Total hip replacement is the golden standard treatment for severe osteoarthritis refractory for conservative treatment. Aseptic loosening and osteolysis are the major long-term complications after total hip replacement. Foreign body giant cells and osteoclasts are locally formed around aseptically loosening implants from precursor cells by cell fusion. When the foreign body response is fully developed, it mediates inflammatory and destructive host responses, such as collagen degradation. In the present study, it was hypothesized that the wear debris and foreign body inflammation are the forces driving local osteoclast formation, peri-implant bone resorption and enhanced tissue remodeling. Therefore the object was to characterize the eventual expression and the role of fusion molecules, ADAMs (an abbreviation for A Disintegrin And Metalloproteinase, ADAM9 and ADAM12) in the fusion of progenitor cells into multinuclear giant cells. For generation of such cells, activated macrophages trying to respond to foreign debris play an important role. Matured osteoclasts together with activated macrophages mediate bone destruction by secreting protons and proteinases, including matrix metalloproteinases (MMPs) and cathepsin K. Thus this study also assessed collagen degradation and its relationship to some of the key collagenolytic proteinases in the aggressive synovial membrane-like interface tissue around aseptically loosened hip replacement implants. ADAMs were found in the interface tissues of revision total hip replacement patients. Increased expression of ADAMs at both transcriptional and translational levels was found in synovial membrane-like interface tissue of revision total hip replacement (THR) samples compared with that in primary THR samples. These studies also demonstrate that multinucleate cell formation from monocytes by stimulation with macrophage-colony stimiulating factor (M-CSF) and receptor activator of nuclear factor kappa B ligand (RANKL) is characterized by time dependent changes of the proportion of ADAMs positive cells. This was observed both in the interface membrane in patients and in two different in vitro models. In addition to an already established MCS-F and RANKL driven model, a new virally (parainfluenza 2) driven model (of human salivary adenocarcinoma (HSY) cells or green monkey kidney (GMK) cells) was developed to study various fusion molecules and their role in cell fusion in general. In interface membranes, collagen was highly degraded and collagen degradation significantly correlated with the number of local cells containing collagenolytic enzymes, particularly cathepsin K. As a conclusion, fusion molecules ADAM9 and ADAM12 seem to be dynamically involved in cell-cell fusion processes and multinucleate cell formation. The highly significant correlation between collagen degradation and collagenolytic enzymes, particularly cathepsin K, indicates that the local acidity of the interface membrane in the pathologic bone and soft tissue destruction. This study provides profound knowledge about cell fusion and mechanism responsible for aseptic loosening as well as increases knowledge helpful for prevention and treatment.

We describe in some detail the process of development of a dynamic model of a three wheeled vehicle using ADAMS-CAR. We first describe the rigid body model, and then the modeling of structural flexibilities. The aim of this report is to document procedural details of such modeling, with a view to presenting more research and development oriented investigations in the future. The contents of this report may also be of interest to practicing engineers engaged in multi-body dynamics modeling of wheeled vehicles.

ENGLISH: One phase of the duties of the Inter-American Tropical Tuna Commission is the gathering and interpretation of data concerning the life history of the commercially important bait species throughout the Eastern Pacific Ocean. During 1958 the Commission established a laboratory in Manta, Ecuador to study tuna. It was subsequently found that this fishery was dependent upon one species of anchovy, Anchoa naso, which was locally referred to as "colorado." During the calendar year 1959 approximately 380,000 scoops of bait were taken by the Manta tuna fleet (Schaefer, 1960), which at that time numbered about 23 vessels. Since then the fleet has increased by about 25 per cent and it is probable that the bait catch has increased also. Virtually nothing has been reported concerning the life history of this species. Hildebrand (1943) reviewed its taxonomy and reported standard lengths ranging from 32 to 135 mm. Peterson (1956) examined specimens from Central America and found them to range from 27 to 66 mm. He also indicated that the species spawned over a long period of time. The present report describes some aspects of the life history of Anchoa naso in Ecuadorian waters. The findings are based on 121 collections taken during the period March 1959 through June 1961. SPANISH: Una fase de las obligaciones de La Comisión Interamericana del Atún Tropical es la obtensión e interpretación de los datos concernientes a la historia natural de las especies de carnada comercialmente importantes en todo el Océano Pacifico Oriental. En el año de 1958 la Comisión estableció un laboratorio en Manta, Ecuador, para estudiar el atún. Se encontró subsecuentemente que esta pesquería dependía de una especie de anchoa, Anchoa naso, conocida localmente con el nombre de colorado. Durante el año calendario de 1959, la flota atunera de Manta, que en ese tiempo alcanzaba a unos 23 barcos, obtuvo aproximadamente 380,000 copas (scoops) de carnada (Schaefer, 1960). Desde entonces la flota ha aumentado en un 25 por ciento, y es probable que la captura de peces-cebo haya aumentado también. Nada se ha informado virtualmente sobre la historia natural de esta especie. Hildebrand (1943) revisó su taxonomia e informó sobre su longitud estándar, que varia entre los 32 y 135 mm. Some aspects of the reproduction of Nematopalaemon hastatus in the artisanal shrimp fishery in the outer Cross River estuary (Nigeria) was investigated. 2 kg samples were taken from the daily catch of the N. hastatus fishery on a twice-weekly basis. The N. hastatus in the sample were then observed for gravidity, using a hand lens, and their post-orbital carapace length (POCL) taken. This was done over a 12-month period. The eggs in formalin-fixed samples showed colour changes from light orange to dark brown with maturation. Gravid females with eggs in different stages of maturation, from light orange to dark brown were observed in the samples. The reproduction occurs all year-round. The proportion of gravid females observed in samples had two distinct peaks in June and November, and two distinct troughs in May and October. The lowest length at which gravidity was observed was 6.0 mm POCL. There were no Penaeus notialis in the samples

A dissertação comenta criticamente as interpretações recentes referentes ao vitalismo no século XVIII, dedicando atenção especial aos Nouveuax Éléments de la Science de lHomme (publicado primeiramente em 1778), de Paul-Joseph Barthez (1734-1806). Até a segunda metade do século XX, como é primeiramente argumentado nesta dissertação, intérpretes do iluminismo entendiam a doutrina mecanicista como a herdeira direta da Revolução Científica, bem como a corrente dominante no mundo das ciências da vida ao longo de todo o século XVIII. Assim, na historiografia do século passado, o vitalismo era ou escassamente mencionado, ou visto como uma retrógrada corrente anti-iluminista. Mais recentemente, vários historiadores e pesquisadores da história das ciências no século XVIII (sobretudo Williams e Reill) entendem o iluminismo de um modo mais amplo e plural, considerando o vitalismo iluminista (um termo proposto por Reill) como parte integrante de um conceito mais dinâmico de iluminismo. A seguir, são apresentados a doutrina mecanicista e seus conceitos centrais, bem como as ideias de alguns dos principais representantes do mecanicismo no século XVII e início do XVIII, no caso, mais especificamente, do mecanicismo newtoniano. Em seguida, são expostos e comentados a doutrina vitalista e seus conceitos, no que é dado destaque ao vitalismo na Universidade de Montpellier. Nesse contexto, são comentados conceitos vitalistas, tal como apresentados nos Nouveuax Éléments de la Science de lHomme, no qual Barthez propõe uma nova fisiologia baseada no princípio vital; nisso são apresentados sua metodologia de pesquisa, o conceito de princípio vital, as forças sensitivas e motrizes do princípio da vida, além dos conceitos de simpatia, sinergia e, por fim, o conceito de temperamento. Esses conceitos ou essa terminologia , tal como é mostrado, não são originalmente concebidos por Barthez, mas foram por ele reapropriados e reformulados em debate com o newtonianismo e demais correntes filosóficas médicas desde a Antiguidade até o século XVIII, assim como com observações e experimentos próprios às investigações médico-científicas da época. Como resultado, é alcançada uma compreensão da doutrina vitalista como um esforço intelectual inovador tanto interagindo quanto integrado com o debate científico contemporâneo, ou seja, os médicos vitalistas se viam e, em geral, eram vistos como atuando segundo os padrões de cientificidade exigidos por seus pares.
