Semen cryopreservation is still considered suboptimal due to lower fertility when compared to fresh semen. The reasons for the loss of fertility are various and related to irreversible damage caused to the cells during the freeze-thaw process. An alternative to conventional cryopreservation represents the use of chilled bull semen, preventing the damage associated with freezing, thereby guaranteeing greater sperm viability. The aim of this study was to describe the use of cooled bull semen as a strategy to increase the pregnancy for Fixed-Time Artificial Insemination (FTAI) of Nellore (Bos indicus) cows. One ejaculate of a select Nellore bull obtained by electroejaculation was used; the semen sample was fractioned into two aliquots: one diluted in Botu-Bov® extender containing 6.4% glycerol for cryopreservation (BB-F, frozen group) and one diluted in the same extender, free from cryoprotectants and used for cooling (BB-C, cooled semen group). The samples in the BB-C group were chilled to 5°C using an isothermic box and maintained for 24 h prior to use. A total of 349 lactating Nellore cows (70-90 days after birth) were synchronized by the insertion of a progesterone releasing device (1.0 g) and estradiol benzoate (2.0 mg i.m.) on a random day of the estrous cycle (Day 0); FTAI was performed 44-48 h after the removal of the device. The pregnancy rates were 45.71 and 61.49% (P<0.05), respectively, for the cryopreserved or chilled bovine semen groups. In conclusion, the use of bull semen cooled for 24 h represents an alternative to conventionally cryopreserved semen, as determined by the increase the pregnancy per artificial insemination in bovine herds. © 2012 Science Publication.

Multiple ovulation (superovulation) and embryo transfer has been used extensively in cattle. In the past decade, superstimulatory treatment protocols that synchronise follicle growth and ovulation, allowing for improved donor management and fixed-time AI (FTAI), have been developed for zebu (Bos indicus) and European (Bos taurus) breeds of cattle. There is evidence that additional stimulus with LH (through the administration of exogenous LH or equine chorionic gonadotrophin (eCG)) on the last day of the superstimulatory treatment protocol, called the 'P-36 protocol' for FTAI, can increase embryo yield compared with conventional protocols that are based on the detection of oestrus. However, inconsistent results with the use of hormones that stimulate LH receptors (LHR) have prompted further studies on the roles of LH and its receptors in ovulatory capacity (acquisition of LHR in granulosa cells), oocyte competence and embryo quality in superstimulated cattle. Recent experiments have shown that superstimulation with FSH increases mRNA expression of LHR and angiotensin AT(2) receptors in granulosa cells of follicles >8 mm in diameter. In addition, FSH decreases mRNA expression of growth differentiation factor 9 (GDF9) and bone morphogenetic protein 15 (BMP15) in oocytes, but increases the expression of both in cumulus cells, without diminishing the capacity of cumulus-oocyte complexes to generate blastocysts. Although these results indicate that superstimulation with FSH is not detrimental to oocyte competence, supplementary studies are warranted to investigate the effects of superstimulation on embryo quality and viability. In addition, experiments comparing the cellular and/or molecular effects of adding eCG to the P-36 treatment protocol are being conducted to elucidate the effects of superstimulatory protocols on the yield of viable embryos.

Four experiments evaluated the effects of vaccination against bovine herpesvirus-1 (BoHV-1), bovine viral diarrhea virus (BVDV), and Leptospira spp. on reproductive performance of lactating dairy cows without (experiments 1, 2, and 3) or with previous vaccination against these diseases (experiment 4). Cows were assigned to a fixed-time AI protocol (FTAI; d -11 to 0) in all experiments, as well as AI 12. h upon estrus detection in experiment 3. Pregnancy status was determined with transrectal ultrasonography on d 30 and 71 (d 60 for experiment 3) after AI. Pregnancy loss was considered in cows pregnant on d 30 but non-pregnant on the subsequent evaluation. In experiment 1, 853 cows received (VAC) or not (CON) vaccination against BoHV-1, BVDV, and Leptospira spp. at the beginning of the FTAI (d -11) and 30. d after AI. Pregnancy loss was reduced (P=0.03) in VAC cows compared with CON. In experiment 2, 287 cows received VAC or CON 30. d prior to (d -41) and at the beginning (d -11) of the FTAI. Pregnancy rates on d 30 and 71 were greater (P≤0.03) in VAC cows compared with CON. In experiment 3, 1680 cows with more than 28. d in milk were randomly assigned to receive VAC or CON with doses administered 14. d apart, and inseminated within 15-135. d after the second dose. Pregnancy rates on d 30 and 60 were greater (P≤0.02) in VAC cows compared with CON. In experiment 4, 820 cows received (REVAC) or not (CON) revaccination against BoHV-1, BVDV, and Leptospira spp. at the beginning of the FTAI protocol (d -11). Pregnancy rates and loss were similar (P≥0.54) between treatments. Hence, vaccinating naïve cows against BoHV-1, BVDV, and Leptospira spp. improved reproductive efficiency in dairy production systems, particularly when both doses were administered prior to AI. © 2013 Elsevier B.V.

Incrementa o uso da Inseminação Artificial em Tempo Fixo (IATF) em búfalas criadas em sistema produtivo extensivo de terras alagadas sazonalmente. O experimento realizado na cidade de Itaubal, no estado do Amapá, mais precisamente na Fazenda Motogeral LTDA, no período de novembro de 2004 a fevereiro de 2005, na estação favorável à reprodução. A propriedade possui um rebanho bubalino mestiço, onde predomina as raças Mediterrâneo e Murrah. Os animais possuem idade média de seis anos, em sua maioria plurípara e todas amamentando. Utilizou-se 93 animais, sendo que 70 animais sincronizados e inseminados em tempo fixo e 23 animais como grupo controle. Todos os animais foram previamente selecionados através de exame ginecológico e escore de condição corporal (ECC). Os animais sincronizados dividiram-se em três grupos: No grupo 1 (G1) com 20 animais utilizou-se o protocolo ovsynch; No grupo 2 (G2) com 29 animais utilizou-se o protocolo DIB-synch; e no grupo 3 (G3), 21 animais compuseram o grupo denominado CLsynch. E os não sincronizados compuseram o grupo controle ou Grupo 4 (G4) composto por 23 animais que foram inseminados 12h após a visualização do cio. Das 93 búfalas inseminadas 52 (55,91%) tornaram-se gestantes. Das 70 búfalas sincronizadas e inseminadas em tempo pré-determinado 43 (61,43%) ficaram gestantes. No G1 12 (60%) búfalas ficaram gestantes, no G2 19 (65,5%) búfalas ficaram gestantes e no G3 12 (57,14%) búfalas ficaram gestantes. No grupo controle a taxa de prenhez foi de 39,13%. Não houve diferença significativa entre os grupos sincronizados e o grupo controle (P=0,27; X²=3,86). Nos grupos sincronizados não se observou diferença significativa entre os grupos (P=0,82; X² =0,38). As variáveis presença de muco (P= 4,11; Fisher=0,99) e contratilidade uterina (P=8,65; Fisher=0,99) influenciaram significativamente na taxa de prenhez. Dos 70 animais sincronizados 34 (48,57%) apresentaram muco e 54 (77,14%) apresentaram contratilidade uterina durante a IATF. Quanto ao ECC apesar de estatisticamente não ter influencia nos resultados, notou-se que os animais com ECC• 2.5 apresentaram as maiores taxas de prenhez. Pode-se concluir que os protocolos utilizados mostram que a IATF em bubalinos, criados de forma extensiva em áreas pantanosas, são uma alternativa promissora para melhorar a eficiência reprodutiva desta espécie na região Amazônica. O protocolo CL-synch precisa de mais estudos, com maior número de animais. Os custos dos tratamentos, não inviabilizam a adoção da técnica em propriedades rurais com sistema de produção extensiva.
