Para diagnosticar la hipocalcemia, que a veces se presenta tras una tiroidectomia, habitualmente se recurre a la medida del ion calcio y a la evaluación de síntomas específicos de hipocalcemia (tetania, signo de Trousseau) o inespecíficos (parestesia, vómitos, etc) en los días posteriores a la intervención. Sería ideal que existiesen magnitudes bioquímicas que permitieran diagnosticar precozmente esta hipocalcemia y sintomatologia. Recientemente, se ha demostrado que las concentraciones intraoperatorias de paratirina (PTH) informan sobre el estado funcional de las glándulas paratiroides tras la tiroidectomia, por lo que éstas serian las magnitudes ideales que buscamos.

Recientemente se ha demostrado que la concentración intraoperatoria de paratirina 10 minutos después de la exéresis (PTH-post) y la diferencia relativa porcentual entre las concentraciones de PTH intraoperatorias preexéresis y 10 minutos después de la exéresls (PTH-decay), parecen predecir el estado funcional de las glándulas paratiroideas tras la tiroidectomía

The improvement of the reliability of the contact between the osseous tissues and the implant materials has been tested by recovering the metallic implants with ceramic materials, usually calcium phosphates. In our study, the calcium phosphate recovering layers were deposited by means of a pulsed-laser deposition technique. Our aim was to to evaluate the tissue interactions established between cortical bone and titanium implants covered by five different layers, ranging from amorphous calcium phosphate to crystalline hydroxyapatite, obtained by altering the parameters of the laser ablation process. The surgical protocol of the study consisted in the simultaneous implantation of the five types of implants in both the tibial dyaphisis of three Beagle dogs, sacrificed respectively one, two and three months after the last surgical procedures. After the sacrifice, the samples were submitted to a scheduled procedure of embedding in plastic polymers without prior decalcification, in order to perform the ultrastructural studies: scanning microscopy with secondary and backscattered electrons (BS-SEM). Our observations show that both in terms of the calcified tissues appearing as a response to the presence of the different coatings and of time of recovering, the implants coated with crystalline calcium phosphate layers by laser ablation present a better result than the amorphous-calcium-phosphate-coated implants. Moreover, the constant presence of chondroid tissue, related with the mechanical induction by forces applied on the recovering area, strongly suggests that the mechanisms implied in osteointegration are related to endomembranous, rather than endochondral ossification processes

Calcium signals trigger the translocation of the Prz1 transcription factor from the cytoplasm to the nucleus. The process is regulated by the calciumactivated phosphatase calcineurin, which activates Prz1 thereby maintaining active transcription during calcium signalling. When calcium signalling ceases, Prz1 is inactivated by phosphorylation and exported to the cytoplasm. In budding yeast and mammalian cells, different kinases have been reported to counter calcineurin activity and regulate nuclear export. Here, we show that the Ca2+/calmodulin-dependent kinase Cmk1 is first phosphorylated and activated by the newly identified kinase CaMKK2 homologue, Ckk2, in response to Ca2+. Then, active Cmk1 binds, phosphorylates and inactivates Prz1 transcription activity whilst at the same time cmk1 expression is enhanced by Prz1 in response to Ca2+. Furthermore, Cdc25 phosphatase is also phosphorylated by Cmk1, inducing cell cycle arrest in response to an increase in Ca2+. Moreover, cmk1 deletion shows a high tolerance to chronic exposure to Ca2+, due to the lack of cell cycle inhibition and elevated Prz1 activity. This work reveals that Cmk1 kinase activated by the newly identified Ckk2 counteracts calcineurin function by negatively regulating Prz1 activity which in turn is involved in activating cmk1 gene transcription. These results are the first insights into Cmk1 and Ckk2 function in Schizosaccharomyces pombe.

Guanylate cyclase activating proteins are EF-hand containing proteins that confer calcium sensitivity to retinal guanylate cyclase at the outer segment discs of photoreceptor cells. By making the rate of cGMP synthesis dependent on the free intracellular calcium levels set by illumination, GCAPs play a fundamental role in the recovery of the light response and light adaptation. The main isoforms GCAP1 and GCAP2 also localize to the synaptic terminal, where their function is not known. Based on the reported interaction of GCAP2 with Ribeye, the major component of synaptic ribbons, it was proposed that GCAP2 could mediate the synaptic ribbon dynamic changes that happen in response to light. We here present a thorough ultrastructural analysis of rod synaptic terminals in loss-of-function (GCAP1/GCAP2 double knockout) and gain-of-function (transgenic overexpression) mouse models of GCAP2. Rod synaptic ribbons in GCAPs−/− mice did not differ from wildtype ribbons when mice were raised in constant darkness, indicating that GCAPs are not required for ribbon early assembly or maturation. Transgenic overexpression of GCAP2 in rods led to a shortening of synaptic ribbons, and to a higher than normal percentage of club-shaped and spherical ribbon morphologies. Restoration of GCAP2 expression in the GCAPs−/− background (GCAP2 expression in the absence of endogenous GCAP1) had the striking result of shortening ribbon length to a much higher degree than overexpression of GCAP2 in the wildtype background, as well as reducing the thickness of the outer plexiform layer without affecting the number of rod photoreceptor cells. These results indicate that preservation of the GCAP1 to GCAP2 relative levels is relevant for maintaining the integrity of the synaptic terminal. Our demonstration of GCAP2 immunolocalization at synaptic ribbons at the ultrastructural level would support a role of GCAPs at mediating the effect of light on morphological remodeling changes of synaptic ribbons.

This article presents the results of a study of the efficiency of silanation process of calcium phosphate glasses particles and its effect on the bioactivity behavior of glasspoly( methyl methacrylate) (PMMA) composites. Two different calcium phosphate glasses: 44.5CaO-44.5P2O5-11Na2O (BV11) and 44.5CaO-44.5P2O5-6Na2O-5TiO2 (G5) were synthesized and treated with silane coupling agent. The glasses obtained were characterized by Microprobe and BET while the efficiency of silanation process was determined using Fourier Transform Infrared Spectroscopy (FTIR), X-ray Photoelectron Spectroscopy (XPS) and Thermal Analysis (DTA and TG)techniques. The content of coupling agent chemically tightly bond to the silanated glasses ascended to 1.69 6 0.02 wt % for BV11sil glass and 0.93 6 0.01 wt % for G5sil glass. The in vitro bioactivity test carried out in Simulated Body Fluid (SBF) revealed certain bioactive performance with the use of both silanated glasses in a 30% (by weight) as filler of the PMMA composites because of a superficial deposition of an apatite-like layer with low content of CO3 22 and HPO4 22 in its structure after soaking for 30 days occurred. Under pathological conditions, microglia, the resident CNS immune cells, become reactive and release pro-inflammatory cytokines and neurotoxic factors. We investigated whether this phenotypic switch includes changes in the expression of the L-type voltage-gated calcium channel (VGCC) in a rat model of N-methyl-d-aspartate-induced hippocampal neurodegeneration. Double immunohistochemistry and confocal microscopy evidenced that activated microglia express the L-type VGCC. We then analyzed whether BV2 microglia express functional L-type VGCC, and investigated the latter's role in microglial cytokine release and phagocytic capacity. Activated BV2 microglia express the CaV1.2 and CaV1.3 subunits of the L-type VGCC determined by reverse transcription-polymerase chain reaction, Western blot and immunocytochemistry. Depolarization with KCl induced a Ca2+ entry facilitated by Bay k8644 and partially blocked with nifedipine, which also reduced TNF-α and NO release by 40%. However, no nifedipine effect on BV2 microglia viability or phagocytic capacity was observed. Our results suggest that in CNS inflammatory processes, the L-type VGCC plays a specific role in the control of microglial secretory activity.

Estudiem les característiques edàfiques del sòl dels boscos joves de pi negre (Pinus uncinata) de l’estatge subalpí del Parc Nacional d’Aigüestortes i Estany de Sant Maurici corresponents a àrees de reforestació entre els anys 1956 i 2008. Aquest estudi s’ha fet de 29 parcel•les d’aquesta zona situades sobre diferents tipus de substrat, orientació i pendent. S’ha caracteritzat el sòl a partir de l’anàlisi de textura, matèria orgànica, nitrogen total, fòsfor, sodi, potassi, magnesi, calci, capacitat d’intercanvi catiònic, acidesa del sòl, relació C/N. Els resultats confirmen que es tracta de sòls àcids amb un elevat contingut de matèria orgànica a l’horitzó superficial (0-5 cm) i majoritàriament tenen humus de tipus moder. Això fa que aquest tingui valors de nitrogen total elevats. Gràcies al pH àcid i els continguts de potassi, calci i magnesi tindrem una bona fertilitat ja que la solubilitat i assimilació dels nutrients del sòl serà bona. Tots els valors obtinguts disminueixen en profunditat i mostren una gran variabilitat entre les parcel•les estudiades.

Introdução: Há poucos dados na literatura sobre a suplementação de vitamina D e cálcio e o desenvolvimento de cálculos renais. Objetivo: Avaliar o efeito de doses elevadas de vitamina D3 (V), com suplemento de cálcio (Ca) no desenvolvimento de litíase em modelo experimental. Métodos: Pastilhas foram inseridas na bexiga de ratos, que receberam V com ou sem Ca. Ratos foram divididos em seis grupos: 1. Sham; 2. Controle com pastilha, 3. Controle com V, 4. Pastilha + V, 5. Pastilha + Ca e 6. Pastilha + Ca + V. Resultados: Observou-se 50% e 17% de redução na formação de cálculos, respectivamente nos grupos 5 e 6 em comparação ao grupo 2 (p < 0,005). Não foram observadas hipercalcemia ou hipercalciúria em todos os grupos. Encontramos no grupo 6 (p = 0,03) uma redução significativa na calciúria. Conclusão: A administração de V associada com Ca diminuiu significantemente a formação de cálculos e reduziu significantemente a calciúria, sugerindo uma interferência benéfica na fisiopatologia litogênica.

La ricerca presentata è un’ampia esplorazione delle possibili applicazioni di concetti, metodi e procedure della Fuzzy Logic all’Ingegneria dei Materiali. Tale nuovo approccio è giustificato dalla inadeguatezza dei risultati conseguiti con i soli metodi tradizionali riguardo alla reologia ed alla durabilità, all’utilizzo di dati di laboratorio nella progettazione e alla necessità di usare un linguaggio (informatizzabile) che consenta una valutazione congiunta degli aspetti tecnici, culturali, economici, paesaggistici della progettazione. – In particolare, la Fuzzy Logic permette di affrontare in modo razionale l’aleatorietà delle variabili e dei dati che, nel settore specifico dei materiali in opera nel costruito dei Beni Culturali, non possono essere trattati con i metodi statistici ordinari. – La scelta di concentrare l’attenzione su materiali e strutture in opera in siti archeologici discende non solo dall’interesse culturale ed economico connesso ai sempre più numerosi interventi in questo nuovo settore di pertinenza dell’Ingegneria dei Materiali, ma anche dal fatto che, in tali contesti, i termini della rappresentatività dei campionamenti, della complessità delle interazioni tra le variabili (fisiche e non), del tempo e quindi della durabilità sono evidenti ed esasperati. – Nell’ambito di questa ricerca si è anche condotto un ampio lavoro sperimentale di laboratorio per l’acquisizione dei dati utilizzati nelle procedure di modellazione fuzzy (fuzzy modeling). In tali situazioni si è operato secondo protocolli sperimentali standard: acquisizione della composizione mineralogica tramite diffrazione di raggi X (XRD), definizione della tessitura microstrutturale con osservazioni microscopiche (OM, SEM) e porosimetria tramite intrusione forzata di mercurio (MIP), determinazioni fisiche quali la velocità di propagazione degli ultrasuoni e rotoviscosimetria, misure tecnologiche di resistenza meccanica a compressione uniassiale, lavorabilità, ecc. – Nell’elaborazione dei dati e nella modellazione in termini fuzzy, la ricerca è articolata su tre livelli: a. quello dei singoli fenomeni chimico-fisici, di natura complessa, che non hanno trovato, a tutt’oggi, una trattazione soddisfacente e di generale consenso; le applicazioni riguardano la reologia delle dispersioni ad alto tenore di solido in acqua (calci, cementi, malte, calcestruzzi SCC), la correlazione della resistenza a compressione, la gelività dei materiali porosi ed alcuni aspetti della durabilità del calcestruzzo armato; b. quello della modellazione della durabilità dei materiali alla scala del sito archeologico; le applicazioni presentate riguardano i centri di cultura nuragica di Su Monte-Sorradile, GennaMaria-Villanovaforru e Is Paras-Isili; c. quello della scelta strategica costituita dalla selezione del miglior progetto di conservazione considerando gli aspetti connessi all’Ingegneria dei Materiali congiuntamente a quelli culturali, paesaggistici ed economici; le applicazioni hanno riguardato due importanti monumenti (Anfiteatro e Terme a Mare) del sito Romano di Nora-Pula.

Compositions and abundances of calcareous nannofossil taxa have been determined in a ca 170 kyrs long time interval across the Paleocene/Eocene boundary at 1-cm to 10-cm resolution from two ODP Sites (1262, 1263) drilled along the flank of the Walvis Ridge in the South Atlantic. The results are compared to published data from ODP Site 690 in the Weddell Sea. The assemblages underwent rapid evolution over a 74 kyrs period, indicating stressed, unstable and/or extreme photic zone environments during the PETM hyperthermal. This rapid evolution, which created 5 distinct stratigraphic horizons, is consistent with the restricted brief occurrences of malformed and/or weakly calcified morphotypes. The production of these aberrant morphotypes is possibly caused by major global scale changes in carbon cycling in the ocean-atmosphere system, affecting also photic zone environments. No marked paleoecologically induced changes are observed in abundances of the genera Discoaster, Fasciculithus and Sphenolithus at the Walvis Ridge sites. Surprisingly, there is no significant correlation in abundance between these three genera, presumed to have had a similar paleoecological preference for warm and oligotrophic conditions.

Ocean acidification poses a serious threat to a broad suite of calcifying organisms. Scleractinian corals and cal- careous algae that occupy shallow, tropical waters are vulnerable to global changes in ocean chemistry be- cause they already are subject to stressful and variable carbon dynamics at the local scale. For example, net heterotrophy increases carbon dioxide concentrations, and pH varies with diurnal fluctuations in photosyn- thesis and respiration. Few researchers, however, have investigated the possibility that carbon dioxide con- sumption during photosynthesis by non-calcifying photoautotrophs, such as seagrasses, can ameliorate deleterious effects of ocean acidi fi cation on sympatric calcareous algae. Naturally occurring variations in the density of seagrasses and associated calcareous algae provide an ecologically relevant test of the hypoth- esis that diel fl uctuations in water chemistry driven by cycles of photosynthesis and respiration within seagrass beds create microenvironments that enhance macroalgal calci fi cation. In Grape Tree Bay off Little Cayman Island BWI, we quanti fi ed net production and characterized calci fi cation for thalli of the calcareous green alga Halimeda incrassata growing within beds of Thalassia testudinum with varying shoot densities. Re- sults indicated that individual H . incrassata thalli were ~6% more calci fi ed in dense seagrass beds. On an areal basis, however, far more calcium carbonate was produced by H . incrassata in areas where seagrasses were less dense due to higher rates of production. In addition, diel pH regimes in vegetated and unvegetated areas within the lagoon were not signi fi cantly different, suggesting a high degree of water exchange and mixing throughout the lagoon. These results suggest that, especially in well-mixed lagoons, carbonate pro- duction by calcareous algae may be more related to biotic interactions between seagrasses and calcareous algae than to seagrass-mediated changes in local water chemistry.
