Hepatozoon species are the most abundant hemoparasites of snakes. Its identification has been based mainly on the morphologic characterization of the gamonts in the peripheral blood of the vertebrate host and also of the cysts found in the internal organs of the vertebrate and invertebrate hosts. Using a computerized image analysis system, we studied five species of Hepatozoon from recently captured snakes in Botucatu, State of São Paulo, Brazil, to evaluate the importance of the morphology and morphometry of the gamonts for the characterization of Hepatozoon species and to analyze the morphologic changes induced in the erythrocytes by the parasite. The studied species were H. terzii of Boa constrictor amarali, Hepatozoon sp. of Crotalus durissusterrificus, H. philodryasi of Philodryas patagoniensis, and H. migonei and H. cyclagrasi of Hydrodynastes gigas. We observed three different groups, one of them including the species H. terzii, H. philodryasi and Hepatozoon sp. of C. durissus terrificus; and the other two consisting of H. migonei and H. cyclagrasi. Degree of alterations in the erythrocytes was variable and it may be useful for characterization of Hepatozoon species.

A reprodução faz parte do ciclo de vida dos animais permitindo a perpetuação e a conservação das espécies. Em serpentes, existe uma escassez de informações técnicas a respeito do ciclo reprodutivo. Este estudo teve o objetivo de avaliar o aparelho reprodutivo por meio da ultrassonografia em serpentes vivíparas cativas da família Boidae, permitindo diagnosticar as diferentes fases reprodutivas. Foram avaliadas ultrassonograficamente onze serpentes adultas de quatro espécies da família Boidae: Eunectes murinus, Boa constrictor constrictor, Corallus hortulanus e Epicrates cenchria pertencentes ao acervo do Museu Biológico do Instituto Butantan, São Paulo Brasil. Para a avaliação ultrassonográfica, as serpentes foram contidas fisicamente com gancho herpetológico e depois manualmente por aproximadamente 15 minutos. A avaliação foi feita aplicando-se gel acústico sobre a pele e posicionando o transdutor na linha lateral-ventral direita e esquerda, em região medial do corpo em sentido crânio-caudal. O exame ultrassonográfico permitiu avaliar todo o ciclo reprodutivo nas serpentes. Nas avaliações ultrassonográficas das fêmeas pode-se definir as fases de desenvolvimento ovariano e ovidutal. Os folículos ovarianos durante a fase pré-vitelogênica foram visualizados como homogêneos e anecogênicos, em forma de "cacho de uva". Já na fase vitelogênica, os folículos estavam maiores e mais ecogênicos seguidos uns dos outros, como um "colar de pérolas". Quando não houve cópula, os folículos foram reabsorvidos dentro do ovário retornando a fase pré-vitelogênica. Na fase pós ovulatória foram visualizados três estágios bem definidos de desenvolvimento fetal dentro do oviduto: 1) logo após a ovulação (e fecundação), somente o vitelo foi visualizado; 2) o vitelo ocupava 60% e o feto 40% do ovo e 3) o feto estava formado e não havia vitelo. Nos machos, os testículos foram visualizados como uma imagem homogênea e hipoecogênica quando se encontravam em estágio reprodutivo. Quando não estavam reprodutivos não era possível visualizar a imagem do testículo devido ao seu tamanho. A avaliação ultrassonográfica do aparelho reprodutor em serpentes demonstrou ser uma técnica de diagnóstico segura, não invasiva e que permite o acompanhamento das principais fases reprodutivas

We quantified the oxygen uptake rates ((V) over dot O-2) and time spent, during the constriction, inspection, and ingestion of prey of different relative sizes, by the prey-constricting boid snake Boa constrictor amarali. Time spent in prey constriction varied from 7.6 to 16.3 min, and (V) over dot O-2 during prey constriction increased 6.8-fold above resting values. This was the most energy expensive predation phase but neither time spent nor metabolic rate during this phase were correlated with prey size. Similarly, prey size did not affect the (V) over dot O-2 or duration of prey inspection. Prey ingestion time, on the other hand, increased linearly with prey size although (V) over dot O-2 during this phase, which increased 4.9-fold above resting levels, was not affected by prey size. The increase in mechanical difficulty of ingesting larger prey, therefore, was associated with longer ingestion times rather than proportional increases in the level of metabolic effort. The data indicate that prey constriction and ingestion are largely sustained by glycolysis and the intervening phase of prey inspection may allow recovery between these two predatory phases with high metabolic demands. The total amount of energy spent by B. c. amarali to constrict, inspect, and ingest prey of sizes varying from 5 to 40% of snake body mass varied inversely from 0.21 to 0.11% of the energy assimilated from the prey, respectively. Thus, prey size was not limited by the energetic cost of predation. on the contrary, snakes feeding on larger prey were rewarded with larger energetic returns, in accordance with explanations of the evolution of snake feeding specializations. We investigated the combined effect of meal size and temperature on the aerobic metabolism and energetics of digestion in Boa constrictor amarali. Oxygen uptake rates ((V) over dot o(2)) and the. duration of the digestion were determined in snakes fed with meals equaling to 5%, 10%, 20%, and 40% of the snake's body mass at 25degrees and 30 degreesC. The maximum (V) over dot o(2) values attained during digestion were greater at 30 degreesC than at 25 degreesC. Both maximal (V) over dot (o2) values and the duration of the specific dynamic action. (SDA) were attained sooner at 30 degreesC than at 25 degreesC. Therefore, the temperature effect on digestion in Boa is characterized by the shortening of the SDA duration at the expense of increased. Energy allocated to SDA was not affected by meal size but. was greater at 25 degreesC compared to 30 degreesC. This indicates that a postprandial thermophilic response can be advantageous not only by decreasing the duration of digestion but also by improving digestive efficiency. Maximal (V) over dot o(2) and SDA duration. increased with meal size at both temperatures.

We present the first formal report on the squamate assemblage from Parque Nacional de Ubajara. This park contains the most important cave complex in the state of Ceara in northeastern Brazil, called Provincia Espeleologica de Ubajara. The material comes from the Urso Fossil cave at Pendurado Hill. All previously reported fossil remains found in this cave are tentatively attributed to the Quaternary (late Pleistocene-early Holocene). Probably only Arctotherium brasiliense represents a relictual fossil bear from the late Pleistocene megafauna. The taxa recognized in this paper belong to Tropidurus sp., Ameiva sp., cf. Epicrates, and cf. Crotalus durissus, adding to the knowledge of the Brazilian Quaternary squamate fauna as a whole, and contribute to a major taxonomic refinement of the squamate assemblages from the early Holocene of northeastern Brazil.

El presente estudio se llevó a cabo en el Centro Experimental ICIDRI Masatepe para realizar un Diseño de un circuito Agroecoturístico con base en los potenciales naturales y productivos del Centro Experimental ICIDRI Masatepe en un área de 10.2 manzanas. Se contemplaron 3 etapas: (I) La de planeación y organización que facilitó a través de una visita previa, los tiempos y formas de recabar la información requerida; (II) Trabajo de campo en la cual se levantó la información en dos visitas al sitio para la delimitación del área, realizar un inventario florístico y de fauna, y de la propuesta de estaciones interpretativas; y en la Etapa III, se logró el análisis e interpretación de toda la información recabada y en conjunto con el equipo de docentes UNA y de ICIDRI de la UPOLI, reviso la propuesta y fue aceptada. El Centro, cuenta con un potencial natural que incluye diversidad de especies arbóreas y fauna, que dan lugar al disfrute de variados paisajes y de clima; con potenciales productivos que brindan la oportunidad a productores la adopción de nuevas prácticas. A partir de estos escenarios, se propuso un circuito Agroecoturístico cerrado con 11 estaciones interpretativas en un tiempo no mayor de 3 horas. Del inventario florístico resultaron 1,284 especies, siendo las más representativas Aguacate (Persea americana Mill.), Cedro Real (Cedrela odorata L.), Laurel (Cordia alliodora (Ruiz & Pav.) Oken.) y Aceituno (Simarouba amara Aubl.). Entre la fauna silvestre se encontró Ardilla (Sciurus variegatoides), Ameiva undulata, (Holcosus undulatus) traga venado (Boa Constrictor), Guarda Barranco (Eumomota superciliosa) y Salta piñuela (Anthracothorax prevostii). Las condiciones de cercanía, topografía, capacidades locales, distancia de recorrido, escenarios, temáticas abordadas en las estaciones e infraestructura básica, hacen del centro una oportunidad para la implementación del circuito. Sin embargo, se debe divulgar su quehacer, rotular los caminos, ubicación de cestos de basura y brindar capacitación al personal para guiar a los diferentes usuarios que lo visiten.

O-GlcNAcylation is a modification that alters the function of numerous proteins. We hypothesized that augmented O-GlcNAcylation levels enhance myosin light chain kinase (MLCK) and reduce myosin light chain phosphatase (MLCP) activity, leading to increased vascular contractile responsiveness. The vascular responses were measured by isometric force displacement. Thoracic aorta and vascular smooth muscle cells (VSMCs) from rats were incubated with vehicle or with PugNAc, which increases O-GlcNAcylation. In addition, we determined whether proteins that play an important role in the regulation of MLCK and MLCP activity are directly affected by O-GlcNAcylation. PugNAc enhanced phenylephrine (PE) responses in rat aortas (maximal effect, 14.2±2 vs 7.9±1 mN for vehicle, n=7). Treatment with an MLCP inhibitor (calyculin A) augmented vascular responses to PE (13.4±2 mN) and abolished the differences in PE-response between the groups. The effect of PugNAc was not observed when vessels were preincubated with ML-9, an MLCK inhibitor (7.3±2 vs 7.5±2 mN for vehicle, n=5). Furthermore, our data showed that differences in the PE-induced contractile response between the groups were abolished by the activator of AMP-activated protein kinase (AICAR; 6.1±2 vs 7.4±2 mN for vehicle, n=5). PugNAc increased phosphorylation of myosin phosphatase target subunit 1 (MYPT-1) and protein kinase C-potentiated inhibitor protein of 17 kDa (CPI-17), which are involved in RhoA/Rho-kinase-mediated inhibition of myosin phosphatase activity. PugNAc incubation produced a time-dependent increase in vascular phosphorylation of myosin light chain and decreased phosphorylation levels of AMP-activated protein kinase, which decreased the affinity of MLCK for Ca2+/calmodulin. Our data suggest that proteins that play an important role in the regulation of MLCK and MLCP activity are directly affected by O-GlcNAcylation, favoring vascular contraction.

O-linked N-acetylglucosaminylation (O-GlcNAcylation) plays a role in many aspects of protein function. Whereas elevated O-GlcNAc levels contribute to diabetes-related end-organ damage, O-GlcNAcylation is also physiologically important. Because proteins that play a role in vascular tone regulation can be O-GlcNAcylated, we hypothesized that O-GlcNAcylation increases vascular reactivity to constrictor stimuli, Aortas front male Sprague-Dawley rats and C57BL/6 mice were incubated for 24 hours with vehicle or PugNAc (O-GlcNAcase inhibitor. 100 mu M). PugNAc incubation significantly increased O-GlcNAc proteins, as determined by Western blot. PugNAc also increased vascular contractions to phenylephrine and serotonin, an effect not observed in the presence of N(omega)-nitro-L-arginine methyl ester or in endothelium-denuded vessels. Acetylcholine-induced relaxation. but not that to sodium nitroprusside, was decreased by PugNAc treatment, an effect accompanied by decreased levels of phosphorylated endothelial nitric oxide synthase (eNOS)(Ser-1177) and Akt(Ser-473). Augmented O-GlcNAcylation increases vascular reactivity to constrictor stimuli, possibly due to its effects oil eNOS expression and activity, reinforcing the concept that O-GlcNAcylation modulates vascular reactivity and may play a role in pathological conditions associated with abnormal vascular function.
