In the event of a terrorist-mediated attack in the United States using radiological or improvised nuclear weapons, it is expected that hundreds of thousands of people could be exposed to life-threatening levels of ionizing radiation. We have recently shown that genome-wide expression analysis of the peripheral blood (PB) can generate gene expression profiles that can predict radiation exposure and distinguish the dose level of exposure following total body irradiation (TBI). However, in the event a radiation-mass casualty scenario, many victims will have heterogeneous exposure due to partial shielding and it is unknown whether PB gene expression profiles would be useful in predicting the status of partially irradiated individuals. Here, we identified gene expression profiles in the PB that were characteristic of anterior hemibody-, posterior hemibody- and single limb-irradiation at 0.5 Gy, 2 Gy and 10 Gy in C57Bl6 mice. These PB signatures predicted the radiation status of partially irradiated mice with a high level of accuracy (range 79-100%) compared to non-irradiated mice. Interestingly, PB signatures of partial body irradiation were poorly predictive of radiation status by site of injury (range 16-43%), suggesting that the PB molecular response to partial body irradiation was anatomic site specific. Importantly, PB gene signatures generated from TBI-treated mice failed completely to predict the radiation status of partially irradiated animals or non-irradiated controls. These data demonstrate that partial body irradiation, even to a single limb, generates a characteristic PB signature of radiation injury and thus may necessitate the use of multiple signatures, both partial body and total body, to accurately assess the status of an individual exposed to radiation.

BACKGROUND: Positive skin prick tests (SPT) for food allergens and specific IgE (sIgE) in serum indicate sensitization but do not enable distinction between sensitized but tolerant and clinically allergic patients. OBJECTIVE: Herein, we evaluate the clinical relevance of basophil activation tests (BATs) for peanut or egg allergy diagnosis. METHODS: Thirty-two peanut-allergic, 14 peanut-sensitized (sIgE(+) and/or SPT(+) to peanuts) but tolerant children and 29 controls with no history of an adverse reaction to peanuts were included. Similarly, 31 egg-allergic, 14 egg-sensitized children (sIgE(+) and/or SPT(+) to egg white) and 22 controls were studied. Flow cytometric analysis of CD63 expression or CD203c upregulation on basophils and the production of leukotrienes (LT) were performed in response to an in vitro crude peanut extract or ovalbumin (OVA) challenge. RESULTS: After in vitro peanut challenge, the basophils from peanut-allergic children showed significantly higher levels of activation than those from controls (P<0.001). After OVA challenge, a similar distinction (P<0.001) was observed between egg-allergics and controls. Interestingly, the majority of egg- or peanut-sensitized children failed to activate basophils, respectively, in response to OVA and peanut challenge. The sensitivity of the CD63, CD203c and LT assay was 86.7%, 89.5% and 76.0% with a specificity of 94.1%, 97.1% and 94.6% for peanut allergy diagnosis. The corresponding performances of BATs applied to egg allergy diagnosis were 88.9%, 62.5% and 77.8% for the sensitivity and 100%, 96.4% and 96.4% for the specificity. CONCLUSION: Neither conventional tests nor BATs are sensitive and specific enough to predict food allergy accurately. However, BATs may helpfully complete conventional tests, especially SPT, allowing improved discrimination between allergic and non-allergic individuals.

This paper presents a statistical-based fault diagnosis scheme for application to internal combustion engines. The scheme relies on an identified model that describes the relationships between a set of recorded engine variables using principal component analysis (PCA). Since combustion cycles are complex in nature and produce nonlinear relationships between the recorded engine variables, the paper proposes the use of nonlinear PCA (NLPCA). The paper further justifies the use of NLPCA by comparing the model accuracy of the NLPCA model with that of a linear PCA model. A new nonlinear variable reconstruction algorithm and bivariate scatter plots are proposed for fault isolation, following the application of NLPCA. The proposed technique allows the diagnosis of different fault types under steady-state operating conditions. More precisely, nonlinear variable reconstruction can remove the fault signature from the recorded engine data, which allows the identification and isolation of the root cause of abnormal engine behaviour. The paper shows that this can lead to (i) an enhanced identification of potential root causes of abnormal events and (ii) the masking of faulty sensor readings. The effectiveness of the enhanced NLPCA based monitoring scheme is illustrated by its application to a sensor fault and a process fault. The sensor fault relates to a drift in the fuel flow reading, whilst the process fault relates to a partial blockage of the intercooler. These faults are introduced to a Volkswagen TDI 1.9 Litre diesel engine mounted on an experimental engine test bench facility.

This brief examines the application of nonlinear statistical process control to the detection and diagnosis of faults in automotive engines. In this statistical framework, the computed score variables may have a complicated nonparametric distri- bution function, which hampers statistical inference, notably for fault detection and diagnosis. This brief shows that introducing the statistical local approach into nonlinear statistical process control produces statistics that follow a normal distribution, thereby enabling a simple statistical inference for fault detection. Further, for fault diagnosis, this brief introduces a compensation scheme that approximates the fault condition signature. Experimental results from a Volkswagen 1.9-L turbo-charged diesel engine are included.

Subspace monitoring has recently been proposed as a condition monitoring tool that requires considerably fewer variables to be analysed compared to dynamic principal component analysis (PCA). This paper analyses subspace monitoring in identifying and isolating fault conditions, which reveals that the existing work suffers from inherent limitations if complex fault senarios arise. Based on the assumption that the fault signature is deterministic while the monitored variables are stochastic, the paper introduces a regression-based reconstruction technique to overcome these limitations. The utility of the proposed fault identification and isolation method is shown using a simulation example and the analysis of experimental data from an industrial reactive distillation unit.

Fundus autofluorescence (AF) imaging by confocal scanning laser ophthalmoscopy has been widely used by ophthalmologists in the diagnosis/monitoring of various retinal disorders. It is believed that fundus AF is derived from lipofuscin in retinal pigment epithelial (RPE) cells; however, direct clinicopathological correlation has not been possible in humans. We examined fundus AF by confocal scanning laser ophthalmoscopy and confocal microscopy in normal C57BL/6 mice of different ages. Increasingly strong AF signals were observed with age in the neuroretina and subretinal/RPE layer by confocal scanning laser ophthalmoscopy. Unlike fundus AF detected in normal human subjects, mouse fundus AF appeared as discrete foci distributed throughout the retina. Most of the AF signals in the neuroretina were distributed around retinal vessels. Confocal microscopy of retinal and choroid/RPE flat mounts demonstrated that most of the AF signals were derived from Iba-1+ perivascular and subretinal microglia. An age-dependent accumulation of Iba-1+ microglia at the subretinal space was observed. Lipofuscin granules were detected in large numbers in subretinal microglia by electron microscopy. The number of AF+ microglia and the amount of AF granules/cell increased with age. AF granules/lipofuscin were also observed in RPE cells in mice older than 12 months, but the number of AF+ RPE cells was very low (1.48 mm-2 and 5.02 mm-2 for 12 and 24 months, respectively) compared to the number of AF+ microglial cells (20.63 mm-2 and 76.36 mm-2 for 6 and 24 months, respectively). The fluorescence emission fingerprints of AF granules in subretinal microglia were the same as those in RPE cells. Our observation suggests that perivascular and subretinal microglia are the main cells producing lipofuscin in normal aged mouse retina and are responsible for in vivo fundus AF. Microglia may play an important role in retinal aging and age-related retinal diseases.

Proton radiography using laser-driven sources has been developed as a diagnostic since the beginning of the decade, and applied successfully to a range of experimental situations. Multi-MeV protons driven from thin foils via the Target Normal Sheath Acceleration mechanism, offer, under optimal conditions, the possibility of probing laser-plasma interactions, and detecting electric and magnetic fields as well as plasma density gradients with similar to ps temporal resolution and similar to 5-10 mu m spatial resolution. In view of these advantages, the use of proton radiography as a diagnostic in experiments of relevance to Inertial Confinement Fusion is currently considered in the main fusion laboratories. This paper will discuss recent advances in the application of laser-driven radiography to experiments of relevance to Inertial Confinement Fusion. In particular we will discuss radiography of hohlraum and gasbag targets following the interaction of intense ns pulses. These experiments were carried out at the HELEN laser facility at AWE (UK), and proved the suitability of this diagnostic for studying, with unprecedented detail, laser-plasma interaction mechanisms of high relevance to Inertial Confinement Fusion. Non-linear solitary structures of relevance to space physics, namely phase space electron holes, have also been highlighted by the measurements. These measurements are discussed and compared to existing models.

This study presents a vibration-based health monitoring of short span bridges by
an inspection vehicle. How to screen health condition of short span bridges in terms of the
drive-by bridge inspection is described. Feasibility of the drive-by bridge inspection is
investigated through a scaled laboratory moving vehicle experiment. The feasibility of using an
instrumented vehicle to detect the natural frequency and changes in structural damping of a
model bridge is observed. Observations also demonstrate possibility of diagnosis of bridges by
comparing patterns of identified dynamic parameters of bridges through a periodical
monitoring. It is confirmed that the method for damage identification under a moving vehicle
identifies the damage location and severity well.

This paper presents the application of the on-load exciting current Extended Park's Vector Approach for diagnosing incipient turn-to-turn winding faults in operating power transformers. Experimental and simulated test results demonstrate the effectiveness of the proposed technique, which is based on the spectral analysis of the AC component of the on-load exciting current Park's Vector modulus.

O presente trabalho apresenta um estudo laboratorial que teve como finalidade o reconhecimento de perda de propriedades da madeira de edifícios antigos quando degradada por carunchos. Foi estudada madeira antiga de pinho e de choupo, com idades compreendidas entre 100 e 200 anos. Como abordagem inicial são apresentadas as características da madeira e dos edifícios pombalinos e gaioleiros onde esta era bastante usada, não só como elemento deacabamento, mas principalmente como elemento estrutural. São apresentados os vários fatores que levam à degradação da madeira assim como alguns métodos de avaliação e diagnóstico dos estados de conservação dos elementos de madeira que se encontram nos edifícios. É também desenvolvido um estudo sobre o caruncho grande e caruncho pequeno, seu ciclo de vida e forma como degrada a madeira, servindo de base ao estudo laboratorial. No desenvolvimento foi avaliado o estado de degradação de provetes de madeira antiga com 30 x 30 x 90 mm e de seguida correlacionado com a sua resistência à compressão, o seu módulo de elasticidade e a extensão em fase plástica. Desta forma pretende-se estudar o modo como os diferentes estados de degradação por caruncho influenciam as caraterísticas mecânicas das peças de madeira.

La schizophrénie est une psychopathologie largement hétérogène caractérisée entre autres par d’importantes défaillances dans le fonctionnement cognitif et émotionnel. En effet, par rapport à la population générale, forte proportion de ces individus présentent une mémoire déficitaire pour les événements émotionnels. À ce jour, le peu d’études qui se sont penchées sur la mémoire émotionnelle épisodique dans la schizophrénie, ont uniquement mis l’emphase sur l'effet de la valence des stimuli (c’est-à-dire le caractère agréable ou désagréable du stimulus). Toutefois, aucune n’a investigué spécifiquement l’intensité de la réaction aux stimuli (c’est-à-dire une faible par rapport à une forte réaction) malgré quantité de preuves faisant montre, dans la population générale, de différents processus de mémoire émotionnelle pour des stimuli suscitant une forte réaction par rapport à ceux évoquant une faible réponse. Ce manque est d’autant plus flagrant étant donné le nombre d’études ayant rapporté un traitement et un encodage atypiques des émotions spécifiquement au niveau de l’intensité de la réponse subjective chez des patients atteints de schizophrénie. Autre fait important, il est étonnant de constater l’absence de recherches sur les différences de sexe dans la mémoire émotionnelle étant donné l’ensemble des divergences entre hommes et femmes atteints de schizophrénie au niveau de la prévalence, de l’âge de diagnostic, de la manifestation clinique, de l’évolution de la maladie, de la réponse au traitement et des structures cérébrales. Pour pallier à ces lacunes, ce mémoire a évalué : (1) l’effet de la valence des stimuli et de l'intensité de la réaction émotionnelle au niveau des fonctions cérébrales correspondant à la mémoire émotionnelle chez des patients atteints de schizophrénie comparativement à des participants sains; et (2) les possibles différences de sexe dans les processus cérébraux impliqués dans la mémoire émotionnelle chez des patients atteints de schizophrénie par rapport à des volontaires sains. Ainsi, la première étude a comparé les activations cérébrales de patients atteints de schizophrénie par rapport à des participants sains au cours d’une tâche de mémoire émotionnelle dont les stimuli variaient à la fois au niveau de la valence et de l'intensité de la réaction subjective. 37 patients atteints de schizophrénie ainsi que 37 participants en bonne santé ont effectué cette tâche de mémoire émotionnelle lors d’une session d’imagerie par résonance magnétique fonctionnelle (IRMf). Pour toutes les conditions étudiées (images négatives, positives, de faible et de forte intensité), le groupe atteint de schizophrénie a performé significativement moins bien que les volontaires sains. Comparativement aux sujets sains, ils ont montré moins d’activations cérébrales dans les régions limbiques et préfrontales lors de la reconnaissance des images négatives, mais ont présenté un patron d'activations similaire à celui des participants sains lors de la reconnaissance des images chargées positivement (activations observées dans le cervelet, le cortex temporal et préfrontal). Enfin, indépendamment de la valence des stimuli, les deux groupes ont démontré une augmentation des activations cérébrales pour les images de forte intensité par rapport à celles de plus faible intensité. La seconde étude a quant à elle exploré les différences de sexe potentielles au niveau des activations cérébrales associées à la mémoire émotionnelle dans la schizophrénie et dans la population en général. Nous avons comparé 41 patients atteints de schizophrénie (20 femmes) à 41 participants en bonne santé (19 femmes) alors qu’ils effectuaient la même tâche de mémoire émotionnelle mentionnée plus haut. Or, pour cette étude, nous nous sommes concentrés sur les conditions suivantes : la reconnaissance d’images positives, négatives et neutres. Nous n'avons pas observé de différences entre les hommes et les femmes au niveau des performances à la tâche de mémoire pour aucune des conditions. En ce qui a trait aux données de neuroimagerie, comparativement aux femmes en bonne santé, celles atteintes de schizophrénie ont montré une diminution des activations cérébrales dans les régions corticales du système limbique (p. ex. cortex cingulaire moyen) et dans les régions sous-corticales (p. ex. amygdale) lors de la reconnaissance d'images négatives. Pour ce qui est de la condition positive, elles ont présenté, comparativement au groupe de femmes saines, des diminutions d’activations spécifiquement dans le cervelet ainsi que dans le gyrus frontal inférieur et moyen. Les hommes atteints de schizophrénie, eux, ont montré une augmentation d’activations par rapport aux hommes sains dans le gyrus préfrontal médian lors de la reconnaissance des stimuli négatifs ; ainsi que dans les régions pariétales, temporales et limbiques lors de la reconnaissance des stimuli positifs. Dans un autre ordre d’idées, notre analyse corrélationnelle a mis en évidence, chez les femmes, un lien significatif entre l’activité cérébrale et les symptômes au cours de la mémoire des stimuli positifs, alors que chez les hommes atteints schizophrénie, ce lien a été observé au cours de la mémoire des stimuli négatifs. Bref, l’ensemble de nos résultats suggère, chez les patients atteints de schizophrénie, un fonctionnement cérébral atypique spécifiquement lors de la reconnaissance d’images négatives, mais un fonctionnement intact lors de la reconnaissance de stimuli positifs. De plus, nous avons mis en évidence la présence de différences de sexe dans les activations cérébrales associées à la mémoire épisodique émotionnelle soulignant ainsi l'importance d’étudier séparément les hommes et les femmes atteints de schizophrénie dans le cadre de recherches sur les plans cognitif et émotionnel.

Estudiar los errores conceptuales cometidos por alumnos de ciclo superior de EGB. 31 alumnos de octavo, 64 de séptimo y 21 de sexto de EGB. En la primera parte, se presenta una síntesis teórica de autores como Wandersee, Mintzes y Novak relativa a los errores conceptuales de los alumnos de ciencias y una síntesis de las principales investigaciones llevadas a cabo en este ámbito como apoyatura conceptual y metodológica del trabajo. La parte experimental consiste en la doble aplicación de un test de 15 cuestiones relativas al agua y sus transformaciones en mayo de 1994 y en junio del curso siguiente, tras un periodo de instrucción específica para detectar los errores conceptuales al final de dos cursos consecutivos. Mapa conceptual de los temas clave para comprender el ciclo del agua y diagrama en V del proceso de cambio de estados del agua. En el cuestionario 93-94 la tasa de error es del 80 por ciento y en el de 94-95 del 60 por ciento. El mismo método se ha empleado para el test de Matemáticas.

Details about the parameters of kinetic systems are crucial for progress in both medical and industrial research, including drug development, clinical diagnosis and biotechnology applications. Such details must be collected by a series of kinetic experiments and investigations. The correct design of the experiment is essential to collecting data suitable for analysis, modelling and deriving the correct information. We have developed a systematic and iterative Bayesian method and sets of rules for the design of enzyme kinetic experiments. Our method selects the optimum design to collect data suitable for accurate modelling and analysis and minimises the error in the parameters estimated. The rules select features of the design such as the substrate range and the number of measurements. We show here that this method can be directly applied to the study of other important kinetic systems, including drug transport, receptor binding, microbial culture and cell transport kinetics. It is possible to reduce the errors in the estimated parameters and, most importantly, increase the efficiency and cost-effectiveness by reducing the necessary amount of experiments and data points measured.
