O presente trabalho teve como objetivos: determinar o método mais adequado para detecção e identificação de fungos associados às sementes (cariopses) de cana-de-açúcar; caracterizar os fungos associados; verificar as incidências nessas sementes e relacionar a incidência fúngica nessas sementes com o ambiente onde foram produzidas. Para detecção do método mais adequado, foram comparados dois substratos, em placas de Petri: agar-água com papel quadriculado e papel de filtro. Utilizaram-se placas de Petri de plástico e de vidro do tipo pirex, para verificar a influência do recipiente. Também foram comparados dois regimes de luz (12 h luz branca fluorescente/12 h escuro e escuro contínuo). As sementes foram mantidas durante sete dias sob temperatura constante de 28 2ºC, quando se procedeu à avaliação. Os requisitos para comparação dos métodos foram sensibilidade, economicidade e praticidade. A partir do método determinado como o mais adequado, foi realizada análise sanitária de 29 cruzamentos dos anos de 2002, 2003 e 2004, caracterizando os fungos associados e verificando as incidências. Posteriormente, compararam-se estas incidências com as condições ambientes, de temperatura e umidade relativa, em que as sementes foram produzidas no programa de melhoramento genético. O método considerado mais adequado, de acordo com os parâmetros analisados, foi o do papel de filtro em placa de Petri de plástico e incubação sob regime de luz (12 h luz branca fluorescente/12 h escuro). Os fungos detectados foram: Alternaria alternata; Aspergillus sp.; Bipolaris sacchari; três grupos morfológicos distintos pertencentes ao gênero Bipolaris; dois grupos morfológicos de Cladosporium; Colletotrichum sp.; três grupos morfológicos de Curvularia; Epicoccum sp.; Fusarium verticillioides; Fusarium semitectum; Leptosphaerulina sp.; Nigrospora sp.; Penicillium sp.; Periconia sp.; Phoma herbarum; Rhizopus sp. e Trichoderma sp. Os mais freqüentemente encontrados foram: Bipolaris sacchari; Bipolaris spp.; Cladosporium spp.; Curvularia spp.; Fusarium verticillioides; Fusarium semitectum e Phoma herbarum. Quando se comparou a porcentagem de incidência dos diversos fungos com o ambiente de produção das sementes, observou-se que não houve relação de temperatura e umidade relativa com incidência fúngica. Supõe-se que as variações de incidência possam estar relacionadas com diferentes fontes de inóculo nos locais de cruzamento ou características genéticas das sementes.

A identificação de espécies fúngicas presentes em grãos e frutos de amendoim (Arachis hypogaea), bem como a caracterização cultural das espécies de Aspergillus detectadas, é um importante passo para a prevenção da presença de micotoxinas no substrato, garantindo a qualidade do produto, tanto para a comercialização in natura quanto já processado. A partir de grãos e frutos de amendoim comercializados, in natura ou processados, no estado de Alagoas foram isolados os fungos Aspergillus flavus, A. niger, A. ochraceus, A. parasiticus, Fusarium verticillioides (= F. moniliforme), F. equiseti, Rhizopus stolonifer, Botrytis cinerea, Penicillium italicum e Colletotrichum sp. De um modo geral, os grãos processados apresentaram menor incidência de fungos, diferindo estatisticamente dos grãos in natura, com destaque para as espécies A. flavus, A. parasiticus, F. verticillioides e F. equiseti. As espécies de Aspergillus foram identificadas com base nas características culturais, exibidas em meio de Czapek-ágar, e morfológicas, através de microscópio ótico. No oitavo dia de crescimento em meio de BDA, cada espécie apresentou diferenças quanto à taxa de crescimento durante o período de incubação, de acordo com a procedência das amostras dos grãos ou frutos.

O trabalho avaliou a interação entre temperaturas do solo (13ºC, 15ºC, 17ºC e 19ºC), profundidades de semeadura (2,5; 5,0 e 7,5 cm) e tratamento de semente com fungicida (captan + tiabendazole) na emergência de plantas de milho. Utilizaram-se sementes do híbrido AS 1565 com 22% de incidência de Fusarium verticillioides e solo de lavoura naturalmente infestado. O ensaio foi conduzido em câmaras climatizadas por 25 dias, com contagem diária da emergência. Houve diferença significativa entre sementes tratadas e não tratadas, sendo que quanto menor a temperatura do solo e maior a profundidade de semeadura, maior a resposta ao tratamento de sementes na emergência de plantas. A estabilidade de emergência ocorreu aos 23, 18, 12 e 12 dias após a semeadura respectivamente para 13ºC, 15ºC, 17ºC e 19ºC, mostrando redução na velocidade de emergência nas temperaturas baixas.

O trabalho teve como objetivo verificar os efeitos do teor de água e da temperatura do ambiente de armazenamento na incidência de fungos e na germinação de sementes de açaí (Euterpe oleracea Mart.). Foram utilizadas sementes da cultivar BRS Pará, com teores de água de 43%, 37%, 30%, 26%, 21% e 12%, que foram acondicionadas em sacos de polietileno, armazenadas sob temperaturas de 20 ºC, 15 ºC e 10 ºC, durante 360 dias e submetidas a avaliações bimensais do teor de água, da germinação e da incidência de Penicillium sp., Aspergillus sp., Fusarium solani e Fusarium verticillioides. Foi adotado o delineamento experimental inteiramente casualizado com quatro repetições para o teste de germinação e cinco repetições para os testes de sanidade. Foi observada maior ocorrência de Penicillium sp., e Aspergillus sp em sementes com teores de água abaixo de 30% e mantidas nas temperaturas de 15 ºC e 20 ºC. Enquanto, as maiores incidências de fungos do gênero Fusarium foram encontradas em sementes armazenadas com graus de umidade de 43% e 37% e mantidas em temperatura de 10 ºC.

The aim of this study was to investigate the effects of nutrients (nitrogen, zinc and boron) on fungal growth and fumonisins production in corn samples obtained at the beginning of grain formation and at harvest. Three nitrogen doses were applied to the corn plants through soil in combination with three zinc doses and two boron doses during sowing. Mycological analysis of grains, using Dichloran Rose-Bengal Chloramphenicol Agar, collected at the beginning of formation demonstrated a fungal population predominantly of yeasts. Analysis of freshly harvested corn revealed a higher frequency of Penicillium spp. (72%) and F verticillioides (27%). High Performance Liquid Chromatography analysis revealed that 100% of grains were contaminated with fumonisins B, at levels ranging from 0.3 to 24.3 mg/kg and 93% contaminated with fumonisin B(2) at levels ranging from 0.05 to 5.42 mg/kg. Nitrogen (50 kg/ha) in combination with boron (0.5 kg/ha) resulted in an increased fumonisin B2 production. (c) 2007 Elsevier Ltd. All rights reserved.

BACKGROUND: The aim of this study was to characterise the mycoflora and the presence of fumonisin in sorghum grains, correlating the results with the environment and abiotic factors. RESULTS: Fifty samples (five collections of ten samples each) of sorghum were analysed. All samples were found to be contaminated with fungi, with higher frequencies of Cladosporium spp. (61.8%) and Helminthosporium spp. (33.4%). Fusarium verticillioides was isolated from 15.1% of the samples, with 38% of them being contaminated with fumonisin B(1) (FB(1)) at levels ranging from 50 to 368.78 ng g(-1). Regarding abiotic factors, temperature, water activity and rainfall showed a positive correlation with the frequency of F. verticillioides and FB(1) production. There was a significant positive correlation between relative air humidity and FB(1) production. The results obtained from sexual crosses between standard F mating tester strains and the isolated strains confirmed that the strains isolated were F. verticillioides. CONCLUSION: It can be concluded that the decrease in F. verticillioides and fumonisin contamination occurred owing to atypical climatic factors during the period of sorghum cultivation, when there was any occurrence of rain and the level of water activity of grains did not reach 0.58. The effect of inoculation of Aspergillus flavus, Fusarium verticillioides, and Penicillium sp. in Dystrophic Red Latosol (DRL) and Eutroferric Red Latosol (ERL) soils with or without glucose on the total carbohydrate content and the dehydrogenase and amylase activities was studied. The fungal growth and spore production in culture medium with and without glucose were also evaluated. A completely randomized design with factorial arrangement was used. The addition of glucose in the culture medium increased the growth rate of A. flavus and Penicillium sp. but not of F. verticillioides. The number of spores increased 1.2 for F. verticillioides and 8.2 times for A. flavus in the medium with glucose, but was reduced 3.5 times for Penicillium sp. The total carbohydrates contents reduced significantly according to first and second degree equations. The consumption of total carbohydrates by A. flavus and Penicillium sp. was higher than the control or soil inoculated with F. verticillioides. The addition of glucose to soils benefited the use of carbohydrates, probably due to the stimulation of fungal growth. Dehydrogenase activity increased between 1.5 to 1.8 times (p <0.05) in soils with glucose and inoculated with the fungi (except F. verticillioides), in relation to soil without glucose. Amylase activity increased 1.3 to 1.5 times due to the addition of glucose in the soil. Increased amylase activity was observed in the DRL soil with glucose and inoculated with A. flavus and Penicillium sp. when compared to control.

Background: Fumonisins produced by Fusarium verticillioides are among the most important medical mycotoxins known. The intake of concentrate based on corn and corn by-products contaminated with fumonisins can cause severe poisoning in horses. The injuries are observed mainly in the white matter of the brain, and the disease is known as Equine Leukoencephalomalacia (ELEM). This study aims to describe and discuss the epidemiological, clinical and diagnostic aspects of an outbreak of ELEM occurred in three farms in the municipalities of Canarana and Agua Boa, in the eastern region of Mato Grosso, Brazil.Materials, Methods & Results: The outbreak occurred between May and August 2010. The disease affected six horses and four mules of different ages and sex. Clinical examination was only possible in animals with chronic evolution of the disease. All the affected animals showed neurological clinical signs such as ataxia and recumbency, which progressed to death or sudden death. Histopathological analysis showed foci of necrosis that predominantly affected the white matter, and the presence of gitter cells. Degenerative lesions were observed in the liver of the animals. Mortality rate ranged from 12.5 to 71%, and lethality reached 100%. The cases were preceded by sudden drops in the weather temperature. Fumonisins levels of 6.6 ppm were detected in the feed of the animals.Discussion: The presumptive diagnosis of leukoencephalomalacia was consistent and based on clinical and epidemiological studies. However, the definitive diagnosis was based upon the histological features of the brain including the presence of extensive areas of malacia. Moreover, the animals were being fed with corn or corn by-products contaminated with fumonisins levels considered to be toxic to equids. The mortality and lethality rates are in agreement with outbreaks described in previous studies. The animals showed neurological signs as the predominant clinical manifestation, with gait ataxia followed by recumbency, prostration and death between 24 h and 29 days. Similarly to other reports, the disease was more frequent in adult animals, which succumbed in 24-48 h. Conversely, the evolution of the disease in young animals was of 10 to 29 days. Sudden death was more prevalent in the mules. Previous studies have shown a predominance of cerebral and brainstem lesions in horses, whereas in mules the clinical signs are related to brainstem lesions. Corn and corn by-products are commonly used as energy supplementation to horses in the southern of Mato Grosso state, but outbreaks of the disease are uncommon. This may be influenced by the prevalence of hot climate conditions, which does not favor the production of toxin by the fungus. The atypical low weather temperatures (9-11 degrees C) observed prior to the outbreak could have contributed to the mycotoxin production by F. verticillioides, which requires temperatures between 8-12 degrees C to produce toxins. However, the disease in the region may be underestimated, considering that the practice of necropsies is not common among field technicians, mainly in the occurrence of sporadic deaths among horses intended for work. Preventive measures include avoiding the use of corn and corn by-products for horses after periods of sudden drops in temperature in the region. Furthermore, clinical and epidemiological surveys and post-mortem and histopathological analyses are undoubtedly important for appropriate differential diagnosis, especially in equids with neurological signs.

Endophytic fungi are considered a rich source of active compounds resulting from their secondary metabolism. Fungi from marine environment grow in a habitat with unique conditions that can contribute to the activation of metabolic pathways of synthesis of different unknown molecules. The production of these compounds may support the adaptation and survival of the fungi in the marine ecosystem. Mangroves are ecosystems situated between land and sea. They are frequently found in tropical and subtropical areas and enclose approximately 18.1 million hectares of the planet. The great biodiversity found in these ecosystems shows the importance of researching them, including studies regarding new compounds derived from the endophytic fungi that inhabit these ecosystems. 3-hydroxypropionic acid (3-HPA) has been isolated from the mangrove endophytic fungus Diaporthe phaseolorum, which was obtained from branches of Laguncularia racemosa. The structure of this compound was elucidated by spectroscopic methods, mainly 1D and 2D NMR. In bioassays, 3-HPA showed antimicrobial activities against both Staphylococcus aureus and Salmonella typhi. The structure of this antibiotic was modified by the chemical reaction of Fischer-Speier esterification to evaluate the biologic activity of its chemical analog. The esterified product, 3-hydroxypropanoic ethyl ester, did not exhibit antibiotic activity, suggesting that the free carboxylic acid group is important to the pharmacological activity. The antibiotic-producing strain was identified with internal transcribed spacer sequence data. To the best of our knowledge, this is the first report of antibacterial activity by 3-HPA against the growth of medically important pathogens.

This study investigated the ability of weevils to transmit Aspergillus flavus and Fusarium verticillioides fungal spores and the consequent production of mycotoxins. For this purpose, corn grain samples were stored in flasks connected to a hose to form a closed system (flasks A and B). Flasks A were inoculated with the following groups: group 1 (corn + weevil); group 2 (corn + A. flavus); group 3 (corn + A. flavus + weevil); group 4 (corn + F. verticillioides); group 5 (corn + E verticillioides + weevil), and group 6 (corn + A. flavus + E verticillioides + weevil). Flasks B contained sterile grains. The samples were incubated for 10, 20 and 30 days, posteriorly, weight, water activity, mycoflora, aflatoxins and fumonisins. The corn grain samples were also submitted to scanning electron microscopy. Our results showed that weevils could enhance corn grains contamination by these fungi, hence, could increase mycotoxins production. These findings demonstrate the importance of weevils as fungal vectors and the need for good manipulation and storage practices of grains. (C) 2012 Elsevier Ltd. All rights reserved.

Background: Sugarcane is one of the most important crops in Brazil, mainly because of its use in biofuel production. Recent studies have sought to determine the role of sugarcane endophytic microbial diversity in microorganism-plant interactions, and their biotechnological potential. Epicoccum nigrum is an important sugarcane endophytic fungus that has been associated with the biological control of phytopathogens, and the production of secondary metabolites. In spite of several studies carried out to define the better conditions to use E. nigrum in different crops, little is known about the establishment of an endophytic interaction, and its potential effects on plant physiology. Methodology/Principal Findings: We report an approach based on inoculation followed by re-isolation, molecular monitoring, microscopic analysis, plant growth responses to fungal colonization, and antimicrobial activity tests to study the basic aspects of the E. nigrum endophytic interaction with sugarcane, and the effects of colonization on plant physiology. The results indicate that E. nigrum was capable of increasing the root system biomass and producing compounds that inhibit the in vitro growth of sugarcane pathogens Fusarium verticillioides, Colletotrichum falcatum, Ceratocystis paradoxa, and Xanthomomas albilineans. In addition, E. nigrum preferentially colonizes the sugarcane surface and, occasionally, the endophytic environment. Conclusions/Significance: Our work demonstrates that E. nigrum has great potential for sugarcane crop application because it is capable of increasing the root system biomass and controlling pathogens. The study of the basic aspects of the interaction of E. nigrum with sugarcane demonstrated the facultative endophytism of E. nigrum and its preference for the phylloplane environment, which should be considered in future studies of biocontrol using this species. In addition, this work contributes to the knowledge of the interaction of this ubiquitous endophyte with the host plant, and also to a better use of microbial endophytes in agriculture.

El presente estudio evalúa el efecto que 6 diferentes géneros hongos aislados a partir de semillas de 54 diferentes cultivares de cardo y sus extractos acuosos tienen sobre la germinación y nascencia de las semillas. Se han realizado pruebas de patogenicidad con dos aislados de cada uno de los seis géneros de mayor frecuencia del inventario (Aspergillus, Penicillium, Fusarium, Rhizopus, Cladosporium y Alternaria), así como de los extractos producidos tras 3, 7 y 11 días de incubación de los micetos. Los resultados de las inoculaciones con los micetos muestran efectos negativos sobre los porcentajes de germinación, con reducciones en la germinación que fueron máximas tras las inoculaciones con Rhizopus stolonifer (29% de disminución) y Fusarium verticillioides (23%). Los porcentajes de emergencia disminuyen tras duplicar la concentración del inóculo, aumentando además drásticamente el número de plántulas dañadas sobre el total de las emergidas. En el significativo caso de la inoculación con Cladosporium la duplicación del inóculo disminuyó la germinación hasta en un 31% respecto al testigo. Las plántulas emergidas tras las inoculaciones con los extractos obtenidos a partir de cultivos líquidos de los hongos ensayados presentaban los mismos síntomas de atrofias y daños sobre raíz y coleóptilo que los descritos para cada hongo. Los extractos acuosos de los géneros estudiados disminuyen también la germinación. Los resultados nos muestran la diferente capacidad parasitaria de cada una de las especies estudiadas apreciándose además diferencias según los diferentes periodos de agitación de los hongos y permiten asegurar que la producción de toxinas está regulada por el hongo, y que no aumenta linealmente con el crecimiento miceliar.

O fungo Fusarium moniliforme (sin. F. verticillioides) é o principal patógeno associado a sementes de milho (Zea mays) no Brasil. O fungo pode ser introduzido pelas sementes em áreas isentas, causando deterioração de sementes, morte de plântulas, podridão radicular, do colmo e da espiga. Com o objetivo de quantificar sua transmissão para as plântulas de milho, foi conduzido um experimento em casa de vegetação utilizando-se sementes do híbrido AG-9020 sem tratamento com fungicida e com incidência natural média de 46%. Realizou-se a semeadura em 04/09/2001, em caixas de madeira contendo substrato isento do fungo constituído pela mistura de solo de horizonte "B", areia grossa e vermiculita na proporção de 3:1:1. As avaliações do número de plantas emersas e a transmissão do fungo foram realizadas aos 15, 30 e 45 dias após a semeadura. Em cada época, coletaram-se, ao acaso, 200 plântulas. As plantas foram lavadas e levadas ao laboratório. De cada planta destacou-se o tegumento remanescente da semente, a raiz primária, o entrenó subcoronal, o coleóptilo e a base das folhas. Após, realizou-se a assepsia do material em hipoclorito de sódio (1%) por 3 min, seguido de lavagem com água esterilizada e plaqueamento em meio ¼ de batata-sacarose-ágar. Os resultados indicaram que o fungo foi detectado em todos os órgãos da plântula isolados e nas três épocas de avaliação. O percentual médio de transmissão foi de 46,1; 34,9; 23,6; 7,2 e 14,6%, respectivamente, para o tegumento remanescente da semente, raiz primária, entrenó subcoronal, coleóptilo e base das folhas.

Avaliou-se a reação das cultivares de soja, IAS-5, FT-Cometa, CAC-1, Monarca e MG/BR 46 (Conquista), consideradas resistentes, e de FT-Estrela e FT-Cristalina, suscetíveis a F. tucumaniae, por dois métodos. Foram inoculadas plântulas e folhas destacadas de soja através do método de grãos de sorgo e grãos de aveia, respectivamente. As avaliações de severidade da doença foram efetuadas semanalmente utilizando-se escalas de notas tanto para sintomas observados na parte aérea das plântulas como para o sistema radicular, e também através da porcentagem de plântulas mortas. Avaliou-se também a altura de plantas e os comprimentos das lesões externa e interna da haste. A relação das cultivares em ordem crescente de porcentagem de plantas mortas foi CAC-1 (47%), Monarca (60%), MG/BR 46 (Conquista) (61%), IAS-5 (64%), FT-Cristalina (84%), FT-Cometa (86%) e FT-Estrela (90%). Em folhas destacadas, inicialmente, o patógeno infectou tecidos das regiões próximas ao pecíolo, porém este método não foi adequado na caracterização dos materiais, uma vez que ocorre senescência precoce das folhas.
