The great utilization of synthetic and persistent xenobiotic cumulative compounds is the main causes of the deterioration of aquatic ecosystems. The one of objective of this work was the determination of the organochlorine levels in the Piracicaba river basin, situated at the center of São Paulo state. Four PCBs congeners, PCP and HCB were determined by GC-ECD in two different matrices: water and sediments. The pattern of the compound distribution indicated a dominance of PCP, HCB and PCB-200. The results indicated contamination in the sampling points located in Campinas, Piracicaba, Santa Bárbara d'Oeste and Sumaré cities.

Sediment contamination is evaluated by determining organic micropollutants (organochlorine compounds - OCs and polycyclic aromatic hydrocarbons - PAHs) in two important Brazilian water reservoirs. Trace levels of OCs were observed in the Santana reservoir (44.8 ng g-1 d.w. of p,p'-DDT), while in the Funil reservoir the levels were below detection level. Forty-eight percent of the found sigmaocs were polychlorinated biphenyls, 29% dichlorodiphenyltrichloroethane (DDT), 18% Drins, and 5% other pesticides (HCB, Heptachlor, Heptachlor-epoxide, gamma-HCH and a-Endosulfan). We observed lower levels of sigmaPAH in the Funil reservoir (1 to 275 ng g-1d.w.) than in the Santana reservoir (2.2 to 26.7 µg g-1 d.w.).

Fueron evaluados los niveles de residuos de pesticidas organoclorados (p,p'-DDE, HCB, gamma-HCH, aldrín) presentes en 123 muestras de tejido adiposo mamario obtenidas de mujeres residentes en Córdoba (España). Además, se investigó la influencia de hábitos alimentarios y edad de las donantes sobre los niveles de organoclorados encontrados en el tejido. El p,p'-DDE fue el compuesto encontrado en concentraciones más altas (1,848myg/g), seguido de HCB (0,235mg/g), aldrín (0,005myg/g) y gamma-HCH (0,003myg/g). Las donantes mayores presentaron niveles de p,p'-DDE, aldrín y HCB superiores que las más jóvenes. Sin embargo, la edad no influyó en los niveles de gamma-HCH. La mayoría de las donantes (40,65%) indicaron las frutas como el alimento más consumido, seguido de la alimentación variada (34,15%), los legumbres (6,50%), los guisos y potajes (6,50%), las verduras (4,88%), la leche (3,25%), el pescado (2,44%) y la carne (1,63%). Una correlación no significativa fue observada entre los niveles de organoclorados en el tejido adiposo y los hábitos alimentarios de las donantes.

Um método de extração de multiresíduos baseado na técnica de dispersão da matriz em fase sólida ("MSPD"), foi otimizado e validado para a extração e análise cromatográfica de 27 agrotóxicos (isômeros a, b, g, d do hexaclorociclohexano (HCH), dieldrin, endrin, heptacloro e seu epóxido (HE), a e b-endosulfan, o,p'-DDD, p,p'-DDD, o,p'-DDE, p,p'-DDE, o,p'-DDT, p,p'-DDT, dicofol, metoxicloro, mirex, hexaclorobenzeno (HCB), clorotalonil, parationa metílica, fenitrotiona, malationa, folpete, diazinona, cis e trans-permetrina em laranjas. A amostra macerada é inicialmente homogeneizada com C18, o homogeneizado é transferido para uma coluna de vidro contendo sílica gel onde se adiciona 10mL de acetato de etila como solvente de eluição. O eluente é concentrado, diluído em isooctano e 1mL deste é injetado no cromatógrafo a gás. Para a separação e quantificação dos 27 agrotóxicos, foi utilizado um CG/DCE. A confirmação dos mesmos foi feita por espectrometria de massas. Para a quantificação dos 27 agrotóxicos utilizou-se a padronização externa. As recuperações dos mesmos variaram de 70 a 120%, considerando-se os níveis de adição agrotóxicos/amostra de 0,02 e 2,0mg/kg. Os limites de quantificação variaram de 0,01 a 0,5mg/kg.

Foram pesquisados resíduos organoclorados em três tecidos humanos: sangue, leite materno e tecido adiposo. Sangue foi colhido de 122 indivíduos, leite materno de 19 lactantes e tecido adiposo de 23 pacientes submetidos a diversas cirurgias. Para cada tecido foram compostos dois grupos: um urbano e outro residente em áreas de intensa atividade agrícola. Os resíduos organoclorados pesquisados foram: α - HCH; γ-HCH; Aldrin; Dieldrin; Endrin; Heptacloro; Heptacloro epóxido; HCB; Mirex; o,p´DDD; o,p´DDT; Oxiclordane; p,p´Metoxicloro; p,p´DDD; p,p´DDE; p,p´DDT e Transnonacloro. Todos os doadores responderam a questionário com quesitos sobre idade, sexo, peso, atividade, local de residência, contato ou não com pesticidas. Foi testada, através de análise estatística, a relação entre presença/ausência de organoclorados nos diferentes tecidos e os dados levantados no questionário. Todas as amostras urbanas foram negativas para os organoclorados analisados. As amostras obtidas nas áreas de atividade agrícola foram positivas, com níveis crescentes de contaminação na seqüência sangue, leite materno, tecido adiposo. Os resultados das análises do presente trabalho foram comparados com resultados préexistentes no Rio Grande do Sul. Entre a pesquisa atual e as pesquisas anteriores usadas para comparação, verificou-se diminuição da incidência de organoclorados em todos os tecidos pesquisados. O OC p,p´DDE foi o único organoclorado encontrado em todos os tecidos e em todas as pesquisas, tanto atuais como anteriores usadas para comparação.

The interaction between dietary energy restriction and low dose of the fungicide hexachlorobenzene (HCB) was evaluated in a rat liver medium-term bioassay for carcinogenesis. Male Wistar rats were fed a control or a 50% energy-restricted diet, both added or not with 50 ppm HCB, for 6 weeks. HCB exposure or energy restriction separately did not exert any influence on the development of glutathione S-transferase placental form (GST-P+) foci of hepatocytes. Simultaneous HCB exposure and energy restriction induced a significant increase in liver centrilobular hypertrophy and GST-P+ foci development. Our findings suggest that energy restriction increases liver response to low dose of HCB, unmasking the promoting potential of this fungicide. (C) 2000 Elsevier B.V. Ireland Ltd. The efficiency of methods for the determination of hexachlorobenzene (HCB) and pentachlorophenol (PCP) in soil samples was evaluated. An on-line method was applied for HCB determination. Soil samples were transferred to chromatographic columns prepacked with alumina. The HCB elution was processed with n-hexane. The PCP was extracted from soil samples with n-hexane-acetone in an ultrasonic bath. After re-extraction with K2CO3 solution PCP was acetylated with acetic anhydride. The pentachlorophenyl acetate derivative was then extracted with n-hexane. The HCB and PCP derivative were analyzed by gas chromatography with electron capture detection (GC-ECD). Mean recoveries obtained from soil samples fortified at levels of 0.5; 4 and 20 ng g(-1) ranged from 91 to 100% for HCB, and for PCP, at levels of 10; 40 and 200 ng g(-1), ranged from 88 to 101%. The promoting activity of the herbicide Diuron was evaluated in a medium-term rat liver carcinogenesis bioassay that uses as endpoint immunohistochemically identified glutathione S-transferase positive (GST-P+) foci. Male Wistar rats were allocated to the following groups: G1 to G6 were initiated for liver carcinogenesis by a single dose of diethylnitrosamine (DEN, 200 mg/kg) while groups G7 and G8 received only 0.9% NaCl (DEN vehicle). From the 2nd week animals were fed a basal diet (G1 and G7) or a diet added with Diuron at 125, 500, 1250, 2500 and 2500 ppm (G2 to G5 and G8, respectively) or 200 ppm Hexaclorobenzene (HCB; G6). The animals were submitted to 70% partial hepatectomy at the 3rd week and sacrificed at the 8th week. The herbicide did not alter ALT or creatinine serum levels. No conspicuous GST-P+ foci development was registered in non-initiated rats fed Diuron at 2500 ppm. While DEN-initiated animals fed Diuron at 1250 or 2500 ppm developed mild centrilobular hypertrophy, DEN-initiated HCB-fed animals showed severe liver centrilobular hypertrophy and significant GST-P+ foci development. These findings indicate that the medium-term assay adopted in this study does not reveal any liver carcinogenesis initiating or promoting potential of Diuron in the rat.

The individual effects of protein deficiency and energy restriction on liver response to low-hexachlorobenzene (HCB) exposure were investigated in adult male Wistar rats. In rats fed either the low-protein or control diet, the only effect caused by HCB was a decrease in paralysis time following an ip injection of zoxazolamine. This decrease was similar for both groups. In the animals subjected to energy restriction, HCB induced a greater decrease in paralysis time, an increase in the size of centrilobular hepatocytes, a lower liver DNA content and an increased concentration of HCB in the adipose tissue, compared with the control and protein-deficient groups. Our data suggest that energy restriction increases liver response to HCB, while protein deficiency does not impair the hepatic reaction to small doses of HCB exposure.

Organochlorine compounds were dumped by chemical industries during the 1970s in many areas of the coastal plain of São Paulo state in Brazil. These dumps, located on hillsides and in valleys, in both rural and urban environments, are responsible for soil and water pollution. The objective of this work was to determine how the pollutants have spread in an area occupied by a spodosol-type soil mantle. The study combines soil morphological observations with soil and water analysis of hexachlorobenzene (HCB) and pentachlorophenol (PCP) in soil toposequences. The results indicate that the highest pollutant concentrations are observed near the dump site and that the compounds contamination is increasing. A map integrating topography and chemical concentrations was created to visualize the spatial distribution of HCB levels in the landscape. Physical and chemical analyses were performed to measure HCB and PCP levels in the soil. Soil water appears to act as a vector of HCB, probably through complexation with and dispersal of dissolved organic matter. The persistence of HCB at the studied site is most likely due to the low pH values in combination with a high content of organic matter. HCB was consistently found in higher concentrations than PCP. It is plausible that the cause of this difference is that PCP is degraded more easily under sunlight than HCB and that degradation of PCP under acid conditions leads to the formation of HCB. © 2003 Published by Elsevier B.V.
