, ovoide o alargada, con presencia de espinas. Se determinó correlación entre las variables cuantitativas, y mediante los tres primeros Componentes Principales se aisló el 60% de la variación total. El análisis de conglomerado mediante el método Ward y la distancia de Gower agrupó a las 35 accesiones recolectadas en 6 grupos sobresaliendo la longitud, espesor y peso de fruto, así como el peso de semilla, forma del fruto, presencia de espinas , costillas en el pericarpio, y el color del fruto. Se concluye que los materiales muestreados presentaron variación en muchas características cuantitativas y cualitativas.

The ontogeny of IgM-producing cells was studied in juvenile mandarin fish Simperca chuatsi, an important fish in China's aquaculture sector. The IgM-producing cells were localised through in situ hybridisation with a probe complementary to the Ig mu-chain in lymphoid-related tissues, including head kidney, spleen, thymus, intestine and gills. In head kidney, transcripts of Ig mu were first detected at 20 days post-hatching (dph) with a few positive signals. and the number of IgM-producing cells increased obviously from 39 dph onwards. At 136 dph, a large amount of positive cells were observed in the entire organ with clusters of these cells located around the blood vessels. In spleen, IgM-producing cells were found from 26 dph onwards, followed by an increase until 67 dph: clusters of positive cells were also detected around blood vessels at 102 dph. In thymus, IgM-producing cells were first observed at 39 dph; thereafter, no obvious increase was detected until 78 dph. The positive cells in thymus were distributed mainly in the outer zone of thymus. A few IgM-producing cells were still observed in thymus of 1-year-old mandarin fish. IgM-producing cells were not detected in the intestine until 87 dph, with several discrete positively stained cells distributed in the lamina propria. IgM-producing cells, scattered mainly in primary gill filaments around blood vessels, were detected in gills from 90 dph. As in other teleosts, these results indicated that the head kidney appears to be the primary organ for IgM production in mandarin fish, and IgM-producing cells exist in all organs examined in the present study, implying their lymphoid role in fish. We report a female child with tetrasomy of the 15q11-q13 chromosomal region, and autistic disorder associated with mental retardation, developmental problems and behavioral disorders. Combining classical and molecular cytogenetic approaches by fluorescence in situ hybridization technique, the karyotype was demonstrated as 47,XX,+mar.ish der(15)(D15Z1++,D15S11++,GABRB3++,PML-). Duplication of the 15q proximal segment represents the most consistent chromosomal abnormality reported in association with autism. The contribution of the GABA receptor subunit genes, and other genes mapped to this region, to the clinical symptoms of the disease is discussed.

A comparative study of holocentric chromosomes in the triatomine species Panstrongylus megistus, Rhodnius pallescens and Triatoma infestans was carried out in order to characterize heterochromatin, rDNA active sites and nucleolar proteins. Cytological preparations of seminiferous tubules were stained by silver impregnation, C banding, fluorochromes CMA 3/DA and DAPI/DA, and fluorescent in situ hybridization (FISH) with Drosophila melanogaster 28S rDNA probe. Our results showed interesting aspects of the organization of chromatin and chromosomes in the meiotic cells of these insects. In R. pallescens, sex chromosomes (X, Y) were distinct from autosomes, when submitted to silver impregnation, C banding, CMA 3 staining, and FISH, confirming that these chromosomes bear nucleolar organizer regions (NORs). In P. megistus, two of the three sex chromosomes were CMA 3/DAPI-; at early meiotic prophase and at diakinesis, silver impregnation corresponded with FISH signals, indicating that in this species, two chromosomes (probably a sex chromosome and an autosome) bear NORs. In T. infestans, silver nitrate and FISH also stained corresponding areas on meiotic chromosomes. Our data suggest that in triatomines, in general, the number and location of NORs are species-specific. These regions may be considered important chromosome markers for comparative studies to improve the understanding of evolutionary mechanisms in these hematophagous insects. ©FUNPEC-RP.

El Convenio sobre la Diversidad Biológica revela el papel primordial que desempeña el mantenimiento de la diversidad biológica y establece como prioritaria la conservación in situ de los recursos genéticos. Los parientes silvestres de los cultivos, son parte de esta biodiversidad y representan recursos imprescindibles y relevantes para abordar las necesidades de seguridad alimentaria. La papa, ancestralmente cultivada, es el principal cultivo hortícola a nivel mundial y cuenta con más de 200 especies silvestres emparentadas que proporcionan diversidad genética para el mejoramiento del cultivo. La conservación in situ en Áreas Protegidas (APs) permite la conservación de recursos genéticos y son el eje central en prácticamente todas las estrategias nacionales e internacionales de conservación. Este trabajo busca promover y destacar la importancia de conservación in situ de especies silvestres de papa en APs de Argentina. El diseño experimental contempló un trabajo de gabinete y otro de campo. La primera aproximación consistió primeramente en la identificación de APs donde crecen especies silvestres de papa, género Solanum sección Petota, en base al solapamiento de coordenadas geográficas de las APs argentinas y las introducciones del Banco de Germoplasma de Papa y Forrajeras INTA-Balcarce, seguido de la consulta de la base de datos del Sistema de Información de Biodiversidad, para posteriormente relevar el estado actual de conservación enviando una encuesta a los responsables de APs seleccionadas en base a la presencia de las especies de interés. La segunda aproximación experimental implicó el análisis de la variabilidad genética presente en poblaciones naturales de la especie silvestre de papa Solanum kurtzianum que crece en la Reserva Natural Villavicencio provincia de Mendoza. A partir del trabajo de gabinete, se identificaron 18 especies de la sección Petota distribuidas en 21 APs ubicadas en 11 provincias argentinas. Tomando como criterio la riqueza de especies, se destacaron las APs correspondientes al Parque Nacional los Cardones, Monumento Natural Laguna de los Pozuelos y Reserva de la Biósfera de Las Yungas en la región norte del país, las cuales tendrían un alto potencial para establecer reservas genéticas para la conservación in situ de estos recursos. Respecto al trabajo de campo, se analizó la variabilidad genética en 22 poblaciones de S. kurtzianum ubicadas en sitios de fácil y difícil acceso (correspondiente al camino aledaño a la ruta y travesía pedestre respectivamente) dentro de la RN Villavicencio empleando marcadores moleculares AFLP (Amplified Fragment Length Polymorphism). La matriz binaria de presencia/ausencia de fragmentos quedó formada por 67 muestras y 214 fragmentos totales. La similitud genética para el coeficiente DICE varió entre 0,66 y 1. En el fenograma, algunas poblaciones se agruparon por origen geográfico, mientras que en otros casos los agrupamientos fueron heterogéneos, conteniendo muestras recolectadas en distintos sitios. El número total de fragmentos (F) por población varió entre 109 y 143. El número de F únicos varió de 0 a 4 entre las poblaciones. El número de F raros compartidos entre poblaciones varió de 1 a 13 y los frecuentes entre 105 y 132. El promedio de acuerdo al sitio de origen, para los F únicos varió entre 0,6 y 1,6 para los raros entre 2,4 y 6,6 y para los frecuentes de 109,8 a 121,4. Al comparar entre si los sitios de fácil y difícil acceso se obtuvo que el promedio de F únicos, raros y frecuentes, en el primer caso, fue de 0,93, 4,07 y 116,33 respectivamente, mientras que para el sitio de difícil acceso correspondió a 1,00; 4,29 y 121,00. El uso del marcador molecular AFLP ha permitido generar una base de datos de los fragmentos AFLP en poblaciones de S. kurtzianum distribuidas en la RN Villavicencio que permitirá el monitoreo a través del tiempo de la variabilidad genética, herramienta indispensable para implementar estrategias de conservación in situ y que podría contribuir para elaborar y evaluar acciones de manejo dentro de la reserva.

En los procesos de mantenimiento y rehabilitación de pavimentos, se deben definir los objetivos, bien sea para realizar unas reparaciones superficiales sobre daños generados por el agua como consecuencia de fallas en los drenajes, o bien realizar una intervención sobre la estructura buscando recuperarla a sus condiciones de diseño originales, la cual se realiza con el proceso de reciclaje el cual tiene en cuenta las características físico-químicas de los materiales existentes los cuales se consideran sufrieron el adecuado proceso que les permitió hacer parte de la estructura. Existiendo cuatro tipos de reciclaje de los cuales se realiza una descripción general de cada proceso, el presente trabajo se concentra en el análisis técnico – económico del proceso ―In situ‖ en frio, método utilizado en la rehabilitación de la vía Sopetrán - Puente de Occidente, en el Departamento de Antioquia, en una longitud de 13 Km. El presente estudio se realiza verificando el cumplimiento de las especificaciones a nivel nacional que para estos procesos estableció el Instituto Nacional de Vías (INVIAS) y verificando el cumplimiento de las normas IDU ET-2005. El reciclaje ―in situ‖ en frio tiene ventajas ecológicas (no necesita afectar las eventuales fuentes de materiales de la zona), económico (bajos costos comparados con reconstrucción) y técnico (los equipos para este proceso han presentado importantes avances tecnológicos) lo cual permitió su aplicación en la vía mencionada, lo cual con la adición de un agente estabilizador (cemento para el presente estudio) permite recuperar las condiciones iniciales de diseño de la estructura intervenida. El árbol de decisiones es una herramienta utilizada para el análisis y selección de la mejor opción de estructura a realizar teniendo en cuenta calidad y costos. El presente trabajo termina con un análisis detallado de las condiciones existentes, revisión de las diferentes opciones de intervención y el estudio económico de las diferentes alternativas de estructura de pavimento para la rehabilitación de la vía referida.

In the marine environment, phytoplankton and bacterioplankton can be physically associated. Such association has recently been hypothesized to be involved in the toxicity of the dinoflagellate genus Alexandrium. However, the methods, which have been used so far to identify, localize, and quantify bacteria associated with phytoplankton, are either destructive, time consuming, or lack precision. In the present study we combined tyramide signal amplification–fluorescent in situ hybridization (TSA-FISH) with confocal microscopy to determine the physical association of dinoflagellate cells with bacteria. Dinoflagellate attached microflora was successfully identified with TSA-FISH, whereas FISH using monolabeled probes failed to detect bacteria, because of the dinoflagellate autofluorescence. Bacteria attached to entire dinoflagellates were further localized and distinguished from those attached to empty theca, by using calcofluor and DAPI, two fluorochromes that stain dinoflagellate theca and DNA, respectively. The contribution of specific bacterial taxa of attached microflora was assessed by double hybridization. Endocytoplasmic and endonuclear bacteria were successfully identified in the nonthecate dinoflagellate Gyrodinium instriatum. In contrast, intracellular bacteria were not observed in either toxic or nontoxic strains of Alexandrium spp. Finally, the method was successfully tested on natural phytoplankton assemblages, suggesting that this combination of techniques could prove a useful tool for the simultaneous identification, localization, and quantification of bacteria physically associated with dinoflagellates and more generally with phytoplankton.
