It is well known that ocean acidification can have profound impacts on marine organisms. However, we know little about the direct and indirect effects of ocean acidification and also how these effects interact with other features of environmental change such as warming and declining consumer pressure. In this study, we tested whether the presence of consumers (invertebrate mesograzers) influenced the interactive effects of ocean acidification and warming on benthic microalgae in a seagrass community mesocosm experiment. Net effects of acidification and warming on benthic microalgal biomass and production, as assessed by analysis of variance, were relatively weak regardless of grazer presence. However, partitioning these net effects into direct and indirect effects using structural equation modeling revealed several strong relationships. In the absence of grazers, benthic microalgae were negatively and indirectly affected by sediment-associated microalgal grazers and macroalgal shading, but directly and positively affected by acidification and warming. Combining indirect and direct effects yielded no or weak net effects. In the presence of grazers, almost all direct and indirect climate effects were nonsignificant. Our analyses highlight that (i) indirect effects of climate change may be at least as strong as direct effects, (ii) grazers are crucial in mediating these effects, and (iii) effects of ocean acidification may be apparent only through indirect effects and in combination with other variables (e.g., warming). These findings highlight the importance of experimental designs and statistical analyses that allow us to separate and quantify the direct and indirect effects of multiple climate variables on natural communities.

Research in conditioning (all the processes of preparation for competition) has used group research designs, where multiple athletes are observed at one or more points in time. However, empirical reports of large inter-individual differences in response to conditioning regimens suggest that applied conditioning research would greatly benefit from single-subject research designs. Single-subject research designs allow us to find out the extent to which a specific conditioning regimen works for a specific athlete, as opposed to the average athlete, who is the focal point of group research designs. The aim of the following review is to outline the strategies and procedures of single-subject research as they pertain to.. the assessment of conditioning for individual athletes. The four main experimental designs in single-subject research are: the AB design, reversal (withdrawal) designs and their extensions, multiple baseline designs and alternating treatment designs. Visual and statistical analyses commonly used to analyse single-subject data, and advantages and limitations are discussed. Modelling of multivariate single-subject data using techniques such as dynamic factor analysis and structural equation modelling may identify individualised models of conditioning leading to better prediction of performance. Despite problems associated with data analyses in single-subject research (e.g. serial dependency), sports scientists should use single-subject research designs in applied conditioning research to understand how well an intervention (e.g. a training method) works and to predict performance for a particular athlete.

Neste trabalho são descritas as técnicas de análise estatística utilizadas e a acessibilidade estatística em uma amostra dos artigos originais publicados no período 1996-2006 em duas revistas de pesquisa na área de fruticultura: a Revista Brasileira de Fruticultura (RBF) e a revista francesa Fruits. No total foram classificados 986 artigos em 16 categorias de análise estatística, ordenadas em grau ascendente de complexidade. No período analisado, foi constatado um aumento no uso de análises mais sofisticadas ao longo do tempo em ambos as revistas. Os trabalhos publicados pela RBF aplicaram com maior freqüência técnicas estatísticas mais complexas, com maior utilização de delineamentos em blocos aleatorizados, arranjos fatoriais, parcelas subdivididas e modelos hierárquicos, e do teste de Tukey para comparações múltiplas de médias. Nos trabalhos publicados pela revista Fruits, predominou o uso de outros testes paramétricos e do teste de Duncan. O pacote estatístico SAS foi o mais utilizado nos artigos publicados em ambas as revistas. Os leitores da revista RBF precisaram de um nível de conhecimento estatístico mais elevado para ter acesso à maior parte dos artigos publicados no período, em comparação com os leitores da revista francesa.

O objetivo deste experimento foi avaliar o efeito da adição de doses crescentes de P2O5 sobre a altura do dossel, o número de perfilhos e a produção de matéria seca de folhas e de colmos do capim-Mombaça, em diferentes idades. Foi conduzido um experimento implantado em um Nitossolo Vermelho Eutrófico. O delineamento experimental usado foi o de blocos completos casualizados, com quatro repetições, cinco doses de P2O5 (30, 60, 90, 120 e 150kg ha-1) e uma testemunha. Para a primeira e segunda coletas, observou-se o efeito linear do fósforo sobre o perfilhamento. Para a terceira e quarta coletas, os dados ajustaram-se ao modelo quadrático. A participação das lâminas foliares na matéria seca da parte aérea diminuiu com as doses de P2O5. Por outro lado, a participação de colmos aumentou com as doses de P2O5. A produção de matéria seca (MS) da parte aérea para primeira, segunda e terceira coletas respondeu de forma linear à aplicação de P2O5 observando-se um aumento estimado de 7, 15 e 19kg ha-1 de MS por kg ha-1 de P2O5, respectivamente. Para a quarta coleta, os dados ajustaram-se ao modelo quadrático de regressão, sendo a máxima produção, 8,3Mg ha-1 de MS, obtida com a aplicação de 103kg ha-1 de P2O5.

This work deals with the use of an aqueous two-phase system (ATPS) of PEG/citrate to remove proteases from a Clostridium perfringens fermentation broth. To plan the experimental tests and evaluate the corresponding results, three successive experimental designs were employed, for which the PEG molar mass (M-PEG) and concentration (C-PEG), the citrate concentration (C-C) and the pH were selected as independent variables, while the purification factor (PF), the partition coefficient (K), the activity yield (Y) and the selectivity (S) were selected as responses. PF of proteases in the top phase was shown to increase with increasing MPEG and decreasing Cc, whereas a completely opposite trend was observed for K. On the other hand, Y was favored by simultaneous decreases in both these variables, while S decreased with increasing Cc. Therefore, selecting a simultaneous increase in PF and Y as the most desirable result, the best performance of the system was obtained using M-PEG = 10-000 g/mol C-PEG = 22% (w/w) and C-c = 8.0% (w/w) at pH 8.5. Under these conditions, the activity yield was very high (131 %) but the purification factor (4.2) and the selectivity (4.3) were lower than those ensured by more selective purification methods. According to these results, the ATPS seems to be an interesting alternative primary concentration/decontamination step for vaccine preparation from C. perfringens fermented broth. Culture conditions (pH, time, temperature, inoculum size, orbital agitation speed and substrate concentration) for an extracellular collagenase produced by Candida albicans URM3622 were studied using three experimental designs (one 2(6-2) fractionary factorial and two 2(3) full factorial). The analysis of the 2(6-2) fractionary design data indicated that agitation speed and substrate concentration had the most significant effect on collagenase production. Based on these results, two successive 2(3) full factorial design experiments were run in which the effects of substrate concentration, orbital agitation speed and pH were further studied. These two sets of experiments showed that all variables chosen were significant for the enzyme production, with the maximum collagenolytic activity of 6.8 +/- 0.4 U achieved at pH 7.0 with an orbital agitation speed of 160 rpm and 2% substrate concentration. Maximum collagenolytic activity was observed at pH 8.2 and 45 degrees C. The collagenase was stable within a pH range of 7.2-8.2 and over a temperature range of 28-45 degrees C. These results clearly indicate that C. albicans URM3622 is a potential resource for collagenase production and could be of interest for pharmaceutical, cosmetic and food industry. Studies were conducted at sites in south-cast Queensland, Australia, to investigate the effect of habitat modification for mosquito control on the distribution of eggshells of the salt marsh mosquito, Ochlerotatus vigilax (Skuse). Modifications were mainly tunnelling, but an Open Marsh Water Management (OMWM) site and a grid-ditched site were also included. There were two separate experimental designs: one was data collected Before and After (BA) modification and the other was for other sites with a Treatment and Control (TC) experimental design. For the BA data, there were significant reductions in eggshells after modification. Eggshells were generally fewer after modification in areas which were close to unrestricted tidal flushing. A sandy substrate and vegetation changes which resulted in reduced Sporobolus virginicus or mixed Sporobolus and Sarcocornia quinqueflora also contributed to the effect. In the TC experiment, there was no effect of modification at the tunnelled site, eggshells were fewer at the OMWM site, but there were more eggshells at the grid-ditched site. There was some general indication that recent oviposition activity was reduced in sites that had been modified, evidenced by a relatively small proportion of young (dark coloured) eggshells.

The aim of this study was to investigate whether the toxicity of saturated and polyunsaturated fatty acids (PUFA) on RINm5F cells is related to the phosphorylation state of Akt, ERK and PKC delta. The regulation of these kinases was compared in three experimental designs: (a) 4 h-exposure, (b) 4 h-exposure and a subsequent withdrawn of the FA for a 20 h period and (c) 24 h-exposure. Saturated and PUFA were toxic to RINm5F cells even at low concentrations. Also, evidence is provided for a late (i.e. the effect only appeared hours after the treatment) and a persistent regulation (i.e. maintenance of the effect for several hours) of Akt, ERK and PKC delta phosphorylation by the FA. Late activation of PKC delta seems important for palmitate cytotoxicity. Persistent activation of the survival proteins Akt and ERK by stearate, oleate and arachidonate might play an important role to prevent the toxic effect of posterior PKC delta activation. The results shown may explain why a short-period exposure to FA is not enough to induce cytotoxicity in pancreatic beta-cells, since survival pathways are activated. Besides, when this activation is persistent, it may overcome a posterior induction of death pathways. (c) 2008 Elsevier Ltd. All rights reserved.

A procura de piscinas para a prática de atividades desportivas, recreativas e/ou terapêuticas tem sofrido um aumento gradual ao longo do tempo. No entanto, nas piscinas existem vários perigos associados à sua utilização. Relativamente aos perigos químicos, a utilização de desinfetantes à base de cloro, bromo ou compostos derivados vai, por um lado, inativar microrganismos patogénicos mas, por outro, dar origem a subprodutos ao reagir com compostos orgânicos presentes na água. Os trihalometanos são um exemplo de subprodutos que se podem formar e, entre os compostos principais, estão o clorofórmio (TCM), bromodiclorometano (BDCM), clorodibromometano (CDBM) e bromofórmio (TBM). Este trabalho teve como objetivo o desenvolvimento de uma metodologia analítica para a determinação de trihalometanos em água e ar de piscinas e a sua aplicação a um conjunto de amostras. Para a análise dos compostos, foi utilizada a microextração em fase sólida no espaço de cabeça (HS-SPME) com posterior quantificação dos compostos por cromatografia gasosa com detetor de captura eletrónica (GC-ECD). Foi realizada uma otimização das condições de extração dos compostos em estudo em amostras de água, através da realização de dois planeamentos experimentais. As condições ótimas são assim obtidas para uma temperatura de extração de 45ºC, um tempo de extração de 25 min e um tempo de dessorção de 5 min. Foram analisadas amostras de águas de piscina cedidas pelo Centro de Estudos de Águas, sendo avaliada a aplicação da técnica HS-SPME e o efeito de matriz. O modo como se manuseiam as soluções que contêm os compostos em estudo influencia os resultados devido ao facto destes serem bastante voláteis. Concluiu-se também que existe efeito de matriz, logo a concentração das amostras deverá ser determinada através do método de adição de padrão. A caraterização da água de piscinas interiores permitiu conhecer a concentração de trihalometanos (THMs). Foram obtidas concentrações de TCM entre 4,5 e 406,5 μg/L sendo que apenas 4 das 27 amostras analisadas ultrapassam o valor limite imposto pelo Decreto-Lei nº306/2007 (100 μg/L) no que diz respeito a águas de consumo humano e que é normalmente utilizado como valor indicativo para a qualidade das águas de piscina. Relativamente à concentração obtida no ar de uma piscina interior, foi detetada uma concentração média de 224 μg/m3 de TCM, valor muito abaixo dos 10000 μg/m3 impostos pelo Decreto-lei nº24/2012, como valor limite para exposição profissional a agentes químicos.

ABSTRACT OBJECTIVE To analyze the scientific literature about the effects of exposure to psychosocial risk factors in work contexts. METHODS A systematic review was performed using the terms “psychosocial factors” AND “COPSOQ” in the databases PubMed, Medline, and Scopus. The period analyzed was from January 1, 2004 to June 30, 2012. We have included articles that used the Copenhagen Psychosocial Questionnaire (COPSOQ) as a measuring instrument of the psychosocial factors and the presentation of quantitative or qualitative results. German articles, psychometric studies or studies that did not analyze individual or work factors were excluded. RESULTS We included 22 articles in the analysis. Individual factors, such as gender, age, and socioeconomic status, were analyzed along with work-related factors such as labor demands, work organization and content, social relationships and leadership, work-individual interface, workplace values, justice and respect, personality, health and well-being, and offensive behaviors. We analyzed the sample type and the applied experimental designs. Some population groups, such as young people and migrants, are more vulnerable. The deteriorated working psychosocial environment is associated with physical health indicators and weak mental health. This environment is also a risk factor for the development of moderate to severe clinical conditions, predicting absenteeism or intention of leaving the job. CONCLUSIONS The literature shows the contribution of exposure to psychosocial risk factors in work environments and their impact on mental health and well-being of workers. It allows the design of practical interventions in the work context to be based on scientific evidences. Investigations in specific populations, such as industry, and studies with more robust designs are lacking.

The impending introduction of lead-free solder in the manufacture of electrical and electronic products has presented the electronics industry with many challenges. European manufacturers must transfer from a tin-lead process to a lead-free process by July 2006 as a result of the publication of two directives from the European Parliament. Tin-lead solders have been used for mechanical and electrical connections on printed circuit boards for over fifty years and considerable process knowledge has been accumulated. Extensive literature reviews were conducted on the topic and as a result it was found there are many implications to be considered with the introduction of lead-free solder. One particular question that requires answering is; can lead-free solder be used in existing manufacturing processes? The purpose of this research is to conduct a comparative study of a tin-lead solder and a lead-free solder in two key surface mount technology (SMT) processes. The two SMT processes in question were the stencil printing process and the reflow soldering process. Unreplicated fractional factorial experimental designs were used to carry out the studies. The quality of paste deposition in terms of height and volume were the characteristics of interest in the stencil printing process. The quality of solder joints produced in the reflow soldering experiment was assessed using x-ray and cross sectional analysis. This provided qualitative data that was then uniquely scored and weighted using a method developed during the research. Nested experimental design techniques were then used to analyse the resulting quantitative data. Predictive models were developed that allowed for the optimisation of both processes. Results from both experiments show that solder joints of comparable quality to those produced using tin-lead solder can be produced using lead-free solder in current SMT processes.

Most leadership and management researchers ignore one key design and estimation problem rendering parameter estimates uninterpretable: Endogeneity. We discuss the problem of endogeneity in depth and explain conditions that engender it using examples grounded in the leadership literature. We show how consistent causal estimates can be derived from the randomized experiment, where endogeneity is eliminated by experimental design. We then review the reasons why estimates may become biased (i.e., inconsistent) in non-experimental designs and present a number of useful remedies for examining causal relations with non-experimental data. We write in intuitive terms using nontechnical language to make this chapter accessible to a large audience.

A method to estimate DSGE models using the raw data is proposed. The approachlinks the observables to the model counterparts via a flexible specification which doesnot require the model-based component to be solely located at business cycle frequencies,allows the non model-based component to take various time series patterns, andpermits model misspecification. Applying standard data transformations induce biasesin structural estimates and distortions in the policy conclusions. The proposed approachrecovers important model-based features in selected experimental designs. Twowidely discussed issues are used to illustrate its practical use.

Schizotypy refers to a constellation of personality traits that are believed to mirror the subclinical expression of schizophrenia in the general population. Evidence from pharmacological studies indicates that dopamine is involved in the aetiology of schizophrenia. Based on the assumption of a continuum between schizophrenia and schizotypy, researchers have begun investigating the association between dopamine and schizotypy using a wide range of methods. In this article, we review published studies on this association from the following areas of work: (1) Experimental investigations of the interactive effects of dopaminergic challenges and schizotypy on cognition, motor control and behaviour, (2) dopaminergically supported cognitive functions, (3) studies of associations between schizotypy and polymorphisms in genes involved in dopaminergic neurotransmission, and (4) molecular imaging studies of the association between schizotypy and markers of the dopamine system. Together, data from these lines of evidence suggest that dopamine is important to the expression and experience of schizotypy and associated behavioural biases. An important observation is that the experimental designs, methods, and manipulations used in this research are highly heterogeneous. Future studies are required to replicate individual observations, to enlighten the link between dopamine and different schizotypy dimensions (positive, negative, cognitive disorganisation), and to guide the search for solid dopamine-sensitive behavioural markers. Such studies are important in order to clarify inconsistencies between studies. More work is also needed to identify differences between dopaminergic alterations in schizotypy compared to the dysfunctions observed in schizophrenia.
