A low molecular mass alkaliphilic extra-cellular lipase of Bacillus cereus MTCC 8372 was purified 35-fold by hydrophobic interaction (Octyl-Sepharose) chromatography. The purified enzyme was found to be electrophoretically pure by denaturing gel electrophoresis and possessed a molecular mass of approximately 8 kDa. It is a homopentamer of 40 kDa as revealed by native-PAGE. The lipase was optimally active at 55 °C and retained approximately half of its original activity after 40 min incubation at 55 °C. The enzyme was maximally active at pH 8.5. Mg 2+ , Cu 2+ , Ca 2+ , Hg 2+ , Al 3+ and Fe 3+ at 1 mM enhanced hydrolytic activity of the lipase. Interestingly, Hg 2+ ions synergized and Zn 2+ and Co 2+ ions antagonized the lipase activity. Among surfactants, Tween 80 promoted the lipase activity. Phenyl methyl sulfonyl fluoride (PMSF, 15 mM) decreased 98% of original activity of lipase. The lipase was highly specific towards p -nitrophenyl palmitate and showed a V max and K m of 0.70 mmol.mg −1 .min −1 and 32 mM for hydrolysis of p NPP.

A wide range of fatty acid esters can be synthesized by esterification and transesterification reactions catalyzed by lipases in non-aqueous systems. In the present study, immobilization of a purified alkaline extra-cellular lipase of Bacillus cereus MTCC 8372 by adsorption on diatomaceous earth (celite) for synthesis of ethyl acetate via transesterification route was investigated. B. cereus lipase was deposited on celite (77% protein binding efficiency) by direct binding from aqueous solution. Immobilized lipase was used to synthesis of ethyl acetate from vinyl acetate and ethanol in n -nonane. Various reaction conditions, such as biocatalyst concentration, substrates concentration, choices of solvents ( n -alkanes), incubation time, temperature, molecular sieves (3Å × 1.5 mm), and water activity(a w ), were optimized. The immobilized lipase (25 mg/ml) was used to perform transesterification in n -alkane(s) that resulted in approximately 73.7 mM of ethyl acetate at 55 °C in n -nonane under shaking (160 rpm) after 15 h, when vinyl acetate and ethanol were used in a equimolar ratio (100 mM each). Addition of molecular sieves (3Å × 1.5 mm) as well as effect of water activity of saturated salt solutions (KI, KCl and KNO 3 ) to the transesterification efficiency has inhibitory effect. Batch operational stability tests indicated that immobilized lipase had retained 50% of its original catalytic activity after four consecutive batches of 15 h each.

Selective production of fragrance fatty acid ester from isopropanol and acetic acid has been achieved using silica-immobilizedlipase of Bacillus cereus MTCC 8372. A purified thermoalkalophilic extracellular lipase was immobilized by adsorption onto the silica. The effects of various parameters like molar ratio of substrates (isopropanol and acetic acid; 25 to 100 mM), concentration ofbiocatalyst (25–125 mg/mL), reaction time, reaction temperature, organic solvents,molecular sieves, and initial water activity werestudied for optimal ester synthesis. Under optimized conditions, 66.0mM of isopropyl acetate was produced when isopropanol and acetic acid were used at 100mM: 75mM in 9h at 55◦C in n-heptane under continuous shaking (160 rpm) using bound lipase(25mg). Addition of molecular sieves (3 °A ×1.5mm) resulted in a marked increase in ester synthesis (73.0mM). Ester synthesiswas enhanced by water activity associated with pre-equilibrated saturated salt solution of LiCl. The immobilized lipase retained more than 50% of its activity after the 6th cycle of reuse.

Abuse is rife in Disgrace by J.M. Coetzee and Cereus Blooms at Night by Shani Mootoo. Sexual violence is in both narratives, part of their richly textured social, emotional and political worlds. Fiction involving various traumas seems bleak, almost hopeless, perhaps weighted by sadness. Yet both these novels, even through depictions of rupturing, disruptive rape, trigger a recognition of possibility and potential among characters and perhaps readers. It is in this open ended potential for betterment of some kind that hope lies. What is the nature of hope and to what extent is it present in these novels? In this paper, I explore the emotion of hope in relation to the notion of becoming as elaborated on by Gilles Deleuze and Felix Guattari particularly in A Thousand Plateaus. They expound on Remy Chauvin‟s term “aparallel evolution” in relation to becoming (Deleuze and Guattari, 11). Deleuze also states that becoming is not a “phenomena of imitation or assimilation”. Rather, it is an encounter, “a double capture” (Deleuze and Parnet 2) between heterogeneous elements. There is no end or destination in becoming; it is constant change. I examine the transformative potential of becoming to elicit signs of hope in these novels. David Lurie, the self-absorbed womaniser and arguably rapist, becomes-dog by the end of Disgrace. How does this contribute to any sense of redemption and consequently hope? And how does hope emanate from the beaten, broken, brutally raped Mala Ramchandin in Cereus Blooms at Night? At heart, this paper is an acknowledgment of the unique relation literature has with life and the enriching insight that it may provide into the expression of hope.

Stevioside, a glycoside present in the leaves of Stevia rebaudiana Bertoni, offers therapeutic benefits such as anti-hyperglycemic, anti-hypertensive, antiinflammatory, anti-tumor, diuretic and immune influencing properties. In this work antimicrobial activity of stevioside against Bacillus cereus, a major source of milk contamination was investigated. The isolate was confirmed by various biochemical and 16S rRNA gene sequencing. The effect of temperature, incubation time and concentration of stevioside was optimized from a central composite response surface design. The standard plate count (SPC) of pasteurized milk was drastically reduced in comparison to toned and fresh milk. The optimal temperature, incubation time and stevioside concentration were observed to be 60.23°C, 21 h, and 275 μg/ mL respectively. The synergism of stevioside with the external factors (temperature and time) against B. cereus was observed. Our studies showed that addition of stevioside in fresh as well as pasteurised milk would control growth of B. cereus in milk.

Foi estudada uma bacteriocina produzida por uma linhagem de B. cereus 8A, isolado de solo da região Sul do Brasil. Na primeira etapa de estudo determinaram-se as condições básicas de produção de bacteriocina com amplo espectro de ação denominada de Cereína 8A. Observou-se que durante a fase estacionária ocorre o máximo da sua produção, iniciando sua síntese no final da fase exponencial. As condições de maior produção foram a 30º C, agitação e contínua e numa faixa de pH de 7,0-8,5. A bacteriocina bruta inibiu várias bactérias indicadoras, como Listeria monocytogenes, Clostridium perfringens e Bacillus cereus. O teste de termoestabilidade mostrou a perda de atividade quando submetida a uma temperatura a partir de 87º C. Verificou-se a resistência da bacteriocina bruta frente à tripsina e papaína, mas não frente à proteinase K e pronase E. B. cereus e L. monocytogenes foram utilizadas como bactérias indicadoras para a determinação do modo de ação, após a determinação da dose bactericida de 200 UA mL-1 e 400 UA mL-1 respectivamente. A Cereína 8A demonstrou uma ação inibidora em culturas de Escherichia coli e Salmonella Enteritidis, quando tratadas com EDTA. A atividade esporicida foi observada contra esporos de B. cereus após tratamento com 400 UA ml -1. A análise da biomassa de L. monocytogenes e B. cereus após tratamento com a Cereína 8A, através da espectrofotometria de infravermelho determinou alteração no perfil, correspondente à fração dos ácidos graxos da membrana celular bacteriana. A substância peptídica foi separada por meio da precipitação com sulfato de amônio, extração com 1-butanol e aplicação em coluna de cromatografia por troca iônica tipo Q-Sepharose. A Cereína 8A purificada mostrou maior sensibilidade a proteases e ao calor e um peso molecular de aproximadamente 26 kDa. O espectro ultravioleta foi típico de um polipeptídeo e o espectro de infravermelho indica presença de grupamentos NH, acil e ligações peptídicas na sua estrutura. Uma hipótese do mecanismo de ação seria a desestruturação da membrana celular pela abertura de poros.

O presente trabalho teve como principal objetivo avaliar a produção de enzimas proteolíticas por Bacillus cereus. As fermentações foram conduzidas em Biorreator Biodesign, com aeração de 1vvm, temperatura de 37°C e agitação de 400 r.p.m. como parâmetros fixos. Dois meios diferentes foram utilizados, Meio Referência (MR) e Meio de Proteína de Soja PS60® (MPS1), em experimentos realizados com 24 horas. Para avaliação da produção de complexos enzimáticos, retirou-se amostras a intervalos de 3 horas para análise de valores de pH, densidade ótica e massa seca. A atividade proteolítica, concentração de proteína solúvel e açúcares redutores, contagem de células viáveis totais e esporos também foram investigadas. Os resultados demonstraram que ambos os meios propiciaram condições para o ótimo desenvolvimento do B. cereus. Os resultados também indicam que, embora os dois meios tenham apresentado um crescimento celular semelhante, o meio composto por proteína de soja propiciou a obtenção de um extrato bruto com atividade proteolítica mais elevada. Nas condições de fermentação empregadas, o meio MPS1 apresentou em 15 horas uma atividade enzimática de 18,7UmL-1.h-1.enquanto que o meio MR apresentou uma atividade proteolítica de 11,8 UmL-1.h-1.
