Two new sesquiterpene evoninate alkaloids, aquifoliunine E-III and aquifoliunine E-IV, were isolated from the root bark of Maytenus aquifolium. The structures of the new compounds were elucidated on the basis of spectroscopic methods, including ES-MS.

A general procedure was developed for the simultaneous separation of flavonoids and naphthopyrones from the polar extracts of the capitula from Brazilian everlasting plants is described. The ethanolic extracts of several species from the Paepalanthus genus (Eriocaulaceae) were fractionated by droplet countercurrent chromatography followed by column chromatography on pvp and sephadex LH-20. The isolated compounds were identified by spectrometric analysis and comparison with literature data. This approach led to the isolation of 9-O-β-D-glucopyranosylpaepalantine (1), 9-O-β-D-glucopyranosyl (1→6)allopyranosylpaepalantine (2), along with the flavonoids 6-methoxykaempferol (3), 3-O-β-D-glucopyranosyl-6-methoxykaempferol (4), patuletin (5), 3-Oβ-D-rutinosylpatuletin (6), 7-O-β-D-glucopyranosylquercetagetin (7), 5,7,4'-trihydroxy-6,3'-dimethoxyflavone (8) and 5,7,4'-trihydroxy-6,3'-dimethoxyflavonol (9).

A new naphthopyrone dimer was isolated from the capitula of Paepalanthus bromelioides by chromatographic procedures. Its structure was deduced from spectrometric data. On colorimetric assay for cytotoxicity the new dimer showed IC50 of 55.9 μM. (C) 2000 Elsevier Science B.V.

Chemical examination of the leaves of Styrax ferrugineus yielded 5-[3'-(β-D-glucopyranosyloxy)propyl]-7-methoxy-2-(3',4'-dime-thoxyphenyl) benzofuran, along with the known nor-lignans 5-(3'-hydroxypropyl)-7-methoxy-2-(3',4'-methylenedioxyphenyl) benzofuran, 5-(3'-hydroxypropyl)-7-methoxy-2-(3',4'-dimethoxyphenyl)benzofuran, 5-[3'-(β-D-glucopyranosyloxy)propyl]-7-methoxy-2-(3', 4'-methylenedioxyphenyl)benzofuran and the lignan, dihydrodehydrodiconiferyl alcohol. All arylpropanoids isolated showed antibacterial and antifungal activities. The structures of the isolates were established by spectroscopic analysis. (C) 2000 Elsevier Science Ltd.

The Influence of Drying Temperatures on the Yield Composition of Citronella (Cymbopogon winterianus Jowitt) essential oil. The studies were carried out to establish more precise parameters for citronella (C. winterianus Jowitt) post-harvest, and to optimize drying time and essential oil quantitative/qualitative yield. Five treatments were designed (30°C, 40°C, 50°C, 60°C and 70°C), with 14 repetitions of the drying process and 12 of the essential oil extraction. Drying at 60°C gave the best results for drying time (48 hours until weight stabilization), and also for extracted oil quantity (1.228 ± 0.127% over dry weight). Essential oil content showed high quantitative variations. The main compound found was neral, except in the 50°C treatment, where citronelal was the main compound.

The immune responses are mediated by a variety cells and molecules that cells secreted. Macrophages are the first cells that participate in the immune response, and, when are activated, release more than hundred compounds at the extracelular medium, such as cytokine (TNF-α) and the intermediate compounds of the nitrogen (NO). In this paper the release of nitric oxide (NO) and necrose tumoral factor (TNF-α) were determined in peritoneal macrophage cultures of mice in the presence of the 70% ethanolic extract obtained from the flowers of the Melampodium divaricatum (Asteraceae) in the concentrations of 20, 10 and 5 mg/mL. The 70% ethanolic extracts from flowers of the Melampodium divaricatum presented higher liberation of NO and TNF-α in the concentration of the 20 mg/mL when compared with LPS. We conclude that this extract is a potente stimulator of macrophage, could be immunomodulatory activity.

This work aimed to determine the best harvest time for biomass production, yield and essential oil composition considering the seasonal variation (spring, summer, autumn and winter) on different plant parts (apical, medial and basal). Essential oils were extracted by hydro-distillation with a Clevenger apparatus for both fresh and dry mass obtained in field and lab conditions respectively. The extracted essential oils were analyzed by GS/MS (Shimadzu, QP-5000). The chemical components were identified by comparing their mass spectrum to the patterns filed in the MS computer memory (Wiley,139,Lib.), to the literature references, and by co-injection with authentic standards. Applying phyto-chemical tests on fresh and dry mass, the chemical component percentages of essential oils were calculated and identified as follows: citral (neral and geranial), myrcene, caryophylene and elemene.

The sun mushroom is the popular name for the Agaricus blazei Murill fungus, a mushroom native to south-eastern Brazil, which has been frequently used in popular medicine mainly in the form of tea to treat various ailments (stress, diabetes, etc.). In the present study, the genotoxic and/or anti-genotoxic effects ofA. blazei on mammalian cells in culture was assessed by checking the increase or reduction of micronucleus (MN) frequency and comets. The sun mushroom (lineage 99/26) was used as aqueous extracts prepared (2.5%) at three different temperatures (60, 25 and 4°C). The in vitro micronucleus (MN) test in binucleated cells and comet assay were used in V79 cells cultivated in HAM-F10+DMEM medium (1:1), supplemented with 10% of fetal bovine serum. The experiments were divided into four treatment types: 1. Negative control; 2. Positive control with MMS; 3. Treatments with the three forms of extracts (60, 25 and 4°C); and 4. Treatments with the extracts in different associations (simultaneous, pre-treatment, post-treatment and simultaneous after pre-incubation for 1 h) with MMS. None of the A. blazei extracts show genotoxic activity. In the comet assay no protecting effect was found. The results obtained in the MN test showed that the three forms of extracts used had protective activity, suggesting that the compound or active ingredients of A. blazei are always present in these extracts. The greater protective efficiency of the simultaneous treatment and simultaneous treatment with pre-incubation mixture with MMS suggests that the extracts have an antimutagenic action of the desmutagenic type. © 2002 Elsevier Science Ltd. An iridoid glucoside: randinoside, along with five known iridoids: galioside, deacetylasperulosidic acid methyl ester, scandoside methyl ester, geniposide and gardenoside, were isolated from the stems of Randia spinosa. A new flavonol 4′-methoxykaempferol-7-(acetyloxy)-3,5-O-α-L- rhamnoside (1), was isolated from the leaves of Chiococca braquiata, along with three known flavonoids apigenin, 7-O-methoxyquercetrin and quercetrin and four triterpenes α-amirin, β-amirin, ursolic and oleanolic acids . Their structures were established on the basis of spectroscopic methods.

Helivypolide G was isolated from leaves of Helianthus annuus L. cv. Stella. In the course of our ongoing research for new allelochemicals from Helianthus annuus, a novel dimeric bioactive sesquiterpene lactone, helivypolide G has been isolated and characterized from the medium polar active fractions of the leaves of cultivar variety Stella. The monomers are connected through carbons C-15 of each unit and an oxygen bridge, forming an enolic oxane ring. © 2004 Elsevier Ltd. All rights reserved.

Propolis (bee glue) is one of the major hive products of bees and is rich in flavonoids, which are known for their antioxidant activities. The aim of this study was to evaluate the hepatoprotective effects of the ethanolic extract of propolis (EEP) against experimental carbon tetrachloride (CCl 4)-induced liver toxicity in rats by means of biochemical indices. The animals were divided into 4 groups: GI= received mineral oil; GII= CCl 4(4mL/kg; Lp., single dose) treated; GIII= CCl4 (4mL/kg; i.p., single dose) treatment followed by ethanolic extract of propolis (100mg/kg) for gavage from the species Tetragonisca angustula, daily for 3 days and GIV= CCl4 (4mL/kg; i.p., single dose) treatment followed by ethanolic extract of propolis (100mg/kg) for gavage from the species Nannotrigonea testaceicornes, daily, for 3 days. The serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), cholesterol and triacylglycerols were estimated after 3 days. CCl 4 caused a maximum increase (p<0,01) above biochemical parameters. As compared to CCl4 group (GII), the EEP (GIII and GIV) showed reduction in cholesterol, triacylglycerol, ALT, AST and alkaline phosphatase activity in the serum. In conclusion, these data indicate that EEP improved the dyslipidaemia, moreover, significantly attenuated increases in serum ALT and AST activities in rats with liver damage induced by carbon tetrachloride.

Communiols E-H (1-4), four new polyketide-derived natural products containing furanocyclopentane, furanocyclopentene, cyclopentene, or γ-lactone moieties, have been isolated from two geographically distinct isolates of the coprophilous fungus Podospora communis. Agaricus blazei Murrill, popularly known as the sun mushroom, is a native mushroom in SP, Brazil, that has been widely used in the treatment of cancer and many other pathologies in different parts of the world. A water-soluble protein-polysaccharide complex (1 → 6)β-D-glucan has been isolated from its fruiting body that showed immune-modulation activity. From organic extracts, linoleic acid has been isolated and determined to be the main substance with antimutagenic activity. Using both the micronucleus (MN) and comet (single cell microgel electrophoresis) assays, this study determined the genotoxic and antigenotoxic potential of A. blazei (AB) obtained from commercial sources or the following strains: a) strains AB 97/29 (young and sporulated phases); b) a mixture taken from AB 96/07, AB 96/09 and AB 97/ 11 strains; and c) commercial mushrooms from Londrina, PR and Piedade, SP, designated as AB PR and AB SP, respectively. The extracts from these mushrooms were isolated in chloroform:methanol (3:1) and used in vitro at three different concentrations. V79 cells (Chinese hamster lung cells) were exposed to the extracts under pre-, simultaneous and post-treatment conditions, combined with methyl methanesulfonate (MMS). Under the circumstances of this study, these organic extracts did not show any genotoxic or mutagenic effects, but did protect cells against the induction of micronuclei by MMS.
