The visual system is an important link between the animal and the environment, com profound influences on the habits and lifestyle in various habitats. Adaptive mechanismsto the temporal niche are present in the visual system of many vertebrates, involving changins in ocular dimensios and design, retinal cell distribution and organization of neurochemical circuits related to the retinal resolution or sensitivity. The sensory system of the eye is represented by the retina, whose organization is responsible by receipty, initial analysis, and transmission of the information to the brain. The knowledge of the position of the eyes in the head and the distribution of retinal cells allow to identify adaptive aspects of each species to its visual field, which is characteristic to the ecological niche it occupies. In this research, we study eye anatomical characteristics and retina neurochemical features of the rock cavy (Kerodon rupestris), a tipical Brazilian rodent from the suborder Hystricomorpha, family Caviidae. The rock cavy has lateral eyes well constitute bony orbit and well differentiated extrinsic muscle. The study of the descriptive and morphometric anatomy of the showed mean values of axial diameter 10.7±0,5mm and equatorial diameter 11.6±0.7mm. The pupil is slit shaped and the lens has mean axial diameter 5.4±0.03 mm, corresponding to ~45% of the axial diameter of the eye. The posterior nodal distance and the retinal magnification factor were estimated at 6.74 mm e 118 μm/grau, respectively. Flat mounts were processed for Nissl stain, and the topographic distribution of ganglion cells showed a moderate visual band, just below the optic disc, with higher density in the ventral retina. Retinal vertical sections and flat mounts were processed for immunohistochemistry to visualize tyrosine hydroxilase (TH) and thus two types of TH+ cells were detected. Type 1 cells had strong TH-immunoreactivity, the body cell varied from 120.047 to 269.373 μm2 stratifying in the sublamina 1 of the IPL. Type 2 cells were weakly TH-imunoreactive, had cell body located mostly in the IPL, varying from 54.848 to 177.142 μm2, constituting ~10% of the TH+ cells. Both cell types exhibited similar topographic distribution with higher density found in a horizontal band along of the naso-temporal axis in the dorsal retina. The total population of dopaminergic cells was 2,156±469,4 cells, occupying an average area of 198,164 μm2. The presence of cones and rods was detected by immunohistochemistry in vertical sections and flat mounts. S cones density is around 10 times smaller than L cones, with different degree of spatial organization. Other retinal neuronal populations of the rock cavy were also detected in vertical sections with specific markers. Comparative analysis of the anatomical characteristics of the rock cavy eye 12 suggest that it was designed to acquire higher sensitivity to light, at expense of image sharpness, compatible with a vision at mesopic conditions. Additionally, the distribution of the 2 subtypes of dopaminergic cells in a naso-temporal band in the dorsal retina seems suitable to a gain in sensitivity, coherent with an animal with predominantly crepuscular activity pattern

T. gondii is an obligate intracellular protozoan and the main cause of retinochoroiditis in humans. The aim of this study was to evaluate the effect of the antipsychotic drugs haloperidol and clozapine on the course of infection by T. gondii of cultured embryonic retinal cells. Embryo retinas of Gallus gallus domesticus (E12) were used for the preparation of mixed monolayer cultures of retinal cells. Cultures were maintained on plates of 96 and 24 wells by 37°C in DMEM medium supplemented with 5% fetal bovine serum for 2 days. After this period, cultures were simultaneously infected with tachyzoites of T. gondii and treated with the antipsychotics haloperidol and clozapine for 48 hours. Treatment effects were determined by both assessing cell viability with the MTT method and evaluating infection outcomes in slides stained with Giemsa. The treatment with haloperidol and clozapine cells infected with T. gondii resulted in higher viability of these cells, suggesting a possible prevention of neuronal degeneration induced by T. gondii. Additionally, intracellular replication of this protozoan in cells treated with haloperidol and clozapine were significantly reduced, possibly by modulation of the parasite s intracellular calcium concentration

As células-tronco adultas (CTA) são células multipotentes e não especializadas encontradas na medula óssea, no sangue periférico, na córnea, na retina, no cérebro, no músculo esquelético, na polpa dental, no fígado, no pâncreas, no epitélio da pele, no sistema digestivo, no cordão umbilical e na placenta. Estas células podem se renovar e reproduzir indefinidamente e, sob certos estímulos, se transformar em células especializadas de diferentes tecidos ou órgãos. O presente trabalho tem como objetivo a obtenção de CTA a partir de tecido epitelial de roedores silvestres de espécies diferentes (Oecomys concolor - um exemplar fêmea, Proechimys roberti - dois exemplares machos, Hylaeamys megacephalus - dois exemplares machos). A metodologia para isolamento e cultivo in vitro de amostras do tecido epitelial foi estabelecida, a partir de protocolos já descritos, avaliando aspectos morfológicos, estabilidade genômica, contagem e análise da viabilidade celular, potencial clonogênico e indução de diferenciação em osteócitos, condrócitos e adipócitos. Todas essas análises foram feitas pós-criopreservação das culturas. As CTA foram caracterizadas como população homogênea de células que proliferam in vitro, como células aderentes à superfície do plástico, tendo morfologia semelhante a fibroblastos e formato fusiforme, com alta taxa de crescimento e proliferação celular por várias passagens sucessivas, onde a autorrenovação celular foi avaliada por ensaios clonogênicos. Na análise para examinar a estabilidade genômica na P3, todas as amostras apresentaram cariótipo com número diplóide normal e estável. A metodologia empregada nos ensaios para diferenciação das CTA em linhagens osteogênica, condrogênica e adipogênica, apresentou resultados satisfatórios, onde as células mostraram a marcação desejada através das colorações Alizarin Red S, Alcian Blue e Oil Red O, respectivamente. Todas as amostras testadas apresentam capacidade de proliferação e diversidade de diferenciação, sendo potencialmente fornecedores de CTA provenientes da pele e podendo ser utilizados como organismos modelos de estudos em CT.

Neste trabalho apresentamos o resultado de um estudo exploratório investigando a implantação numa obra horizontal a aplicação do conceito de “célula móvel de produção” para a Construção Civil. Esta investigação foi realizada num processo evolutivo entre uma obra de características verticais e horizontais. O estudo direcionou-se para Células Móveis de Produção, ou seja, células que retém todas as características da célula de manufatura tradicional, porém com a característica da mobilidade através do produto, assim o trabalho compara e ilustra os resultados obtidos com esta forma de organização dos trabalhadores diante dos dados de produtividade e custo oriundos de obras executadas anteriormente.

To quantify the effects of methylmercury (MeHg) on amacrine and on ON-bipolar cells in the retina, experiments were performed in MeHg-exposed groups of adult trahiras (Hoplias malabaricus) at two dose levels (2 and 6 µg/g, ip). The retinas of test and control groups were processed by mouse anti-parvalbumin and rabbit anti-aprotein kinase C (aPKC) immunocytochemistry. Morphology and soma location in the inner nuclear layer were used to identify immunoreactive parvalbumin (PV-IR) and aPKC (aPKC-IR) in wholemount preparations. Cell density, topography and isodensity maps were estimated using confocal images. PV-IR was detected in amacrine cells in the inner nuclear layer and in displaced amacrine cells from the ganglion cell layer, and aPKC-IR was detected in ON-bipolar cells. The MeHg-treated group (6 µg/g) showed significant reduction of the ON-bipolar aPKC-IR cell density (mean density = 1306 ± 393 cells/mm2) compared to control (1886 ± 892 cells/mm2; P < 0.001). The mean densities found for amacrine PV-IR cells in MeHg-treated retinas were 1040 ± 56 cells/mm2 (2 µg/g) and 845 ± 82 cells/mm2 (6 µg/g), also lower than control (1312 ± 31 cells/mm2; P < 0.05), differently from the data observed in displaced PV-IR amacrine cells. These results show that MeHg changed the PV-IR amacrine cell density in a dose-dependent way, and reduced the density of aKC-IR bipolar cells at the dose of 6 µg/g. Further studies are needed to identify the physiological impact of these findings on visual function.

El objetivo del presente trabajo es el de presentar la situación actual de las posibles relaciones entre los comportamientos de las células lógicas, empleadas en Computación Óptica, y algunos comportamientos no lineales obtenidos en sistemas complejos. Como se mostrará, las arquitecturas empleadas en sistemas de cálculo, y más en concreto, las unidades básicas de que están compuestas, pueden dar lugar a situaciones no previstas de antemano y, como consecuencia, generar procesos ajenos a los inicialmente previstos. En concreto se mostrará como de una célula lógica, pueden obtenerse comportamientos caóticos. Este estudio se extenderá al análisis de redes neuronales biológicas y a su posible modelización con las anteriores técnicas. Como caso concreto de estudio se ofrecerá una simulación de la retina de los vertebrados, obtenida mediante las células lógicas presentadas anteriormente.

El glaucoma es una de las causas más comunes de discapacidad visual y una de las enfermedades neurodegenerativas oculares más frecuentes de pérdida irreversible de visión. La afectación originada en la retina se caracteriza por la degeneración de las células ganglionares y la pérdida de axones. La presión intraocular es un factor de riesgo importante en el glaucoma, entre otros factores, implicando mecanismos bioquímicos que desencadenan la muerte de las células ganglionares. El ratón DBA/2J es un modelo de hipertensión ocular y de degeneración de las células ganglionares de la retina (CGR). Las características principales de éste son la dispersión del pigmento del iris (IPD) y la atrofia del estroma del iris (ISA) que conducen a la patogénesis del glaucoma. Los mecanismos bioquímicos que comprometen al sistema purinérgico en procesos patológicos como la degeneración glaucomatosa han sido estudiados en los últimos años, siendo de gran relevancia como posibles dianas farmacológicas para el tratamiento de diferentes neuropatías. Los receptores P2X comprenden una familia de siete canales iónicos de membrana activados por ligando (P2X1-7) que se activan por el ATP extracelular (ATPe). En particular, los receptores P2X7 podrían estar involucrados en la regulación de la transmisión sináptica y la muerte neuronal en la retina. Además, la excitotoxicidad mediada por ATP a través de la activación del receptor P2X7 sugiere su posible implicación en la degeneración neuronal y la pérdida de la función visual en las retinas glaucomatosas. Tan importante como la presencia de este receptor purinérgico es estudiar los niveles de ATP extracelular de la retina, así como evaluar los cambios en la expresión del transportador de nucleótidos vesicular (VNUT) y los niveles de ecto-nucleotidasa (E-NPP1) en este modelo murino de glaucoma durante el desarrollo de la enfermedad...

Purpose: To investigate early functional changes of local retinal defects in type II diabetic patients using the global flash multifocal electroretinogram (MOFO mfERG). Methods: Thirty-eight diabetic patients and 14 age-matched controls were recruited. Nine of the diabetics were free from diabetic retinopathy (DR), while the remainder had mild to moderate non-proliferative diabetic retinopathy. The MOFO mfERG was performed at high (98%) and low (46%) contrast levels. MfERG responses were grouped into 35 regions for comparison with DR classification at those locations. Z-scores of the regional mfERG responses were compared across different types of DR defects. Results: The mfERG waveform consisted of the direct component (DC) and the induced component (IC). Local reduction in DC and IC amplitudes were found in diabetic patients with and without DR. With increasing severity of retinopathy, there was a further deterioration in amplitude of both components. Under MOFO mfERG paradigm, amplitude was a useful screening parameter. Conclusion: The MOFO mfERG can help in detecting early functional anomalies before the appearance of visible signs, and may assist in monitoring further functional deterioration in diabetic patients.

This project provides a foundation for the use of silk membranes in a tissue engineered therapy for the treatment of devastating retinal diseases such as age-related macular degeneration. The three-dimensional tissue model described in this thesis has great potential for use in basic research of retinal pathologies, and the potential to be implemented into clinical approaches after appropriate refinement.
