Did SBS chief executive Michael Ebeid score a well-timed free kick or an own goal in his attack on the ABC this week? The ABC recently secured the free-to-air television rights for the Asian Cup football tournament to be held in Australia early next year, together with tonight’s match between the Socceroos and Japan. A lower bid by SBS – still in some circles fondly known as the “Soccer Broadcasting Service” – was rejected, dealing a significant blow to the smaller public broadcaster. The ABC was reportedly asked to make a bid by Football Federation Australia. The FFA presumably believes the ABC’s coverage will attract larger audiences to the game. This is despite SBS’s long-term success with the sport. It should not be forgotten, however, that while SBS has largely been defined by its long connection with the world game, ABC was the home of football from the late 1950s until the 1980s. But the stoush is only partly about football. It was surely no coincidence that it comes on the eve of the government’s formal announcement of the size of the cuts to public broadcasting...

Clustering techniques are used in regional flood frequency analysis (RFFA) to partition watersheds into natural groups or regions with similar hydrologic responses. The linear Kohonen's self‐organizing feature map (SOFM) has been applied as a clustering technique for RFFA in several recent studies. However, it is seldom possible to interpret clusters from the output of an SOFM, irrespective of its size and dimensionality. In this study, we demonstrate that SOFMs may, however, serve as a useful precursor to clustering algorithms. We present a two‐level. SOFM‐based clustering approach to form regions for FFA. In the first level, the SOFM is used to form a two‐dimensional feature map. In the second level, the output nodes of SOFM are clustered using Fuzzy c‐means algorithm to form regions. The optimal number of regions is based on fuzzy cluster validation measures. Effectiveness of the proposed approach in forming homogeneous regions for FFA is illustrated through application to data from watersheds in Indiana, USA. Results show that the performance of the proposed approach to form regions is better than that based on classical SOFM.

Recent focus of flood frequency analysis (FFA) studies has been on development of methods to model joint distributions of variables such as peak flow, volume, and duration that characterize a flood event, as comprehensive knowledge of flood event is often necessary in hydrological applications. Diffusion process based adaptive kernel (D-kernel) is suggested in this paper for this purpose. It is data driven, flexible and unlike most kernel density estimators, always yields a bona fide probability density function. It overcomes shortcomings associated with the use of conventional kernel density estimators in FFA, such as boundary leakage problem and normal reference rule. The potential of the D-kernel is demonstrated by application to synthetic samples of various sizes drawn from known unimodal and bimodal populations, and five typical peak flow records from different parts of the world. It is shown to be effective when compared to conventional Gaussian kernel and the best of seven commonly used copulas (Gumbel-Hougaard, Frank, Clayton, Joe, Normal, Plackett, and Student's T) in estimating joint distribution of peak flow characteristics and extrapolating beyond historical maxima. Selection of optimum number of bins is found to be critical in modeling with D-kernel.

Background: This study was performed to understand the possible therapeutic activity of Terminalia paniculata ethanolic extract (TPEE) on non alcoholic fatty liver in rats fed with high fat diet. Methods: Thirty six SD rats were divided into 6 groups (n = 6): Normal control (NC), high fat diet (HFD), remaining four groups were fed on HFD along with different doses of TPEE (100,150 and 200 mg/kg b.wt) or orlistat, for ten weeks. Liver tissue was homogenized and analyzed for lipid profiles, activities of superoxide dismutase (SOD), catalase (CAT) and malondialdehyde (MDA) content. Further, the expression levels of FAS and AMPK-1 alpha were also studied in addition to histopathology examination of liver tissue in all the groups. Results: HFD significantly increased hepatic liver total cholesterol (TC), triglycerides (TG), free fatty acids (FFA) and MDA but decreased the activities of SOD and CAT which were subsequently reversed by supplementation with TPEE in a dose-dependent manner. In addition, TPEE administration significantly down regulated hepatic mRNA expression of FAS but up regulated AMPK-1 alpha compared to HFD alone fed group. Furthermore, western blot analysis of FAS has clearly demonstrated decreased expression of FAS in HFD + TPEE (200 mg/kg b. wt) treated group when compared to HFD group at protein level. Conclusions: Our biochemical studies on hepatic lipid profiles and antioxidant enzyme activities supported by histological and expression studies suggest a potential therapeutic role for TPEE in regulating obesity through FAS.

The principal objective of this study is to evaluate the benefits that accrue to the national economies as a result of different longline operational models and licensing regimes, computed, for comparability reasons, on a per tonne basis. The primary source for the derivation of these per tonne returns was individual fisheries enterprises currently operating in the sector in the countries chosen for this study. In each case, the company disclosed to the consultant detailed financial, operational and marketing data which has enabled the findings to be based upon actual empirical data, rather than assumptions or interpretations of current practice and benefit outcomes. The cystic fibrosis transmembrane conductance regulator (CFTR) is a chloride channel member of the ATP-binding cassette (ABC) superfamily of membrane proteins. CFTR has two homologous halves, each consisting of six transmembrane spanning domains (TM) followed by a nucleotide binding fold, connected by a regulatory (R) domain. This thesis addresses the question of which domains are responsible for Cl^- selectivity, i.e., which domains line the channel pore.
To address this question, novel blockers of CFTR were characterized. CFTR was heterologously expressed in Xenopus oocytes to study the mechanism of block by two closely related arylaminobenzoates, diphenylamine-2-carboxylic acid (DPC) and flufenamic acid (FFA). Block by both is voltage-dependent, with a binding site ≈ 40% through the electric field of the membrane. DPC and FFA can both reach their binding site from either side of the membrane to produce a flickering block of CFTR single channels. In addition, DPC block is influenced by Cl^- concentration, and DPC blocks with a bimolecular forward binding rate and a unimolecular dissociation rate. Therefore, DPC and FFA are open-channel blockers of CFTR, and a residue of CFTR whose mutation affects their binding must line the pore.
Screening of site-directed mutants for altered DPC binding affinity reveals that TM-6 and TM-12 line the pore. Mutation of residue 5341 in TM-6 abolishes most DPC block, greatly reduces single-channel conductance, and alters the direction of current rectification. Additional residues are found in TM-6 (K335) and TM-12 (T1134) whose mutations weaken or strengthen DPC block; other mutations move the DPC binding site from TM-6 to TM-12. The strengthened block and lower conductance due to mutation T1134F is quantitated at the single-channel level. The geometry of DPC and of the residues mutated suggest α-helical structures for TM-6 and TM-12. Evidence is presented that the effects of the mutations are due to direct side-chain interaction, and not to allosteric effects propagated through the protein. Mutations are also made in TM-11, including mutation S1118F, which gives voltage-dependent current relaxations. The results may guide future studies on permeation through ABC transporters and through other Cl^- channels.

As mudanças nos hábitos alimentares têm causado efeitos impressionantes na saúde pública, diretamente relacionados ao aumento da ingestão de refeições ricas em gorduras, principalmente gorduras saturadas. A principal consequência desse consumo é o estado prolongado e excessivo da lipemia pós-prandial (LPP), considerada um dos fatores relacionados às anormalidades metabólicas e aos danos vasculares. O objetivo do estudo foiavaliar o efeito da sobrecarga lipídica na reatividade microvascular em mulheres obesas. Das 41 participantes deste estudo, 21 apresentavam o diagnóstico de obesidade, com IMC de 32,41,6 kg/m2 (média SD) e idade 31,65 anos e 20 mulheres saudáveis, com IMC de 21,91,7 kg/m2 e idade 27,25,5 anos. Após a avaliação clínica e laboratorial, as participantes tiveram a microcirculação examinada por dois métodos: a dinâmica do leito periungueal, para avaliação da densidade capilar funcional (DCF), velocidade de deslocamento das hemácias no basal (VDH) e após uma isquemia de 1 min (VDHmax) e tempo de reperfusão (TVDHmax). A segunda técnica foi a do dorso do dedo para avaliação da DCF no repouso, durante a hiperemia reativa e após oclusão venosa. Foi feita a coleta de sangue para avaliação do colesterol total (CT), triglicerídeos (TG), HDL-c e ácidos graxos livres (AGL), glicose, insulina e viscosidade plasmática em 30 e 50 rotações por minuto (rpm). Também foram medidas a pressão arterial sistólica (PAS), diastólica (PAD) e frequência cardíaca (FC). Após essas análises no repouso, todas as participantes receberam uma refeição rica em lipídios, e após 30, 60, 120 e 180 minutos da ingestão da refeição, os exames de videocapilaroscopia e a coleta de sangue foram novamente realizados.As participantes com obesidade apresentaram, após a sobrecarga lipídica, valores significativamente menores do que no jejum para: DCF basal do dorso do dedo (p=0,02); DCF durante hiperemia reativa (p=0,02), DCF pós-oclusão venosa (p=0,02), HDL-c (p<0,0001), LDL-C (p<0,0001) e AGL (p<0,0001) e valores elevados para: VDH (p<0,0001), VDHmax(p=0,003), TVDHmax (p=0,004), glicose (p<0,0001), insulina (p<0,001), CT (p=0,03), TG (p<0,0001) e FC (p=0,03). Alterações na viscosidade não foram observadas no grupo OB após a refeição quando comparado aos seus valores basais em 30 e 50 rpm (p=0,87 e p=0,42, respectivamente). A PAS foi elevada nas participantes OB após a sobrecarga quando comparada às saudáveis em todo tempo de estudo. Concluímos que alimentos ricos em lipídios podem aumentar ainda mais a disfunção microcirculatória e as alterações metabólicas já presentes em mulheres obesas.

磷脂酰甘油(PG)是植物类囊体膜中唯一的磷脂,在它的sn-2位上总是连着一个棕榈酸(16:0)或反式十六碳烯酸(16:1 trans)。由于PG的分子结构独特,对它的功能已有了很多研究,目前认为PG在维持类囊体膜的结构与功能方面具有非常重要的作用。缺磷胁迫下,蓝藻、衣藻及拟南芥、大麦等物种中均检测到了PG含量的下降。对这一现象的常见解释是缺磷导致了PG生物合成受阻,从而引起了其含量的降低。但迄今为止尚没有试验证据支持。本研究比较了缺磷对不同叶龄的小麦与烟草叶片中PG含量与PG水解酶的活性的影响,同时对缺磷叶片酶粗提液水解外源PG后的主要产物、几种磷脂酶抑制剂对上述酶反应的影响等进行了研究,以阐明缺磷条件下叶片中PG含量下降的主要原 因。 缺磷小麦第一叶完全展开时,PG含量与PG水解酶活性均与对照相似;而第三叶完全展开时,尽管缺磷第三叶中PG水解酶活性也与对照相似,但其PG含量低于对照。这一结果表明,在小麦叶片完全展开之前,缺磷条件未影响叶片中的PG水解酶活性,第三叶中较低的PG含量应由PG的生物合成受阻引起。并且,由于缺磷植株第一叶完全展开时PG含量未受影响而第三叶中却表现出了轻微降低,可以推测叶片萌发越晚,PG生物合成受到的抑制就会越严重。 为了研究叶片衰老过程中PG含量下降的原因,我们比较了6,10,14与18日龄时缺磷与对照小麦植株第一叶中PG的相对含量与PG水解酶活性。研究发现:6日龄时,刚刚完全展开的缺磷和对照小麦第一叶中无论是PG含量还是PG水解酶活性都较为相似;而随着叶片的逐渐衰老,缺磷植株第一叶中PG含量大幅度下降,同时伴随着PG水解酶活性的急剧上升。18日龄时,缺磷小麦第一叶中的PG含量较对照降低了69.1%,其PG水解酶活性也远高于对照,37ºC下温育30min后,缺磷叶片的酶粗提液使外源PG含量降低了74.16%,而对照中只降低了13.7%。上述结果表明,缺磷条件下,小麦叶片中PG含量降低的程度与PG水解酶活性的强弱密切相关,PG水解加剧是导致老叶中PG含量降低的一个重要原因。 磷脂酶是水解磷脂的主要酶类。目前在植物体中发现的磷脂酶种类主要有磷脂酶D(PLD)、磷脂酶C(PLC)与磷脂酶A(PLA)。通过薄层层析(TLC),我们发现缺磷小麦叶片的酶粗提液水解外源PG后的主要产物是磷脂酸(PA)、二脂酰甘油(DAG)与游离脂肪酸(FFA)。将n-丁醇加入到缺磷小麦叶片的体外酶反应体系中后,观察到PA、DAG与FFA的生成量均表现出一定程度的降低。由于n-丁醇是PA经PLD途径生成的抑制剂,因此,上述结果表明PLD参与了缺磷条件下小麦叶片中PG的水解。硫酸新霉素是PLC的非特异性抑制剂,低浓度的硫酸新霉素(100μM 和 200μM )加入到缺磷小麦叶片的体外酶反应体系后,三种产物的生成受到了严重抑制,表明PLC也与缺磷叶片中PG的降解密切相关。 为了进一步分析缺磷导致PG含量降低的原因,我们以烟草为试验材料,检测了缺磷胁迫对烟草嫩叶和老叶中的PG含量、PG水解酶活性、与PG降解相关的酶的种类及PLC、PLDα、PLDβ与PAT-1基因在mRNA上表达水平的的影响。结果表明,缺磷烟草叶片中PG含量的降低由PG生物合成受阻与PG降解加剧共同导致,PLC和PLD活性与烟草叶片中PG的降解有关。缺磷植株老叶中PG水解酶活性及PLC、PLDα、PLDβ基因在mRNA水平上的表达量均高于对照,表明在磷胁迫条件下,老叶中PG水解酶活性可能受到转录水平上的调节, PLC、PLDα、PLDβ转录活性的增强导致了PLC、PLD活性加强,从而引起PG降解的加剧,最终导致了PG含量的降低。与PLC、PLDα和PLDβ不同,缺磷胁迫对patatin蛋白(表现PLA2活性)的编码基因PAT-1在转录水平上的表达无影响,TLC分析PG的水解产物也未检测到溶血磷脂酰甘油(LPG)的生成。由此可见,PLA活性可能与缺磷条件下PG的降解无关。

Shark livers are considered as an important raw material providing a quality fish oil. It has been reported to aid white — blood-cell production and act as an active ingredient in hemorrhoid treatments. It is also reported that liver oil as a good supplement of vitamin A and poly-unsaturated fatty acids which are important to the development of brain cells in human. Freshness of livers is very important to extract better quality oil. In Sri Lanka, the annual shark production amounts to 8000t, however the quality of livers collected from landing sites has not being measured yet. Present study was conducted to evaluate the quality of silky (Charcarninus fakiformis) shark livers available in Negombo and Beruwala landing sites in the West Coast of Sri Lanka and also to study the relationship between organoleptic and bio-chemical correlation on freshness of shark livers. Liver samples which were collected from landing sites in the West coast of Sri Lanka, were evaluated for external and internal colour, texture and odour. Total volatile nitrogen (TVN), pH value, free fatty acid (FFA%) and peroxide (PV) values of livers were also determined to assess quality. According to the organoleptic scoring system 4.3% of liver samples were categorized as best in quality while 30.4%, 56.5% and 8.7% rated as good, medium and poor in quality respectively at the Negombo and Beruwala landing sites. Bio-chemical analysis showed that the better quality livers had the highest score for sensory evaluation and low values for TVN, FFA and peroxide value while low quality livers gave low score for sensory evaluation and high TVN, FFA, peroxide values. Correlation coefficient of organoleptic scores against total volatile nitrogen value, pH value, free fatty acid % and peroxide value of shark livers were determined by statistical analysis. Proximate composition, lipid and fatty acid components of dried mussel and changes in lipids during 1 year storage were studied. Male mussel contained lower fat contents and higher contents of polyunsaturated fatty acids of C20:5n-3, and C22:6n-3. High percentages of Cl6:1, Cl7:1, Cl8:3n-3, C20:3n-8 existed in NL and C!6:0, C18:0, Cl8:1n-9, C20:2n-6, C20:5n- 3, C22:6n-3 were very rich in PL. Triglycerides phosphatidylcholine, cholesterol were major components of mussel lipids. Free fatty acids (FFA) increased greatly and phospholipids decreased during storage, saturated fatty acids showed an increase trend and polyunsaturated fatty adds decreased differently. Dried mussels were vacuum packed and air packed and packaging methods had a great influence on the oxidation of mussellip,ids, indicating preference of vacuum packaging.

The influence of different pre-freezing ice storage periods on the biochemical and organoleptic qualities of Indian oil sardines (Sardinella longiceps) in the individual quick frozen (IQF) and block frozen (BF) forms and frozen storage at temperatures of -12°C and -23°C was studied. The shelf-life of the sardines varied between 24 and 2 weeks for samples iced for 0 to 5 days prior to freezing. The deterioration in quality was accompanied by considerable increase in the peroxide value (PV) and free fatty acid (FFA) content and decrease in salt extractability of the proteins. These changes were more rapid at -12°C than at -23°C. BF sardines appeared to be better than IQF samples with respect to the biochemical changes although the differences in overall organoleptic quality were not significant.

The effect of betel leaf (Piper betle Linn.) extract on control of autoxidation of fat in dry fish has been studied. Oil sardine has been selected for experiments since it contains very high amount of fat. The treatments were given with 5% (w/v) betel leaf extract in water at different stages of salt curing. FFA, PV and TVN values of the samples were determined periodically to assess the keeping quality and autoxidation. The sample, prepared by dipping the fish in the extract immediately after salting and then drying as usual, was found to have better keeping qualities.

Quality of analog fishery products invariably depends on the gel characteristics and nutritional status of minced meat. With an objective to find out the effect of water washing on kamaboko gel, the minced meat from Croaker fish was washed for four times (5 minutes each) using chilled water at a temperature of 8-10°C. Results reflected noticeable improvement in folding test and SSN% of kamaboko with essential decrease in fat content, water soluble proteins, expressible water and quality parameters like NPN, VBN, TMA, FFA and PV denoting superior gel quality than control sample after repeated washing. The results indicated that there was a definite improvement in functional properties such as gel forming ability, expressible water content of the croaker minced meat essential decrease in fat content, water soluble proteins, expressible water and quality parameters after each wash, but two washes of 5 minutes duration each was necessary to achieve satisfactory results.

The present research was done with the aim of studing the effects of some medicinal plant essential oils against Lactococcus garvieae and on some spoilage indexes in rainbow trout fillets in 4°C during 18 days. Essential oils of Mentha longifola (20, 40 and 80 μg g-1), Satureja bachtiarica (0.125, 0.25 and 0.50 μg g-1) and Satrureja khuzestanica (0.06, 0.12, 0.25 and 0.50 μg g-1) were used for this study. According to the results, the highest and the lowest microbial load were observed in control group and the groups contained 80 and 40 μg g-1 of Mentha longifola, respectively (P≤0.05). The spoilage indexes were increased after 18 days showing a statistical difference for protein, fat, TVN, FFA and TBA (P≤0.05). The most and the least changes were observed in the the control group and the fillets containing 80 μg g-1 of Mentha longifola, respectively (P≤0.05). The results indicated that, in comparison with control group, using herbal essential oils leads to reduction of Lactococcus garvieae population and spoilage process of the fillets through decreasing in TVN, FFA and TBA.
