BACKGROUND: The bacterial flagellum is the most important organelle of motility in bacteria and plays a key role in many bacterial lifestyles, including virulence. The flagellum also provides a paradigm of how hierarchical gene regulation, intricate protein-protein interactions and controlled protein secretion can result in the assembly of a complex multi-protein structure tightly orchestrated in time and space. As if to stress its importance, plants and animals produce receptors specifically dedicated to the recognition of flagella. Aside from motility, the flagellum also moonlights as an adhesion and has been adapted by humans as a tool for peptide display. Flagellar sequence variation constitutes a marker with widespread potential uses for studies of population genetics and phylogeny of bacterial species. RESULTS: We sequenced the complete flagellin gene (flaA) in 18 different species and subspecies of Aeromonas. Sequences ranged in size from 870 (A. allosaccharophila) to 921 nucleotides (A. popoffii). The multiple alignment displayed 924 sites, 66 of which presented alignment gaps. The phylogenetic tree revealed the existence of two groups of species exhibiting different FlaA flagellins (FlaA1 and FlaA2). Maximum likelihood models of codon substitution were used to analyze flaA sequences. Likelihood ratio tests suggested a low variation in selective pressure among lineages, with an omega ratio of less than 1 indicating the presence of purifying selection in almost all cases. Only one site under potential diversifying selection was identified (isoleucine in position 179). However, 17 amino acid positions were inferred as sites that are likely to be under positive selection using the branch-site model. Ancestral reconstruction revealed that these 17 amino acids were among the amino acid changes detected in the ancestral sequence. CONCLUSION: The models applied to our set of sequences allowed us to determine the possible evolutionary pathway followed by the flaA gene in Aeromonas, suggesting that this gene have probably been evolving independently in the two groups of Aeromonas species since the divergence of a distant common ancestor after one or several episodes of positive selection. Horizontal acquisition of DNA by bacteria dramatically increases genetic diversity and hence successful bacterial colonization of several niches, including the human host. A relevant issue is how this newly acquired DNA interacts and integrates in the regulatory networks of the bacterial cell. The global modulator H-NS targets both core genome and HGT genes and silences gene expression in response to external stimuli such as osmolarity and temperature. Here we provide evidence that H-NS discriminates and differentially modulates core and HGT DNA. As an example of this, plasmid R27-encoded H-NS protein has evolved to selectively silence HGT genes and does not interfere with core genome regulation. In turn, differential regulation of both gene lineages by resident chromosomal H-NS requires a helper protein: the Hha protein. Tight silencing of HGT DNA is accomplished by H-NS-Hha complexes. In contrast, core genes are modulated by H-NS homoligomers. Remarkably, the presence of Hha-like proteins is restricted to the Enterobacteriaceae. In addition, conjugative plasmids encoding H-NS variants have hitherto been isolated only from members of the family. Thus, the H-NS system in enteric bacteria presents unique evolutionary features. The capacity to selectively discriminate between core and HGT DNA may help to maintain horizontally transmitted DNA in silent form and may give these bacteria a competitive advantage in adapting to new environments, including host colonization.

The bacteriophage life cycle has an important role in Shiga toxin (Stx) expression. The induction of Shiga toxin-encoding phages (Stx phages) increases toxin production as a result of replication of the phage genome, and phage lysis of the host cell also provides a means of Stx toxin to exit the cell. Previous studies suggested that prophage induction might also occur in the absence of SOS response, independently of RecA.

We present here the first genome sequence of the Aeromonas diversa type strain (CECT 4254T). This strain was isolated from the leg wound of a patient in New Orleans (Louisiana, USA) and was originally described as Enteric Group 501 and distinguished from A. schubertii by DNADNA hybridization and phenotypical characterization.

Protein glycosylation had been considered as an eccentricity of a few bacteria. However, through advances in analytical methods and genome sequencing, it is now established that bacteria possess both N-linked and O-linked glycosylation pathways. Both glycosylation pathways can modify multiple proteins, flagellins from Archaea and Eubacteria being one of these. Flagella O-glycosylation has been demonstrated in many polar flagellins from Gram-negative bacteria and in only the Gram-positive genera Clostridium and Listeria. Furthermore, O-glycosylation has also been demonstrated in a limited number of lateral flagellins. In this work, we revised the current advances in flagellar glycosylation from Gram-negative bacteria, focusing on the structural diversity of glycans, the O-linked pathway and the biological function of flagella glycosylation.

Marine microorganisms, including Aeromonas, are a source of compounds for drug development that have generated great expectations in the last decades. Aeromonas infections produce septicaemia, and ulcerative and haemorrhagic diseases in fish. Among the pathogenic factors associated with Aeromonas, the lipopolysaccharides (LPS), a surface glyconconjugate unique to Gram-negative bacteria consisting of lipid A (lipid anchor of the molecule), core oligosaccharide and O-specific polysaccharide (O antigen), are key elicitors of innate immune responses. The chemical structure of these three parts has been characterized in Aeromonas. Based on the high variability of repeated units of O-polysaccharides, a total of 97 O-serogroups have been described in Aeromonas species, of which four of them (O:11; O:16; O:18 and O:34) account for more than 60% of the septicemia cases. The core of LPS is subdivided into two regions, the inner (highly conserved) and the outer core. The inner core of Aeromonas LPS is characterized by the presence of 3-deoxy-D-manno-oct-2-ulosonic (ketodeoxyoctonic) acid (Kdo) and L-glycero-D-manno-Heptoses (L,D-Hep), which are linked to the outer core, characterized by the presence of Glc, GlcN, Gal, and GalNAc (in Aeromonas salmonicida), D,D-Hep (in Aeromonas salmonicida), and L,D-Hep (in Aeromonas hydrophila). The biological relevance of these differences in the distal part of the outer core among these species has not been fully assessed to date. The inner core is attached to the lipid A, a highly conserved structure that confers endotoxic properties to the LPS when the molecule is released in blood from lysed bacteria, thus inducing a major systemic inflammatory response known as septic or endotoxic shock. In Aeromonas salmonicida subsp. salmonicida the Lipid A components contain three major lipid A molecules, differing in acylation patterns corresponding to tetra-, penta- and hexaacylated lipid A species and comprising of 4′-monophosphorylated β-2-amino-2-deoxy-D-glucopyranose-(1→6)-2-amino-2-deoxy-D-glucopyranose disaccharide. In the present review, we discuss the structure-activity relationships of Aeromonas LPS, focusing on its role in bacterial pathogenesis and its possible applications.

We experimentally identified the activities of six predicted heptosyltransferases in Actinobacillus pleuropneumoniae genome serotype 5b strain L20 and serotype 3 strain JL03. The initial identification was based on a bioinformatic analysis of the amino acid similarity between these putative heptosyltrasferases with others of known function from enteric bacteria and Aeromonas. The putative functions of all the Actinobacillus pleuropneumoniae heptosyltrasferases were determined by using surrogate LPS acceptor molecules from well-defined A. hydrophyla AH-3 and A. salmonicida A450 mutants. Our results show that heptosyltransferases APL_0981 and APJL_1001 are responsible for the transfer of the terminal outer core D-glycero-D-manno-heptose (D,D-Hep) residue although they are not currently included in the CAZY glycosyltransferase 9 family. The WahF heptosyltransferase group signature sequence [S(T/S)(GA)XXH] differs from the heptosyltransferases consensus signature sequence [D(TS)(GA)XXH], because of the substitution of D(261) for S(261), being unique.

Este proyecto se ha realizado en el Area Genética de la Universidad de León. Los componentes del equipo de trabajo son profesores de este Area, que imparten clases en las diversas materias de Genética en las titulaciones de Biología, Ciencias Ambientales, Ingeniero Agrónomo, Ingeniería Técnica Agrícola e Ingeniería Técnica Forestal. La docencia en Genética debe desarrollarse de modo equilibrado entre teoría y prácticas, pero determinadas prácticas exceden la duración de un curso académico, especialmente las de Genética de Poblaciones y las de Mejora; pero también en Genética Bacteriana determinadas prácticas requerirían un volumen de experimentos simultáneos difícil de realizar en la práctica con grupos numerosos de alumnos. Así se han desarrollado programas de simulación de estos tres ámbitos que han sido empleados durante el presente curso. Resultados preliminares indican que facilitan el aprendizaje de los alumnos en estos campos y por tanto ayudan a disminuir el fracaso escolar. Además potencian la eficacia de las prácticas en casos de especial dificultad. Se han elaborado tres programas en entorno Windows de fácil utilización, y unos manuales que permiten familiarizarse rápidamente con su manejo. El presente software está aún sin publicar.

Elaborar programas de ordenador para uso docente de Ciencias (Bioestadística, Genética, Física y Geología/ que sustituyan o complementen las sesiones tradicionales de laboratorio. Desarrollar una serie de clases prácticas basadas o apoyadas en ordenadores, acordes a las características de cada una de las asignaturas implicadas en el proyecto. El proyecto consta de programas para la enseñanza de la Bioestadística, Física, Genética y Geología. Las pautas son: han de constituir simulaciones dinámicas e interactivas de los fenómenos que describen, han de apoyarse fundamentalmente en las representaciones gráficas, a ser posible animadas, y han de limitar a su mínima expresión la aparición de texto en pantalla. Los programas suponen un cierto conocimiento por el alumno del tema que se aborda, pero no exigen en absoluto nociones acerca del funcionamiento de ordenadores. Es idea común que el ordenador ha de ser limitado a una función complementaria. 1. Se han elaborado dos programas para la enseñanza de la Bioestadística: Uno de simulación de datos para procesado con variables cuantitativas o cualitativas y otro de cálculo que comprende el agrupamiento de datos, cálculo de estadísticos y la elaboración de pruebas estadísticas. 2. Los programas para la enseñanza de la Física tratan de conseguir que el ordenador se convierta en una forma de enseñanza programada. Toma como ejemplo la práctica denominada 'Kepler'. 3. Los dos programas para la enseñanza de la Genética bacteriana son la conjugacion bacteriana y la prueba de la fluctuación de Luria y Delbruck. Ambos son autónomos y no requieren la presencia de un profesor. 4. Los programas para la enseñanza de la Geología son: cálculo de constantes reticulares en diagramas de polvo de rayos X dirigido a la investigación pura; cálculo de índices de Miller en diagramas de polvo de rayos X con fines docentes; cálculo de ángulos entre caras cristalinas, es útil en sistemas de baja simetría; uso del ordenador en mineralogía descriptiva, que ayuda al alumno con las relaciones zonales entre las caras cristalinas y la proyección estereográfica. Ordenadores Olivetti M20. Los resultados obtenidos en la aplicación de las prácticas asistidas por ordenador dentro del área de Bioestadística son alentadores, pero se aprecia la falta de programas, que faciliten al alumno la comunicación con el ordenador. Hay una cierta tendencia a no seguir el procedimiento de trabajo, achacable a una posible rutina en el uso del ordenador. El alumno asiste a las prácticas sin haber realizado cálculos manuales. En general creen que unos programas bien estructurados, ejecutables en modo interactivo en máquinas poderosas y fáciles de usar, pueden constituir un excelente estímulo de actitudes activas para el alumno. Se han desarrollado con fecundidad las iniciativas interdisciplinares. El papel del profesor como asesor cobra un valor fundamental. Los programas han explotado adecuadamente la potencialidad específica del ordenador que le convierte en un instrumento diferente de los clásicamente utilizados en la enseñanza.

Some non-pathogenic trypanosomatids maintain a mutualistic relationship with a betaproteobacterium of the Alcaligenaceae family. Intensive nutritional exchanges have been reported between the two partners, indicating that these protozoa are excellent biological models to study metabolic co-evolution. We previously sequenced and herein investigate the entire genomes of five trypanosomatids which harbor a symbiotic bacterium (SHTs for Symbiont-Haboring Trypanosomatids) and the respective bacteria (TPEs for Trypanosomatid Proteobacterial Endosymbiont), as well as two trypanosomatids without symbionts (RTs for Regular Trypanosomatids), for the presence of genes of the classical pathways for vitamin biosynthesis. Our data show that genes for the biosynthetic pathways of thiamine, biotin, and nicotinic acid are absent from all trypanosomatid genomes. This is in agreement with the absolute growth requirement for these vitamins in all protozoa of the family. Also absent from the genomes of RTs are the genes for the synthesis of pantothenic acid, folic acid, riboflavin, and vitamin B6. This is also in agreement with the available data showing that RTs are auxotrophic for these essential vitamins. On the other hand, SHTs are autotrophic for such vitamins. Indeed, all the genes of the corresponding biosynthetic pathways were identified, most of them in the symbiont genomes, while a few genes, mostly of eukaryotic origin, were found in the host genomes. The only exceptions to the latter are: the gene coding for the enzyme ketopantoate reductase (EC:1.1.1.169) which is related instead to the Firmicutes bacteria; and two other genes, one involved in the salvage pathway of pantothenic acid and the other in the synthesis of ubiquinone, that are related to Gammaproteobacteria. Their presence in trypanosomatids may result from lateral gene transfer. Taken together, our results reinforce the idea that the low nutritional requirement of SHTs is associated with the presence of the symbiotic bacterium, which contains most genes for vitamin production.

Objetivo: presentar el estado del arte de las investigaciones que, hasta el momento, relacionan el polimorfismo genético del paciente con la evolución de la sepsis, como herramienta diagnóstica y un nuevo enfoque terapéutico de esta condición. Los conceptos actuales basados en investigaciones sostienen que el polimorfismo genético del individuo es relevante en la evolución de la enfermedad y en la respuesta efectiva al tratamiento del paciente en estado crítico, en especial con sepsis bacteriana y choque séptico. Materiales y métodos: se revisó literatura indexada que relaciona los factores genéticos con la evolución de algunas enfermedades del paciente en estado crítico. Resultados: las características particulares de la enfermedad estarían influenciadas por el acervo genético del paciente, condicionando en gran medida la respuesta patofisiológica. Se ha evidenciado la susceptibilidad genética de algunos individuos a desarrollar infección; estos individuos con un tratamiento similar no evolucionan de igual forma, desencadenándose una sepsis bacteriana grave y choque séptico. El polimorfismo en los genes que codifican por el factor de necrosis tumoral -α (TNF-α) las interlucinas- 1 (IL-1), IL-6, IL-10, el factor soluble CD-14, los receptores similares a Toll y el inhibidor tipo 1 del activador del plasminógeno estaría asociado con el desarrollo de sepsis grave y choque séptico, en particular las mutaciones TNF-α 308 G/A, PAI-1 4G/4G, IL-6 174 G/C. Conclusiones: el conocimiento de la susceptibilidad genética, los factores de riesgo y el buen funcionamiento del sistema inmune de cada persona ayudan a reducir y compensar las complicaciones de la sepsis bacteriana. Es claro que el tratamiento oportuno individualizado en los pacientes con sepsis se asocia con disminución de la mortalidad y con reducción en el deterioro de la respuesta inflamatoria.

Um dos principais entraves em cultivar o tomateiro no trópico úmido está relacionado à presença do patógeno Ralstonia solanacearum, pois este é de ocorrência natural nos solos da região Amazônica. Foram estimados os níveis de resistência genética à bactéria R. solanacearum de linhagens de tomate do grupo Yoshimatsu e a produtividade em comparação a outras variedades disponíveis no mercado. O ensaio foi realizado em uma unidade de produção de um agricultor familiar na qual foram avaliados 8 genótipos de tomate: Santa Cruz Kada, Yoshimatsu 4-11, (L1-2002), (L2-2002), (L3-2002), (L4-2002), (L5-2002) e (L6-2002). Foram avaliados o índice de sanidade (IS) e a produção total dos frutos. A cultivar Santa Cruz Kada apresentou maiores níveis de incidência ao patógeno em relação às demais cultivares. No entanto, ao avaliar o parâmetro de produtividade, a cultivar (L6-2006) foi superior às demais.
