A bactéria Xylella fastidiosa possui uma ampla gama de plantas hospedeiras que inclui espécies de pelo menos 28 famílias de mono e dicotiledôneas. Em cafeeiro (Coffea spp.), a ocorrência dessa bactéria foi relatada previamente em cultivares da espécie Coffea arabica. Estudos foram realizados para determinar a presença de X. fastidiosa em diferentes espécies e híbridos interespecficos de cafeeiro. As amostragens foram realizadas em dois anos consecutivos. As espécies de cafeeiro examinadas foram: C. kapakata, C. canephora, C. racemosa, C. arabica, C. dewevrei, C. stenophylla e C. eugenioides. Também foram incluídos neste estudo híbridos interespecficos de C. arabica: C. arabica x C. dewevrei, C. arabica x C. eugenioides, C. arabica x C. racemosa e C. arabica x C. robusta. Foram coletadas amostras de ramos plagiotrópicos de diferentes plantas para cada espécie e híbrido. A detecão de X. fastidiosa nas amostras foi realizada utilizando os testes serológicos de DAS-ELISA e imunofluorescncia indireta. A bactéria foi detectada nas sete espécies e nos quatro híbridos de cafeeiro examinados. Entretanto, as plantas aparentemente não apresentavam sintomas de infecão por X. fastidiosa. A espécie C. arabica apresentou a maior proporção de amostras positivas e maiores valores de absorbância no teste de DAS-ELISA. Em contraste, as espécies C. racemosa e C. dewevrei foram as que apresentaram menores proporções de amostras positivas para presença de X. fastidiosa, como também menores valores de absorbância no teste de DAS-ELISA.

A resistência do cafeeiro (Coffea arabica) Iapar-59 a Meloidogyne exigua foi comparada com a cultivar IAC-Apoatã, empregada como padrão de resistência, e com as cultivares suscetíveis Mundo Novo (IAC 379-19), Catuaí (IAC 62) e Tupi (IAC 1669-33). Plantas de um ano de idade foram inoculadas com 10.000 ovos de M. exigua. Aos 93 dias da inoculação avaliou-se a reprodução de M. exigua através do número de galhas (NG), número de ovos (NO), fator de reprodução (FR), reduco no fator de reprodução e comportamento dos cafeeiros. Menor reprodução de M. exigua foi igualmente (P<0,05) observada nos cafeeiros Iapar-59 e Apoatã, nos quais o IG, NO e FR foram menores do que aqueles observados nas cultivares Mundo Novo, Catuaí e Tupi. Maior redução no FR de M. exigua ocorreu nos cafeeiros Apoatã e Iapar-59, indicando a reação de resistência do Iapar-59.

O café é um dos principais produtos agrícolas, sendo considerado o segundo item em importância do comércio internacional de commodities. O gênero Coffea pertence à família Rubiaceae que também inclui outras plantas importantes. Este gênero contém aproximadamente 100 espécies, mas a produção comercial é baseada somente em duas espécies, Coffea arabica e Coffea canephora, que representam aproximadamente 70 % e 30 % do mercado total de café, respectivamente. O Projeto Genoma Café Brasileiro foi desenvolvido com o objetivo de disponibilizar os modernos recursos da genômica à comunidade científica e aos diferentes segmentos da cadeia produtiva do café. Para isso, foram seqüenciados 214.964 clones escolhidos aleatoriamente de 37 bibliotecas de cDNA de C. arabica, C. canephora e C. racemosa representando estádios especficos do desenvolvimento de clulas e de tecidos do cafeeiro, resultando em 130.792, 12.381 e 10.566 seqüências de cada espécie, respectivamente, após processo de trimagem. Os ESTs foram agrupados em 17.982 contigs e em 32.155 singletons. A comparação destas seqüências pelo programa BLAST revelou que 22 % não tiveram nenhuma similaridade significativa às seqüências no banco de dados do National Center for Biotechnology Information (de função conhecida ou desconhecida). A base de dados de ESTs do cafeeiro resultou na identificação de cerca de 33.000 unigenes diferentes. Os resultados de anotação das seqüências foram armazenados em base de dados online em camp.br/cafe>http://www.lge.ibi.unicamp.br/cafe. Os recursos desenvolvidos por este projeto disponibilizam ferramentas genéticas e genômicas que podem ser decisivas para a sustentabilidade, a competitividade e a futura viabilidade da agroindústria cafeeira nos mercados interno e externo.

A cochonilha Planococcus citri (Risso, 1813) suga a seiva dos botões florais e frutos do cafeeiro, atacando as rosetas desde a floração até a colheita. Embora seja relatada há alguns anos na cafeicultura, são escassas as informações sobre o desenvolvimento dessa cochonilha em cafeeiros. Assim, o presente trabalho teve por objetivos avaliar alguns aspectos biológicos da fase ninfal de P. citri em plantas de café. Ovos dessa cochonilha foram retirados de uma criação em laboratório, isolados em placas de Petri contendo discos foliares de Coffea arabica L., das cultivares Acaiá Cerrado, Mundo Novo e Catuaí Vermelho e de C. canephora Pierre & Froenher, cultivar Apoatã. As placas foram mantidas em cmara climatizada a 25 ± 1ºC, 70 ± 10% de umidade relativa e 12h de fotofase. Constatou-se que a cultivar Catuaí Vermelho foi a que proporcionou maior duração do período ninfal das fêmeas, porém, não foram constatadas diferenças na mortalidade. Essa cochonilha se desenvolveu satisfatoriamente em todas as cultivares de café estudadas e os resultados não mostraram diferenças claras de susceptibilidade.

Flavonoid compounds were analyzed in ripe fruit pulp of ten species of Coffea, including two cultivars of C. arabica and two of C. canephora. Three coefficients of similarity: Simple-Matching, Jaccard and Ochiai and three different clustering methods, Single Linkage, Complete Linkage and Unweighted Pair Group, Using Arithmetic Averages (UPGMA), were used to analyze the data.Jaccard and Ochiai's coefficients of association showed a more coherent result, when compared with taxonomic and hybridization studies. Inclusion of Psilanthopsis kapakata in the genus Coffea, as C. kapakata, is justified by the similarity of this species with other studied species, and clusters clearly approximate the species C. arabica and C. eugenioides. The latter is one of the possible parents of the allotetraploid species C. arabica, C. congensis is the only species whose position remains ambiguous, probably due to the fact that the plants of this species that were introduced into the Campinas collections, were hybrids and not typical of C. congensis.

A descafeinação do café canephora é vantajosa tanto para a obtenção da cafeína quanto para valorização desta espécie. A cafeína, subproduto da descafeinação, atua como estimulante e diurético no organismo e é usado pelas indústrias de bebidas de refrigerante e farmacutica. Existem patentes industriais aplicando CO2 supercrítico na extração da cafeína do café. Porém pouco é divulgado sobre a extração da trigonelina (alcalóide importante na formação da vitamina niacina), que atua no sistema nervoso central, na secreção da bile e no intestino. O objetivo deste trabalho foi levantar subsídios experimentais e teóricos para a extração de alcalóides dos grãos de café canephora, variedade robusta, usando CO2 supercrítico. Os dados foram obtidos num aparelho de extração a altas pressões onde as variáveis termodinâmicas são independentemente controladas. Os alcalóides foram analisados por Cromatografia Liquida de Alta Eficiência (CLAE). As solubilidades da cafeína pura e a obtida dos grãos de café no CO2 supercrítico foram determinadas a 313, 323 e 343 K, de 9,5 a 23,5 MPa. Os resultados revelam a existência de um comportamento retrógrado para a solubilidade da cafeína pura e a obtida dos grãos de café; também mostram a seletividade do CO2 supercrítico pela cafeína quando comparada a trigonelina. A modelagem termodinâmica do equilíbrio sólido-fluido usando uma equação de estado de quarta ordem correlacionou satisfatoriamente as solubilidades da cafeína pura no solvente supercrítico.

To produce porcelain tiles fluxing agents are used in order to obtain a liquid phase during firing. This liquid phase fills the pores decreasing porosity, water absorption and contributes to material densification. In the porcelain tiles industry, feldspar is the main flux material used, with quantities ranging between 35 and 50%. Studies focus on the discovery of materials with flux characteristics that can reduce the consumption of feldspar by porcelain tiles industry. In this context, the coffee husk ashes, a residue obtained when coffee husks are burned to produce heat for the dryers during the processing of the green fruit, have as main chemical constituents potassium, calcium and magnesium, giving them characteristics of fluxing material. Brazil is the largest coffee producer in the world and is responsible for over 30% of the world s production. In this work a physical treatment of coffee husk ash was carried out in order to eliminate the organic matter and, after this, two by-products were obtained: residual wastes R1 and R2. Both residues were added separately as single fluxes and also in association with feldspar in mixtures with raw materials collected in a porcelain industry located in Dias d Ávila-Ba. The addition of these residues aimed to contribute to the reduction of the consumption of feldspar in the production of porcelain tiles. Specimens were produced with dimensions of 60 mm x 20 mm x 6 mm in an uniaxial die with compacting pressure of 45 MPa. The samples were heated to a temperature of 1200 °C, for 8 minutes. Tests were performed to characterize the raw materials by XRF, XRD, particle size analysis, DTA and TGA and, additionally, the results of the physical properties of water absorption, apparent porosity, linear shrinkage, density, dilatometry, flexural strength and SEM of sintered body were analyzed. Additions of less than 8% of the residue R1 contributed to the decrease of porosity, but the mechanical strength of the samples was not satisfactory. Additions of 5% the R2 residue contributed significantly to decrease the water absorption and apparent porosity, and also to increase the mechanical strength. Samples with addition of feldspar associated with the R2 residue, in proportions of 6.7% of R2 and 6.7% of feldspar, led to results of water absorption of 0.12% and mechanical strength of 46 MPa, having parameters normalized to the manufacture of porcelain stoneware tiles

Mutations in the FGFR3 gene cause the phenotypic spectrum of FGFR3 chondrodysplasias ranging from lethal forms to the milder phenotype seen in hypochondroplasia (Hch). The p.N540K mutation in the FGFR3 gene occurs in ∼70% of individuals with Hch, and nearly 30% of individuals with the Hch phenotype have no mutations in the FGFR3, which suggests genetic heterogeneity. The identification of a severe case of Hch associated with the typical mutation c.1620C > A and the occurrence of a c.1150T > C change that resulted in a p.F384L in exon 10, together with the suspicion that this second change could be a modulator of the phenotype, prompted us to investigate this hypothesis in a cohort of patients. An analysis of 48 patients with FGFR3 chondrodysplasia phenotypes and 330 healthy (control) individuals revealed no significant difference in the frequency of the C allele at the c.1150 position (p = 0.34). One patient carrying the combination `pathogenic mutation plus the allelic variant c.1150T > C' had a typical achondroplasia (Ach) phenotype. In addition, three other patients with atypical phenotypes showed no association with the allelic variant. Together, these results do not support the hypothesis of a modulatory role for the c.1150T > C change in the FGFR3 gene.

The p23 protein is a chaperone widely involved in protein homeostasis, well known as an Hsp90 co-chaperone since it also controls the Hsp90 chaperone cycle. Human p23 includes a β-sheet domain, responsible for interacting with Hsp90; and a charged C-terminal region whose function is not clear, but seems to be natively unfolded. p23 can undergo caspase-dependent proteolytic cleavage to form p19 (p231-142), which is involved in apoptosis, while p23 has anti-apoptotic activity. To better elucidate the function of the human p23 C-terminal region, we studied comparatively the full-length human p23 and three C-terminal truncation mutants: p23₁₋₁₁₇; p23₁₋₁₃₁ and p23₁₋₁₄₂. Our data indicate that p23 and p19 have distinct characteristics, whereas the other two truncations behave similarly, with some differences to p23 and p19. We found that part of the C-terminal region can fold in an α-helix conformation and slightly contributes to p23 thermal-stability, suggesting that the C-terminal interacts with the β-sheet domain. As a whole, our results suggest that the C-terminal region of p23 is critical for its structure-function relationship. A mechanism where the human p23 C-terminal region behaves as an activation/inhibition module for different p23 activities is proposed.

Vitamin C stability and concentration was evaluated in isotonic beverages and B group vitamins (B1, B2, B3, B5 and B6) in power beverages. The amount of vitamins was found to be above of that declared on the labels, even after the shelf life had been exceeded. A small decrease in the amount of B group vitamins was observed during the shelf life of the products. In the case of vitamin C this decrease was slightly higher. The present research shows the need of increased quality control and inspection.
