The 32-bp deletion in the HIV-1 co-receptor CCR5 confers a high degree of resistance to HIV-1 infection in homozygous individuals for the deleted allele and partial protection against HIV-1 during disease progression in heterozygotes. Natural ligands for CCR5, MIP-1alpha, MIP-1ß and RANTES, have been shown to inhibit HIV replication in CD4+ T cells. In the present study, we examined the CCR5 genotype by PCR and the plasma levels of RANTES and MIP-1alpha by ELISA among blood donors (N = 26) and among HIV-1-infected individuals (N = 129). The control group consisted of healthy adult volunteers and HIV-1-infected subjects were an asymptomatic and heterogeneous group of individuals with regard to immunologic and virologic markers of HIV-1 disease. The frequency of the CCR5 mutant allele (delta32ccr5) in this population was 0.032; however, no delta32ccr5 homozygote was detected. These results could be related to the intense ethnic admixture of the Brazilian population. There was no correlation between circulating ß-chemokines (MIP-1alpha, RANTES) and viral load in HIV-infected individuals. RANTES concentrations in plasma samples from HIV+ patients carrying the homozygous CCR5 allele (CCR5/CCR5) (28.23 ng/ml) were higher than in the control samples (16.07 ng/ml; P<0.05); however, this HIV+ patient group (mean 26.23 pg/ml) had significantly lower concentrations of MIP-1alpha than those observed in control samples (mean 31.20 pg/ml; P<0.05). Both HIV-1-infected and uninfected individuals heterozygous for the delta32ccr5 allele had significantly lower concentrations of circulating RANTES (mean 16.07 and 6.11 ng/ml, respectively) than CCR5/CCR5 individuals (mean 28.23 and 16.07 ng/ml, respectively; P<0.05). These findings suggest that the CCR5 allele and ß-chemokine production may affect the immunopathogenesis of HIV-1.

The effects of grain-based diets from C3 or C4-cycle plants on muscle delta C-13 change process in Nile tilapia (Oreochromis niloticus) fingerlings were investigated. Two groups of sex reversal males Nile tilapia fingerlings were fed with isoproteic (32.0% DP) and isocaloric (3200 kcal DE/kg) diets, differing from each other by their delta C-13. Muscle samples were collected and the carbon isotopic composition was measured. For C4 diet, the formula for the muscle delta C-13 change related to the intake time of a new diet was delta C-13=- 14.88 - 9.2 1 e(-0.0209t) and the half-life (T) of the muscle carbon was 33.2 days. For C3 diet, the formula was delta C-13 = - 25.43 + 8.59e(-0.0533), with T = 13 days. The C3 diet was considered more appropriate based on its palatability and consequent larger food intake than the C4 diet, resulting in an increased muscle delta C-13 change rate. However, for future studies, would be necessary to mix both the C3 and C4 feedstuffs to formulate diets nutritionally appropriated, with contrasting stable isotopes signatures. Tissue delta C-13 change rate is therefore indicated as a promising tool to better understand the biotic and abiotic factors that influence nutrients utilization from the diet and animal growth. The Amazonian lowlands include large patches of open vegetation which contrast sharply with the rainforest, and the origin of these patches has been debated. This study focuses on a large area of open vegetation in northern Brazil, where d13C and, in some instances, C/N analyses of the organic matter preserved in late Quaternary sediments were used to achieve floristic reconstructions over time. The main goal was to determine when the modern open vegetation started to develop in this area. The variability in d13C data derived from nine cores ranges from -32.2 to -19.6 parts per thousand, but with nearly 60% of data above -26.5 parts per thousand. The most enriched values were detected only in ecotone and open vegetated areas. The development of open vegetation communities was asynchronous, varying between estimated ages of 6400 and 3000 cal a BP. This suggests that the origin of the studied patches of open vegetation might be linked to sedimentary dynamics of a late Quaternary megafan system. As sedimentation ended, this vegetation type became established over the megafan surface. In addition, the data presented here show that the presence of C4 plants must be used carefully as a proxy to interpret dry paleoclimatic episodes in Amazonian areas. From cultures of thermophilic soil fungus Humicola grisea var thermoidea, a delta-lactam derivative (3-(2-(4-hydroxyphenyl)-2-oxoethyl)-5,6-dihydropyridin-2( 1H)-one) that displayed anti-allergic activity was isolated, which was predicted by in silico computational chemistry approaches. The in vitro anti-allergic activity was investigated by beta-hexosaminidase release assay in rat basophilic leukaemia RBL-2H3 cells. The delta-lactam derivative exhibited similar anti-allergic activity (IC50 = 18.7 +/- 6.7 mu M) in comparison with ketotifen fumarate (IC50 = 15.0 +/- 1.3 mu M) and stronger anti-allergic activity than azelastine (IC50 = 32.0 mu M). Also, the MTT cytotoxicity assay with RBL-2H3 cells showed that delta-lactam does not display cytotoxicity at concentrations lower than 50 mu M. This study suggests that the delta-lactam derivative has the potential to be used as a lead compound in the development of anti-allergic drugs for clinical use in humans.

The Lena River Delta, which is the largest delta in the Arctic, extends over an area of 32 000 km**2 and likely holds more than half of the entire soil organic carbon (SOC) mass stored in the seven major deltas in the northern permafrost regions. The geomorphic units of the Lena River Delta which were formed by true deltaic sedimentation processes are a Holocene river terrace and the active floodplains. Their mean SOC stocks for the upper 1 m of soils were estimated at 29 kg/m**2 ± 10 kg/m**2 and at 14 kg/m**2 ± 7 kg/m**2, respectively. For the depth of 1 m, the total SOC pool of the Holocene river terrace was estimated at 121 Tg ± 43 Tg, and the SOC pool of the active floodplains was estimated at 120 Tg ± 66 Tg. The mass of SOC stored within the observed seasonally thawed active layer was estimated at about 127 Tg assuming an average maximum active layer depth of 50 cm. The SOC mass which is stored in the perennially frozen ground at the increment 50-100 cm soil depth, which is currently excluded from intense biogeochemical exchange with the atmosphere, was estimated at 113 Tg. The mean nitrogen (N) stocks for the upper 1 m of soils were estimated at 1.2 kg/m**2 ± 0.4 kg/m**2 for the Holocene river terrace and at 0.9 kg/m**2 ± 0.4 kg/m**2 for the active floodplain levels, respectively. For the depth of 1 m, the total N pool of the river terrace was estimated at 4.8 Tg ± 1.5 Tg, and the total N pool of the floodplains was estimated at 7.7 Tg ± 3.6 Tg. Considering the projections for deepening of the seasonally thawed active layer up to 120 cm in the Lena River Delta region within the 21st century, these large carbon and nitrogen stocks could become increasingly available for decomposition and mineralization processes.

Particulate organic matter (POM) derived from permafrost soils and transported by the Lena River represents a quantitatively important terrestrial carbon pool exported to Laptev Sea sediments (next to POM derived from coastal erosion). Its fate in a future warming Arctic, i.e., its remobilization and remineralization after permafrost thawing as well as its transport pathways to and sequestration in marine sediments, is currently under debate. We present one of the first radiocarbon (14C) data sets for surface water POM within the Lena Delta sampled in the summers of 2009 - 2010 and spring 2011 (n = 30 samples). The bulk D14C values varied from -55 to -391 per mil translating into 14C ages of 395 to 3920 years BP. We further estimated the fraction of soil-derived POM to our samples based on (1) particulate organic carbon to particulate nitrogen ratios (POC : PN) and (2) on the stable carbon isotope (d13C) composition of our samples. Assuming that this phytoplankton POM has a modern 14C concentration, we inferred the 14C concentrations of the soil-derived POM fractions. The results ranged from -322 to -884 per mil (i.e., 3060 to 17 250 14C years BP) for the POC : PN-based scenario and from -261 to -944 per mil (i.e., 2370 to 23 100 14C years BP) for the d13C-based scenario. Despite the limitations of our approach, the estimated D14C values of the soil-derived POM fractions seem to reflect the heterogeneous 14C concentrations of the Lena River catchment soils covering a range from Holocene to Pleistocene ages better than the bulk POM D14C values. We further used a dual-carbon-isotope three-end-member mixing model to distinguish between POM contributions from Holocene soils and Pleistocene Ice Complex (IC) deposits to our soil-derived POM fraction. IC contributions are comparatively low (mean of 0.14) compared to Holocene soils (mean of 0.32) and riverine phytoplankton (mean of 0.55), which could be explained with the restricted spatial distribution of IC deposits within the Lena catchment. Based on our newly calculated soil-derived POM D14C values, we propose an isotopic range for the riverine soil-derived POM end member with D14C of -495 ± 153 per mil deduced from our d13C-based binary mixing model and d13C of -26.6 ± 1 per mil deduced from our data of Lena Delta soils and literature values. These estimates can help to improve the dual-carbon-isotope simulations used to quantify contributions from riverine soil POM, Pleistocene IC POM from coastal erosion, and marine POM in Siberian shelf sediments.
