A chain-forming diatom Skeletonema costatum (Greville) Cleve collected from Yatsushiro Sea, Japan was cultured to determine the optimum level of some physico-chemical factors for their growth under laboratory conditions. Filtered and sterilized aged sea water enriched by adding nutrient solution (Provasoli 1968) was used as the culture medium. The plankton could tolerate a wide range of salinities (3-55 ppt). Optimum growth was observed at salinities of 20-35 ppt, temperatures of 20-25°C, light intensities of 80-120µE mˉ² secˉ¹ and pH between 7.5 and 8.0. Growth did not occur at salinities below 3 ppt and at temperatures above 30°C. From the present study, it is concluded that S. costatum was extremely euryhaline and tolerable to very low salinities.

Diatom culture and larval feeding experiments were conducted to test the viability and acceptability of preserved algal concentrates. C. calcitrans is characterised by the presence of setae which keep them suspended in cultures and make autoflocculation very difficult. Flocculation was induced by the addition of a floc-forming chemical. Using the optimum conditions, it was possible to harvest the algae within 1-h settling time and with about 84% recovery. The viability of frozen Chaetoceros was determined by actual cell reproduction. Preliminary feeding experiments showed that Chaetoceros can be successfully used as a substitute for fresh diatoms as feed for Penaeus monodon larvae. Simple freezing techniques, with or without the use of protectants has been found convenient for preserving algal concentrates in small volumes for both feeding and culture purposes.

Batch cultures of C. calcitrans were maintained indoors at a temperature range of 21 to 25 C and continuously illuminated by 40-watt daylight fluorescent lights. Cultures were exposed to 5 different intensities from 200 to 25,000 lux.Population counts show that light intensity affects growth and reproduction of the algae cultivated. A comparison of population peak growths showed cultures illuminated by 12,000 lux to have higher cell counts than those exposed to higher or lower light intensities.

A brief account is given of experiments undertaken rearing Penaeus monodon larvae fed on diatom (Chaetoceros calcitran) and fermented vegetable trash, which included fruits and their peels, vegetables and rice. The possible use of high protein content trash materials as a feed substitute is examined briefly.

A study was undertaken to determine the most suitable food for Penaeus monodon zoea out of mixed diatoms, Skeletonema costatum, Chaetoceros calcitrans and C. gracilis. Results indicate that mixed diatoms and C. gracilis are the better food. Addition of zooplankton, e.g.rotifers, is also recommended as early as Z3 since the larvae start to be omnivorous at this stage.

The growth of Skeletonema costatum in two artificial nutrient media was studied using various culture vessels. Skeletonema costatum was collected from the Cox's Bazar coast around the Bay of Bengal. Different growths stages i.e. lag phase, exponential phase, prestationary phase, stationary and death phase were observed during the culture period. The number of cells increased during the active division period and decreased after the beginning of the prestationary phase. The average densities of S. costatum in primary and secondary cultures were 0.55 x 10 super(6) cells mlˉ¹ and 0.93x10 super(6) cells mlˉ¹, respectively. In mass culture of S. costatum two, types of media were used. Highest cells densities of S. costatum cement tank culture were recorded 1.23x10 super(6) cell mlˉ¹ and 0.78x10 super(6) cells mlˉ¹ in their respective f/4 medium and commercial fertilizer medium. In the cement tanks culture fertilizer medium was found to be the best medium for mass culture of S. costatum in respect of production efficiency and culture stability.

Fish introduction, eutrophication and disappearance of aquatic vegetation are important disturbances of aquatic ecosystems, especially in plateau lakes, which are generally considered to be very vulnerable. Fish were introduced to Lake Dianchi, a eutrophic plateau lake in southwest China, in the late 1950s and 1970s. After the introduction, invasive fish became the dominant species, and the total fish yield increased. Meanwhile, the trophic level of Lake Dianchi had a tendency to increase in the past decades because of the increases in human activities in the watershed area. In addition, the area of aquatic vegetation decreased from more than 90 to 1.8% of the lake area from the 1950s to 2000. This study investigated the effects of fish introduction, eutrophication and aquatic vegetation on the diatom community of Lake Dianchi by examining the changes of microfossil diatom assemblage and abundance. Results showed that the absolute abundance and diatom assemblages changed after fish were introduced. The endemic species, Cyclotella rohomboideo-elliptica, disappeared with the introduction of fish and increasing trophic levels after 1958. Fragilaria crotonensis entered into the lake with the introduction of fish and gradually thrived in the lake after 1958. Diatom species numbers also decreased gradually from 21 to 9 from the past to present. Epiphytic diatoms disappeared with the decrease of aquatic vegetation after 1985. Our study indicated that eutrophication was the most important process determining diatom abundance, and fish introduction was a secondary process determining diatom abundance, while aquatic vegetation had a more important role in structuring the diatom community in this eutrophic plateau lake.

We investigated dynamics of the phytoplankton community and abiotic factors in Xiangxi Bay of the Three-Gorge Reservoir, China, by daily sampling, a specific site during a spring algal bloom (February 23-April 28, 2005). Among the 76 taxa observed, Asterionella formosa and Cyclotella spp. were the dominants, accounting for 47.2% and 29.9% of the total abundance, respectively. We determined the five distinct developing phases of the bloom by analyzing the dissimilarity of physicochemical parameters. Simultaneously, six phytoplankton community groups were distinguished by TWINSPAN classifications. The pattern for algal community succession was similar to that for the bloom phase shift, and the structural complexity of communities significantly decreased over time. Water temperature and silicate were the main factors that related to the development of the bloom and the shifts of the phytoplankton community.

Phytoplanktonic species acclimated to high light are known to show less photoinhibition. However, little has been documented on how cells grown under indoor conditions for decades without exposure to UV radiation (UVR, 280-400 nm) would respond differently to solar UVR compared to those in situ grown under natural solar radiation. Here, we have shown the comparative photosynthetic and growth responses to solar UVR in an indoor-(IS) and a naturally grown (WS) Skeletonema costatum type. In short-term experiment (<1 day), phi(PSII) and photosynthetic carbon fixation rate were more inhibited by UVR in the IS than in the WS cells. The rate of UVR-induced damages of PSII was faster and their repair was significantly slower in IS than in WS. Even under changing solar radiation simulated for vertical mixing, solar UVR-induced higher inhibition of photosynthetic rate in IS than in WS cells. During long-term (10 days) exposures to solar radiation, the specific growth rate was much lower in IS than WS at the beginning, then increased 3 days later to reach an equivalent level as that of WS. UVR-induced inhibition of photosynthetic carbon fixation in the IS was identical with that of WS at the end of the long-term exposure. The photosynthetic acclimation was not accompanied with increased contents of UV-absorbing compounds, indicating that repair processes for UVR-induced damages must have been accelerated or upgraded. We report on taxa from the diatom genera Gomphonema and Cymbella (Cymbellales, Bacillariophyta) found in the Jolmolungma Mountain region of China. This region is unique for its diversity of habitats, which include rivers, springs, moist soil, snowfields, swamps and lakes. We re-examine diatom taxa found in samples first documented in 1973 (Jao et A, 1973) and incorporate recent taxonomic revisions from the literature. In the genera Gomphonema and Cymbella we report 113 species and varieties. Of these 113 taxa, 59 are new record for China; 1 new combination, Encyonema jolmolungmensis, is made. Morphometric data and habitat features are reported for each taxon. Their distribution is strongly correlated to their microhabitats.

Assessment method for ecological condition of Xiangxi River system was studied by using 13 candidate metrics of epilithic diatom which can reflect conditions in pH, salinity, nitrogen uptake metabolism, oxygen requirements, saprobity, trophic state, morphological character and pollution tolerant capability etc. By one-way ANOVA, the metrics of relative abundance of acidobiontic algae (ACID), freshwater algae (FRESH), high oxygen requirement (HIGH-O), eutraphentic state (EUTRA) and mobile taxa ( MOBILE) were suitable for distinguishing sites in different conditions. Then, the river diatom index (RDI) composed of these five metrics was used to evaluate ecological condition of the river. The results showed that the healthiest sites were in the Guanmenshan Natural Reserve ( with the mean RDI of 79.73). The sites located in tributary of Jiuchong River also owned excellent state (mean RDI of 78.25). Mean RDI of another tributary - Gufu River and the main river were 70.85 and 68.45 respectively, and the unhealthiest tributary was Gaolan River (with mean RDI of 65.64). The mean RDI for all the 51 sites was 71.40. The competence of RDI was discussed with comparison of evaluation results of DAIpo and TDI, it can be concluded that multimetrics is more competent in assessment task.

Photosynthetic performance was examined in Skeletonema costatum (Greville) Cleve. under 12: 12-h light: dark (LD) cycle at ambient CO2 (350 muL L-1) and elevated CO2 (1000 muL L-1). At ambient CO2, the cellular chlorophyll a content, the light-saturated photosynthetic rate (P-m), the initial slope of the light saturation curves ( a), the photochemical efficiency of PSII (F-v/F-m), the apparent carboxylating efficiency (ACE) and the photosynthetic affinity for CO2 [1/K-m (CO2)] all showed rhythmical changes with different amplitudes during the light period. The P-m had similar changing pattern in the light period with the ACE and 1/K-m (CO2) rather than with the alpha and F-v/F-m, indicating that rhythmical changes of photosynthetic capacity may be mainly controlled by the activity of C- reduction associated with CO2 uptake during the light period. The CO2 enrichment reduced the ACE and the affinity to CO2, and increased the a, cellular chlorophyll a content and P m based on cell number. By contrast, the changing patterns of all photosynthetic parameters examined here during the light period had almost the same for cells grown at ambient CO2 and elevated CO2, suggesting that the photosynthetic rhythms of S. costatum are not affected by CO2 enrichment.

The growth and activity of photosynthetic CO2 uptake and extracellular carbonic anhydrase (CA(ext)) of the marine diatom Skeletonema costatum were investigated while cultured at different levels of CO2 in order to see its physiological response to different CO2 concentrations under either a low (30 mumol . m(-2) . s(-1)) or high (210 mumol . m(-2) . s(-1)) irradiance. The changes in CO2 concentrations (4-31 mumol/L) affected the growth and net photosynthesis to a greater extent under the low than under the high light regime. CAext was detected in the cells grown at 4 mumol/L CO2 but not at 31 and 12 mumol/L CO2, with its activity being about 2.5-fold higher at the high than at the low irradiance. Photosynthetic CO2 affinity (1/K-1/2(CO2)) of the cells decreased with increased CO2 concentrations in culture. The cells cultured under the high-light show significantly higher photosynthetic CO2 affinity than those grown at the low-light level. It is concluded that the regulations of CA(ext) activity and photosynthetic CO2 affinity are dependent not only on CO2 concentration but also on light availability, and that the development of higher CA(ext) activity and CO2 affinity under higher light level could sufficiently support the photosynthetic demand for CO2 even at low level of CO2.

The digestibility of algae by stomachless filter-feeding fish has been debated for decades. Results from gut contents and digestive enzyme analysis suggest poor utilization, while the measurement of food assimilation using radiolabeled isotope techniques indicate reasonable assimilation efficiency. Phytoplankton in the gut contents of the planktivorous filter-feeding silver carp were studied during March-May. The fish were cultured in a large net cage in a shallow, hypertrophic subtropical Chinese lake, Lake Donghu. In terms of biomass, the dominant phytoplankton in the fore-gut contents were Cyclotella (average 77.8%, range 69.7-93.5%) and Cryptomonas (average 9.57%, range 0-20.4%). The Ivlev's electivity index E of silver carp was much higher for Cyclotella (1.54) than for Cryptomonas (0.56). The majority of the phytoplankton found in the intestines of silver carp were 8-20 mu m, but they were also able to collect particles as small as 4.5-10 mu m, smaller than their filtering net meshes, suggesting that the secretion of mucus may play an important role in collecting such small particles. We conclude that disruption of cell walls is principally by the pharyngeal teeth, explaining previous contradictory conclusions. (C) 1999 Elsevier Science B.V. All rights reserved.

The theoretical model of collisional quantum interference (CQI) in intramolecular rotational energy transfer is described in an atom-diatom system, based on the first Born approximation of time-dependent perturbation theory and considering a long-range interaction potential. The relation between differential and integral interference angles is obtained. For the CO A(1)Pi (v = 0)/e(3)Sigma (-)(v = 1)-He collision system, the calculated integral interference angles are consistent with the experimental values. The physical significance of interference angle and the essential factors it depends on as well as the influence of the short-range interaction on CQI are discussed. (C) 2001 Elsevier Science B.V.
