Elevated expression of the marORAB multiple antibiotic-resistance operon enhances the resistance of Escherichia coli to various medically significant antibiotics. Transcription of the operon is repressed in vivo by the marR-encoded protein, MarR, and derepressed by salicylate and certain antibiotics. The possibility that repression results from MarR interacting with the marO operator-promoter region was studied in vitro using purified MarR and a DNA fragment containing marO. MarR formed at least two complexes with marO DNA, bound > 30-fold more tightly to it than to salmon sperm DNA, and protected two separate 21-bp sites within marO from digestion by DNase I. Site I abuts the downstream side of the putative -35 transcription-start signal and includes 4 bp of the -10 signal. Site II begins 13 bp downstream of site I, ending immediately before the first base pair of marR. Site II, approximately 80% homologous to site I, is not required for repression since a site II-deleted mutant (marO133) was repressed in trans by wild-type MarR. The absence of site II did not prevent MarR from complexing with the site I of marO133. Salicylate bound to MarR (Kd approximately 0.5 mM) and weakened the interaction of MarR with sites I and II. Thus, repression of the mar operon, which curbs the antibiotic resistance of E. coli, correlates with the formation of MarR-site I complexes. Salicylate appears to induce the mar operon by binding to MarR and inhibiting complex formation, whereas tetracycline and chloramphenicol, which neither bind MarR nor inhibit complex formation, must induce by an indirect mechanism.

"Whe' yu' from?" The question was put to me as I wandered, camera in hand, in the old square of Spanish Town, Jamaica's former capital. The local man, lounging in the shade of one of the colonial Georgian buildings that enclose the square, was mildly curious about what he took to be a typical white tourish photgraphing the sights of the decayed historic town. At that time, my home was in Kingston where i lived with my wife and baby son. I was then working in the Jamaican Government Town Planning Department in a job that took me all over the island. Turning to my questioner, I replied, "Kingston". There was a brief pause, and then the man spoke again: "No Man! Whe' yu' really from?" I still have difficulties when asked this question. Where am I from? What does this question mean? Does it refer to where I was born, where I spent my previous life or where I live now? Does it have a broader meaning, an enquiry about my origins in terms of background and previous experience? The following chapters are my attempt to answer these questions for my own satisfaction and, I hope, for the amusement of others who may be interested in the life of an ordinary English boy whose dream to travel and see the world was realized in ways he could not possibly have imagined. Finding an appropriate title for this book was difficult. Thursday's Child, North and South and War and Peace all came to mind but, unfortunately for me, those titles had been appropriated by other writers. Thursdays's Child is quite a popular book title, presumably because people who were born on that day and, in the words of the nursery rhyme, had 'far to go', are especially likely to have travellers' tales to tell or life stories of the rags-to-riches variety. Born on a Thursday, I have travelled a lot and I suppose that I have gone far in life. Coming from a working class family, I 'got on' by 'getting a good education' and a 'good job'. I decided against adding to the list of Thursday's Children. North and South would have reflected my life in Britain, spent in both the North and South of England, and my later years, divided between the Northern and Southern Hemispheres of the globe, as well as in countries commonly referred to as the 'advanced' North and the 'underdeveloped' South. North and South has already been appropriated by Mrs Gaskell, something that did not deter one popular American writer from using the title for a book of his. My memories of World War Two and the years afterwards made War and Peace a possible candidate, but readers expectnig an epic tale of Tolstoyan proportions may have been disappointed. To my knowledge, no other book has the title "Whe' Yu' From?". I am grateful to the Jamaican man whose question lingered in my memory and provided the title of this memoir, written decades later. This book is a word picture. It is, in a sense, a self-portrait, and like all portraits, it captures something of the character, it attempts to tell the truth, but it is not the whole truth. This is because it is not my intention to write my entire life story; rather I wish to tell about some of the things in my experience of life that have seemed important or interesting to me. Unlike a painted portrait, the picture I have created is intended to suggest the passage of time. While, for most of us in Western society, time is linear and unidirectional, like the flight of an arrov or the trajectory of a bullet, memory rearranges things, calling up images of the past in no particular order, making connections that may link events in various patterns, circular, web-like, superimposed. The stream of consciousness is very unlike that of streams we encounter in the physical world. Connections are made in all directions; thoughts hop back and forth in time and space, from topic to topic. My book is a composition drawn from periods, events and thoughts as I remember them. Like life itself, it is made up of patches, some good, some bad, but in my experience, always fascinating. In recording my memories, I have been as accurate as possible. Little of what I have written is about spectacular sights and strange customs. Much of it focuses on my more modest explorations includng observations of everyday things that have attracted my attention. Reading through the chapters, I am struck by my childhood freedom to roam and engage in 'dangerous' activities like climbing trees and playing beside streams, things that many children today are no longer allowed to enjoy. Also noticeable is the survival of traditions and superstitions from the distant past. Obvious too, is my preoccupation with place names, both official ones that appear on maps and sign boards and those used by locals and children, names rarely seen in print. If there is any uniting theme to be found in what I have written, it must be my education in the fields, woods and streets of my English homeland, in the various other countries in which I have lived and travelled, as well as more formally from books and in classrooms. Much of my book is concerned with people and places. Many of the people I mention are among those who have been, and often have remained, important and close to me. Others I remember from only the briefest of encounters, but they remain in my memory because of some specific incident or circumstance that fixed a lasting image in my mind. Some of my closest friends and relatives, however, appear nowhere in these pages or they receive only the slightest mention. This is not because they played an unimportant roles in my life. It is because this book is not the whole story. Among those whe receive little or no mention are some who are especially close to me, with whom I have shared happy and sad times and who have shown me and my family much kindness, giving support when this was needed. Some I have known since childhood and have popped up at various times in my life, often in different parts of the world. Although years may pass without me seeing them, in an important sense they are always with me. These people know who they are. I hope that they know how much I love and appreciate them. When writing my memoir, I consulted a few of the people mentioned in this book, but in the main, I have relied on my own memory, asided by daiary and notebook entries and old correspondence. In the preparation of this manuscript, I benefited greatly from the expert advice and encouragement of Neil Marr of BeWrite Books. My wife Anne, inspiration for this book, also contributed in the valuable role of critic. She has my undying gratitude.

The vacuolating autotransporter (AT) toxin (Vat) contributes to Uropathogenic Escherichia coli (UPEC) fitness during systemic infection. Here we characterised Vat and investigated its regulation in UPEC. We assessed the prevalence of vat in a collection of 45 UPEC urosepsis strains and showed that it was present in 31 (68%) of the isolates. The isolates containing the vat gene corresponded to three major E. coli sequence types (ST12, 73 and 95) and these strains secreted the Vat protein. Further analysis of the vat genomic locus identified a conserved gene located directly downstream of vat that encodes a putative MarR-like transcriptional regulator, which we termed vatX. The vat-vatX genes were present in the UPEC reference strain CFT073 and RT-PCR revealed both genes are co-transcribed. Over-expression of vatX in CFT073 led to a 3-fold increase in vat gene transcription. The vat promoter region contained three putative nucleation sites for the global transcriptional regulator H-NS; thus the hns gene was mutated in CFT073 (to generate CFT073hns). Western blot analysis using a Vat-specific antibody revealed a significant increase in Vat expression in CFT073hns compared to wild-type CFT073. Direct H-NS binding to the vat promoter region was demonstrated using purified H-NS in combination with electrophoresis mobility shift assays. Finally, Vat-specific antibodies were detected in plasma samples from urosepsis patients infected by vat-containing UPEC strains, demonstrating Vat is expressed during infection. Overall, this study has demonstrated that Vat is a highly prevalent and tightly regulated immunogenic SPATE secreted by UPEC during infection.

In a number of applications of computerized tomography, the ultimate goal is to detect and characterize objects within a cross section. Detection of edges of different contrast regions yields the required information. The problem of detecting edges from projection data is addressed. It is shown that the class of linear edge detection operators used on images can be used for detection of edges directly from projection data. This not only reduces the computational burden but also avoids the difficulties of postprocessing a reconstructed image. This is accomplished by a convolution backprojection operation. For example, with the Marr-Hildreth edge detection operator, the filtering function that is to be used on the projection data is the Radon transform of the Laplacian of the 2-D Gaussian function which is combined with the reconstruction filter. Simulation results showing the efficacy of the proposed method and a comparison with edges detected from the reconstructed image are presented

ENGLISH: Knowledge of spawning habits is useful in the elucidation of the life history, ecology and population structure of tropical tunas, and is essential to the sound management of these resources. Until recently, little was known concerning the spawning of tunas, or about the distribution of their larval and juvenile stages, in the Eastern Pacific Ocean. Nichols and Murphy (1944) reported the capture off Colombia of young scombroids ultimately identified as frigate mackerel, Auxis thazard (Schaefer and Marr, 1948a). Fowler (1944) reported the capture off Manzanillo, Mexico of two young tunas, one of which is definitely and the other most likely Neothunnus macropterus (Klawe, 1959). In 1947, young of N. macropterus, K. pelamis, A. thazard and E. lineatus were caught offshore from Central America (Schaefer and Marr, 1948a, 1948b, and Schaefer, 1948). Further collections of young N. macropterus, A. thazard and E. lineatus were made in the same general area in the spring of 1949 (Mead, 1951). In January and February 1955, Clemens (1956) carried Out experiments in rearing young tunas, E. lineatus and A. thazard, in shipboard aquaria, using fish caught off Central America. Matsumoto (1958) reported captures of larval N. macropterus and K. pelamis in the area along the 120th meridian of west longitude. Klawe (1958 and 1961b) reported captures of larval N. macropterus and Auxis from the Revillagigedo Islands. Captures of young Auxis and E. lineatus in the Gulf of Panama in January 1922 during the Dana Expedition have recently been reported by Matsumoto (1959). Capture of juveniles of K. pelamis, E. lineatus and Auxis in the area off tropical Mexico and in the area of outlying islands during the SCOT Expedition has been reported by Klawe (1960a). SPANISH: El conocimiento sobre los hábitos del desove es útil para el esclarecimiento de la historia natural, ecología y estructura de las poblaciones de atunes tropicales, y es esencial para la acertada administración de estos recursos. Hasta hace poco tiempo no se sabía mucho sobre el desove de los atunes o acerca de la distribución de sus larvas y juveniles en el Océano Pacífico Oriental. Nichols y Murphy (1944) informaron sobre la captura frente a Colombia de escómbridos jóvenes últimamente identificados como melva, Auxis thazard (Schaefer y Marr, 1948a). Fowler (1944) también informó sobre la captura de dos atunes jóvenes frente a Manzanillo, México, uno de los cuales era definitivamente Neothunnus macropterus y el otro era lo más probable que también lo fuera (Klawe, 1959). Matsumoto (1959) ha informado recientemente acerca de capturas de ejemplares jóvenes de Auxis y E. lineatus en el Golfo de Panamá en enero de 1922 durante la Expedición Dana. Klawe (1960a) informó así mismo que durante la Expedición SCOT se capturaron juveniles de K. pelamis, E. lineatus y Auxis en el área frente a la zona tropical de México y en la región de las islas alejadas del continente.

ENGLISH: Knowledge of the size and age at maturity, spawning seasons, and spawning areas of the tropical tunas supporting the fishery in the Eastern Pacific is an important part of the basic information required for understanding their life history, population structure, and fishery dynamics. Until a few years ago nothing was known of these matters. In 1947 the senior author and one of his colleagues (Schaefer and Marr 1948, Schaefer 1948) were able to demonstrate that both yellowfin tuna and skipjack spawn offshore from Central America at least during the late winter and spring months. During January to April many yellowfin tuna over about 70 cm. total length in commercial catches from that region were found to have gonads in advanced stages of maturity, and specimens caught during late June were found to be spent. Maturing skipjack were collected in late February, and spawned-out fish were observed in late March. Numerous very young juveniles of the yellowfin, down to 10 mm. in length, and two very young juvenile skipjack, were captured in this area between January and May. SPANISH: El conocimiento del tamaño y la edad que corresponden a la primera madurez sexual, así como de las estaciones y áreas de desove de los atunes tropicales que mantienen las pesquerías del Pacífico Oriental, constituyen parte importante de la información que es menester para comprender la historia natural, la estructura de la población y la dinámica de la pesquería. Hasta hace pocos años nada se sabía sobre el particular. En 1947 el autor principal y uno de sus colegas (Schaefer y Marr, 1948; Schaefer, 1948) pudieron demostrar que tanto el atún aleta amarilla como el barrilete desovan en el mar abierto, frente a América Central, por lo menos durante la última parte del invierno y en la primavera. De enero a abril encontraron que muchos de los atunes aleta amarilla de más de 70 cm. de longitud total, procedentes de las pescas comerciales de dicha región; tenían gónadas en avanzados estados de madurez, mientras que ejemplares pescados hacia fines de junio ya habían desovado. Se recolectaron barriletes en vías de maduración a fines de febrero, al paso que en los últimos días de marzo se encontraron especímenes que ya habían desovado. Numerosos ejemplares muy juveniles del atún aleta amarilla, tan pequeños como 10 mm., y dos barriletes también muy juveniles, fueron pescados en esta región entre enero y mayo. On July 12-15, 2008, researchers and resource managers met in Jupiter, Florida to discuss and review the state of knowledge regarding mesophotic coral ecosystems, develop a working definition for these ecosystems, identify critical resource management information needs, and develop a Mesophotic Coral Ecosystems Research Strategy to assist the U.S. National Oceanic and Atmospheric Administration (NOAA) and other agencies and institutions in their research prioritization and strategic planning for mesophotic coral ecosystems. Workshop participants included representatives from international, Federal, and state governments; academia; and nongovernmental organizations. The Mesophotic Coral Ecosystems Workshop was hosted by the Perry Institute for Marine Science (PIMS) and organized by NOAA and the U.S. Geological Survey (USGS). The workshop goals, objectives, schedule, and products were governed by a Steering Committee consisting of members from NOAA (National Centers for Coastal Ocean Science’s Center for Sponsored Coastal Ocean Research, the Office of Ocean Exploration and Research’s NOAA Undersea Research Program, and the National Marine Fisheries Service), USGS, PIMS, the Caribbean Coral Reef Institute, and the Bishop Museum.

The study of biogenic proxy of tropical and subtropical regions provides important evidence about the process and history of vegetation and environmental changes, and is of globally importance for understanding the dynamic mechanism of paleoclimatic and paleoenvironmental changes. The sediments from the Huguangyan Maar lake in Guangdong Province offer a continuous high-resolution record of the past 55 ka about environmental and vegetational changes. The studies of chronology, and physical, chemical environmental proxies have provided much important information about the paleoenvironmental and paleoclimatic histories. The phytolith, a new biogenicl proxy, has been used to determine the nature and types of plants in this area since the last 55 ka. This study presents a preliminary result about the characteristics of phytolith shapes, the variations of the fossils assemblages, and their significance for environmental changes. Moreover, the author probes the process of special specie evolution and their relationship to climatic parameters. The history of fire has been reconstructed based on the variations in charcoals. The main results and conclusions include: 28 types of phytoliths from 233 samples have been identified. Their environmental meanings are investigated in detail. Based on the variations in phytolith associations, the history and process of climatic and environmental changes in the last 55 kaBP have been established for this region. Climatic changes experienced eight intervals during this period, showing the variations of hot-humid to cool-try climate in the ten thousands years scale, and a shorter dry-hot climate condition in millennial scale. The history of palm plant has been established in this region. Two peaks appeared from 55-39 ka and since the Holocene. Plants in Bambusoideae have been growing in this area all the period, representing the impact of the East Asian summer monsoon. Bamboo plants have similar tendency in their abundance to palm plants, but with a lag of 1-2 ka BP. Panicoideae plants, the representative of C4 plants, have 6 flourishing periods occurred at 54.5, 44, 41.5, 32.5, 14, and 10 kaBP, respectively, reflecting 6 times short-term arid events. Charcoal record from the Huguang Marr lake reveals the history of nature fire, that mostly happened in dry period of last glacial from 55-10 kaBP, centered at 50-45, 40-35, 30-25, and 20-15kaBP, showing about a cycle of 10,000 years.

We review the progress made in computational vision, as represented by Marr's approach, in the last fifteen years. First, we briefly outline computational theories developed for low, middle and high-level vision. We then discuss in more detail solutions proposed to three representative problems in vision, each dealing with a different level of visual processing. Finally, we discuss modifications to the currently established computational paradigm that appear to be dictated by the recent developments in vision.

This report describes the implementation of a theory of edge detection, proposed by Marr and Hildreth (1979). According to this theory, the image is first processed independently through a set of different size filters, whose shape is the Laplacian of a Gaussian, ***. Zero-crossings in the output of these filters mark the positions of intensity changes at different resolutions. Information about these zero-crossings is then used for deriving a full symbolic description of changes in intensity in the image, called the raw primal sketch. The theory is closely tied with early processing in the human visual systems. In this report, we first examine the critical properties of the initial filters used in the edge detection process, both from a theoretical and practical standpoint. The implementation is then used as a test bed for exploring aspects of the human visual system; in particular, acuity and hyperacuity. Finally, we present some preliminary results concerning the relationship between zero-crossings detected at different resolutions, and some observations relevant to the process by which the human visual system integrates descriptions of intensity changes obtained at different resolutions.
