Septins are an evolutionarily conserved group of GTP-binding and filament-forming proteins that belong to the large superclass of P-loop GTPases. While originally discovered in yeast as cell division cycle mutants with cytokinesis defects, they are now known to have diverse cellular roles which include polarity determination, cytoskeletal reorganization, membrane dynamics, vesicle trafficking, and exocytosis. Septin proteins form homo- and hetero-oligomeric polymers which can assemble into higher-order filaments. They are also known to interact with components of the cytoskeleton, ie actin and tubulin. The precise role of GTP binding is not clear but a current model suggests that it is associated with conformational changes which alter binding to other proteins. There are at least 12 human septin genes, and although information on expression patterns is limited, most undergo complex alternative splicing with some degree of tissue specificity. Nevertheless, an increasing body of data implicates the septin family in the pathogenesis of diverse disease states including neoplasia, neurodegenerative conditions, and infections. Here the known biochemical properties of mammalian septins are reviewed in the light of the data from yeast and other model organisms. The data implicating septins in human disease are considered and a model linking these data is proposed. It is posited that septins can act as regulatable scaffolds where the stoichiometry of septin associations, modifications, GTP status, and the interactions with other proteins allow the regulation of key cellular processes including polarity determination. Derangements of such septin scaffolds thus explain the role of septins in disease states. Copyright © 2004 Pathological Society of Great Britain and Ireland.

Support for family caregivers, including bereavement follow-up, is a core function of palliative care. Many caregivers acknowledge positive aspects associated with the role; however a considerable proportion will experience poor psychological, social, financial, spiritual, and physical well-being and some will suffer from complicated grief. Many family caregivers have unmet needs and would like more information, preparation, and support to assist them in the caregiving role. There is a shortage of evidence-based strategies to guide health professionals in providing optimal support while the caregiver is providing care and after the patient's death.

The Woodruff Family Collection: From the time the Woodruff Family came to Canada from the United States in 1795, they took an active role in the forming of their communities both in a civic and social manner. This is evident through the documents contained in this collection. The Woodruffs played an active role in the battles fought in Upper Canada and they were an integral part of the Village of St. Davids. They were educated, business-minded and socially engaged. They accumulated much of their fortune through land dealings. Much of this collection focuses on Samuel DeVeaux Woodruff who was principally a businessman. His dedication to his work is shown through his numerous undertakings. He made his mark on the Niagara Peninsula through his work on the railways, roads, marsh land revisions, canals and the paper industry. He was also involved with the founding of the Long Point Company and he took control of building DeVeaux Hall down to the last detail. His offspring inherited his work ethic and his business acumen. The people who married into the Woodruff Family also possessed key social, political and business ties. Anne and Margaret Clement were from a staunch Loyalist background. Samuel Zimmerman was instrumental to the founding of Niagara Falls and Judge Samuel DeVeaux left behind a legacy for poor and homeless boys in Niagara Falls, New York. The Woodruff Family undoubtedly left a mark on the Niagara Peninsula. This collection brings to light many endeavours of the family and their varied contributions.

Karyotypes are defined for two nearctic species of marmots, Marmota olympus (2n = 40) and M. vancouverensis (2n = 42), and supplemental information is included on the karyotypes of M. flaviventris, M. monax ochracea, and M. marmota. The six North American species of Marmota (NF = 66) comprise a distinct group as compared with the middle Asian species (NF = 70) for which the karyotypes are known. Karyologic findings and zoogeographic evidence based upon the distribution of two nearctic species of host-specific cestodes indicate that M. broweri, in northern Alaska, is a pre-Würm relict. Its affinities appear to be with the North American caligata-group rather than with the northeastern Siberian M. camtschatica. The occurrence on M. broweri of the Asian flea, Oropsylla silantiewi, has not been explained. Some ecological and behavioral characteristics of M. broweri are briefly described and compared with those of other species. Family groups of M. broweri hibernate together in single winter dens that are plugged at the entrance; copulation takes place before the animals emerge from the winter den, near mid-May; face-glands are utilized in marking of territory. French abstract: Les auteurs définissent les caryotypes de deux espèces néarctiques de marmottes, Marmota olympus (2n=40) et M. vancouverensis (2n=42), et donnent des précisions sur les caryotypes de M. flaviventris, M. monax ochracea et M. marmota. Les six especes de Marmota (NF=66) d'Amérique du Nord forment un groupe distinct des espèces d'Asie centrale (NF=70) dont Ie caryotype est connu. Les données caryologiques et les preuves zoogéographiques basées sur la répartition de deux espèces néarctiques de cestodes spécifiques de I'hôte démontrent que M. broweri, dans l'Alaska septentrional, est une relicte du pré-Würm. Elle semble avoir plus d'affinités avec Ie groupe nord américain de caligata qu'avec M. camtschatica du nord de la Sibérie. La présence sur M. broweri de la puce asiatique, Oropsylla silantiewi, n'est pas expliquée. Quelques caractéristiques écologiques et éthologiques de M. broweri sont décrites brièvement et comparées avec celles d'autres espèces. Les groupes familiaux de M. broweri hibernent dans un meme terrier dont l'entrée est bouchée; la copulation à lieu avant que les animaux sortent de leur abri hivernal, it la mi-mai ; ils se servent de leurs glandes faciales pour marquer leur territoire.

The purpose of this study was to identify guanine nucleotide-binding proteins (G proteins) involved in the agonist- and guanosine 5'-[gamma-thio]triphosphate (GTP[gamma-S])-induced increase in the Ca2+ sensitivity of 20-kDa myosin light chain (MLC20) phosphorylation and contraction in smooth muscle. A constitutively active, recombinant val14p21rhoA.GTP expressed in the baculovirus/Sf9 system, but not the protein expressed without posttranslational modification in Escherichia coli, induced at constant Ca2+ (pCa 6.4) a slow contraction associated with increased MLC20 phosphorylation from 19.8% to 29.5% (P < 0.05) in smooth muscle permeabilized with beta-esein. The effect of val14p21rhoA.GTP was inhibited by ADP-ribosylation of the protein and was absent in smooth muscle extensively permeabilized with Triton X-100. ADP-ribosylation of endogenous p21rho with epidermal cell differentiation inhibitor (EDIN) inhibited Ca2+ sensitization induced by GTP [in rabbit mesenteric artery (RMA) and rabbit ileum smooth muscles], by carbachol (in rabbit ileum), and by endothelin (in RMA), but not by phenylephrine (in RMA), and only slowed the rate without reducing the amplitude of contractions induced in RMA by 1 microM GTP[gamma-S] at constant Ca2+ concentrations. AlF(4-)-induced Ca2+ sensitization was inhibited by both guanosine 5'-[beta-thio]diphosphate (GDP[beta-S]) and by EDIN. EDIN also inhibited, to a lesser extent, contractions induced by Ca2+ alone (pCa 6.4) in both RMA and rabbit ileum. ADP-ribosylation of trimeric G proteins with pertussis toxin did not inhibit Ca2+ sensitization. We conclude that p21rho may play a role in physiological Ca2+ sensitization as a cofactor with other messengers, rather than as a sole direct inhibitor of smooth muscle MLC20 phosphatase.
