Rockfishes (Sebastes spp.) support one of the most economically important f isheries of the Pacific Northwest and it is essential for sustainable management that age estimation procedures be validated for these species. Atmospheric testing of thermonuclear devices during the 1950s and 1960s created a global radiocarbon (14C) signal in the ocean environment that scientists have identified as a useful tracer and chronological marker in natural systems. In this study, we first demonstrated that fewer samples are necessary for age validation using the bomb-generated 14C signal by emphasizing the utility of the time-specific marker created by the initial rise of bomb-14C. Second, the bomb-generated 14C signal retained in fish otoliths was used to validate the age and age estimation method of the quillback rockfish (Sebastes maliger) in the waters of southeast Alaska. Radiocarbon values from the first year’s growth of quillback rockfish otoliths were plotted against estimated birth year to produce a 14C time series spanning 1950 to 1985. The initial rise in bomb-14C from prebomb levels (~ –90‰) occurred in 1959 [±1 year] and 14C levels rose relatively rapidly to peak Δ14C values in 1967 (+105.4‰) and subsequently declined through the end of the time series in 1985 (+15.4‰). The agreement between the year of initial rise of 14C levels from the quillback rockfish time series and the chronology determined for the waters of southeast Alaska from yelloweye rockfish (S. ruberrimus) otoliths validated the aging method for the quillback rockfish. The concordance of the entire quillback rockfish 14C time series with the yelloweye rockfish time series demonstrated the effectiveness of this age validation technique, confirmed the longevity of the quillback rockfish up to a minimum of 43 years, and strongly confirms higher age estimates of up

Over 70% of nosocomial infections in the United States are resistant to one or more traditional antibiotics, necessitating research for alternative treatment options. This study aims to chelate gallium (Ga) onto a bacterial siderophore, desferrioxamine (DFO), to retard bacterial growth. By exploiting natural bacterial pathways, metal-siderophore treatments are hypothesized to circumvent traditional resistance mechanisms. Additionally, the GaDFO complex will be tested against several bacterial species to determine the specificity of DFO uptake. This research aims to prove the feasibility of siderophore piracy as an alternative to antibiotics. In showing the feasibility of siderophore piracy mechanisms, this research will enable the development of future avenues for protecting against resistant nosocomial infections.

Rationale: Delirium is common in intensive care unit (ICU) patients and is a predictor of worse outcomes and neuroinflammation is a possible mechanism. The antiinflammatory actions of statins may reduce delirium.

Objectives: To determine whether critically ill patients receiving statin therapy had a reduced risk of delirium than those not on statins.

Methods: A prospective cohort analysis of data from consecutive ICU patients admitted to a UK mixed medical and surgical critical care unit between August 2011 and February 2012; the Confusion Assessment Method for ICU was used to determine the days each patient was assessed as being free of delirium during ICU admission.

Measurements and Main Results: Delirium-free days, daily administration of statins, and serum C-reactive protein (CRP) were recorded. Four hundred and seventy consecutive critical care patients were followed, of whom 151 patients received statins. Using randomeffects multivariable logistic regression, statin administration the previous evening was associated with the patient being assessed as free of delirium (odds ratio, 2.28; confidence interval, 1.01-5.13; P , 0.05) and with lower CRP (b = 20.52; P , 0.01) the following day. When the association between statin and being assessed as free of delirium was controlled for CRP, the effect size became nonsignificant (odds ratio, 1.56; confidence interval, 0.64-3.79; P = 0.32).

Conclusions: Ongoing statin therapy is associated with a lower daily risk of delirium in critically ill patients. An ongoing clinical trial, informed by this study, is investigating if statins are a potential therapy for delirium in the critically ill.Copyright © 2014 by the American Thoracic Society.

Les contrevenants de la conduite avec capacités affaiblies (CCA) n’entrent pas tous dans les registres de la sécurité routière avec le même risque de récidive. Pour pallier cette hétérogénéité, cette thèse propose de modéliser les interrelations entre les traits de personnalité et les comportements à risque associés à la récidive et de détecter un sous-groupe de contrevenants au risque de récidive élevé à l’aide de l’axe hypothalamo-hypophyso-surrénalien (HHS). Plus particulièrement, les trois articles de cette thèse s’intéressent au cortisol, l’hormone du stress. Le premier article élabore un modèle théorique réconciliant les connaissances sur l’axe HHS issues du domaine de la CCA et de domaines connexes. Lors de précédentes études, le nombre de condamnations antérieures pour CCA a été associé négativement à la réactivité du cortisol à la suite d’une situation stressante. Chez les récidivistes, cette faible réactivité s’explique partiellement par la recherche d’expériences, une dimension de la recherche de sensations. Au-delà ce trait de personnalité désinhibiteur, une faible activité de l’axe HHS a été associée à d’autres traits (c.-à-d. impulsivité et tendances antisociales) et d’autres comportements à risque (c.-à-d. infractions routières, arrestations criminelles et consommation problématique de substances psychoactives). Ce modèle intégrant la réactivité du cortisol permet une conceptualisation approfondie des diverses caractéristiques des contrevenants de la CCA et explique hypothétiquement la répétition des comportements à risque. Les deux articles suivants se penchent sur l’intérêt empirique d’utiliser l’axe HHS pour déterminer un sous-groupe de contrevenants à risque élevé de récidive. Plus précisément, le deuxième article émet l’hypothèse que les récidivistes (n = 30) ayant une faible activité de leur cortisol (c.-à-d. médiane de la surface sous la courbe relative au niveau de base et relative à la réactivité) ont davantage de traits de personnalité désinhibiteurs et de comportements à risque que les récidivistes ayant une forte activité. L’hypothèse n’a pas été confirmée. Au contraire, les récidivistes présentant une faible réactivité commettent moins d’infractions routières et d’arrestations criminelles que ceux ayant une forte réactivité. Quant à lui, le troisième article investigue une hypothèse similaire auprès des contrevenants primaires (n = 139). Les contrevenants manifestant une faible réactivité du cortisol (c.-à-d. différence entre prélèvements post-stress et pré-stress) ont davantage d’impulsivité attentionnelle, de non-planification, d’arrestations criminelles et de cigarettes fumées par jour que les contrevenants ayant une forte réactivité. Lors d’analyses exploratoires, la présence d’une variété de traits de personnalité désinhibiteurs et de comportements à risque chez les contrevenants primaires présentant une faible réactivité lorsque comparé au groupe contrôle (n = 31) suggère encore une fois leur risque élevé de récidive. L’intérêt d’ajouter un mécanisme neurobiologique pour modéliser les interrelations entre les traits de personnalité désinhibiteurs et les comportements à risque des contrevenants de la CCA a été exploré dans cette thèse. La détermination d’un sous-groupe de contrevenants présentant un risque élevé de récidive, à l’aide de l’axe HHS, semble davantage profitable auprès de l’hétérogène population des contrevenants primaires. En contrepartie, l’axe HHS ne permet pas de déterminer un sous-groupe ayant une problématique sévère auprès des récidivistes à l’extrême du continuum du risque.

Driving while impaired (DWI) is a grave and persistent high-risk behavior. Previous work demonstrated that DWI recidivists had attenuated cortisol reactivity compared to non-DWI drivers. This suggests that cortisol is a neurobiological marker of high-risk driving. The present study tested the hypothesis that this initial finding would extend to first-time DWI (fDWI) offenders compared to non-DWI drivers. Male fDWI offenders (n = 139) and non-DWI drivers (n = 31) were exposed to a stress task, and their salivary cortisol activity (total output and reactivity) was measured. Participants also completed questionnaires on sensation seeking, impulsivity, substance use, and engagement in risky and criminal behaviors. As hypothesized, fDWI offenders, compared to non-DWI drivers, had lower cortisol reactivity; fDWI offenders also showed lower total output. In addition, cortisol activity was the most important predictor of group membership, after accounting for alcohol misuse patterns and consequences and other personality and problem behavior characteristics. The findings indicate that attenuated cortisol activity is an independent factor associated with DWI offending risk at an earlier stage in the DWI trajectory than previously detected.

Dating lake sediments by accelerator mass spectrometry (AMS) 14C analysis of terrestrial plant macrofossils overcomes one of the main problems associated with dating bulk sediment samples, i.e., the presence of old organic matter. Even so, many AMS dates from arctic and boreal sites appear to misrepresent the age of the sediment. To understand the nature of these apparent dating anomalies better, we conducted a series of 14C dating experiments using samples from Alaskan and Siberian lake-sediment cores. First, to test whether our analytical procedures introduced a sample-mass bias, we obtained 14C dates for different-sized pieces of single woody macrofossils. In these sample-mass experiments, statistically equivalent ages were found for samples as small as 0.05 mg C. Secondly, to assess whether macrofossil type influenced dating results, we conducted sample-type experiments in which 14C dates were obtained for different macrofossil types sieved from the same depth in the sediment. We dated materials from multiple levels in sediment cores from Upper Capsule Lake (North Slope, northern Alaska) and Grizzly Lake (Copper River Basin, southern Alaska) and from single depths in other records from northern Alaska. In several of the experiments there were significant discrepancies between dates for different plant tissues, and in most cases wood and charcoal were older than other macrofossil types, usually by several hundred years. This pattern suggests that 14C dates for woody macrofossils may misrepresent the age of the sediment by centuries, perhaps because of their longer terrestrial residence time and the potential in-built age of longlived plants. This study identifies why some 14C dates appear to be inconsistent with the overall age-depth trend of a lake-sediment record, and it may guide the selection of 14C samples in future studies.

The formation of Lake Melkoe (64°51'30''N, 175°14'E, altitude 36 m), one of the largest lakes of the Anadyr Lowland, is related to the moraine left by the Tyellakh Glacier, which originated on the Pekul'nei Ridge. The lake (6 km long and 4.4 km wide) extends in the northwestern direction. The Kholmy Priozernye moraine (16 km long along the arc, 1.5 km wide, and 92-103 masl) surrounds the lake in the west and south. The lake coasts are covered by sand with pebbles and shingle. The flat lake bottom dips toward its central part to a depth of 160 cm. In distinction from many other lakes of the Anadyr Lowland, the thickness of the upper layer of water-saturated sediments overlying compact aleurites in Lake Melkoe is only 5-6 cm. Such a peculiarity of the bottom is explained by the large size of the lake, low sedimentation rates, and frequent storms caused by strong winds. Regional and local vegetation corresponds to a mosaic tundra represented by high shrubs Pinus pumila, Duschekia fruticosa , and hummocky Betula - Ericales - Eriophorum communities. Pinus pumila and Alnus form thickets on the banks of the Anadyr River, coasts of lakes, and moraine slopes.
