Background: Laboratory studies of host-seeking olfactory behaviour in sandflies have largely been restricted to the American visceral leishmaniasis vector Lutzomyia longipalpis. In comparison, almost nothing is known about the chemical ecology of related species, which transmit American cutaneous leishmaniasis (ACL), due in part to difficulties in raising these insects in the laboratory. Understanding how ACL vectors locate their hosts will be essential to developing new vector control strategies to combat this debilitating disease.Methods: This study examined host-odour seeking behaviour of the ACL vector Nyssomyia neivai (Pinto) (=Lutzomyia neivai) using a wind tunnel olfactometer. The primary aim was to determine whether field-collected female N. neivai would respond to host odours in the laboratory, thereby eliminating the need to maintain colonies of these insects for behavioural experiments. Responses to two key host odour components, 1-octen-3-ol and lactic acid, and a commercially-available mosquito lure (BG-Lure (TM)) were assessed and compared relative to an air control. We also tested whether trials could be conducted outside of the normal evening activity period of N. neivai without impacting on fly behaviour, and whether the same flies could be used to assess baseline responses to air without affecting responses to octenol, thereby reducing the number of flies required for experiments.Results: Octenol was found to both activate host-seeking behaviour and attract female N. neivai in the wind tunnel, while lactic acid elicited weaker responses of activation and attractiveness under identical conditions. The BG-Lure did not activate or attract N. neivai under test conditions. Further experiments showed that sandfly behaviour in the wind tunnel was not affected by time of day, such that experiments need not be restricted to nocturnal hours. Moreover, using the same flies to measure both baseline responses to air and attraction to test compounds did not affect odour-seeking behaviour.Conclusions: The results of this study demonstrate that N. neivai taken from the field are suitable for use in laboratory olfactometer experiments. It is hoped this work will facilitate further research into chemical ecology of this species, and other ACL vectors.

The aim was to analyze the prevalence of the stages of behavior change (BCS) and the perceived barriers to physical activity in older people residents in a medium size city of São Paulo state, Brazil. Seventy-nine older people participated of the study. The BCS was assessed by the Questionnaire of Behavior Chance Stages and those on perception of barriers by the Questionnaire on Barriers to the Practice of Physical Activity in Older People (QBPPAE). Statistical analyses was comprised by the Chi-square test (p<0.05). Data showed that: a) 64,6% of the older people do not practice physical activity and only 22,8% of them have been practicing it on a regular basis for more than 6 months; b) The most perceived barriers were: "Active enough", "Too old"; "Need to rest" and; "Disease or injury"; c) The BCS seems modulate significantly the perception of barriers (p<0.05). We can conclude that: a) most of the older people are in the pre-contemplation, contemplation and preparation stages of the BCS, yet; b) gender influences almost nothing with perception of barriers and; c) the BCS interferes with the type of perceived barriers. Such conclusions support the suggestions that it is necessary to develop specific strategies for different stages of behavior change if it is to minimize the barriers. This may help to find the most effective way to change to more active lifestyle in the elderly.

The invasive fire ant Solenopsis invicta is medically important because its venom is highly potent. However, almost nothing is known about fire ant venom proteins because obtaining even milligram-amounts of these proteins has been prohibitively challenging. We present a simple and fast method of obtaining whole venom compounds from large quantities of fire ants. For this, we separate the ants are from the nest soil, immerse them in dual-phase mixture of apolar organic solvent and water, and evaporate each solvent phase in separate. The remaining extract from the aqueous phase is largely made up of ant venom proteins. We confirmed this by using 2D gel electrophoresis while also demonstrating that our new approach yields the same proteins obtained by other authors using less efficient traditional methods. © 2013 Elsevier Ltd.

For Estonia and its people social work is one of the vitally important fields that had to be built up from almost nothing since independence was regained in 1991. During Soviet times social work and social workers did not receive the necessary attention. Severe social problems were denied and kept hidden since according to official communist ideology, life in the Soviet Union was the best in the world and getting better all the time. Social workers did not receive specialised education and their functions were to be carried out by the workers of trade unions and the party, by teachers and by the workers of the personnel departments. In the 1990s big changes, having also an effect on social life, took place in the development of Estonian society. Concepts such as social work and social worker were rediscovered in Estonia. There are certain prerequisites for the success of any activity (including social work). One of the most important ones is being a professional, a worker with thorough preparation. Social work as an occupation requires specialised academic education, which is based on theoretical knowledge and practical skills that have been acquired through theoretical knowledge. Specialised knowledge is a foundation for attaining a specialised qualification. However, at the same time one has to keep in mind that social work as an occupation is constantly changing, there is no absolute knowledge - everything is relative, dynamic and changing (Tamm, 1998). The changing nature of the activity requires reflection by a social worker, who also has to be able to evaluate his/her work and its basis and learn from experiences. Academic specialised education implies also the development of a new professional identity and higher levels of competence. This underlines the necessity of specialised education.

Neurons and their precursor cells are formed in different regions within the developing CNS, but they migrate and occupy very specific sites in the mature CNS. The ultimate position of neurons is crucial for establishing proper synaptic connectivity in the brain. In Drosophila, despite its extensive use as a model system to study neurogenesis, we know almost nothing about neuronal migration or its regulation. In this paper, I show that one of the most studied neuronal pairs in the Drosophila nerve cord, RP2/sib, has a complicated migratory route. Based on my studies on Wingless (Wg) signaling, I report that the neuronal migratory pattern is determined at the precursor cell stage level. The results show that Wg activity in the precursor neuroectodermal and neuroblast levels specify neuronal migratory pattern two divisions later, thus, well ahead of the actual migratory event. Moreover, at least two downstream genes, Cut and Zfh1, are involved in this process but their role is at the downstream neuronal level. The functional importance of normal neuronal migration and the requirement of Wg signaling for the process are indicated by the finding that mislocated RP2 neurons in embryos mutant for Wg-signaling fail to properly send out their axon projection.

Lower Cretaceous sediments are frequently characterized by a well expressed cyclicity. While the processes influencing environments above the carbonate compensation depth (CCD) are reasonably well understood, almost nothing is known about the deep ocean. Cretaceous sub-CCD sediments from the Tethys and Atlantic Oceans typically show rhythmic black/green shale successions. To gain insight into the nature of these black/green shale cycles, we performed detailed geochemical analyses (X-ray fluorescence, Rock-Eval and reactive iron analysis) on a 3 m long section of latest Aptian age. The major-element distribution of the analyzed shale sequence indicates a periodic change from a high-productivity and well-oxygenated green shale mode to a low-productivity oxygen-deficient black shale mode. It is proposed here that the preservation of organic matter was dependent on the strength of salinity-driven deepwater generation. Furthermore, the data show that the Corg content covaries with changes in the detrital composition. Therefore we hypothesize that Tethyan deepwater circulation was sensitive to changes in the monsoonal system. Time series analysis suggests that these changes are periodic in nature, although we are currently unable to prove that the dominant periodicity is related to the precession component of the Milankovitch frequencies.

Well, I firmly believe that, as a professor, one has the duty, the obligation to show and explain his work. Not, of course, as a model to imitate, but rather as an opportunity to reflect upon that work. So, I will try to explain some of the considerations that were crucial in the development of each project. It is my hope that these reflections may be of interest to you, and due to my optimistic nature, that you may even enjoy it. Today I am going to present some projects, from a specific point of view: that of MATERIALITY. This is why I decided to title this presentation ARCHITEXTURE: that is, architecture from the point of view of texture, the quality of its material. Our architecture wants to pay attention to materials; we like to use very physical materials. We try to explore the expressive possibilities of different materials. That allows us, on the one hand, to try to master different techniques, and exploit their potential. On the other hand, it also avoids the dangers of style. Style is the death of an architect. When he starts repeating formulas, avoiding experimentation, copying himself, he dies of boredom, of intellectual boredom. Nevertheless, I don’t believe that the material itself determines anything. Architecture is an exercise of the freedom of an architect. Almost nothing is a given. Of course, there is the law of gravity and economic restraints, and even the overwhelming building code. But the most determinant factor is always the freedom of the architect, derived from his mastery of knowledge and culture, and his decision to innovate and to take risks

Phase changing flows are being considered for thermal management in space platforms. The resulting flow patterns are very complicated and extremely sensitive to gravity action. Concerning fluid flow in ducts, the available evidence indicates that although the pressure loss does not depend too much on the fluid flow pattern,the heat transfer (and resulting phase change) does. A simple exercise to illustrate this point is presented in this paper. It deals with condensing flow in straight circular cross-sectional ducts. Two extreme configurations are considered here, one corresponds to a stratified flow and the other to an annular flow. Both types of flow patterns have been extensively considered in the past and from this point of view almost nothing is new in the paper, but past results look conflictive and this could be due to the limitations and computational intricacies of the models used. Thus the problem has been reformulated from the onset and the results are presented as the evolution of the vapor quality (vapor to total mass flow rate) along the duct, in typical cases. The results presented here indicate that within the validity of the present models and the assumed ranges of mass flow rate, duct diameter, thermal conditions and fluid characteristics,the length of the ducts required to achieve complete condensation under zero gravity are an order of magnitude larger than in horizontal tubes under normal terrestrial conditions.

Cell-wall mechanical properties play an integral part in the growth and form of Saccharomyces cerevisiae. In contrast to the tremendous knowledge on the genetics of S. cerevisiae, almost nothing is known about its mechanical properties. We have developed a micromanipulation technique to measure the force required to burst single cells and have recently established a mathematical model to extract the mechanical properties of the cell wall from such data. Here we determine the average surface modulus of the S. cerevisiae cell wall to be 11.1 ± 0.6 N/m and 12.9 ± 0.7 N/m in exponential and stationary phases, respectively, giving corresponding Young's moduli of 112 ± 6 MPa and 107 ± 6 MPa. This result demonstrates that yeast cell populations strengthen as they enter stationary phase by increasing wall thickness and hence the surface modulus, without altering the average elastic properties of the cell-wall material. We also determined the average breaking strain of the cell wall to be 82% ± 3% in exponential phase and 80% ± 3% in stationary phase. This finding provides a failure criterion that can be used to predict when applied stresses (e.g., because of fluid flow) will lead to wall rupture. This work analyzes yeast compression experiments in different growth phases by using engineering methodology.

The maturation of 5S RNA in Escherichia coli is poorly understood. Although it is known that large precursors of 5S RNA accumulate in mutant cells lacking the endoribonuclease-RNase E, almost nothing is known about how the mature 5' and 3' termini of these molecules are generated. We have examined 5S RNA maturation in wild-type and single- or multiple-exoribonuclease-deficient cells by Northern blot and primer-extension analysis. Our results indicate that no mature 5S RNA is made in RNase T-deficient strains. Rather, 5S RNA precursors containing predominantly 2 extra nucleotides at the 3' end accumulate. Apparently, these 5S RNAs are functional inasmuch as mutant cells are viable, growing only slightly slower than wild type. Purified RNase T can remove the extra 3' residues, showing that it is directly involved in the trimming reaction. In contrast, mutations affecting other 3' exoribonucleases have no effect on 5S RNA maturation. Approximately 90% of the 5S RNAs in both wild-type and RNase T- cells contain mature 5' termini, indicating that 5' processing is independent of RNase T action. These data identify the enzyme responsible for generating the mature 3' terminus of 5S RNA molecules and also demonstrate that a completely processed 5S RNA molecule is not essential for cell survival.

El texto que a continuación se presenta se corresponde de manera bastante fidedigna con el texto que el autor leyó en el acto de defensa de su tesis doctoral, realizada en la Universidad de Alicante el 14 de junio de 2012. El tribunal estuvo compuesto por Remedios Zafra, Federico Soriano, Jessica Jaques, Miguel Angel Hernández y Juan Freire. Los directores del trabajo fueron José María Torres Nadal y Pedro Alberto Cruz. Después de lo vivido, lo pensado, y lo contado, descubro perplejo al final de esta investigación que la relevancia de lo que hemos realizado junto con otrxs muchxs en la Universidad de Alicante, radica precisamente en una obsesión infantil por organizar lo prescindible, todo aquello que no nos es requerido para el cumplimiento de nuestras estrictas funciones académicas. O por utilizar otros términos, descubro con un cierto horror que casi nada de lo que en algún momento nos ha llegado a excitar parece atravesado por los dispositivos maquínicos institucionales.
