En aquest projecte es desenvolupa una distribució GNU/Linux adaptada als laboratoris de docència del departament d’enginyeria de la informació i de les comunicacions. Partint de una anàlisi exhaustiu de les necessitats del departament i gràcies a la col·laboració dels seus membres s’ha realitzat la distribució dEICbian, un sistema que permetrà facilitar les tasques docents als laboratoris del departament. Amb l’objectiu de mantenir el sistema actualitzat també s’ha desenvolupat un generador de noves versions de la distribució dEICbian per tal de garantir tenir, sempre, un sistema modern i lliure d’errors als laboratoris del departament. L’opinió dels usuaris, expressada mitjançant enquestes, ha estat favorable i ara només resta el darrer pas: la seva implementació als laboratoris.

UEV proteins are enzymatically inactive variants of the E2 ubiquitin-conjugating enzymes that regulate noncanonical elongation of ubiquitin chains. In Saccharomyces cerevisiae, UEV is part of the RAD6-mediated error-free DNA repair pathway. In mammalian cells, UEV proteins can modulate c-FOS transcription and the G2-M transition of the cell cycle. Here we show that the UEV genes from phylogenetically distant organisms present a remarkable conservation in their exon–intron structure. We also show that the human UEV1 gene is fused with the previously unknown gene Kua. In Caenorhabditis elegans and Drosophila melanogaster, Kua and UEV are in separated loci, and are expressed as independent transcripts and proteins. In humans, Kua and UEV1 are adjacent genes, expressed either as separate transcripts encoding independent Kua and UEV1 proteins, or as a hybrid Kua–UEV transcript, encoding a two-domain protein. Kua proteins represent a novel class of conserved proteins with juxtamembrane histidine-rich motifs. Experiments with epitope-tagged proteins show that UEV1A is a nuclear protein, whereas both Kua and Kua–UEV localize to cytoplasmic structures, indicating that the Kua domain determines the cytoplasmic localization of Kua–UEV. Therefore, the addition of a Kua domain to UEV in the fused Kua–UEV protein confers new biological properties to this regulator of variant polyubiquitination.[Kua cDNAs isolated by RT-PCR and described in this paper have been deposited in the GenBank data library under accession nos. We investigate the problem of finding minimum-distortion policies for streaming delay-sensitive but distortion-tolerant data. We consider cross-layer approaches which exploit the coupling between presentation and transport layers. We make the natural assumption that the distortion function is convex and decreasing. We focus on a single source-destination pair and analytically find the optimum transmission policy when the transmission is done over an error-free channel. This optimum policy turns out to be independent of the exact form of the convex and decreasing distortion function. Then, for a packet-erasure channel, we analytically find the optimum open-loop transmission policy, which is also independent of the form of the convex distortion function. We then find computationally efficient closed-loop heuristic policies and show, through numerical evaluation, that they outperform the open-loop policy and have near optimal performance.

Under the Dynamic Model of Multilingualism multilinguals are especially vulnerable to language attrition. It was the aim of the present study to verify if this was the case and to observe whether the different linguistic skills (receptive vs. descriptive) and the different linguistic levels (syntactic, lexical, morphological, etc.) would be affected equally.Data were gathered longitudinally by means of a language test for the subject’s reading, writing, listening and speaking skills as well as her knowledge of grammar and vocabulary. Although the overall accuracy remained intact and no proof for attrition in the receptive skills was found, the productive skills - mainly fluency - were shown to have suffered from language attrition. This was demonstrated by an increase in the number of pauses, hesitations, repetitions and self-corrections among others and decrease in the percentage of error-free clauses and decrease in the clause length, in oral and written fluency respectively.

In eukaryotes, Rad51 protein is responsible for the recombinational repair of double-strand DNA breaks. Rad51 monomers cooperatively assemble on exonuclease-processed broken ends forming helical nucleo-protein filaments that can pair with homologous regions of sister chromatids. Homologous pairing allows the broken ends to be reunited in a complex but error-free repair process. Rad51 protein has ATPase activity but its role is poorly understood, as homologous pairing is independent of adenosine triphosphate (ATP) hydrolysis. Here we use magnetic tweezers and electron microscopy to investigate how changes of DNA twist affect the structure of Rad51-DNA complexes and how ATP hydrolysis participates in this process. We show that Rad51 protein can bind to double-stranded DNA in two different modes depending on the enforced DNA twist. The stretching mode is observed when DNA is unwound towards a helical repeat of 18.6 bp/turn, whereas a non-stretching mode is observed when DNA molecules are not permitted to change their native helical repeat. We also show that the two forms of complexes are interconvertible and that by enforcing changes of DNA twist one can induce transitions between the two forms. Our observations permit a better understanding of the role of ATP hydrolysis in Rad51-mediated homologous pairing and strand exchange.

Due to the helical structure of DNA the process of DNA replication is topologically complex. Freshly replicated DNA molecules are catenated with each other and are frequently knotted. For proper functioning of DNA it is necessary to remove all of these entanglements. This is done by DNA topoisomerases that pass DNA segments through each other. However, it has been a riddle how DNA topoisomerases select the sites of their action. In highly crowded DNA in living cells random passages between contacting segments would only increase the extent of entanglement. Using molecular dynamics simulations we observed that in actively supercoiled DNA molecules the entanglements resulting from DNA knotting or catenation spontaneously approach sites of nicks and gaps in the DNA. Type I topoisomerases, that preferentially act at sites of nick and gaps, are thus naturally provided with DNA-DNA juxtapositions where a passage results in an error-free DNA unknotting or DNA decatenation.

La déubiquitinase BAP1 (« BRCA1-Associated Protein1 ») a initialement été isolée pour sa capacité de promouvoir la fonction suppressive de tumeurs de BRCA1. BAP1 est muté de manière homozygote dans plusieurs cancers (tel que le cancer du rein, de la peau, de l’oeil et du sein) suggérant fortement que cette déubiquitinase est un suppresseur de tumeurs. Effectivement, la surexpression de BAP1 réduit la prolifération cellulaire et la croissance tumorale dans des modèles de xénogreffe de souris. Toutefois, la fonction biologique et le mécanisme d’action de cette déubiquitinase restent encore marginalement connus. Ainsi, les objectifs de cette thèse sont de caractériser la fonction biologique de BAP1 et de révéler les bases moléculaires de sa fonction suppressive de tumeurs. Pour déterminer la fonction biologique de BAP1, nous avons immuno-purifié et identifié les protéines associées à BAP1, qui s’avèrent être principalement des facteurs et co-facteurs de transcription. Ensuite, nous avons démontré que BAP1 est un régulateur de la transcription. Parallèlement, un autre groupe a montré que BAP1 chez la drosophile, Calypso, régule l’ubiquitination de H2A et la transcription génique. D’autre part, nos résultats d’analyse d’expression génique globale suggèrent que BAP1 jouerait un rôle important dans la réponse aux dommages à l’ADN. Effectivement, des expériences de gain et de perte de fonction (méthode de l’ARNi, modèle de cellules KO en BAP1 et de cellules déficientes en BAP1 re-exprimant BAP1) ont révélé que cette déubiquitinase régule la réponse aux bris double brin d’ADN par la recombinaison homologue. Nos résultats suggèrent que BAP1 exerce sa fonction suppressive de tumeurs en contrôlant la réparation sans erreur de l’ADN via la recombinaison homologue. En cas d’inactivation de BAP1, les cellules deviendront plus dépendantes du mécanisme de réparation par jonction d'extrémités non-homologues, qui est potentiellement mutagénique causant ainsi l’instabilité génomique. D’autres études seront nécessaires afin de déterminer le rôle exact de BAP1 dans la transcription et de comprendre comment la dérégulation de l’ubiquitination de H2A contribue au développement du cancer. Définir les mécanismes de suppression tumorale est de grand intérêt, non seulement pour comprendre la carcinogénèse mais également pour le développement de nouvelles thérapies contre cette maladie.

RMS measuring device is a nonlinear device consisting of linear and nonlinear devices. The performance of rms measurement is influenced by a number of factors; i) signal characteristics, 2) the measurement technique used and 3) the device characteristics. RMS measurement is not simple, particularly when the signals are complex and unknown. The problem of rms measurement on high crest-factor signals is fully discussed and a solution to this problem is presented in this thesis. The problem of rms measurement is systematically analized and found to have mainly three types of errors: (1) amplitude or waveform error 2) Frequency error and (3) averaging error. Various rms measurement techniques are studied and compared. On the basis of this study the rms -measurement is reclassified three categories: (1) Wave-form-error-free measurement (2) High-frequncy-error measurement and (3) Low-frequency error-free measurement. In modern digital sampled-data systems the signals are complex and waveform-error-free rms measurement is highly appreciated. Among the three basic blocks of rms measuring device the squarer is the most important one. A squaring technique is selected, that permits shaping of the squarer error characteristic in such a way as to achieve waveform-errob free rms measurement. The squarer is designed, fabricated and tested. A hybrid rms measurement using an analog rms computing device and digital display combines the speed of analog techniques and the resolution and ease of measurement of digital techniques. An A/D converter is modified to perform the square-rooting operation. A 10-V rms voltmeter using the developed rms detector is fabricated and tested. The chapters two, three and four analyse the problems involved in rms measurement and present a comparative study of rms computing techniques and devices. The fifth chapter gives the details of the developed rms detector that permits wave-form-error free rms measurement. The sixth chapter, enumerates the the highlights of the thesis and suggests a list of future projects

Garment information tracking is required for clean room garment management. In this paper, we present a camera-based robust system with implementation of Optical Character Reconition (OCR) techniques to fulfill garment label recognition. In the system, a camera is used for image capturing; an adaptive thresholding algorithm is employed to generate binary images; Connected Component Labelling (CCL) is then adopted for object detection in the binary image as a part of finding the ROI (Region of Interest); Artificial Neural Networks (ANNs) with the BP (Back Propagation) learning algorithm are used for digit recognition; and finally the system is verified by a system database. The system has been tested. The results show that it is capable of coping with variance of lighting, digit twisting, background complexity, and font orientations. The system performance with association to the digit recognition rate has met the design requirement. It has achieved real-time and error-free garment information tracking during the testing.

The overall operation and internal complexity of a particular production machinery can be depicted in terms of clusters of multidimensional points which describe the process states, the value in each point dimension representing a measured variable from the machinery. The paper describes a new cluster analysis technique for use with manufacturing processes, to illustrate how machine behaviour can be categorised and how regions of good and poor machine behaviour can be identified. The cluster algorithm presented is the novel mean-tracking algorithm, capable of locating N-dimensional clusters in a large data space in which a considerable amount of noise is present. Implementation of the algorithm on a real-world high-speed machinery application is described, with clusters being formed from machinery data to indicate machinery error regions and error-free regions. This analysis is seen to provide a promising step ahead in the field of multivariable control of manufacturing systems.

In recent years, ZigBee has been proven to be an excellent solution to create scalable and flexible home automation networks. In a home automation network, consumer devices typically collect data from a home monitoring environment and then transmit the data to an end user through multi-hop communication without the need for any human intervention. However, due to the presence of typical obstacles in a home environment, error-free reception may not be possible, particularly for power constrained devices. A mobile sink based data transmission scheme can be one solution but obstacles create significant complexities for the sink movement path determination process. Therefore, an obstacle avoidance data routing scheme is of vital importance to the design of an efficient home automation system. This paper presents a mobile sink based obstacle avoidance routing scheme for a home monitoring system. The mobile sink collects data by traversing through the obstacle avoidance path. Through ZigBee based hardware implementation and verification, the proposed scheme successfully transmits data through the obstacle avoidance path to improve network performance in terms of life span, energy consumption and reliability. The application of this work can be applied to a wide range of intelligent pervasive consumer products and services including robotic vacuum cleaners and personal security robots1.

The rapid development of data transfer through internet made it easier to send the data accurate and faster to the destination. There are many transmission media to transfer the data to destination like e-mails; at the same time it is may be easier to modify and misuse the valuable information through hacking. So, in order to transfer the data securely to the destination without any modifications, there are many approaches like cryptography and steganography. This paper deals with the image steganography as well as with the different security issues, general overview of cryptography, steganography and digital watermarking approaches. The problem of copyright violation of multimedia data has increased due to the enormous growth of computer networks that provides fast and error free transmission of any unauthorized duplicate and possibly manipulated copy of multimedia information. In order to be effective for copyright protection, digital watermark must be robust which are difficult to remove from the object in which they are embedded despite a variety of possible attacks. The message to be send safe and secure, we use watermarking. We use invisible watermarking to embed the message using LSB (Least Significant Bit) steganographic technique. The standard LSB technique embed the message in every pixel, but my contribution for this proposed watermarking, works with the hint for embedding the message only on the image edges alone. If the hacker knows that the system uses LSB technique also, it cannot decrypt correct message. To make my system robust and secure, we added cryptography algorithm as Vigenere square. Whereas the message is transmitted in cipher text and its added advantage to the proposed system. The standard Vigenere square algorithm works with either lower case or upper case. The proposed cryptography algorithm is Vigenere square with extension of numbers also. We can keep the crypto key with combination of characters and numbers. So by using these modifications and updating in this existing algorithm and combination of cryptography and steganography method we develop a secure and strong watermarking method. Performance of this watermarking scheme has been analyzed by evaluating the robustness of the algorithm with PSNR (Peak Signal to Noise Ratio) and MSE (Mean Square Error) against the quality of the image for large amount of data. While coming to see results of the proposed encryption, higher value of 89dB of PSNR with small value of MSE is 0.0017. Then it seems the proposed watermarking system is secure and robust for hiding secure information in any digital system, because this system collect the properties of both steganography and cryptography sciences.

A faithful depiction of the tropical atmosphere requires three-dimensional sets of observations. Despite the increasing amount of observations presently available, these will hardly ever encompass the entire atmosphere and, in addition, observations have errors. Additional (background) information will always be required to complete the picture. Valuable added information comes from the physical laws governing the flow, usually mediated via a numerical weather prediction (NWP) model. These models are, however, never going to be error-free, why a reliable estimate of their errors poses a real challenge since the whole truth will never be within our grasp. The present thesis addresses the question of improving the analysis procedures for NWP in the tropics. Improvements are sought by addressing the following issues: - the efficiency of the internal model adjustment, - the potential of the reliable background-error information, as compared to observations, - the impact of a new, space-borne line-of-sight wind measurements, and - the usefulness of multivariate relationships for data assimilation in the tropics. Most NWP assimilation schemes are effectively univariate near the equator. In this thesis, a multivariate formulation of the variational data assimilation in the tropics has been developed. The proposed background-error model supports the mass-wind coupling based on convectively-coupled equatorial waves. The resulting assimilation model produces balanced analysis increments and hereby increases the efficiency of all types of observations. Idealized adjustment and multivariate analysis experiments highlight the importance of direct wind measurements in the tropics. In particular, the presented results confirm the superiority of wind observations compared to mass data, in spite of the exact multivariate relationships available from the background information. The internal model adjustment is also more efficient for wind observations than for mass data. In accordance with these findings, new satellite wind observations are expected to contribute towards the improvement of NWP and climate modeling in the tropics. Although incomplete, the new wind-field information has the potential to reduce uncertainties in the tropical dynamical fields, if used together with the existing satellite mass-field measurements. The results obtained by applying the new background-error representation to the tropical short-range forecast errors of a state-of-art NWP model suggest that achieving useful tropical multivariate relationships may be feasible within an operational NWP environment.

Atmosphärische Partikel beeinflussen das Klima durch Prozesse wie Streuung, Reflexion und Absorption. Zusätzlich fungiert ein Teil der Aerosolpartikel als Wolkenkondensationskeime (CCN), die sich auf die optischen Eigenschaften sowie die Rückstreukraft der Wolken und folglich den Strahlungshaushalt auswirken. Ob ein Aerosolpartikel Eigenschaften eines Wolkenkondensationskeims aufweist, ist vor allem von der Partikelgröße sowie der chemischen Zusammensetzung abhängig. Daher wurde die Methode der Einzelpartikel-Laserablations-Massenspektrometrie angewandt, die eine größenaufgelöste chemische Analyse von Einzelpartikeln erlaubt und zum Verständnis der ablaufenden multiphasenchemischen Prozesse innerhalb der Wolke beitragen soll.rnIm Rahmen dieser Arbeit wurde zur Charakterisierung von atmosphärischem Aerosol sowie von Wolkenresidualpartikel das Einzelpartikel-Massenspektrometer ALABAMA (Aircraft-based Laser Ablation Aerosol Mass Spectrometer) verwendet. Zusätzlich wurde zur Analyse der Partikelgröße sowie der Anzahlkonzentration ein optischer Partikelzähler betrieben. rnZur Bestimmung einer geeigneten Auswertemethode, die die Einzelpartikelmassenspektren automatisch in Gruppen ähnlich aussehender Spektren sortieren soll, wurden die beiden Algorithmen k-means und fuzzy c-means auf ihrer Richtigkeit überprüft. Es stellte sich heraus, dass beide Algorithmen keine fehlerfreien Ergebnisse lieferten, was u.a. von den Startbedingungen abhängig ist. Der fuzzy c-means lieferte jedoch zuverlässigere Ergebnisse. Darüber hinaus wurden die Massenspektren anhand auftretender charakteristischer chemischer Merkmale (Nitrat, Sulfat, Metalle) analysiert.rnIm Herbst 2010 fand die Feldkampagne HCCT (Hill Cap Cloud Thuringia) im Thüringer Wald statt, bei der die Veränderung von Aerosolpartikeln beim Passieren einer orographischen Wolke sowie ablaufende Prozesse innerhalb der Wolke untersucht wurden. Ein Vergleich der chemischen Zusammensetzung von Hintergrundaerosol und Wolkenresidualpartikeln zeigte, dass die relativen Anteile von Massenspektren der Partikeltypen Ruß und Amine für Wolkenresidualpartikel erhöht waren. Dies lässt sich durch eine gute CCN-Aktivität der intern gemischten Rußpartikel mit Nitrat und Sulfat bzw. auf einen begünstigten Übergang der Aminverbindungen aus der Gas- in die Partikelphase bei hohen relativen Luftfeuchten und tiefen Temperaturen erklären. Darüber hinaus stellte sich heraus, dass bereits mehr als 99% der Partikel des Hintergrundaerosols intern mit Nitrat und/oder Sulfat gemischt waren. Eine detaillierte Analyse des Mischungszustands der Aerosolpartikel zeigte, dass sich sowohl der Nitratgehalt als auch der Sulfatgehalt der Partikel beim Passieren der Wolke erhöhte. rn

Abnormal activation of cellular DNA repair pathways by deregulated signaling of receptor tyrosine kinase systems has broad implications for both cancer biology and treatment. Recent studies suggest a potential link between DNA repair and aberrant activation of the hepatocyte growth factor receptor Mesenchymal-Epithelial Transition (MET), an oncogene that is overexpressed in numerous types of human tumors and considered a prime target in clinical oncology. Using the homologous recombination (HR) direct-repeat direct-repeat green fluorescent protein ((DR)-GFP) system, we show that MET inhibition in tumor cells with deregulated MET activity by the small molecule PHA665752 significantly impairs in a dose-dependent manner HR. Using cells that express MET-mutated variants that respond differentially to PHA665752, we confirm that the observed HR inhibition is indeed MET-dependent. Furthermore, our data also suggest that decline in HR-dependent DNA repair activity is not a secondary effect due to cell cycle alterations caused by PHA665752. Mechanistically, we show that MET inhibition affects the formation of the RAD51-BRCA2 complex, which is crucial for error-free HR repair of double strand DNA lesions, presumably via downregulation and impaired translocation of RAD51 into the nucleus. Taken together, these findings assist to further support the role of MET in the cellular DNA damage response and highlight the potential future benefit of MET inhibitors for the sensitization of tumor cells to DNA damaging agents.
