This work is about the role that environment plays in the production of evolutionary significant variations. It starts with an historical introduction about the concept of variation and the role of environment in its production. Then, I show how a lack of attention to these topics may lead to serious mistakes in data interpretation. A statistical re-analysis of published data on the effects of malnutrition on dental eruption, shows that what has been interpreted as an increase in the mean value, is actually linked to increase of variability. In Chapter 3 I present the topic of development as a link between variability and environmental influence, giving a review of the possible mechanisms by which development influences evolutionary dynamics. Chapter 4 is the core chapter of the thesis; I investigated the role of environment in the development of dental morphology. I used dental hypoplasia as a marker of stress, characterizing two groups. Comparing the morphology of upper molars in the two groups, three major results came out: (i) there is a significant effect of environmental stressors on the overall morphology of upper molars; (ii) the developmental response increases morphological variability of the stressed population; (iii) increase of variability is directional: stressed individuals have increased cusps dimensions and number. I also hypothesized the molecular mechanisms that could be responsible of the observed effects. In Chapter 5, I present future perspectives for developing this research. The direction of dental development response is the same direction of the trend in mammalian dental evolution. Since malnutrition triggers the developmental response, and this particular kind of stressor must have been very common in our class evolutionary history, I propose the possibility that environmental stress actively influenced mammals evolution. Moreover, I discuss the possibility of reconsidering the role of natural selection in the evolution of dental morphology.

Our studies of the teeth and faces of Australian twins commenced at the School of Dentistry, The University of Adelaide in the early 1980s. There are now over 900 pairs of twins enrolled in our continuing investigations, together with 1200 relatives. There are 3 main cohorts of participants. The first cohort comprises around 300 pairs of teenage twins for whom various records have been collected, including dental casts, facial photographs, finger and palm prints and information on laterality, including handedness. The second cohort comprises around 300 pairs of twins who have been examined at 3 stages of dental development from approximately 4 years of age to about 14 years: at primary, mixed, and permanent dentition (excluding 3rd molars) stages. The most recent study of tooth emergence and oral health, for which we are currently recruiting twins, will provide a third cohort of around 500 twin pairs aged from around birth to 3 to 4 years of age. Our broad aim in these studies has been to improve our understanding of how genetic and environmental factors contribute to variation in dental and facial features, and to oral health. We have also used our data to investigate aspects of the determination of laterality, particularly the fascinating phenomenon of mirror imaging. We plan to maximize the use of the longitudinal data and DNA we have collected, and continue to collect, by performing genome-wide scans for putative genetic linkage peaks for a range of dental features, and then to test for association between a series of likely candidate genes and our phenotypes.

O objetivo deste estudo retrospectivo foi comparar a eficiência oclusal do tratamento ortopédico com os aparelhos funcionais Regulador de Função Fränkel-2 e Bionator de Balters em um estágio de desenvolvimento dental diferente e comparar com um grupo controle. A amostra constituiu-se de 45 registros de documentações, pertencentes ao arquivo do programa de pós-graduação em Odontologia, área de concentração Ortodontia, da Universidade Metodista de São Paulo, com má oclusão inicial de Classe II bilateral, divisão 1, sendo 15 pacientes provenientes do grupo tratados com Bionator (grupo 1) com média de idade incial de 8,56 anos e com 80% dos casos em um estágio de desenvolvimento dental-2 (DS 2), 15 pacientes tratados com RF-2 (grupo 2) com média de idade inicial de 10,71 anos e com 80% dos casos em um estágio de desenvolvimento dental-3 (DS 3), e 15 pacientes controle (grupo 3) com media de idade incial de 10,03 anos e com estágio de desenvolvimento dental compatível com os grupos 1 e 2. Os grupos foram divididos em duas fases, de acordo com o período de avaliação: T1:início de tratamento e T2: final de tratamento, totalizando 90 pares de modelos. As avaliações oclusais foram realizadas em modelos de gesso, utilizando o Índice PAR com auxílio da régua PAR e de um paquímetro digital devidamente calibrado. Para comparação entre os três grupos foi utilizado Análise de Variância a um critério e em seguida o Teste de Tukey. A severidade da má oclusão (PAR Inicial) foi semelhante em ambos os grupos, porém, o PAR final apresentou uma diferença estatisticamente significante onde o percentual de redução do índice PAR para o grupo 1 foi de 20,72%, para o grupo 2 foi de 60,06% e no grupo 3 não houve alteração significante do valor do Índice PAR. O presente estudo conclui que o tratamento da má oclusão de Classe II, 1a divisão é mais eficiente quando iniciado no estágio de desenvolvimento dental 3 (DS 3) do que no estágio de desenvolvimento dental 2 (DS2). Além disso, ressalta-se a importância do uso mais prolongado do aparelho ortopédico, já que os pacientes do grupo 2 apresentaram melhores resultados oclusais.(AU)

Our long-term objective is to devise reliable methods to generate biological replacement teeth exhibiting the physical properties and functions of naturally formed human teeth. Previously, we demonstrated the successful use of tissue engineering approaches to generate small, bioengineered tooth crowns from harvested pig and rat postnatal dental stem cells (DSCs). To facilitate characterizations of human DSCs, we have developed a novel radiographic staging system to accurately correlate human third molar tooth developmental stage with anticipated harvested DSC yield. Our results demonstrated that DSC yields were higher in less developed teeth (Stages 1 and 2), and lower in more developed teeth (Stages 3, 4, and 5). The greatest cell yields and colony-forming units (CFUs) capability was obtained from Stages 1 and 2 tooth dental pulp. We conclude that radiographic developmental staging can be used to accurately assess the utility of harvested human teeth for future dental tissue engineering applications.

This paper presents a novel design of a virtual dental training system (hapTEL) using haptic technology. The system allows dental students to learn and practice procedures such as dental drilling, caries removal and cavity preparation for tooth restoration. This paper focuses on the hardware design, development and evaluation aspects in relation to the dental training and educational requirements. Detailed discussions on how the system offers dental students a natural operational position are documented. An innovative design of measuring and connecting the dental tools to the haptic device is also shown. Evaluation of the impact on teaching and learning is discussed.

Two high-performance liquid chromatographic methods for determination of residual monomer in dental acrylic resins are described. Monomers were detected by their UV absorbance at 230 nm, on a Nucleosil((R)) C-18 (5 mu m particle size, 100 angstrom pore size, 15 x 0.46 cm i.d.) column. The separation was performed using acetonitrile-water (55:45 v/v) containing 0.01% triethylamine (TEA) for methyl methacrylate and butyl methacrylate, and acetonitrile-water (60:40 v/v) containing 0.01% TEA for isobutyl methacrylate and 1,6-hexanediol dimethacrylate as mobile phases, at a flow rate of 0.8 mL/min. Good linear relationships were obtained in the concentration range 5.0-80.0 mu g/mL for methyl methacrylate, 10.0-160.0 mu g/mL for butyl methacrylate, 50.0-500.0 mu g/mL for isobutyl methacrylate and 2.5-180.0 mu g/mL for 1,6-hexanediol dimethacrylate. Adequate assay for intra- and inter-day precision and accuracy was observed during the validation process. An extraction procedure to remove residual monomer from the acrylic resins was also established. Residual monomer was obtained from broken specimens of acrylic disks using methanol as extraction solvent for 2 h in an ice-bath. The developed methods and the extraction procedure were applied to dental acrylic resins, tested with or without post-polymerization treatments, and proved to be accurate and precise for the determination of residual monomer content of the materials evaluated. This article discusses the development of a test bench and a methodology for the study of composite resin abrasive wear. To evaluate the operation of the test bench and to compare the proposed methodology with other existing ones, a study was made of the five composites most commonly used by dentists.The one-way ANOVA method and the Tukey test were used to statistically analyze the results by multiple comparisons of the groups of resins tested. Using the proposed methodology, these resins were classified in an increasing order of abrasive wear strength, as follows: Charisma (the lowest abrasive wear strength), Tetric, TPH, Herculite and Z-100 (the highest abrasive wear strength) (P < 0.05). In comparison to other methodologies, the results of the proposed methodology presented the lowest coefficient of variation. (C) 2002 Elsevier B.V. B.V. All rights reserved.
