This paper presents a global-optimisation frame-work for the design of a manipulator for harvesting capsicum(peppers) in the field. The framework uses a simulated capsicum scenario with automatically generated robot models based on DH parameters. Each automatically generated robot model is then placed in the simulated capsicum scenario and the ability of the robot model to get to several goals (capsicum with varying orientations and positions) is rated using two criteria:the length of a collision-free path and the dexterity of the end-effector. These criteria form the basis of the objective function used to perform a global optimisation. The paper shows a preliminary analysis and results that demonstrate the potential of this method to choose suitable robot models with varying degrees of freedom.

In 2002 at Virginia, South Australia, capsicum cultivars having the Tsw resistance gene against Tomato spotted wilt virus (TSWV) developed symptoms typical of TSWV infection and several glasshouse-grown crops were almost 100% infected. Samples reacted with TSWV antibodies in ELISA. Virus isolates from infected plants induced severe systemic symptoms, rather than a hypersensitive reaction, when inoculated onto capsicum cultivars and Capsicum chinense genotypes ( PI 152225 and PI 159236) that carry the Tsw resistance gene. Isolates virulent towards the Tsw gene had molecular and biological properties very similar to standard TSWV isolates, including a hypersensitive reaction in Sw-5 (TSWV-resistant) tomato genotypes. Tsw-virulent isolates were found during surveys at Virginia in 2002 and 2004 in both TSWV-resistant and susceptible cultivars of capsicum and tomato.

The efficacy of insecticides in controlling Helicoverpa spp., predominantly H. armigera (Hubner), on capsicum and zucchini was tested in small plot trials. Indoxacarb, methoxyfenozide, spinosad, emamectin benzoate and novaluron provided control, as measured by the percentage of damaged fruit, equal to or better than standard treatments of methomyl or methomyl alternated with methamidophos on capsicum. The Helicoverpa nucleopolyhedrovirus gave control equivalent to the standard treatment, as did Bacillus thuringiensis aizawai, but B. thuringiensis kurstaki was ineffective. Helicoverpa armigera larvae were present in zucchini flowers but did little damage to the fruit. None of the insecticides significantly reduced the percentage of damaged zucchini fruit compared with the untreated control. Bifenthrin, spinosad, emamectin benzoate and methoxyfenozide were effective in controlling larvae in flowers, while methomyl, B. thuringiensis aizawai, B. thuringiensis kurstaki and novaluron were not effective. Data indicated that all the insecticides effectively controlled larvae of Diaphania indica (Saunders), cucumber moth, in the zucchini flowers. There has been a limited range of insecticides available to manage Helicoverpa spp. in these vegetable crops, but these trials demonstrate the effectiveness of a number of newer insecticides that could be used and that would be compatible with integrated pest management programs in the crops.

Capsicum chlorosis virus (CaCV) was detected in field grown Capsicum annuum from Kununurra in northeast Western Australia. Identification of the Kununurra isolate (WA-99) was confirmed using sap transmission to indicator hosts, positive reactions with tospovirus serogroup IV-specific antibodies and CaCV-specific primers, and amino acid sequence comparisons that showed >97% identity with published CaCV nucleocapsid gene sequences. The reactions of indicator hosts to infection with WA-99 often differed from those of the type isolate from Queensland. The virus multiplied best when test plants were grown at warm temperatures. CaCV was not detected in samples collected in a survey of C. annuum crops planted in the Perth Metropolitan area.

Thrips can be important pests of capsicum and chilli crops, causing damage through their feeding and by vectoring viral diseases. As different species vary in their ability to transmit viruses and in their susceptibility to insecticides, it is important to know which species are present in a crop. The seasonal occurrence of thrips in capsicum and chilli crops in the Bundaberg district of south-east Queensland was investigated from July 2002 to June 2003. Fifty flowers were collected weekly from crops on seven farms and the adult thrips extracted and identified. Thrips palmi Karny and Frankliniella occidentalis (Pergande) were collected in the greatest numbers, with T. palmi predominant in autumn crops (March to July) and F. occidentalis predominant in spring crops (August to November). Pseudanaphothrips achaetus (Bagnall) was common, while Thrips tabaci Lindeman, Thrips imaginis Bagnall and Frankliniella schultzei (Trybom) were collected in low numbers.

The first complete genome sequence of capsicum chlorosis virus (CaCV) from Australia was determined using a combination of Illumina HiSeq RNA and Sanger sequencing technologies. Australian CaCV had a tripartite genome structure like other CaCV isolates. The large (L) RNA was 8913 nucleotides (nt) in length and contained a single open reading frame (ORF) of 8634 nt encoding a predicted RNA-dependent RNA polymerase (RdRp) in the viral-complementary (vc) sense. The medium (M) and small (S) RNA segments were 4846 and 3944 nt in length, respectively, each containing two non-overlapping ORFs in ambisense orientation, separated by intergenic regions (IGR). The M segment contained ORFs encoding the predicted non-structural movement protein (NSm; 927 nt) and precursor of glycoproteins (GP; 3366 nt) in the viral sense (v) and vc strand, respectively, separated by a 449-nt IGR. The S segment coded for the predicted nucleocapsid (N) protein (828 nt) and non-structural suppressor of silencing protein (NSs; 1320 nt) in the vc and v strand, respectively. The S RNA contained an IGR of 1663 nt, being the largest IGR of all CaCV isolates sequenced so far. Comparison of the Australian CaCV genome with complete CaCV genome sequences from other geographic regions showed highest sequence identity with a Taiwanese isolate. Genome sequence comparisons and phylogeny of all available CaCV isolates provided evidence for at least two highly diverged groups of CaCV isolates that may warrant re-classification of AIT-Thailand and CP-China isolates as unique tospoviruses, separate from CaCV.

Plant regeneration from mesophyll protoplasts of pepper, Capsicum annuum L. cv. California Wonder has been demonstrated via shoot organogenesis, Protoplasts isolated from fully expanded leaves of 3-week-old axenic shoots when cultured in TM medium supplemented with 1 mgl(-1) NAA, 1 mgl(-1) 2, 4-D, 0.5 mgl(-1) BAP (CM 1) resulted in divisions with a frequency ranging from 20-25%. Antioxidant ascorbic acid and polyvinylpyrrolidone (PVP) in the medium and incubation in the dark helped overcome browning of protoplasts. Microcalli and macrocalli were formed in TM medium containing 2 mgl(-1) NAA and 0.5 mgl(-1) BAP (CM II) and MS gelled medium containing 2 mgl(-1) NAA and 0.5 mgl(-1) BAP (CM III), respectively, Regeneration of plantlets was possible via caulogenesis, Microshoots, 2-5 per callus appeared on MS gelled medium enriched with 0.5 mgl(-1) IAA, 2 mgl(-1) GA and 10 mgl(-1) BAP (CM IVc). Rooting of microshoots was obtained on half strength gelled medium containing 1 mgl(-1) NAA and 0.5 mgl(-1) BAP, Protoplasts isolated from cotyledons failed to divide and degenerated eventually.
