The paper presents bighead carp broodstock management, an integral part of hatchery techniques as it will determine the quality of resulting fry and fingerlings. Proper methods on the selection and management of broodstock are discussed.

As a prelude to strain selection for domestication and future marker assisted selection, genetic variation revealed by microsatellite DNA was evaluated in yellow perch, Perca flavescens, from four wild North American populations collected in 2003-2004 (Maine, New York, North Carolina, and Pennsylvania,), and two captive populations (Michigan and Ohio). For the loci examined, levels of heterozygosity ranged from H-e=0.04 to 0.88, genetic differentiation was highly significant among all population pairs, and effective migration ranged from low (N(e)m=0.3) to high (N(e)m=4.5). Deviation from Hardy-Weinberg equilibrium was regularly observed indicating significant departures from random mating. Instantaneous measures of inbreeding within these populations ranged from near zero to moderate (median F=0.16) and overall inbreeding levels averaged F-IS=0.18. Estimates of genetic diversity, Phi(ST), and genetic distance were highest between Michigan and all other broodstock groups and lowest between New York and Ohio. Genetic differentiation among groups did not correlate with geographic distance. Overall, the patterns of variation exhibited by the captive (Michigan and Ohio) populations were similar to patterns exhibited by the other wild populations, indicating that spawning and management practices to date have not significantly reduced levels of genetic variation. Tor tambroides and T. douronensis, locally referred to as empurau and semah, respectively, are high valued mahseer species, indigenous to Sarawak, East Malaysia, with an aquaculture potential and of conservational value. Direct sequencing of mitochondrial DNA (mtDNA) 16S rRNA gene region (542 bp) was used to investigate genetic variation of T. tambroides and T. douronensis broodstock collected from different geographic locations in Sarawak and maintained at the Indigenous Fish Research and Production Center (IFRPC), Tarat, Sarawak, Malaysia. A total of 11 unique haplotypes were identified, of which six were detected in T. tambroides, and five in T. douronensis. Overall, nucleotide diversity (π) was low, ranging from 0.000 to 0.006, and haplotype diversity (h) ranged from 0.000 to 0.599. Although the analysis failed to detect genetic variation amongst populations of T. tambroides (significant pairwise FST was found for only one test, but pairwise haplotype frequencies were not statistically significant), substantial inter-population divergence among T. douronensis was recognised, especially those originating from different river systems (pairwise FST = 0.754 to 1.000, P < 0.05). Fixed haplotype differences were found in one population of T. douronensis. Average nucleotide divergence between T. tambroides and T. douronensis was 0.018, similar to the amount recognised between T. tambroides and the outgroup T. khudree (0.017). In addition, phylogenetic analysis revealed that the T. douronensis mtDNA consisted of two highly divergent clusters (0.020), one of which is more closely related to T. tambroides rather than with the other group of haplotypes of the conspecifics. The findings from the present study have important implications for aquaculture, management and conservation of these two species. The data also raise some concerns regarding the taxonomic status of T. douronensis, which needs to be addressed.

A stressful environment induces cortisol that might affect fish breeding and reproduction. In the present work, which aimed to mimic aquacultural conditions of the jundia (Rhamdia quelen) hatcheries in southern South America, females were submitted to normal or stressful handling and the effects of cortisol on serum levels of 17β-estradiol (E2) and testosterone (T) were determined. In addition, the effect of stress on reproductive parameters such as eggs and swim-up fry production was also measured. Eight females from a group submitted to stressful handling (SH) conditions and eight females from a group with normal handling (NH) conditions were captured for blood sampling at D 0 and at D 1, 10, 20, 30, and 40 of the experiment. A typical cortisol response was observed in the SH females group in that they presented higher cortisol level in contrast to the NH female group, in all days sampled, except at D 0. In the 10th and 20th d, the E2 levels were lower in SH females, but cortisol levels were higher, suggesting an effect of cortisol on E2 production and/or release. Stressful handling appeared to affect both the number and the quality of the gametes because a lower number of oocytes was stripped from SH females, and from SH fertilized eggs, a lower number of viable swim-up fry was obtained to be transferred to earthen larviculture ponds. Taken together, the results indicated that stressful handling of broodstock impairs R. quelen reproduction. © Copyright by the World Aquaculture Society 2008.
