Estudaram-se clones de porta-enxertos de citros (Citrus spp.), híbridos e genitores quanto à tolerância das raízes a Phytophthora nicotianae através de inoculações em substrato de argila expandida. Investigaram-se progênies nucelares dos clones Poncirus trifoliata 'Rich 16-6', citrumelo 'Swingle' (C. paradisi x P. trifoliolata ), tangerinas 'Cleópatra' (Citrus reshni) e 'Suen Kat' (C. sunki), limão'Volkameriano' (C. volkameriana), dos genitores tangerina 'Sunki' (C. sunki) (S), limão 'Cravo' (C. limonia) (C), laranja 'Azeda' (C. aurantium) ((A), Poncirus trifoliata 'Davis A' (T), e progênies nucelares de híbridos entre eles, totalizando 2303 plântulas. Avaliou-se a taxa de sobrevivência, redução de raízes e partes aéreas, peso de raízes e de partes aéreas, diâmetro do caule e altura, comparando-se plântulas inoculadas e não inoculadas. Atribuíram-se também notas subjetivas para volume de raízes, enfolhamento, coloração das folhas e altura. Desenvolveu-se um índice total de redução (ITR) baseado na taxa de sobrevivência e nos parâmetros mencionados. Mostraram-se altamente tolerantes os trifoliatas 'Davis A' e 'Rich 16-6', o citrumelo 'Swingle', três híbridos TxS, dois SxT e dois SxA, com ITR < 20%. Laranja 'Azeda', tangerina 'Suen Kat', limões 'Cravo' e 'Volkameriano', oito híbridos TxS, quatro SxT, dois TxA e um SxA mostraram-se tolerantes, com ITR entre 20 e 40%. Três híbridos SxA mostraram-se moderadamente tolerantes, com ITR entre 40 e 60%. Tangerinas 'Sunki' e 'Cleópatra', dois híbridos SxA, um TxA e dois SxC mostraram-se intolerantes, com ITR entre 60 e 80%. Cinco híbridos SxC mostraram-se altamente intolerantes, com ITR > 80%. A metodologia de inoculação e avaliação discriminou com precisão progênies nucelares dos clones e dos híbridos, evidenciando o potencial de seleção principalmente dos híbridos SxT e recprocos.

Broadness of leaf petiole wing (WB) was investigated as a morphological marker for screening hybrids of the very narrow-winged species Citrus limonia and C. sunki with broad-winged species C. aurantium and C. sinensis. Controlled polinizations produced over 500 reciprocal hybrids with potential in the ongoing rootstock breeding program identified by the Pgi-1 and Prxa-1 isozyme loci. Measurement ratios WB/leaf length, WB/leaf broadness and WB/petiole length identified 86 to 91% of the reciprocal hybrids produced. However, visual classification of WB was an equally efficient but much easier and faster method. It can be very useful in breeding programs when large number of plants have to be screened or when isozyme, RFLP or RAPD laboratories are not available.

O presente estudo foi desenvolvido na UNESP/Botucatu, São Paulo, Brasil, num solo identificado como Oxisol, onde foram plantadas mudas de limoeiro Siciliano (C. limon) enxertadas em laranjeira Azeda (C. aurantium) e em limoeiro Cravo (C. limonia). O experimento consistiu em três níveis de saturação por bases (50%, 70% e 90%) e três doses de boro (0,5; 1,5 e 4,5 mg dm-3) no plantio em esquema fatorial de 3x3x2, com quatro repetições. Houve diferentes comportamentos entre os porta-enxertos no que se refere à composição mineral de folhas de limoeiro Siciliano , bem como, no desenvolvimento do sistema radicular, sendo maior na laranjeira azeda em relação ao limoeiro cravo. Não houve nenhum efeito na interação de níveis de saturação por bases e doses de boro para nenhum dos parâmetros avaliados.

Estudou-se a variação diurna da transpiração de quatro porta -enxertos de citros, durante 12 horas, em intervalos de 60 minutos, pelo método das pesagens. Verificou-se que não ocorreu diferenças estatísticas na marcha diária da transpiração, entre Poncirus trifoliata, Citrus aurantium (laranja Azeda), Citrus sinensis (laranja Caipira) e Citrus limonia (limão Cravo). O limoeiro Cravo mostrou, nas condições estudadas, área foliar superior à Poncirus, laranja Azeda e laranja Caipira. Observou-se ainda uma correlação positiva entre a área foliar e a transpiração dos quatro porta-enxertos de citros.

China is an important center of origin for the genus Citrus L. of the family Rutaceae and is rich in wild Citrus species. The taxonomy of Citrus has been a subject of controversy for more than a half century. We propose that the metabolite profiles of Chinese native Citrus species can be used for classification and understanding of the taxonomic relationships within the Citrus germplasm. In this study, triplicate gas chromatography-mass spectrometry (GC-MS) metabolite profiles of 20 Citrus species/varieties were acquired, including 10 native varieties originating in China. R-(+)-limonene, alpha-pinene, sabinene and alpha-terpinene were found to be major characteristic components of the essential oils analyzed in this study, and these compounds contributed greatly to the metabolic classification. The three basic species of the subgenus Eucitrus (Swingle's system), i.e., C. reticulata Blanco, C. medica L. and C. grandis Osb., were clearly differentiated based upon their metabolite profiles using hierarchical cluster analysis (HCA) and partial least square-discriminant analysis (PLS-DA). All the presumed hybrid genotypes, including sweet orange (C. sinensis Osb.), sour orange (C. aurantium L.), lemon (C. limon Burm.f.), rough lemon (C. jambhiri Lush.), rangpur lime (C. limonia Osb.) and grapefruit (C. paradisi Macf.), were grouped closely together with one of their suggested parent species in the HCA-dendrogram and the PLS-DA score plot. These results clearly demonstrated that the metabolite profiles of Citrus species could be utilized for the taxonomic classification of the genus and are complementary to the existing taxonomic evidence, especially for the identification and differentiation of hybrid species.

Citrus aurantium L. is popularly used to treat anxiety, among other indications suggesting central nervous system action. Previous studies showed anxiolytic effect in the essential oil from peel in mice evaluated on the elevated plus maze [Carvalho-Freitas, M.I.R., Costa, M., 2002. Anxiolytic and sedative effects of extracts and essential oil from Citrus aurantium L. Biological and Pharmaceutical Bulletin 25, 1629-1633.]. In order to better characterize the activity of the essential oil, it was evaluated in two other experimental models: the light-dark box and the marble-burying test, respectively related to generalized anxiety disorder and to obsessive compulsive disorder. Mice were treated acutely by oral route 30 min (single dose) or once a day for 15 days (repeated doses) before experimental procedures. In light-dark box test, single treatment with essential oil augmented the time spent by mice in the light chamber and the number of transitions between the two compartments. There were no observed alterations in the parameters evaluated in light-dark box after repeated treatment. Otherwise, single and repeated treatments with essential oil were able to suppress marble-burying behavior. At effective doses in the behavioral tests, mice showed no impairment on rotarod procedure after both single and repeated treatments with essential oil, denoting absence of motor deficit. Results observed in marble-burying test, related to obsessive compulsive disorder, appear more consistent than those observed in light-dark box. (c) 2005 Elsevier B.V. All rights reserved.

The biosynthetic origin of the dichloroimine functional group in the marine sponge terpene metabolites stylotellanes A ( 3) and B ( 4) was probed by the use of [C-14]-labelled precursor experiments. Incubation of the sponge Stylotella aurantium with [C-14]-labelled cyanide or thiocyanate resulted in radioactive terpenes in which the radiolabel was shown by hydrolytic chemical degradation to be associated specifically with the dichloroimine carbons. Additionally, label from both precursors was incorporated into farnesyl isothiocyanate ( 2). A time course experiment with [ 14C]cyanide revealed that the specific activity for farnesyl isothiocyanate decreases over time, but increases for stylotellane B ( 4), consistent with the rapid formation of farnesyl isothiocyanate ( 2) from inorganic precursors followed by a slower conversion to stylotellane B ( 4). The advanced precursors farnesyl isothiocyanate ( 2) and farnesyl isocyanide ( 5) were supplied to S. aurantium, and shown to be incorporated efficiently into stylotellane A ( 3) and B ( 4). Feeding of [C-14]-farnesyl isothiocyanate ( 2) resulted in a higher incorporation of label than with [C-14]-farnesyl isocyanide ( 5). Farnesyl isocyanide was incorporated into farnesyl isothiocyanate in agreement with labelling studies in other marine sponges. Both farnesyl isocyanide and isothiocyanate were further incorporated into axinyssamide A ( 11) as well as the cyclized dichloroimines (12)-(14), ( 16) that represent more advanced biosynthetic products of this pathway. These results identify the likely biosynthetic pathway leading to the major metabolites of S. aurantium.

Mutations in the FGFR3 gene cause the phenotypic spectrum of FGFR3 chondrodysplasias ranging from lethal forms to the milder phenotype seen in hypochondroplasia (Hch). The p.N540K mutation in the FGFR3 gene occurs in ∼70% of individuals with Hch, and nearly 30% of individuals with the Hch phenotype have no mutations in the FGFR3, which suggests genetic heterogeneity. The identification of a severe case of Hch associated with the typical mutation c.1620C > A and the occurrence of a c.1150T > C change that resulted in a p.F384L in exon 10, together with the suspicion that this second change could be a modulator of the phenotype, prompted us to investigate this hypothesis in a cohort of patients. An analysis of 48 patients with FGFR3 chondrodysplasia phenotypes and 330 healthy (control) individuals revealed no significant difference in the frequency of the C allele at the c.1150 position (p = 0.34). One patient carrying the combination `pathogenic mutation plus the allelic variant c.1150T > C' had a typical achondroplasia (Ach) phenotype. In addition, three other patients with atypical phenotypes showed no association with the allelic variant. Together, these results do not support the hypothesis of a modulatory role for the c.1150T > C change in the FGFR3 gene.

The p23 protein is a chaperone widely involved in protein homeostasis, well known as an Hsp90 co-chaperone since it also controls the Hsp90 chaperone cycle. Human p23 includes a β-sheet domain, responsible for interacting with Hsp90; and a charged C-terminal region whose function is not clear, but seems to be natively unfolded. p23 can undergo caspase-dependent proteolytic cleavage to form p19 (p231-142), which is involved in apoptosis, while p23 has anti-apoptotic activity. To better elucidate the function of the human p23 C-terminal region, we studied comparatively the full-length human p23 and three C-terminal truncation mutants: p23₁₋₁₁₇; p23₁₋₁₃₁ and p23₁₋₁₄₂. Our data indicate that p23 and p19 have distinct characteristics, whereas the other two truncations behave similarly, with some differences to p23 and p19. We found that part of the C-terminal region can fold in an α-helix conformation and slightly contributes to p23 thermal-stability, suggesting that the C-terminal interacts with the β-sheet domain. As a whole, our results suggest that the C-terminal region of p23 is critical for its structure-function relationship. A mechanism where the human p23 C-terminal region behaves as an activation/inhibition module for different p23 activities is proposed.

Vitamin C stability and concentration was evaluated in isotonic beverages and B group vitamins (B1, B2, B3, B5 and B6) in power beverages. The amount of vitamins was found to be above of that declared on the labels, even after the shelf life had been exceeded. A small decrease in the amount of B group vitamins was observed during the shelf life of the products. In the case of vitamin C this decrease was slightly higher. The present research shows the need of increased quality control and inspection.

Edibles films are an alternative to synthetic materials used for packing food products. Barbados cherry is rich in vitamin C and carotenoids. The aim of this study was to characterize and develop films by casting from cassava starch, lyophilized Barbados cherry pulp and glycerol. The films were characterized with respect to thickness, water vapor permeability (WVP), water solubility, vitamin C, carotene and mechanical properties. The interaction of pulp and glycerol reduced film thickness. An increase in pulp concentration up to 60% increased WVP but beyond this concentration reduced both WVP and solubility leading to an increased level of vitamin C and β carotene in the films.
