Análise morfométrica da transição esôfago-gástrica de eqüinos submetidos a diferentes manejos alimentares e atividade física

O objetivo deste trabalho foi identificar se diferentes tipos de manejo físico e alimentar determinaram mudanças adaptativas na morfometria da região de transição esôfago-gástrica (TEG) em eqüinos. Foram utilizados 15 conjuntos de animais adultos com raça, sexo e idade não identificados, divididos em três grupos de acordo com a dieta e a atividade física: grupo I - composto por cinco conjuntos da TEG de eqüinos em treinamento intensivo e alimentados principalmente à base de concentrado; grupo II - com cinco conjuntos da TEG de eqüinos que não desempenhavam atividade física intensa e eram alimentados principalmente à base de concentrado, e o grupo III - composto por cinco conjuntos da TEG de eqüinos mantidos a pasto e que não desempenhavam atividade física intensa. A região de transição esôfago-gástrica teve a estrutura do seu epitélio avaliada morfometricamente, onde foram feitas mensurações para identificar a espessura do epitélio estratificado pavimentoso queratinizado. A espessura do grupo I teve como média 122,3µm, do grupo II foi de 173,4µm e do grupo III foi de 281,7µm. Os resultados encontrados para a espessura do epitélio estratificado pavimentoso queratinizado dos eqüinos mostraram a existência de diferenças estatisticamente significativas dos eqüinos do grupo I, quando comparados aos eqüinos do grupo III.

Evaluation of laser induced breakdown spectrometry for the determination of macro and micronutrients in pharmaceutical tablets

The aim of this work is to demonstrate the feasibility of laser induced breakdown spectrometry (LIBS) for the determination of macro and micronutrients in multielement tablets. The experimental setup was designed by using a laser Q-switch (Nd:YAG, 10 Hz, lambda = 1064 nm) and the emission signals were collected by lenses into an optical fiber coupled to an echelle spectrometer equipped with a high-resolution intensified charge coupled device (ICCD). Tablets were cryogenically ground and thereafter pelletized before LIBS analysis. Calibration curves were made by employing samples and mixtures of commercial multielement tablets with binders at different ratios. Best results were achieved by using the following experimental conditions: 29 J cm(-2) laser fluence, 165 mm lens to sample distance (f = 200 mm), 2.0 mu s delay time, 5.0 mu s integration time and 5 accumulated laser pulses. In general, the results obtained by the proposed LIBS procedure were in agreement with those obtained by ICP OES from the corresponding acid digests and coefficients variation of LIBS measurements varied from 2 to 16%. The metrological figures of merit indicate that LIBS fits for the intended purposes, and can be recommended for the analysis of multielement tablets and similar matrices aiming the determination of Ca, Cu, Fe, Mg, Mn, P and Zn.

Atrial Coronary Arteries in Areas Involved in Atrial Fibrillation Catheter Ablation

Background-The proximity to vascular structures is a limiting factor during radiofrequency ablation. However, little or no attention has been given to the atrial arterial circulation during the development of atrial fibrillation (AF) catheter ablation techniques. Methods and Results-We examined the atrial arterial circulation in areas involved in AF ablation in 24 heart specimens by colored resin injection and careful dissection. The sinus node artery (SNA) arose from the circumflex artery in 42% of case; proximal to the LA appendage in 29%, crossing the left atrium (LA) anterior wall; and after the LA appendage in the remaining 13%, crossing the mitral isthmus and passing close to the left pulmonary veins (PVs), the LA roof, and the right superior PV. In 58%, the SNA arose from the right coronary artery. Major arteries (>= 1 mm in external diameter) were found in the mitral isthmus in 54%, at the LA roof in 54%, and at the LA anterior wall in 29%. Around the left PV ostia, there were areas with major arteries in up to 37% (at the roof and inferior segments) and around the right PV ostia in up to 29% (at the roof segment). Conclusions-Major atrial coronary arteries, including the SNA, were commonly found in the areas involved in AF ablation and could cause difficulties in obtaining transmural lesions and electric isolation or even lead to ischemic sinus node or atrial dysfunction. Acute Pancreatitis Hypertonic Saline Increases Heat Shock Proteins 70 and 90 and Reduces Neutrophil Infiltration in Lung Injury

Objectives: Acute pancreatitis (AP) protease release induces lung parenchymal destruction via matrix metalloproteinases (MMPs), a neutrophil (polymorphonuclear leukocyte)-dependent process. Recent studies in hemorrhagic shock revealed that hypertonic saline (HTS) has an anti-inflammatory effect and can inhibit a variety of neutrophil functions. The aim of this study was to determine whether HTS and its actions in the pathway of neutrophil migration, MMPs, and heat shock proteins (HSPs) are effective in protecting the lung from injury associated with AP. Methods: We determined neutrophil infiltration and expressions of MMPs and HSPs in the lung tissue after AP induced by retrograde infusion of 2.5% of sodium taurocholate. Results: Animals submitted to AP that received HTS compared with those who received normal saline presented with increased HSP70 and HSP90 expressions and reduced myeloperoxidase levels and MMP-9 expression and activity. Conclusions: Our data raised the hypothesis that a sequence of HTS lung protection events increases HSP70 and HSP90, inhibiting infiltration of neutrophils and their protease actions in the lung.

Gene expression reprogramming protects macrophage from septic-induced cell death

Sepsis induces a systemic inflammatory response leading to tissue damage and cell death. LPS tolerance affects inflammatory response. To comprehend potential new mechanisms of immune regulation in endotoxemia, we examined macrophage mRNA expression by macroarray affected by LPS tolerance. LPS tolerance was induced with subcutaneous administration of 1 mg/kg/day of LPS over 5 days. Macrophages were isolated from the spleen and the expression of 1200 genes was quantitatively analyzed by the macroarray technique. The tolerant group displayed relevant changes in the expression of 84 mRNA when compared to naive mice. A functional group of genes related to cell death regulation was identified. PARP-1, caspase 3, FASL and TRAIL genes were confirmed by RT-PCR to present lower expression in tolerant mice. In addition, reduced expression of the pro-inflammatory genes TNF-alpha and IFN-gamma in the tolerant group was demonstrated. Following this, animals were challenged with polymicrobial sepsis. Flow cytometry analysis showed reduced necrosis and apoptosis in macrophages from the tolerant group compared to the naive group. Finally, a survival study showed a significant reduction in mortality in the tolerant group. Thus, in the current study we provide evidence for the selective reprogramming of the gene expression of cell death pathways during LPS tolerance and link these changes to protection from cell death and enhanced survival rates. Modelling the Mechanical Behavior of Polymer-Based Nanocomposites

Molecular dynamics simulations were employed to analyze the mechanical properties of polymer-based nanocomposites with varying nanofiber network parameters. The study was focused on nanofiber aspect ratio, concentration and initial orientation. The reinforcing phase affects the behavior of the polymeric nanocomposite. Simulations have shown that the fiber concentration has a significant effect on the properties, with higher loadings resulting in higher stress levels and higher stiffness, matching the general behavior from experimental knowledge in this field. The results also indicate that, within the studied range, the observed effect of the aspect ratio and initial orientation is smaller than that of the concentration, and that these two parameters are interrelated.

Crescimento e produção da bananeira 'Thap Maeo' (AAB) durante dois ciclos de produção no Vale do São Francisco

A avaliação de genótipos de bananeira, durante ciclos sucessivos de produção em diferentes ecossistemas, é essencial não só ao programa de melhoramento genético, mas, também, para indicar cultivares promissores para o sistema produtivo local. O objetivo deste trabalho foi avaliar o crescimento e a produção da bananeira 'Thap Maeo' (AAB), no Vale do São Francisco, durante o primeiro e o segundo ciclos de produção (da colheita da planta-mãe à colheita da planta-filha) e os atributos de qualidade de seus frutos. As mudas de 'Thap Maeo' foram produzidas pelo processo de cultura de tecidos e transplantadas ao campo aos seis meses de idade, em espaçamento 3,0 m x 3,0 m. As características avaliadas foram: ciclos de produção; em cada ciclo, a altura de planta, o perímetro do pseudocaule, o número de folhas por planta, o número de dias entre plantio e colheita, a massa da matéria fresca do cacho, o número de pencas e de frutos por cacho, a massa da matéria fresca da segunda penca; número, comprimento e diâmetro de frutos da segunda penca, relação polpa/casca, firmeza, pH, sólidos solúveis (SS), acidez titulável (AT) e relação SS/AT. A bananeira 'Thap Maeo' apresentou, no segundo ciclo, maior crescimento, melhor desempenho dos caracteres que expressam a produção e qualidade dos frutos em pós-colheita.

Caracterização agronômica e pós-colheita das bananeiras 'Maravilha' e 'Preciosa' no Submédio do Vale São Francisco

RESUMO A caracterização e a avaliação de novos genótipos de bananeira é uma etapa importante, tanto para os programas de melhoramento genético quanto para a indicação aos produtores. Assim, o objetivo desta pesquisa foi avaliar a produção e a qualidade de frutos, em pós-colheita, das bananeiras 'Maravilha' e 'Preciosa', no primeiro e segundo ciclo de produção, cultivadas no Submédio do Vale do São Francisco. As mudas, produzidas por cultura de tecidos, foram transplantadas para o campo com seis meses de idade, cultivadas no espaçamento de 3 x 3 m. Nos dois ciclos avaliados, para todas as características, foi adotado o delineamento experimental inteiramente casualizado, com dois tratamentos (cultivares), dez repetições e duas plantas úteis por parcela, com bordadura externa ao ensaio. Foram avaliadas as seguintes variáveis: ciclo de produção (dias entre plantio e colheita), altura de planta, perímetro do pseudocaule, número de folhas adultas, massa da matéria fresca do cacho, número de pencas e de frutos por cacho, massa da matéria fresca da segunda penca; número, comprimento e diâmetro de frutos da segunda penca, relação polpa/casca, firmeza, pH, teor de sólidos solúveis (SS), acidez titulável (AT) e relação SS/AT. O cultivar 'Maravilha' apresentou características de planta e de produção superiores às do 'Preciosa'; quanto às características físico-químicas, o 'Preciosa' apresentou frutos com maiores teores de açúcares, todavia, menores em tamanho; para as demais características, os dois cultivares apresentaram comportamentos semelhantes entre si, no primeiro e no segundo ciclo de produção.
