Many studies on environmental ecosystems quality related to polycyclic aromatic hydrocarbons (PAH) have been carried out routinely due to their ubiquotus presence worldwide and to their potential toxicity after its biotransformation. PAH may be introduced into the environmet by natural and anthropogenic processes from direct runoff and discharges and indirect atmospheric deposition. Sources of naturally occurring PAHs include natural fires, natural oil seepage and recent biological or diagenetic processes. Anthropogenic sources of PAHs, acute or chronic, are combustion of organic matter (petroleum, coal, wood), waste and releases/spills of petroleum and derivatives (river runoff, sewage outfalls, maritime transport, pipelines). Besides the co-existence of multiples sources of PAH in the environmental samples, these compounds are subject to many processes that lead to geochemical fates (physical-chemical transformation, biodegradation and photo-oxidation), which leads to an alteration of their composition. All these facts make the identification of the hydrocarbons sources, if petrogenic, pyrolytic or natural, a challenge. One of the objectives of this study is to establish tools to identify the origin of hydrocarbons in environmental samples. PAH diagnostic ratios and PAH principal component analysis were tested on a critical area: Guanabara Bay sediments. Guanabara Bay is located in a complex urban area of Rio de Janeiro with a high anthropogenic influence, being an endpoint of chronic pollution from the Greater Rio and it was the scenario of an acute event of oil release in January 2000. It were quantified 38 compounds, parental and alkylated PAH, in 21 sediment samples collected in two surveys: 2000 and 2003. The PAH levels varied from 400 to 58439 ng g-1. Both tested techniques for origin identification of hydrocarbons have shown their applicability, being able to discriminate the PAH sources for the majority of the samples analysed. The bay sediments were separated into two big clusters: sediments with a clear pattern of petrogenic introduction of hydrocarbons (from intertidal area) and sediments with combustion characteristics (from subtidal region). Only a minority of the samples could not display a clear contribution of petrogenic or pyrolytic input. The diagnostic ratios that have exhibited high ability to distinguish combustion- and petroleum-derived PAH inputs for Guanabara Bay sediments were Phenanthrene+Anthracene/(Phenanthrene+Anthracene+C1Phenanthrene); Fluorantene/(Fluorantene+Pyrene); Σ (other 3-6 ring PAHs)/ Σ (5 alkylated PAH series). The PCA results prooved to be a useful tool for PAH source identification in the environment, corroborating the diagnostic indexes. In relation to the temporal evaluation carried out in this study, it was not verified significant changes on the class of predominant source of the samples. This result indicates that the hydrocarbons present in the Guanabara Bay sediments are mainly related to the long-term anthropogenic input and not directly related to acute events such as the oil spill of January 2000. This findings were similar to various international estuarine sites. Finally, this work had a complementary objective of evaluating the level of hydrocarbons exposure of the aquatic organisms of Guanabara Bay. It was a preliminary study in which a quantification of 12 individual biliar metabolites of PAH was performed in four demersal fish representing three different families. The analysed metabolites were 1-hydroxynaphtalene, 2-hidroxinaphtalene, 1hydroxyphenanthrene, 9-hydroxyphenanthrene, 2-hydroxyphenanthrene, 1hydroxypyrene, 3-hidroxibiphenil, 3- hydroxyphenanthrene, 1-hydroxychrysene, 9hydroxyfluorene, 4-hydroxyphenanthrene, 3-hydroxybenz(a)pyrene. The metabolites concentrations were found to be high, ranging from 13 to 177 µg g-1, however they were similar to worldwide regions under high anthropogenic input. Besides the metabolites established by the used protocol, it was possible to verified high concentrations of three other compounds not yet reported in the literature. They were related to pyrolytic PAH contribution to Guanabara Bay aquatic biota: 1-hydroxypyrine and 3-hydroxybenz(a)pyrine isomers

Efluentes industriais e domésticos podem conter agentes químicos e biológicos que, em elevadas concentrações, causam danos aos ecossistemas aquáticos e à saúde ambiental. Um dos efeitos mais nocivos desses agentes poluidores é a sua capacidade de induzir alterações celulares. O uso de testes específicos capazes de detectar o potencial tóxico de substâncias químicas caracteriza-se em uma importante estratégia para avaliação ambiental. Neste contexto, o presente estudo teve como objetivo avaliar os potenciais citotóxico, genotóxico e mutagênico de amostras de água de recursos hídricos da cidade de Santa Gertrudes-SP, relacionada com recebimento de efluentes de atividade ceramista, por meio de ensaios realizados com o sistema-teste de Allium cepa. Foram realizados testes com amostras de água coletadas no córrego Itaqui, após despejos de efluente de indústria cerâmica e em duas nascentes, tanto em período chuvoso como de seca. Para a análise dos parâmetros de toxicidade descritos acima, sementes de A. cepa foram expostas à germinação nas amostras das águas coletadas. Quando as radículas atingiram 2 cm de comprimento, os meristemas foram fixados em solução de Carnoy 3:1, para serem utilizadas nas avaliações das possíveis alterações dos índices de germinação e mitóticos, bem como de indução de aberrações nucleares e cromossômicas. Além disso, também foram avaliadas, como outro parâmetro indicativo de mutagenicidade, as frequências de micronúcleos em células F1 de A. cepa. O controle negativo foi realizado em água (osmose reversa) e o controle positivo em metilmetanosulfonato (MMS). O material fixado foi corado pela reação de Feulgen e as lâminas foram preparadas, utilizando as porções meristemática e F1 das raízes de A. cepa. Devido à alta ação antrópica em rios, lagos e mares, a ictiofauna sofre sérios impactos que podem prejudicar sua dinâmica populacional. No interior do estado de São Paulo, muitos municípios captam águas superficiais para o abastecimento urbano e lançam seus efluentes à jusante da captação. Pode-se dizer que o mesmo acontece com o município de Rio Claro. O presente trabalho de conclusão de curso avaliou os efeitos da poluição de despejos orgânicos sobre a diversidade da ictiofauna e a alimentação, a reprodução e o fator de condição de Astyanax fasciatus, levando em consideração a sazonalidade e a variação de parâmetros físico-químicos da água comparando dois pontos do Ribeirão Claro (Rio Claro – SP) no interior da “Floresta Estadual Edmundo Navarro de Andrade”: um à jusante da captação d’água da cidade (área menos poluída) e outro logo após o despejo dos esgotos (área mais poluída). Foram coletados no total 1274 exemplares distribuídos em 26 espécies de peixes nas quatro coletas realizadas. No ponto 1 – área menos poluída, foram coletados 1121 exemplares de 26 espécies nas estações cheia e seca. Na estação cheia foram capturados 401 indivíduos pertencentes a 19 espécies, enquanto que na estação seca foram coletados 720 indivíduos distribuídos em 24 espécies. No ponto 2 – área mais poluída, foram coletados 153 exemplares de 14 espécies. Na estação cheia, foram capturados 141 indivíduos distribuídos em 11 espécies, enquanto que na estação seca foram coletados 12 indivíduos pertencentes a 6 espécies. Dentre os parâmetros estudados, oxigênio dissolvido e coliformes fecais foram um dos mais discrepantes entre os dois pontos e entre as coletas, auxiliando assim, na caracterização do ambiente estudado. The growing interest in the use of groundwater resources is directly related to the economic advantages that the groundwater exploitation offers when compared to surface waters. This happens especially in large urban centers, such as the city of Americana / SP, where the rivers are increasingly contaminated by household and industrial waste. Therefore, this study aimed to characterize the Tubarão Aquifer System, in the city of Americana, to identify and evaluate the spatial distribution of different hydrogeochemical facies as well as understand the rock-fluid interaction through the construction of a conceptual hydrogeochemical model. This study was made based on the recognition of the possible chemical reactions that print the chemical characteristics of groundwater in the area. To do the job, there were two water sampling campaigns of all deep wells used by the City of Americana public water supply. From the results of hydrochemical, classification of water was made by Piper and Stiff diagrams as well as geostatistical data using cluster analysis of principal components. Based on information from the profiles obtained from the survey SIAGAS as well as in geological profiles provided by the city of Americana, we sought to detail the subsurface geology of the Subgroup Itararé in the city of Americana. The results obtained allowed the identification of three hydrochemical types in the study area: Bicarbonated calcium-sodium (1), bicarbonate sodium (2) and sodium chloride (3). The waters have bicarbonate alkaline pH to alkaline and can be considered weakly saline, with electrical conductivity values of around 161 mS / cm. Samples classified as sodium bicarbonate average of 174.99 mS / cm. The pH values ranging from 6.74 to 7.99, averaging 7.52. The industrial effluents are one of the main sources of water pollution. For an appropriate characterization and control of their discharges, the most efficient strategy is the integrated use of chemical, physical and ecotoxicological analyses. The aims of this study were to asses the efficiency of the treatment plant of a textile industry performing acute toxicity tests and physical-chemical analyses of the effluents before and after the treatment, besides evaluate the toxicity of the effluents of the Treatment System of Liquids Effluents (Sistema de Tratamento de Efluentes Líquidos - SITEL) of Distrito Industrial de Natal (DIN) and some of their physical-chemical variables. The species used in the ecotoxicological tests was the fish Danio rerio. The results showed that the treatment plant reduced significantly (around 50%) the toxicity of the raw textile effluent in only three of the seven tests but, in general, it promoted the reduction of the physical-chemical parameters analyzed. The toxicity and the physical-chemical factors of the effluents of SITEL of DIN varied among the tests and show the importance of monitoring their discharges in the Potengi river, one of the most important rivers of the Rio Grande do Norte state

Methomyl (Lannate®) is an insecticide from the carbamate group, frequently used in pest control in various types of crops. This compound works inhibiting the activity of the enzyme acetylcholinesterase. The use of physicochemical and ecotoxicological analysis is the most efficient strategy for the correct characterization and control of residues of metomil. The main objectives of this study were to evaluate the acute toxicity of methomyl in 96 hours of exposure and, through a sublethal assay of 5 hours, to assess its effect on the activity of acetylcholinesterase present in brain and squeletic dorsal muscle of the Danio rerio fish. The results showed that the LC50-96 found to D. rerio was 3.4 mg/L and it was found through the average of four definitive tests. In vitro assays were used to test the inhibitory action of methomyl directly over soluble AChE, extracted from the squeletic dorsal muscle, with maximum inhibition of 68.57% to the insecticide concentrations of 0.2 mg/L. In sublethal tests with D. rerio, inhibitory effect of methomyl was found over the soluble form of AChE in the squeletic dorsal muscle, both in one and five hours of fish exposure to the insecticide. In both period, the average values of inhibition were around 61%. In the same condition, no significant inhibitory effect of methomyl soluble and membrane AChE of the D. rerio was observed in the 0.42, 0.85, 1.70 and 2.50 mg/L concentrations and in both times of fish exposure
