Fungal metabolism of halogenated and related steroids was investigated. The fungi Aspergillus niger ATCC 9142, Curvularia lunata NRRL 2380 and Rhizopus stolonifer ATCC6227b were studied in this regard. 2l-Fluoro-, 2l-chloro, 2l-bromo- and 2l-methyl-pregn-4-ene-3,20diones were prepared and incubated with ~ niger (a C-2l-hydroxylator) in order to observe the effect of the C-2l substituent on the metabolism of these substrates. In all four cases, the C-2l substituent prevented any significant metabolism of these substrates. llB-Fluoropregn-4-ene-3,20-dione was prepared and incubated with C. lunata (an llB-hydroxylator) and ~ stolonifer (an lla-hydroxylator). With ~ lunata, the ll-fluoro- substituent prevent hydroxylation at the 11 position, but diverted it to a site remote from the fluorine atom. In contrast, with ~ stolonifer the llB-fluoro- substituent, although slowing the apparent rate of hydroxylation, did not prevent its occurrence at the 11a- position. llB-Hydroxypregn-4-ene-3,20-dione was also incubated with R. stolonifer. The llB-hydroxy-;group did not appear to have any significant effect on hydroxylation at the lla- position. The incubation of a substrate, unsaturated at a favoured site of hydroxylation with Rhizopus arrhizus ATCC 11145 provided a complex mixture of products; among them were both the a and S epoxides. The formation of these products is rationalized as arising because of the lack of regio- and stereospecificity of the hydroxylase enzyme(s) involved.

The purpose of the study was to determine the ability of certain fungi to biotransform morphine alkaloids into medicinally relevant intermediates. Fungal strains screened for their ability to affect biotransformation of morphine alkaloids include Cunninghamella echinulata, Helicostylum pirijorme, Pycnoporus sanguinea, Pycnoporus cinnabarina, Curvularia lunata and Sporotrichum sulfurescens. The research demonstrated that Cunninghamella echinulata N-demethylated thebaine, hydrocodone, codeine, oripavine and oxycodone into corresponding nor-compounds in varying yields. The study further focused on the characterization of the enzyme responsible for the biotransformation of thebaine into northebaine by Cunninghamella echinulata. The study clearly showed that incubation of the fungal culture with thebaine over a period of 48 hours was required to activate the biotransformation process. The biotransformation studies with [14C] labeled thebaine showed that Ndemethylation by Cunningham ella echinulata does not involve O-demethylation followed by methyl group transfer as suggested in previous studies.

O conhecimento dos fungos anemófilos em determinada cidade ou região é importante para o diagnóstico etiológico e o tratamento específico das manifestações alérgicas respiratórias. Várias técnicas quantitativas e qualitativas são preconizadas para coleta e identificação desses fungos na dependência do local estudado. Nesta pesquisa, sobre esporos de fungos do ar, foi utilizado o equipamento Rotorod Sampler®, que retira a amostra do ar através de um bastão de plástico preso a um motor elétrico que o faz girar rapidamente, sendo as partículas suspensas no ar recolhidas pelo bastão. As amostras foram coletadas uma vez por semana, durante 24 horas, correspondendo a um ciclo de coleta. Foram realizadas 52 coletas entre abril de 2000 e março de 2001. Os resultados mostraram prevalência de ascosporos (50,49%), Cladosporium (17,86%), Aspergillus/Penicillium (15,03%), basidiosporos (3,84%), rusts (3,82%), Helminthosporium (2,49%), Botrytis (1,22%), Alternaria (1,19%), smuts (0,90%), Curvularia (0,87%), Nigrospora (0,61%) e Fusarium (0,08%). Não foi possível identificar 1,59% dos esporos de fungos coletados no período. O maior número de esporos foi observado nos meses de verão e o menor, durante o outono. Utilizando provas in vivo e in vitro, avaliou-se a hipersensibilidade a fungos entre 39 pacientes atópicos sofrendo de rinite e ou asma brônquica. Os testes cutâneos identificaram sensibilização em 17,94% dos pacientes, enquanto as provas sorológicas caracterizaram presença de IgE específica em 12,82% dos casos avaliados. A detecção de significativo número de esporos de fungos no ar de Porto Alegre, com muitas espécies comprovadamente alergênicas, e os índices de sensibilização observados em indivíduos atópicos confirmam a importância do estudo dos fungos anemófilos nessa cidade, com vistas a aprimorar o diagnóstico e o manejo de pacientes com manifestações alérgicas respiratórias.

Este trabalho teve por objetivo determinar a ocorrência e a freqüência de fungos em banana 'Prata anã' e elucidar o agente causal das podridões em pós-colheita de frutos provenientes do norte de Minas Gerais. Dois métodos de isolamento foram adotados: diluição em placas, a partir da lavagem de frutos verdes, e direto de frutos maduros. Os fungos Colletotrichum musae, Trichoderma harzianum, Fusarium equisetii, Penicillium sp. Aspergillus parasiticus, Trichothecium roseum, Colletotrichum acutatum, Alternaria sp., Cladosporium musae e Curvularia lunata foram os mais freqüentemente associados aos frutos. A patogenicidade desses fungos foi testada pela substituição de discos da casca de frutos verdes por discos de micélio. Colletotrichum musae apresentou área média lesionada em torno do ponto de inoculação igual a 5,8 cm², enquanto para os demais fungos testados não passou de 1,50 cm². Os resultados mostraram que C. musae é o agente primário das podridões dos frutos examinados com 100 % de incidência e os demais fungos limitaram-se a necrosar os ferimentos em torno do ponto de inoculação. O modo de infecção latente, causada por C. musae, parece favorecer, primeiramente, a colonização interna dos tecidos e, posteriormente, a ação dos fungos oportunistas, que aceleram as podridões nos frutos e na coroa.

Objectives To evaluate the presence of fungi on contact electrodes and ultrasound transducers from physiotherapy clinics.Design Quantitative study conducted at the Laboratory of Microbiology and Immunology, Faculty of Health Sciences and Technology of Piaui - NOVAFAPI, Teresina, Brazil.Setting Sample collection was performed in 10 clinics ( 20 ultrasound transducers and 20 contact electrodes).Main outcome measures Swabs were soaked with saline solution, inoculated in culture and incubated for filamentous fungi and yeast growth.Results Fourteen taxons were identified: Acremomium hyalinulum (Sacc.), Aspergillus terreus, Candida albicans, Cladosporium cladosporioides, Cladosporium elatum, Cladosporium oxysporum, Cladosporium sphaerospermum, Cladosphialophora bantiana, Curvularia clavata, Curvularia senegalensis, Fusarium oxysporum, Penicillium decumbens, Scopulariopsis candida and Sporothrix schenckii. Aspergillus terreus, Cladosporium oxysporum, Sporothrix shenckii and Candida albicans were found most often on contact electrodes, and Penicillium decumbens and Cladosporium cladosporioides were found most often on ultrasound transducers.Conclusion Fungi were found on all of the contact electrodes and ultrasound transducers. Neste trabalho, objetivou-se avaliar o efeito de métodos de superação de dormência e do ambiente de armazenamento sobre a qualidade fisiológica e fitopatológica das sementes de canafístula (Peltophorum dubium). As sementes foram submetidas aos seguintes tratamentos de superação de dormência: escarificação com lixa (200); imersão em água na temperatura ambiente, durante 24 e 72 h; imersão em ácido sulfúrico por 2, 6, 10, 15, 20 e 30 min; imersão em água quente (70, 80 e 90 C); e umedecimento do substrato com solução de KNO3 (0,2%). As sementes foram armazenadas na temperatura ambiente e a 10 C por 210 dias. Os efeitos dos tratamentos e do armazenamento foram avaliados por meio do teor de água, teste de germinação (cinco repetições de 30 sementes), de comprimento de plântulas e sanidade (400 sementes), com incubação por oito dias (22-25 C). Na análise estatística dos dados, utilizou-se o delineamento experimental inteiramente casualizado em esquema fatorial 2 x 14 (condições de armazenamento x tratamentos para a superação da dormência). As médias foram comparadas pelo teste de Tukey (P>0,5). Com relação às sementes não armazenadas, os melhores tratamentos para superar a dormência e promover a germinação foram escarificação com lixa ou ácido sulfúrico por 15 a 30 min; quanto às sementes armazenadas, houve a imersão em água quente (70 a 80 ºC). Os fungos detectados nas sementes foram Pestalotia sp., Alternaria sp., Rhizopus sp., Nigrospora sp., Curvularia sp., Fusarium sp., Rhizoctonia sp., Aspergillus sp., Cladosporium sp. e Fusarium semitectum.

The experiment was conducted with grama seda (Cynodon dactylon (L.) Pers.) hays stored with a low moisture content (12-15%) and without chemical treatment, and hays stored with a high moisture content (20-25%) and treated with anhydrous ammonia (NH3) al 0.5 and 1.0% of DM, and urea at 0.9 and 1.8% of DM. At 65 days after treatment (AT) under a plastic cover, the bales were opened and samples were taken at 3, 15 and 30 days to determine the chemical composition and in vitro digestibility (IVDMD) of the hays. For the identification of fungi, samples were taken at 0, 15 and 30 days AT. The data were analyzed according to a split-plot design with the effects of the chemical treatments studied in the main plot and the effects of the periods of post-treatment studied in the sub-plots, Fourteen genera of fungi were observed in the hays, not treated and treated with NH3 and urea, with a higher occurrence of Cladosporium, Curvularia, Aspergillus, and Penicillium. Treatment with anhydrous ammonia and 1.8% urea controlled the occurrence of Aspergillus; however, Penicillium decreased in hays treated with ammonia 30 days AT. Ammoniation did not influence the contents of ADF, cellulose and lignin in the hays, but NDF and hemicellulose decreased with the use of ammonia 30 days AT. The CP contents and the IVDMD increased with ammoniation. The CP contents decreased in hays treated with NH3 as days AT increase, while hays treated with urea did not change.

This study was conducted to evaluate the effects of anhydrous ammonia (NH3) or urea treatment on the occurrence of fungi in hays of Brachiaria decumbens Stapf cv. Basilisk, baled and stored with different moisture contents. The following treatments were evaluated: T-1 = green forage, soon after the harvest; T-2 = hay with 25% of moisture, with evaluation before baling; T-3 = hay with 13% of moisture and untreated; T-4 = hay with 25% of moisture + .5% of NH3 in the DM T-5 = hay with 25% of moisture +1% of NH3 in the DM; T-6 = hay with 25% of moisture +.9% of urea in the DM and T-7 = hay With 25% of moisture +1.8% of urea in the DM. The treated hays stayed under plastic cover during 75 days, and samples were collected soon after the opening of the bales piles,and they were immediately analyzed in the laboratory. Eleven fungi were identified in different treatments, with high incidence of Aspergillus in the untreated hay, in the hay stored with 25% of moisture and treated with .5% NH3. The ammoniation totally reduced the occurrence of Helminthosporium and Nigrospora, but it did not control the occurrence of Cladosposrium and caused the occurrence of Penicillium in large intensity. The occurrence of Epicoccum, Curvularia, Phitomyces and Aspergillus genus were totally controlled by the treatment of hay with 1% of NH3; .9 and 1.8% of urea.
Qualidade fisiológica e sanitária de sementes de mamona submetidas a diferentes tratamentos químicos

The use of fungicides for treating seeds has become an essential measure in the control of pathogens. However, the effectiveness of many products is not well-known to some crops. The objective of this research was to evaluate the effect of different doses of carbendazin and carboxin + thiram fungicides for controlling pathogens and their interference on the physiologic quality of castor bean seeds. The test consisted of the following treatments (g i.a./100 kg of seeds): carbendazin (15, 30, and 60), carboxin + thiram (25 + 25, 50 + 50, and 100 + 100), formol 0.15% (v/v), and control (distilled water). The physiologic quality of the seeds was determined by germination, first counting, field emergence, speed emergence index, length, fresh and dry weight of the aerial part, and electrical conductivity. There were no statistical differences for germination and fresh and dry matter index. The treatment with formol provided lower values for first counting and field emergence. Seeds treated with fungicides presented higher values for electrical conductivity, but did not differ, statistically, among themselves. The fungi Cladosporium spp.; Bipolaris spp.; Curvularia spp.; Aspergillus flavus; Aspergillus niger; Rhizopus sp.; Penicillium sp.; Rhizoctonia sp.; Verticillium sp.; Fusarium sp.;Arthrobotrys sp.; and Epicocum sp. were identified. All tested products, independently of the dose, provided efficient control for pathogens.

Palm trees belong to the Arecaceae family and are widely utilized in landscaping of tropical countries. Although there are many species, just a few are used in Brazil's landscaping, the majority coming from other countries. So, this study had the objective to characterize the plants in the collection of palm trees at UNESP/FCAV, Campus of Jaboticabal, Sao Paulo State, Brazil. The germination of seeds was also studied to increase the production of seedlings for use in landscaping, as this is the biggest difficulty for spreading new species. The introduction of new species in the collection was accomplished. It was also made a survey of major pests and diseases that occur in the collection. The most common pests found were: Brassolis sophorae, Parisoschoenus obesulus, Rhynchophorus palmarum and Coraliomela sp., and the most common disease causing fungi: Colletrotrichum sp., Curvularia sp., Bipolaris sp., Helminthosporium sp., Alternaria sp., Phyllosticta sp., Pestalotia sp., Exosporium palmivorum and Ceratocystis pardoxa.
