The glucocorticoid (GC) receptor (GR) and Kruppel-like factor Klf4 are transcription factors that play major roles in skin homeostasis. However, whether these transcription factors cooperate in binding genomic regulatory regions in epidermal keratinocytes was not known. Here, we show that in dexamethasone-treated keratinocytes GR and Klf4 are recruited to genomic regions containing adjacent GR and KLF binding motifs to control transcription of the anti-inflammatory genes Tsc22d3 and Zfp36. GR- and Klf4 loss of function experiments showed total GR but partial Klf4 requirement for full gene induction in response to dexamethasone. In wild type keratinocytes induced to differentiate, GR and Klf4 protein expression increased concomitant with Tsc22d3 and Zfp36 up-regulation. In contrast, GR-deficient cells failed to differentiate or fully induce Klf4, Tsc22d3 and Zfp36 correlating with increased expression of the epithelium-specific Trp63, a known transcriptional repressor of Klf4. The identified transcriptional cooperation between GR and Klf4 may determine cell-type specific regulation and have implications for developing therapies for skin diseases.

The Sp/KLF transcription factors perform a variety of biological functions, but are related in that they bind GC-box and CACCC-box sequences in DNA via a highly conserved DNA-binding domain. A database homology search, using the zinc finger DNA-binding domain characteristic of the family, has identified human KLF17 as a new family member that is most closely related to KLFs 1-8 and 12. KLF17 appears to be the human orthologue of the previously reported mouse gene, zinc finger protein 393 (Zfp393), although it has diverged significantly. The DNA-binding domain is the most conserved region, suggesting that both the murine and the human forms recognize the same binding sites in DNA and may retain similar functions. We show that human KLF17 can bind G/C-rich sites via its zinc fingers and is able to activate transcription from CACCC-box elements. This is the first report of the DNA-binding characteristics and transactivation activity of human KLF17, which, together with the homology it displays to other KLF proteins, put it in the Sp/KLF family. Gene knockout studies of Kruppel-like factors (KLFs) in mice have shown essential roles in organogenesis. A screen for KLF family members in zebrafish identified many KLFs. One of these, zebrafish KLF4 (zKLF4) is the homologue of neptune, a Xenopus laevis KLF. zKLF4 is expressed from approximately 80% epiboly a patch of dorsal/anterior mesendodermal cells called the pre-polster and, subsequently, in the polster and hatching gland. Here we investigate the function of zKLF4 using morpholino-based antisense oligonucleotides. Knockdown of zKLF4 resulted in complete absence of hatching gland formation and subsequent hatching in zebrafish. In addition, there was early knockdown of expression of the pre-polster/anterior mesendoderm markers CatL, cap1, and BMP4. These results indicate zKLF4 is expressed within the pre-polster, an early mesendodermal site, and that it plays a critical role in the differentiation of these cells into hatching gland cells.

Except for the complement C1q, the immunological functions of other C1q family members have remained unclear. Here we describe zebrafish C1q-like, whose transcription and translation display a uniform distribution in early embryos, and are restricted to mid-hind brain and eye in later embryos. In vitro studies showed that C1q-like could inhibit the apoptosis induced by ActD and CHX in EPC cells, through repressing caspase 3/9 activities. Moreover, its physiological roles were studied by morpholino-mediated knockdown in zebrafish embryogenesis. In comparison with control embryos, the C1q-like knockdown embryos display obvious defects in the head and cramofacial development mediated through p53-induced apoptosis, which was confirmed by the in vitro transcribed C1q-like mRNA or p53 MO co-injection. TUNEL assays revealed extensive cell death, and caspase 3/9 activity measurement also revealed about two folds increase in C1q-like morphant embryos, which was inhibited by p53 MO co-injection. Real-time quantitative PCR showed the up-regulation expression of several apoptosis regulators such as p53, mdm2, p21, Box and caspase 3, and down-regulation expression of hbae1 in the C1q-like morphant embryos. Knockdown of C1q-like in zebrafish embryos decreased hemoglobin production and impaired the organization of mesencephalic vein and other brain blood vessels. Interestingly, exposure of zebrafish embryos to UV resulted in an increase in mRNA expression of C1q-like, whereas over-expression of C1q-like was not enough resist to the damage. Furthermore, C1q-like transcription was up-regulated in response to pathogen Aeromonas hydrophila, and embryo survival significantly decreased in the C1q-like morphants after exposure to the bacteria. The data suggested that C1q-like might play an antiapoptotic and protective role in inhibiting p53-dependent and caspase 3/9-mediated apoptosis during embryogenesis, especially in the brain development, and C1q-like should be a novel regulator of cell survival during zebrafish embryogenesis. Extracellular superoxide dismutase (ECSOD) is a major extracellular antioxidant enzyme that protects organs from damage by reactive oxygen species (ROS). We cloned a novel ECSOD from the bay scallop Argopecten irradians (AiECSOD) by 3' and 5' RACE. The full-length cDNA of AiECSOD was 893 bp with a 657 bp open reading frame encoding 218 amino acids. The deduced amino acid sequence contained a putative signal peptide of 20 amino acids, and sequence comparison showed that AiECSOD had low degree of homology to ECSODs of other organisms. The genomic length of the AiECSOD gene was about 5276 bp containing five exons and six introns. The promoter region contained many putative transcription factor binding sites such as c-Myb, Oct-1, Sp1, Kruppel-like, c-ETS, NF kappa B, GATA-1, AP-1, and Ubx binding sites. Furthermore, tissue-specific expressions of AiECSOD and temporal expressions of AiECSOD in haemocytes of bay scallops challenged with bacteria Vibrio anguillarum were quantified using qRT-PCR. High levels of expression were detected in haemocytes, but not in gonad and mantle. The expression of AiECSOD reached the highest level at 12 h post-injection with V. anguillarum and then returned to normal between 24 h and 48 h post-injection. These results indicated that AiECSOD was an inducible protein and that it may play an important role in the immune responses against V anguillarum. Crown Copyright (C) 2008 Published by Elsevier Ltd. Cells are fundamental units of life, but little is known about evolution of cell states. Induced pluripotent stem cells (iPSCs) are once differentiated cells that have been re-programmed to an embryonic stem cell-like state, providing a powerful platform for biology and medicine. However, they have been limited to a few mammalian species. Here we found that a set of four mammalian transcription factor genes used to generate iPSCs in mouse and humans can induce a partially reprogrammed pluripotent stem cell (PRPSCs) state in vertebrate and invertebrate model organisms, in mammals, birds, fish, and fly, which span 550 million years from a common ancestor. These findings are one of the first to show cross-lineage stem cell-like induction, and to generate pluripotent-like cells for several of these species with in vivo chimeras. LYRIC/AEG-1 and its altered expression have been linked to carcinogenesis in prostate, brain and melanoma as well as promoting chemoresistance and metastasis in breast cancer. LYRIC/AEG-1 function remains unclear, although LYRIC/AEG-1 is activated by oncogenic HA-RAS, through binding of c-myc to its promoter, which in turn regulates the key components of the PI3-kinase and nuclear factor-kappaB pathways. We have identified the transcriptional repressor PLZF as an interacting protein of LYRIC/AEG through a yeast two-hybrid screen. PLZF regulates the expression of genes involved in cell growth and apoptosis including c-myc. Coexpression of LYRIC/AEG-1 with PLZF leads to a reduction in PLZF-mediated repression by reducing PLZF binding to promoters. We have confirmed that nuclear LYRIC/AEG-1 and PLZF interact in mammalian cells via the N- and C termini of LYRIC/AEG-1 and a region C terminal to the RD2 domain of PLZF. Both proteins colocalize to nuclear bodies containing histone deacetylases, which are known to promote PLZF-mediated repression. Our data suggest one mechanism for cells with altered LYRIC/AEG-1 expression to evade apoptosis and increase cell growth during tumourigenesis through the regulation of PLZF repression.

La régulation transcriptionnelle des gènes est cruciale pour permettre le bon fonctionnement des cellules. Afin que les cellules puissent accomplir leurs fonctions, les gènes doivent être exprimés adéquatement dans le bon type cellulaire et au stade de développement et de différenciation approprié. Un dérèglement dans l’expression de un ou plusieurs gènes peut entraîner de graves conséquences sur le destin de la cellule. Divers éléments en cis (ex : promoteurs et enhancers) et en trans (machinerie transcriptionnelle et facteurs de transcription) sont impliqués dans la régulation de la transcription. Les gènes du locus humain beta-globine (hub) sont exprimés dans les cellules érythroïdes et sont finenement régulés lors du développement et de la différenciation. Des mutations dans différentes régions du locus causent entre autres les beta-thalassémies. Nous avons utilisé ce modèle bien caractérisé afin d’étudier différents mécanismes de régulation favorisés par les facteurs de transcription qui sont exprimés dans les cellules érythroïdes. Nous nous sommes intéressés à l’importance de l’élément en cis HS2 du Locus control region. Cet élément possède plusieurs sites de liaison pour des facteurs de transcription impliqués dans la régulation des gènes du locus hub. Nos résultats montrent que HS2 possède un rôle dans l’organisation de la chromatine du locus qui peut être dissocié de son rôle d’enhancer. De plus, HS2 n’est pas essentiel pour l’expression à haut niveau du gène beta alors qu’il est important pour l’expression des gènes gamma. Ceci suggère que le recrutement des différents facteurs au site HS2 lors du développement influence différement les gènes du locus. Dans un deuxième temps, nous avons investigué l’importance de HS2 lors de la différenciation des cellules érythroïdes. Il avait été rapporté que l’absence de HS2 influence grandement la potentialisation de la chromatine du gène beta. La potentialisation dans les cellules progénitrices favorise l’activation transcriptionnelle du gène dans les cellules matures. Nous avons caractérisé le recrutement de différents facteurs de transcription au site HS2 et au promoteur beta dans les cellules progénitrices hématopoïétiques (CPH) ainsi que dans les cellules érythroïdes matures. Nos résultats montrent que le facteur EKLF est impliqué dans la potentialisation de la chromatine et favorise le recrutement des facteurs BRG1, p45 et CBP dans les CPH. L’expression de GATA-1 dans les cellules érythroïdes matures permet le recrutement de GATA-1 au locus hub dans ces cellules. Ces données suggèrent que la combinaison de EKLF et GATA-1 est requise pour permettre une activation maximale du gène beta dans les cellules érythroïdes matures. Un autre facteur impliqué dans la régulation du locus hub est Ikaros. Nous avons étudié son recrutement au locus hub et avons observé que Ikaros est impliqué dans la répression des gènes gamma. Nos résultats montrent aussi que GATA-1 est impliqué dans la répression de ces gènes et qu’il interagit avec Ikaros. Ensemble, Ikaros et GATA-1 favorisent la formation d’un complexe de répression aux promoteurs gamma. Cette étude nous a aussi permis d’observer que Ikaros et GATA-1 sont impliqués dans la répression du gène Gata2. De façon intéressante, nous avons caractérisé le mécanisme de répression du gène Hes1 (un gène cible de la voie Notch) lors de la différenciation érythroïde. Similairement à ce qui a été observé pour les gènes gamma, Hes1 est aussi réprimé par Ikaros et GATA-1. Ces résultats suggèrent donc que la combinaison de Ikaros et GATA-1 est associée à la répression de plusieurs de gènes dans les cellules érythroïdes. Globalement cette thèse rapporte de nouveaux mécanismes d’action de différents facteurs de transcription dans les cellules érythroïdes. Particulièrement, nos travaux ont permis de proposer un modèle pour la régulation des gènes du locus hub lors du développement et de la différenciation. De plus, nous rapportons pour la première fois l’importance de la collaboration entre les facteurs Ikaros et GATA-1 dans la régulation transcriptionnelle de gènes dans les cellules érythroïdes. Des mutations associées à certains des facteurs étudiés ont été rapportées dans des cas de beta-thalassémies ainsi que de leucémies. Nos travaux serviront donc à avoir une meilleure compréhension des mécanismes d’action de ces facteurs afin de potentiellement pouvoir les utiliser comme cibles thérapeutiques.

Introduction: L'arthrose est caractérisée par une destruction progressive du cartilage, une inflammation synoviale, et un remodelage de l’os sous-chondral avec une production excessive des médiateurs inflammatoires et cataboliques. Nous avons démontré que le niveau du 4-hydroxynonénal (4-HNE), un produit de la peroxydation lipidique, est augmenté dans le cartilage humain arthrosique sans qu’on sache le mécanisme exacte impliqué dans l’augmentation de cette molécule. Des données de la littérature indiquent que l’accumulation du HNE est contrôlée par l’action de la glutathione S-transférase A4-4 (GSTA4-4), une enzyme impliquée dans la détoxification du HNE. Au niveau transcriptionel, l’expression de cette enzyme est régulée par la transactivation du facteur de transcription Nrf2. Objectif: L’objectif de cette étude vise à démontrer que l’augmentation du HNE dans le cartilage arthrosique est attribuée, en partie, à l’altération de l’expression de la GSTA4-4 et de Nrf2. Méthode: Le niveau d’expression de la GSTA4-4 et de Nrf2 a été mesurée par Western blot et par PCR en temps réel dans le cartilage humain arthrosique et dans le cartilage provenant des souris atteintes d’arthrose. Pour démontrer le rôle du Nrf2 dans l’arthrose, les chondrocytes humains arthrosiques ont été traités par l’interleukine 1beta (IL-1β) ou par le H2O2 en présence ou en absence des activateurs du Nrf2 tels que le Protandim®, AI, et du 6-Gingérol. Par ailleurs, les chondrocytes ont été transfectés par un vecteur d’expression de Nrf2 puis traités par l’IL-β. En utilisant le modèle d’arthrose chez la souris, les animaux ont été traités par voie orale de 10 mg/kg/jour de Protandim® pendant 8 semaines. Résultats: Nous avons observé une diminution significative de l’expression de la GSTA4-4 et de Nrf2 dans le cartilage humain et murin arthrosique. L'activation de Nrf2 bloque la stimulation de la métalloprotéinase-13 (MMP-13), la prostaglandine E2 (PGE2) et de l'oxyde nitrique (NO) par l’IL-1β. En outre, nous avons montré que l'activation Nrf2 protège les cellules contre la mort cellulaire induite par H2O2. Fait intéressant, l'administration orale de Protandim® réduit la production du HNE par l'intermédiaire de l’activation de la GSTA4. Nous avons démontré que le niveau d’expression de la GSTA4-4 et de Nrf2 diminue dans le cartilage provenant des patients et des souris atteints d’arthrose. De plus, la surexpression de ce facteur nucléaire Nrf2 empêche la production du HNE et la MMP-13 et l’inactivation de la GSTA4-4. Dans notre modèle expérimental d’arthrose induite par déstabilisation du ménisque médial chez la souris, nous avons trouvé que l'administration orale de Protandim® à 10 mg / kg / jour réduit les lésions du cartilage. Conclusion: Cette étude est de la première pour démontrer le rôle physiopathologique du Nrf2 in vitro et in vivo. Nos résultats démontrent que l’activation du Nrf2 est essentielle afin de maintenir l’expression de la GSTA4-4 et de réduire le niveau du HNE. Le fait que les activateurs du Nrf2 abolissent la production de la HNE et aussi un certain nombre de facteurs connus pour être impliqués dans la pathogenèse de l’arthrose les rend des agents cliniquement utiles pour la prévention de la maladie.

MicroRNA miR-199a-5p impairs tight junction formation leading to increased urothelial permeability in bladder pain syndrome. Now using transcriptome analysis in urothelial TEU-2 cells we implicate it in the regulation of cell cycle, cytoskeleton remodeling, TGF and Wnt signaling pathways. MiR-199a-5p is highly expressed in the smooth muscle layer of the bladder and we altered its levels in bladder smooth muscle cells (SMC) to validate the pathway analysis. Inhibition of miR-199a-5p with antimiR increased SMC proliferation, reduced cell size and up-regulated miR-199a-5p targets, including Wnt2. Overexpression of Wnt2 protein or treating SMCs with recombinant Wnt2 closely mimicked the miR-199a-5p inhibition, whereas down-regulation of Wnt2 in antimiR-expressing SMCs with shRNA restored cell phenotype and proliferation rates. Overexpression of miR-199a-5p in the bladder SMCs significantly increased cell size and up-regulated SM22, SM alpha-actin and SM myosin heavy chain mRNA and protein levels. These changes, as well as increased expression of ACTG2, TGFB1I1, and CDKN1A were mediated by up-regulation of smooth muscle-specific transcriptional activator myocardin at mRNA and protein levels. Myocardin-related transcription factor (MRTF-A) downstream targets Id3 and MYL9 were also induced. Up-regulation of myocardin was accompanied by down-regulation of Wnt-dependent inhibitory Kruppel-like transcription factor 4 (KLF4) in miR-199a-5p overexpressing cells. In contrast, KLF4 was induced in antimiR-expressing cells following the activation of Wnt2 signaling, leading to repression of myocardin-dependent genes. MiR-199a-5p plays a critical role in the Wnt2-mediated regulation of proliferative and differentiation processes in the smooth muscle and may behave as a key modulator of smooth muscle hypertrophy, relevant for organ remodeling.

The mammalian Forkhead Box (Fox) transcription factor (FoxM1) is implicated in tumorgenesis. However, the role and regulation of FoxM1 in gastric cancer remain unknown.^ I examined FoxM1 expression in 86 cases of primary gastric cancer and 57 normal gastric tissue specimens. I found weak expression of FoxM1 protein in normal gastric mucosa, whereas I observed strong staining for FoxM1 in tumor-cell nuclei in various gastric tumors and lymph node metastases. The aberrant FoxM1 expression is associated with VEGF expression and increased angiogenesis in human gastric cancer. A Cox proportional hazards model revealed that FoxM1 expression was an independent prognostic factor in multivariate analysis. Furthermore, overexpression of FoxM1 by gene transfer significantly promoted the growth and metastasis of gastric cancer cells in orthotopic mouse models, whereas knockdown of FoxM1 expression by small interfering RNA did the opposite. Next, I observed that alteration of tumor growth and metastasis by elevated FoxM1 expression was directly correlated with alteration of VEGF expression and angiogenesis. In addition, promotion of gastric tumorigenesis by FoxM1 directly and significantly correlated with transactivation of vascular endothelial growth factor (VEGF) expression and elevation of angiogenesis. ^ To further investigate the underlying mechanisms that result in FoxM1 overexpression in gastric cancer, I investigated FoxM1 and Krüppel-like factor 4 (KLF4) expressions in primary gastric cancer and normal gastric tissue specimens. Concomitance of increased expression of FoxM1 protein and decreased expression of KLF4 protein was evident in human gastric cancer. Enforced KLF4 expression suppressed FoxM1 protein expression. Moreover, a region within the proximal FoxM1 promoter was identified to have KLF4-binding sites. Finally, I found an increased FoxM1 expression in gastric mucosa of villin-Cre -directed tissue specific Klf4-null mice.^ In summary, I offered both clinical and mechanistic evidence that dysregulated expression of FoxM1 play an important role in gastric cancer development and progression, while KLF4 mediates negative regulation of FoxM1 expression and its loss significantly contributes to FoxM1 dysregulation. ^

Nitric oxide (NO) is known to have various biologic and pathophysiologic effects on organisms. The molecular mechanisms by which NO exerts harmful effects are unknown, although various O2 radicals and ions that result from reactivity of NO are presumed to be involved. Here we report that adaptive cellular response controlled by the transcription factor hypoxia-inducible factor 1 (HIF-1) in hypoxia is suppressed by NO. Induction of erythropoietin and glycolytic aldolase A mRNAs in hypoxically cultured Hep3B cells, a human hepatoma cell line, was completely and partially inhibited, respectively, by the addition of sodium nitroprusside (SNP), which spontaneously releases NO. A reporter plasmid carrying four hypoxia-response element sequences connected to the luciferase structural gene was constructed and transfected into Hep3B cells. Inducibly expressed luciferase activity in hypoxia was inhibited by the addition of SNP and two other structurally different NO donors, S-nitroso-l-glutathione and 3-morpholinosydnonimine, giving IC50 values of 7.8, 211, and 490 μM, respectively. Inhibition by SNP was also observed in Neuro 2A and HeLa cells, indicating that the inhibition was not cell-type-specific. The vascular endothelial growth factor promoter activity that is controlled by HIF-1 was also inhibited by SNP (IC50 = 6.6 μM). Induction generated by the addition of cobalt ion (this treatment mimics hypoxia) was also inhibited by SNP (IC50 = 2.5 μM). Increased luciferase activity expressed by cotransfection of effector plasmids for HIF-1α or HIF-1α-like factor in hypoxia was also inhibited by the NO donor. We also showed that the inhibition was performed by blocking an activation step of HIF-1α to a DNA-binding form.

To explore the possible involvement of STAT factors ("signal transducers and activators of transcription") in the interleukin 2 receptor (IL-2R) signaling cascade, murine HT-2 cells expressing chimeric receptors composed of the extracellular domain of the erythropoietin receptor fused to the cytoplasmic domains of the IL-2R beta or -gamma c chains were prepared. Erythropoietin or IL-2 activation of these cells resulted in rapid nuclear expression of a DNA-binding activity that reacted with select STAT response elements. Based on reactivity with specific anti-STAT antibodies, this DNA-binding activity was identified as a murine homologue of STAT-5. Induction of nuclear expression of this STAT-5-like factor was blocked by the addition of herbimycin A, a tyrosine kinase inhibitor, but not by rapamycin, an immunophilin-binding antagonist of IL-2-induced proliferation. The IL-2R beta chain appeared critical for IL-2-induced activation of STAT-5, since a mutant beta chain lacking all cytoplasmic tyrosine residues was incapable of inducing this DNA binding. In contrast, a gamma c mutant lacking all of its cytoplasmic tyrosine residues proved fully competent for the induction of STAT-5. Physical binding of STAT-5 to functionally important tyrosine residues within IL-2R beta was supported by the finding that phosphorylated, but not nonphosphorylated, peptides corresponding to sequences spanning Y392 and Y510 of the IL-2R beta tail specifically inhibited STAT-5 DNA binding.

Insulin-like peptide 3 (INSL3), a member of the relaxin peptide family, is produced in testicular Leydig cells and ovarian thecal cells. Gene knock-out experiments have identified a key biological role in initiating testes descent during fetal development. Additionally, INSL3 has an important function in mediating male and female germ cell function. These actions are elicited via its recently identified receptor, LGR8, a member of the leucine-rich repeat-containing G-protein- coupled receptor family. To identify the structural features that are responsible for the interaction of INSL3 with its receptor, its solution structure was determined by NMR spectroscopy together with in vitro assays of a series of B-chain alanine-substituted analogs. Synthetic human INSL3 was found to adopt a characteristic relaxin/ insulin-like fold in solution but is a highly dynamic molecule. The four termini of this two-chain peptide are disordered, and additional conformational exchange is evident in the molecular core. Alanine-substituted analogs were used to identify the key residues of INSL3 that are responsible for the interaction with the ectodomain of LGR8. These include Arg(B16) and Val(B19), with His(B12) and Arg(B20) playing a secondary role, as evident from the synergistic effect on the activity in double and triple mutants involving these residues. Together, these amino acids combine with the previously identified critical residue, Trp(B27), to form the receptor binding surface. The current results provide clear direction for the design of novel specific agonists and antagonists of this receptor.

L'exposition aux ultraviolets, à la pollution, aux substances carcinogènes, à la fumée de cigarette et la pratique intensive d'activités sportives sont autant de causes de stress oxydant pour l'organisme. Ces dommages sont associés à plusieurs maladies et conditions pathologiques telles que cancers, diabètes, infections pulmonaires et maladies neurodégénératives. L'élément de réponse antioxydant (ARE) est un des composants principaux des défenses de la cellule contre ce phénomène. Ce promoteur agit sous le contrôle de Nrf2 (Nuclear factor erythroid 2-related factor 2). Une stratégie populaire pour l'activation de ce mécanisme est l'utilisation d'inducteurs covalents. Ces molécules agissent par la formation de liens covalents avec les nombreux résidus cystéine de Keap1 (Kelch-like ECH-associated protein 1), une protéine chaperonne qui contrôle l'activité de Nrf2. Cette thèse présente la synthèse, les propriétés biologiques et l'étude des relations structure-activité d'une librairie d'électrophiles capables d'induire la transcription des gènes cibles de la voie de signalisation Keap1/Nrf2/ARE. Le premier volet fait état de la comparaison d'une variété d'électrophiles simples pour étudier les préférences de la cible. Le deuxième volet montre que la présence d'une seconde fonction capable de piéger un résidu cystéine fournit des analogues très puissants.
