Ro09-0198 is a tetracyclic polypeptide of 19 amino acids that recognizes strictly the structure of phosphatidylethanolamine (PE) and forms a tight equimolar complex with PE on biological membranes. Using the cyclic peptide coupled with fluorescence-labeled streptavidin, we have analyzed the cell surface localization of PE in dividing Chinese hamster ovary cells. We found that PE was exposed on the cell surface specifically at the cleavage furrow during the late telophase of cytokinesis. PE was exposed on the cell surface only during the late telophase and no alteration in the distribution of the plasma membrane-bound cyclic peptide was observed during the cytokinesis, suggesting that the surface exposure of PE reflects the enhanced scrambling of PE at the cleavage furrow. Furthermore, cell surface immobilization of PE induced by adding the cyclic peptide coupled with streptavidin to prometaphase cells effectively blocked the cytokinesis at late telophase. The peptide-streptavidin complex treatment had no effect on furrowing, rearrangement of microtubules, and nuclear reconstitution, but specifically inhibited both actin filament disassembly at the cleavage furrow and subsequent membrane fusion. These results suggest that the redistribution of the plasma membrane phospholipids is a crucial step for cytokinesis and the cell surface PE may play a pivotal role in mediating a coordinate movement between the contractile ring and plasma membrane to achieve successful cell division.

While astral microtubules are believed to be primarily responsible for the stimulation of cytokinesis in Echinoderm embryos, it has been suggested that a signal emanating from the chromosomal region and mediated by the interzonal microtubules stimulates cytokinesis in cultured mammalian cells. To test this hypothesis, we examined cytokinesis in normal rat kidney cells treated with an inhibitor of topoisomerase II, (+)-1,2-bis(3,5-dioxopiperaz-inyl-1-yl)propane, which prevents the separation of sister chromatids and the formation of a spindle interzone. The majority of treated cells showed various degrees of abnormality in cytokinesis. Furrows frequently deviated from the equatorial plane, twisting daughter cells into irregular shapes. Some cells developed furrows in regions outside the equator or far away from the spindle. In addition, F-actin and myosin II accumulated at the lateral ingressing margins but did not form a continuous band along the equator as in control cells. Imaging of microinjected 5- (and 6-) carboxymtetramethylrhodamine-tubulin revealed that a unique set of microtubules projected out from the chromosomal vicinity upon anaphase onset. These microtubules emanated toward the lateral cortex, where they delineated sites of microtubule bundle formation, cortical ingression, and F-actin and myosin II accumulation. As centrosome integrity and astral microtubules appeared unperturbed by (+)-1,2-bis(3,5-dioxopiperaz-inyl-1-yl)propane treatment, the present observations cannot be easily explained by the conventional model involving astral microtubules. We suggest that in cultured epithelial cells the organization of the chromosomes dictates the organization of midzone microtubules, which in turn determines and maintains the cleavage activity.

During Drosophila development, nuclear and cell divisions are coordinated in response to developmental signals. In yeast and mammalian cells, signals that control cell division regulate the activity of cyclin-dependent kinases (Cdks) through proteins such as cyclins that interact with the Cdks. Here we describe two Drosophila cyclins identified from a set of Cdk-interacting proteins. One, cyclin J, is of a distinctive sequence type; its exclusive maternal expression pattern suggests that it may regulate oogenesis or the early nuclear divisions of embryogenesis. The other belongs to the D class of cyclins, previously identified in mammalian cells. We show that Drosophila cyclin D is expressed in early embryos and in imaginal disc cells in a pattern that anticipates cell divisions. Expression in the developing eye disc at the anterior edge of the morphogenetic furrow suggests that cyclin D acts early, prior to cyclin E, in inducing G1-arrested cells to enter S phase. Our results also suggest that, although cyclin D may be necessary, its expression alone is not sufficient to initiate the events leading to S phase.

Rare earth element concentrations in ferromanganese concretions sampled from Slupsk Furrow in the Polish Exclusive Economic Zone are similar to those of concretions from the Gulf of Bothnia. The lack of positive Ce anomalies in the concretions from Slupsk Furrow indicates that they are formed under less oxidizing conditions than spheroidal concretions from the Gulf of Bothnia. Mossbauer studies indicate that poorly crystalline lepidocrosite is the principal iron oxyhydroxide mineral present in these concretions.

During the "Polarstern"-expeditions ARK-IX/4 (1993) and ARK-XI/1 (1995), organised by the Alfred Wegener Institute (AWI), acoustic subbottom profiles (Parasound) have been collected in the Laptev Sea Shelf, Siberia. These data have been interpreted as an indicator of ice scours frequency and off-shore permafrost patterns. An additional acoustic profile data-base was available by the results of the expedition of the Federal Institute for Geosciences and Natural Resources (BGR) of the year 1994. The area of the expedition was located closer to the shelf, therefore supports a better understanding of ice scours frequency in shallower marine environments. The data-file consists of a 2930 km Parasound-traverse and has been subdivided into 586 working profiles. They are characterised by their location, number of ice scours, interpreted patterns of reflection and their extension and morphology. The data have been evaluated statistically and graphically and were presented in a map. Different patterns of sea floor reflection were established by different environments, outer influences (e.g. size of the icebergs, direction of the drift of icebergs) and the climatic history of the region. In the north-westerly region of the Laptev Sea at the continental slope of Severnaya Zemlya the sea floor in shallower depths has been ploughed intensely by recent icebergs. In some regions (40-60m), as an effect of intensely ploughing, the sea floor is hardly defined in acoustic profiles come along with relocation of marine deposits. Glacial diamiet deposits prevented the development of deep scours. Up to 355m deeper scours result from lower sea levels. The marginal north-easterly region of the Laptev Sea is characterised exclusively by this type of scour. Morphology and depth of these scours can be compared with those of the westerly Vilkitsky-Street so that similar conditions of development may be expected. Both, the north-easterly Laptev Sea and the Vilkitsky-Street, are not dominated by patterns ofrecent icebergs. In contrary the shelf-regions north-easterly ofthe Taimyr peninsula and north-westerly of the New Siberian Islands have been modified evidently by recent icebergs, which drifted with prevalent currents anticlockwise along the shelf edge of the Laptev Sea and cause the deepest scours of the whole region. The off-shore permafrost at the inner shelf regions has an important influence on the scours intensity. The permafrost layer can be recognised by the maximum depth of ice scours. It is represented by a Parasound reflector that can be made up for distances. The age of the ice scours cannot be determined absolutely by Parasound data but a relative order can be estimated whenever two scours are situated close to each other. When the Parasound-traverse ofthe expedition ARK-IX/4 (1993) (77°24'N 133°30'E-77°30'N 133°40'E) was repeated partially in expedition ARK-XI/l (1995) the ice scours of 1993 remained unchanged and uneroded and no new ice scours had been detected. It can be concluded that scours persist for a long time in the Laptev Sea, though after all with an average of 3 ice scours per kilometer there are not many at all in the Laptev Sea.

The concentration of Zn, Cu, Pb, Cd, Ni, Co, Ag, Mn, Fe, Ca, Mg, K and Na in molluscs Macoma balthica, Mya arenaria, Cardium glaucum, Mytilus edulis and Astarte borealis from the southern Baltic was determined. The surface sediments and ferromanganese concretions associated with the molluscs were also analysed for concentration of these metals. Species- and region-dependent differences in the metal levels of the organisms were observed. The properties of molluscs analysed which have a tendency toward elevated biological tolerance of selected trace metals were specified. The interelement relationship between metal concentrations in the soft tissue and the shell was estimated and was discussed.

Scarcity of freshwater due to recurrent drought threatens the sustainable crop production in semi-arid regions of Ethiopia. Deficit irrigation is thought to be one of the promising strategies to increase water use efficiency (WUE) under scarce water resources. A study was carried out to investigate the effect of alternate furrow irrigation (AFI), deficit irrigation (DI) and full irrigation (FI) on marketable fruit yield, WUE and physio-chemical quality of four fresh-market tomato cultivars (Fetan, Chali, Cochoro and ARP Tomato d2) in 2013 and 2014. The results showed that marketable yield, numbers of fruits per plant and fruit size were not significantly affected by AFI and DI irrigations. WUE under AFI and DI increased by 36.7% and 26.1%, respectively with close to 30% irrigation water savings achieved. A different response of cultivars to irrigation treatments was found for marketable yield, number of fruits and fruit size, WUE, total soluble solids (TSS) of the fruit juice, titratable acids (TA) and skin thickness. Cochoro and Fetan performed well under both deficit irrigation treatments exhibited by bigger fruit size which led to higher WUE. ARP Tomato d2 showed good yields under well-watered conditions. Chali had consistently lower marketable fruit yield and WUE. TSS and TA tended to increase under deficit irrigation; however, the overall variations were more explained by irrigation treatments than by cultivars. It was shown that AFI is a suitable deficit irrigation practice to increase fresh yield, WUE and quality of tomato in areas with low water availability. However, AFI requires suitable cultivars in order to exploit its water saving potential.

Understanding the variation in physiological response to deficit irrigation together with better knowledge on physiological characteristics of different genotypes that contribute to drought adaptation mechanisms would be helpful in transferring different irrigation technologies to farmers. A field experiment was carried to investigate the physiological response of four tomato cultivars (Fetan, Chali, Cochoro and ARP Tomato d2) to moderate water deficit induced by alternate furrow irrigation (AFI) and deficit irrigation (DI) under semi-arid condition of Ethiopia during 2013 and 2014. The study also aimed at identifying physiological attributes to the fruit yield of tomato under different deficit irrigation techniques. A factorial combination of irrigation treatments and cultivar were arranged in a complete randomized design with three replicates. Results showed that stomatal conductance (g_s) was significantly reduced while photosynthetic performance measured as chlorophyll fluorescence (Fv’/Fm’), relative water content (RWC) and leaf ash content remained unaffected under deficit irrigations. Significant differences among cultivars were found for water use efficiency (WUE), g_s, chlorophyll content (Chl_SPAD), normal difference vegetation index (NDVI), leaf ash content and fruit growth rate. However, cultivar differences in WUE were more accounted for by the regulation of g_s, therefore, g_s could be useful for breeders for screening large numbers of genotypes with higher WUE under deficit irrigation condition. The study result also demonstrated that cultivar with traits that contribute to achieve higher yields under deficit irrigation strategies has the potential to increase WUE.

Relationships among floral biology, floral micromorphology and pollinator behaviour in bird-pollinated orchids are important issues to understand the evolution of the huge flower diversity within Orchidaceae. We aimed to investigate floral mechanisms underlying the interaction with pollinators in two hummingbird-pollinated orchids occurring in the Atlantic forest. We assessed floral biology, nectar traits, nectary and column micromorphologies, breeding systems and pollinators. In both species, nectar is secreted by lip calli through spaces between the medial lamellar surfaces of epidermal cells. Such form of floral nectar secretion has not been previously described. Both species present functional protandry and are self-compatible yet pollinator-dependent. Fruit sets in hand-pollination experiments were more than twice those under natural conditions, evidencing pollen limitation. The absence of fruit set in interspecific crosses suggests the existence of post-pollination barriers between these synchronopatric species. In Elleanthus brasiliensis, fruits resulting from cross-pollination and natural conditions were heavier than those resulting from self-pollination, suggesting advantages to cross-pollination. Hummingbirds pollinated both species, which share at least one pollinator species. Species differences in floral morphologies led to distinct pollination mechanisms. In E. brasiliensis, attachment of pollinaria to the hummingbird bill occurs through a lever apparatus formed by an appendage in the column, another novelty to the knowledge of orchids. In E. crinipes, pollinaria attachment occurs by simple contact with the bill during insertion into the flower tube, which fits tightly around the bill. As a way to reduce water losses in furrow irrigation systems, used in fresh market tomato production, farmers are improperly distributing water into the field using plastic hose. The objective of this work was to study the suitability of using quick coupler sprinkler as hose connectors for water distribution in tomato plantation. The first step of the study was to assess the current hose field operation for tomato growers. Subsequently, four models of quick couplers sprinklers available in the market were tested in laboratory to determine the coefficient of resistance, the equivalent tube length, the head loss curve and the linking efficiency. As result, a structural design for hose connectors was presented using the model of coupling with the best hydraulic performance. Additionally, some technical recommendations on its use in irrigation water distribution are highlighted. Despite the requirement for additional field trials, the proposed system has potential to optimize the water use efficiency, to improve workers ergonomic conditions, and ensure good profitability to the producer.

Slumping of hardsetting seedbeds upon wetting is likely to determine the shrinking and development of strength on drying. Different processes have been invoked, including aggregate disruption, material relocation, and compaction. To gain a better understanding of the role played by compaction compared with aggregate disruption in seedbed slumping and shrinking, mechanical analysis was combined with previous morphogenetical description. The global structural behavior of repacked seedbeds of a hardsetting sandy loam soil was studied after wetting and again after subsequent drying. Bulk density was measured in 5-mm-depth increments using gamma attenuation, and water content was determined at 10-mm-depth increments. Various wetting conditions were used to simulate a range of climatic and management conditions, including flood irrigation, furrow irrigation of a formed seedbed, drip irrigation, and rainfall. Aggregate coalescence under overburden pressure played the main role in slumping, even though microcracking enhanced coalescence. Most of the slumping occurred at calculated effective stress > 1.1 kPa. Intense aggregate breakdown at the top of seedbeds under fast wetting led to slight slumping because the resulting clogging of the initial interaggregate packing voids was balanced, in part, by the increase in microporosity resulting from aggregate disruption. However, aggregate coalescence induced by overburden pressure developing at the seedbed bottom often resulted in a strong decrease in total porosity. The effect of rainfall kinetic energy on crust bulk density was strong compared with the effect of fast wetting (bulk density increase of about 0.07 Mg m(-3) and 0.03 Mg m(-3), respectively) and could be ascribed to compaction rather than to aggregate breakdown. Shrinking on drying was related to the continuity of the microstructure resulting from wetting rather than to the intensity of slumping. Aggregate breakdown led to more shrinking than did aggregate coalescence.

Atualmente, tem-se difundido a aplicação de inoculante no sulco de semeadura na cultura da soja, mas há poucas informações que dão suporte a essa prática e comprovam sua eficiência em diferentes ambientes manejados sob plantio direto. Este trabalho teve como objetivo avaliar a viabilidade da aplicação de inoculantes na cultura da soja, via semente e sulco de semeadura, em solo já cultivado ou não com soja. Foram realizados dois experimentos em campo a partir de dezembro de 2004 em Latossolo Vermelho-Amarelo, seguindo o mesmo método e tratamentos, porém em dois locais distintos, com ou sem cultivo anterior de soja. Foram testados oito tratamentos: (1) inoculação via semente (inoculante + fungicida + micronutriente); (2) sem inoculação (fungicida + micronutriente); (3) testemunha (semente pura, sem tratamento); (4) aplicação no sulco-dose 1 (dose do inoculante recomendada no sulco); (5) aplicação no sulco-dose 2 (duas vezes a dose recomendada no sulco); (6) aplicação no sulco-dose 3 (três vezes a dose recomendada no sulco); (7) sulco-dose 1 + inoculação via semente; e (8) adubação com N (200 kg ha-1 N). Foram avaliados massa de matéria seca de nódulos e número de nódulos totais e nódulos viáveis e não-viáveis aos 30 e 75 dias após emergência. A melhor nodulação foi obtida com aplicação de inoculante + fungicida + micronutriente via semente no solo ainda não cultivado. No solo previamente cultivado com soja, destacaram-se os tratamentos uma e duas vezes a dose do inoculante no sulco. Menores valores de massa seca de nódulos na soja foram obtidos no tratamento com adubação mineral. A aplicação via sulco do inoculante mostrou-se uma prática viável, em razão da semelhança dos resultados obtidos com a aplicação tradicional via semente.
